摘    要:利用双单倍体技术可加快杂交后代目标性状的纯合。为了选育双抗烟草花叶病毒(TMV)和马铃薯Y病毒(PVY)的烟草材料,本文采用Coker176与NC55的F1与非洲烟杂交获得单倍体苗,经过苗期抗性鉴定、分子标记辅助选择和叶脉培养加倍,获得母本来源的双抗TMV和PVY的双单倍体。通过人工接种抗性鉴定、标记检测和田间株系比较,获得抗性、植物学性状稳定的双单倍体第三代种子。与常规杂交系统选育相比较,双单倍体方法的获得后代变异更大、性状更容易稳定。

Tobacco lines resistant to TMV and PVY obtained by maternal-derived haploid breeding methods
Abstract:Doubled haploid(DH) technology can accerelate hybridization offspring traits stabilization. In this paper, to breeding tobacco resistant to Tobacco mosaic virus (TMV) and Potato virus Y (PVY), Haploids were obtained by F1 hybrids between Coker176 and NC55 crossing as female to N. african. Resistant to TMV and PVY haploids were selected by seedling resistance identification and molecular marker test. DH plants were regenerated from leaf mid-vein culture method. By artificial inoculated resistance evaluation, markers detection, and field lines compare, the third generation seed were harvested of DH lines which showed stable resistance and botanical characters. Compared to conventional hybrid breeding methods, doubled haploid breeding method show greater variation between offspring lines and the objective traits are relative easier to stabilize.
