摘    要:目的探讨缺氧条件培养下诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)抑制剂1400 W对人舌鳞癌CAL-27细胞株iNOS的影响。方法常规培养人舌鳞癌细胞系CAL-27后,实验组分别加入iNOS抑制剂1400W 50(50μmol.L-1组)、100(100μmol.L-1组)、200(200μmol.L-1组)、400(400μmol.L-1组)μmol.L-1的培养液缺氧条件下培养;对照组不加iNOS抑制剂1400 W培养液缺氧条件下培养。在缺氧条件下培养24、48、72、96 h后,采用硝酸还原酶法检测细胞培养上清液NO含量。结果 1400 W作用于人舌鳞癌CAL-27细胞后,实验各组细胞培养上清液NO含量与对照组比较明显下降(P〈0.05),并且随作用时间和剂量的增加而相应递减。结论选择性iNOS抑制剂1400 W可显著抑制缺氧条件下培养的人舌鳞癌CAL-27细胞株iNOS的表达。

Abstract:Objective To investigate the effect of inducible nitric oxide synthase(iNOS) inhibitor 1400 W on the expression of iNOS in hypoxic human tongue squamous carcinoma cell line CAL-27.Methods CAL-27 cells were treated with 1400 W(50~400 μmol·L-1) in hypoxia for 24,48,72 and 96 hours.The control cells only received the medium in hypoxia.The levels of NO in the cell culture supernate were determined by using nitrate reducase method.Results Compared with the control,the levels of NO were decreased after treatment with 1400 W(P0.05) and were progressingly reduced with the increasing of the time and the dose.Conclusion 1 400 W may inhibit the expression of iNOS in hypoxic human tongue squamous carcinoma cell line CAL-27.
