不同品种烤烟衰老期碳氮代谢及相关基因表达的差异分析

摘    要:  目的  研究不同品种烤烟叶片衰老期碳氮代谢及相关基因表达的差异及关系。  方法  以田间成熟落黄表现不同的4个烤烟品种中部叶为材料,分析叶片成熟衰老过程中质体色素、碳氮代谢中间产物含量的变化,检测了相关基因的表达量。  结果  ① 豫烟10号叶片叶绿素和类胡萝卜素的含量较低,并随着叶龄的增加降低的幅度最大,CCD表达量较高,NC89的降低的幅度较小,含量较高,CCD表达量较低。②叶片淀粉含量在叶龄50~60 d时达到峰值后开始下降,从峰值到叶龄70 d,降幅较大的豫烟10号(38.65%),淀粉合成相关基因GBSS1和SBE的表达量相应较低,总糖和还原糖含量从峰值至叶龄70 d也表现较大的降幅。③ NC89的淀粉、总糖、还原糖和总碳含量均相对较高,云烟87和K326的碳代谢指标则居豫烟10号和NC89之间。④豫烟10号叶片成熟衰老过程中总氮、碳氮比、烟碱含量显著低于NC89,这与豫烟10号氮素转移相关的GS1-3基因表达量在叶龄40 d之后显著高于NC89,参与氮同化相关的GS2、GDH1和NR基因表达量相对较低的特征一致。  结论  叶片衰老速度快的品种具有淀粉合成代谢弱,淀粉含量较低,氮素同化代谢较弱而再利用和再转移代谢能力较强,总氮、碳氮比较低的碳氮代谢特征。

Differences of carbon and nitrogen metabolism and related gene expression during leaf senescence phase for different flue-cured tobacco varieties

Abstract:  Background  This study aims to study the difference and relationship of carbon and nitrogen metabolism and related gene expression in the leaf senescence stage of different flue-cured tobacco varieties.  Methods  Taking the middle leaves of four flue-cured tobacco varieties with different yellowing performances in the field as materials, this paper analyzed the changes of the content of plastid pigments and intermediates of carbon and nitrogen metabolism during the process from leaf maturation to senescence, and detected the expression levels of related genes.  Results  The content of chlorophyll and carotenoid in the leaves of Yuyan10 was low, and the decrement reached the maximum with the increase of leaf age. The carotenoid cleavage dioxygenase gene (CCD) expression level was high, the decrement of NC89 was smaller, and the content was higher, and the CCD expression level was lower. The content of starch in leaves reached a peak at 50-60 d of leaf age, and then began to decline. From the peak to the 70 d of leaf age, the decrement in starch content of Yuyan 10 (38.65%) was relatively large, and the expression levels of its starch synthesization-related genes, including granulebound starch synthase gene (GBSS1) and starch branching enzyme gene (SBE), were relatively low. The contents of total sugar and reducing sugar also decreased significantly from the peak to the 70 d of leaf age. The contents of starch, total sugar, reducing sugar and total carbon in NC89 were relatively high. The carbon metabolism indexes of Yunyan87 and K326 were in the middle level between those of Yuyan 10 and those of NC89. The total nitrogen, C/N ratio and nicotine content in Yuyan 10 leaves from leaf maturation to senescence were significantly lower than those of NC89. Consistently, nitrogen transfer related cytoplasmic glutamine synthase gene (GS1-3) had significantly higher expression level in Yuyan10 than in NC89 after 40 days of leaf age, and nitrogen assimilation related chloroplast glutamine synthase gene (GS2), glutamate dehydrogenase gene (GDH1) and nitrate reductase gene (NR) had the relatively low expression levels in Yuyan10.  Conclusions  The senescence characteristics of different flue-cured tobacco varieties are related to carbon and nitrogen metabolism. The varieties with rapid leaf senescence rate have weaker starch synthesis and metabolism, lower starch content, weaker nitrogen assimilation, stronger ability of nitrogen reutilization and retransfer metabolism, and lower carbon and nitrogen metabolism.
