A new optical fiber sensor based on surface plasmon resonance is described. It uses an optical fiber with an inverted graded-index profile. A theoretical analysis of the optical propagation when a point light source was used and a computation of the optical power transmitted by the fiber were performed. Experiments were carried out to measure changes of the transmitted power caused by refractive-index variations of the surrounding dielectric medium. Both the simulation and experiments have shown that the sensor exhibits high sensitivity for changes of the surrounding medium in a refractive index range from 1.33 to 1.39.  相似文献

We present a configuration for surface plasmon resonance sensors based on uniform-waist tapered optical fibers and reflective elements. Once the fiber is tapered fulfilling the adiabatic criterion, a multilayer including a metallic medium is asymmetrically deposited on the uniform waist of the fiber. This feature provides the resonant excitation of multiple surface plasma waves. In addition, a mirror is produced at the fiber tip by a chemical Tollens reaction. In this way, the sensor operates in a reflective mode, more convenient for dip probes. When these sensors are spectrally interrogated, a high sensitivity of 10-4 refractive index units per nanometer is attained. These devices can be advantageously used for any kind of chemical sensing and biosensing.  相似文献

A surface plasmon resonance biosensor based on large size square-lattice photonic crystal fiber has been designed and simulated by finite element method. The square-lattice airholes are first coated with a calcium fluoride layer to provide mode confinement, then a nanoscale gold layer is deposited to excite the plasmon mode, and finally, the sample is infiltrated into the holes. The numerical results reveal that the resonance properties are easily affected by many parameters. The refractive index resolution of corresponding sensor can reach 4.3 × 10?6 RIU when the optimum parameters are set as the radius of curvature of the airhole r = 2 μm, the thickness of the core struts c = 200 nm, the auxiliary dielectric layer s = 1 μm, and the gold film d = 40 nm. In addition, the effective area and nonlinear coefficient are calculated.  相似文献

An oscillating wave sensor based on Fabry-Perot resonance modes has been developed. Different from the surface plasmon resonance sensors and the waveguide mode sensors in which the sample is located in the evanescent field region, the proposed device contains the sample in the core region that supports the oscillating field. Owing to the strong concentration of the electromagnetic field in the sensing medium, the proposed device exhibits unusual sensitivity enhancement, which has never been exploited in any other devices.  相似文献

Graphene is a zero band-gap semi-metal with remarkable electromagnetic and mechanical characteristics. This study is the first ever attempt to use graphene in the surface plasmon resonance (SPR) sensor as replacement material for gold/silver. Graphene, comprised of a single atomic layer of carbon, is a purely two-dimensional material and it is an ideal candidate for use as a biosensor because of its high surface-to-volume ratio. This sensor is based on the resonance occasion of the surface plasmon wave (SPW) according to the dielectric constants of each metal film and detected material in gas or aqueous phase. Graphene in the SPR sensor is expected to enlarge the range of analyte to bio-aerosols based on the superior electromagnetic properties of graphene. In this study, a SPR-based fiber optic sensor coated with multi-layered graphene is described. The multi-layered graphene film synthesized by chemical vapor deposition (CVD) on Ni substrate was transferred on the sensing region of an optical fiber. The graphene coated SPR sensor is used to analyze the interaction between structured DNA biotin and Streptavidin is analyzed. Transmitted light after passing through the sensing region is measured by a spectrometer and multimeter. As the light source, blue light which of 450 to 460 nm in wavelength was used. We observed the SPR phenomena in the sensor and show the contrary trends between bare fiber and graphene coated fiber. The fabricated graphene based fiber optic sensor shows excellent detection sensitivity of the interaction between structured DNA and Streptavidin.  相似文献

Surface plasmon resonance (SPR) biosensors prepared using optical fibers can be used as a cost-effective and relatively simple-to-implement alternative to well established biosensor platforms for monitoring biomolecular interactions in situ or possibly in vivo. The fiber biosensor presented in this study utilizes an in-fiber tilted Bragg grating to excite the SPR on the surface of the sensor over a large range of external medium refractive indices, with minimal cross-sensitivity to temperature and without compromising the structural integrity of the fiber. The label-free biorecognition scheme used demonstrates that the sensor relies on the functionalization of the gold-coated fiber with aptamers, synthetic DNA sequences that bind with high specificity to a given target. In addition to monitoring the functionalization of the fiber by the aptamers in real-time, the results also show how the fiber biosensor can detect the presence of the aptamer's target, in various concentrations of thrombin in buffer and serum solutions. The findings also show how the SPR biosensor can be used to evaluate the dissociation constant (K(d)), as the binding constant agrees with values already reported in the literature.  相似文献

A highly sensitive surface plasmon resonance (SPR) biosensor employing magnetic nanoparticle (MNP) assays is presented. In the reported approach, MNPs simultaneously served as "vehicles" for rapid delivery of target analyte from a sample to the sensor surface and as labels increasing the measured refractive index changes that are associated with the binding of target analyte. An optical setup based on grating-coupled surface plasmon resonance (GC-SPR) was used with a magnetic field gradient applied through the sensor chip for manipulating with MNPs on its surface. Iron oxide MNPs and a sensor surface with metallic diffraction grating were modified with antibodies that specifically recognize different epitopes of the analyte of interest. The sensitivity of the biosensor was investigated as a function of mass transport of the analyte to the sensor surface driven by diffusion (free analyte) or by the magnetic field gradient (analyte bound to MNPs). Immunoassay-based detection of β human chorionic gonadotropin (βhCG) was implemented to evaluate the sensitivity of the MNP-enhanced GC-SPR biosensor scheme. The results reveal that the sensitivity of βhCG detection was improved by 4 orders of magnitude compared with the regular SPR sensor with direct detection format, and a limit of detection below pM was achieved.  相似文献

Two surface plasmon resonance dips in reflectance angular spectrum for a p-polarized incident beam of a [prism/Ag/SiO2 helical thin film] Kretschmann configuration are measured and compared with simulations. The simulation also shows that the angular positions of resonances due to surface plasmon waves in reflectance spectrum are sensitive to the variation of principal refractive indices of helical films. It indicates that multiple surface plasmon waves at the [Ag/SiO2 helical thin film] interface is more attractive than the traditional method of producing only one surface plasmon wave for chemical- and bio-sensing applications.  相似文献

We address a structure for surface plasmon resonance (SPR) sensing supporting a symmetric bound surface plasmon, which results in a SPR feature narrower by a factor of 2 compared with that for the conventional configuration. We demonstrate that it enables a low-cost and low-power-consumption LED to be used as a polychromatic light source, which leads to a decrease in the sensor cost and an increase in the sensor miniaturization potential. Further, we show that these advancements are not at the expense of sensor performance in terms of its sensitivity and resolution. We show that the sensor can be designed to have similar sensitivity and even better resolution compared with those for a conventional configuration.  相似文献

In this report, we developed a new optical biosensor in connection with a gold-deposited porous anodic alumina (PAA) layer chip. In our sensor, we observed that the gold deposition onto the chip surface formed a "caplike" layer on the top of the oxide nanostructures in an orderly fashion, so we called this new surface formation a "gold-capped oxide nanostructure". As a result of its interferometric and localized surface plasmon resonance properties, the relative reflected intensity (RRI) at surface of the chip resulted in an optical pattern that was highly sensitive to the changes in the effective thickness of the biomolecular layer. We demonstrated the method on the detection of picomolar quantities of untagged oligonucleotides and the hybridization with synthetic and PCR-amplified DNA samples. The detection limit of our PAA layer chip was determined as 10 pM synthetic target DNA. The capability of observing both RRI increment and wavelength shift upon biomolecular interactions promises to make our chip widely applicable in various analytical tests.  相似文献

A side-polished multimode fiber sensor based on surface plasmon resonance (SPR) as the transducing element with a halogen light source is proposed. The SPR fiber sensor is side polished until half the core is closed and coated with a 37 nm gold thin film by dc sputtering. The SPR curve on the optical spectrum is described by an optical spectrum analyzer and can sense a range of widths in wavelengths of SPR effects. The measurement system using the halogen light source is constructed for several real-time detections that are carried out for the measurement of the index liquid detections for the sensitivity analysis. The sensing fiber is demonstrated with a series of refractive index (RI) liquids and set for several experiments, including the stability, repeatability, and resolution calibration. The results for the halogen light source with the resolution of the measurement based on wavelength interrogation were 3 x 10(-6) refractive index units (RIUs). The SPR dip shifted in wavelength is used as a measure of the RI change at a surface, and this RI change varies directly with the number of biomolecules at the surface. The SPR dip shift in wavelength, which was hybridized at 0.1 microM of the target DNA to the probe DNA, was 8.66 nm. The all-fiber multimode SPR sensor, which has the advantages of being low cost, being disposable, having high stability and linearity, being free of labeling, and having potential for real-time detection, permit the sensor and system to be used in biochemical sensing and environmental monitoring.  相似文献

This paper describes simulation and experimental methods for designing a D-shaped surface plasmon resonance (SPR) fibre sensor. The sensor consists of two set-up approaches. Finite element method is used in simulation on the fibre sensor device. Two experimental methods for detecting relative intensity are used by varying the wavelength of the optical signal sources and the thickness of gold layer coated on the D-shaped fibre. In the first method, the sensor device works by detecting the relative intensity of two optical signal sources having different wavelengths. In the second set-up, the relative intensity between two D-shaped fibres coated with different thicknesses of gold is measured when a single signal source is launched at the input. The difference in intensities of the signal outputs is used to estimate the refractive index at the sensing region. A prototype SPR D-shaped fibre sensor has been fabricated and the experimental results show good agreement with simulation.  相似文献

We explore a nanowire-based localized surface plasmon resonance (LSPR) sensor system using an effective medium for the nanowire layer. The effective medium is obtained based on the far-field characteristics of the nanowire-based LSPR system. Near-field properties as well as the sensitivity performance of the effective medium-based SPR structure are compared to exact results of the nanowire-based LSPR system. The results indicate that an effective medium can reproduce the far-field and near-field characteristics of nanowires fairly well, while it represents the nanowire-based LSPR on a limited basis in terms of sensitivity characteristics, particularly when the LSPR is significantly enhanced.  相似文献

The efficiency of the transmission of surface plasmon waves by use of a dielectric diffraction grating is discussed. The Kretschmann device allows us to obtain a surface plasmon resonance that consists of an absorption peak in the reflection spectrum. When surface plasmon resonance occurs, the TM-polarization mode of the incident electromagnetic wave is neither transmitted nor reflected. The procedure to transform an 4bsorption peak into a transmission peak is described. Transmittivity of 68% is obtained for a simple structure that consists of a thin-film layer of Ag coated on a volume diffraction grating and embedded between two dielectric media. The results presented herein were obtained by numerical simulations that were carried out by use of an algorithm based on the rigorous coupled-wave theory.  相似文献

The shot noise limitation as well as other factors that influence the sensitivity of measurements with a surface plasmon resonance (SPR) sensor are considered. It is demonstrated that minute changes in the refractive index of a medium close to the surface of a metal film can be detected owing to a shift in the resonance angle. In particular, changes in the adsorption layer of only a fraction of a biomolecular monolayer could be measured. Data for SPR are presented with adjacent media of air, water, as well as aqueous solutions of ethanol and sodium chloride at different concentrations. The immobilization of the protein bovine serum albumin to a specially prepared surface was monitored with the SPR technique. Specific responses to changes in the concentration and thickness of the adsorption layer were determined. The angular resolution of the present apparatus is approximately 1 millidegree, corresponding to a detection limit for an adsorbed protein layer of 15 pg/mm(2), which is still 2 to 3 orders of magnitude larger than the shot-noise limit, and therefore a further improvement in sensitivity is possible.  相似文献

针对表面等离子体共振原理以及光纤传感的优越特性,本文提出并实现了一种基于光纤表面等离子体共振效应的传感检测系统,给出了系统数据分析的处理模型.该方法根据偏度-峰值检验,采用高斯分布检验对共振区域附近的测试数据进行了均值处理,通过理论分析进一步指出采用均值估计的线性模型可以有效地估计共振波长;同时,在保证检测精度的前提下给出了数据处理的最小运算长度.结果表明,可以使系统数据计算的工作量降低至原来的2%,从而提高了系统的处理效率.  相似文献

In this paper a fiber optic surface plasmon resonance (FO-SPR) sensor was used to analyze the melting process of DNA linked to silica nanoparticles. Real-time monitoring of a DNA melting process has rarely been studied using surface plasmon resonance (SPR), since most commercial SPR setups do not allow for dynamic and accurate temperature control above 50 °C. The FO-SPR sensor platform, with silica nanobead signal amplification, allows sensing inside a standard PCR thermocycler, which makes high resolution DNA melting curve analysis possible. This innovative combination was used to characterize the hybridization and melting events between DNA immobilized on the sensor surface and DNA probes on silica nanoparticles. At optimized hybridization conditions complementary DNA strands of different lengths could be distinguished. While the real-time FO-SPR analysis of DNA hybridization did not result in significant variances, the analysis of DNA melting determined the exact length of overlap and the matching Gibbs energy.  相似文献

Existing methods for the optical detection of single molecules require the molecules to absorb light to produce fluorescence or direct absorption signals. This limits the range of species that can be detected, because most molecules are purely refractive. Metal nanoparticles or dielectric resonators can be used to detect non-absorbing molecules because local changes in the refractive index produce a resonance shift. However, current approaches only detect single molecules when the resonance shift is amplified by a highly polarizable label or by a localized precipitation reaction on the surface of a nanoparticle. Without such amplification, single-molecule events can only be identified in a statistical way. Here, we report the plasmonic detection of single molecules in real time without the need for labelling or amplification. Our sensor consists of a single gold nanorod coated with biotin receptors, and the binding of single proteins is detected by monitoring the plasmon resonance of the nanorod with a sensitive photothermal assay. The sensitivity of our device is ~700 times higher than state-of-the-art plasmon sensors and is intrinsically limited by spectral diffusion of the surface plasmon resonance.  相似文献

A surface plasmon resonance sensor array based upon a grating substrate was developed for the detection of biomolecular interactions. The substrate consisted of a gold grating prepared by wet chemical treatment of a commercial recordable compact disk. A custom-built floating pin microspotter was constructed to deliver solutions containing omega-functionalized linear alkanethiols to the grating surface and produce an array of sensor elements with different exposed functional end groups. This array platform can be used to study biomolecular interactions in a label-free, sensitive, and high-throughput format. To illustrate the performance of this device, a test protein (bovine serum albumin) was exposed to sensor elements containing an array of functionalized alkanethiols possessing either activated carboxylic acid-, amine-, or hydroxyl-terminated regions. Local changes in plasmon resonance were monitored in a fixed-angle imaging configuration. Plasmon images clearly distinguish the degree of protein attachment at the various surfaces. The molecular binding events on the grating were also confirmed by ellipsometry. This grating-based SPR imaging platform represents a simple and robust method for performing label-free, high-sensitivity, and high-throughput detection of biomolecular interactions.  相似文献
