烟草白粉病抗性连锁分子标记开发及育种利用

摘    要:  目的  提高烟草白粉病抗病育种效率,有效防止抗性丢失。  方法  通过烟草种质资源白粉病抗性鉴定获得抗源材料,利用F2和BCF1分离群体进行白粉病抗性遗传规律和分子标记连锁分析,开发与烟草白粉病抗性紧密连锁的分子标记。  结果  获得白粉病抗源材料2份,分别为台烟7号和KE1,其中台烟7号的白粉病抗性符合双基因隐性遗传规律。根据抗感亲本的NtMLO基因的差异开发了分子标记,利用分离群体证明该标记与白粉病抗性共分离,并应用于主栽品种的抗性回交改良。  结论  开发了与烟草Ntmlo白粉病抗性共分离的分子标记,可显著提高烟草白粉病抗病育种效率。

Development and breeding utilization of molecular markers linked to powdery mildew resistance in tobacco

Abstract:  Objective  This study aims to improve the breeding efficiency of tobacco with powdery mildew resistance and prevent loss of resistance locus.  Methods  The powdery mildew resistance of tobacco germplasm resources was identified to obtain the resistant materials. The inheritance analyses of powdery mildew resistance were carried out in F2 and BC1F1 segregating populations. The molecular markers closely linked to tobacco powdery mildew resistance were developed according to the sequence difference of NtMLO genes.  Result  Two powdery mildew resistant materials were obtained. The powdery mildew resistance of Taiyan No.7 was inherited by two recessive genes. The molecular markers were proved to be cosegregated with powdery mildew resistance in segregation population, and have been applied to the backcross improvement of resistance of main cultivars.  Conclusion  Molecular markers cosegregated with Ntmlo resistance were developed, which could significantly improve the breeding efficiency of tobacco with powdery mildew resistance.
