Surface-based assays have been used extensively for the functional and structural analysis of biomolecules such as DNA or proteins. These experiments are established by the analysis of binding between acceptor molecules and immobilized receptors on a platform. Site-specific printing of receptor molecules on gold, glass, or polycarbonate (PC) surfaces is conventionally performed by the chemical derivatization of a surface, priming it to covalently bind to subsequently deposited receptor molecules. Unlike conventional methods, we have developed a new fabrication method for bioactive PC surfaces by directly molding PC granules doped with receptor molecules. PC-based receptor molecules were synthesized and commercially available PC granules were doped with these synthesized molecules. In our proof-of-concept study, PC doped with dye 1 (Fig. 1) was used as the receptor molecule. Using an aluminum mold and a hot press machine, PC-based objects were manufactured through compression molding using doped PC granules. Affinity analysis was evaluated by monitoring the localization of a monoclonal antibody elicited against dye 1 to the surface of the molded platforms by fluorescence microscopy. The results illustrated effective binding of an anti-dye 1 monoclonal antibody to the surface, substantiating successful display of assemblies of molecular receptors on the surface through compression molding. Although conventional surface functionalization methods impose limited applications and alter desired opto-mechanical properties of the polymer, our investigation provides a versatile means for the fabrication of bioactive PC-based platforms. The application of protein (or peptide) biomarkers in clinical studies is a dynamic, ever‐growing field. The introduction of clinical proteomics/peptidomics, such as mass spectrometry–based assays and multiplexed antibody–based protein arrays, has reshaped the landscape of biomarker identification and validation, allowing the discovery of novel biomarkers at an unprecedented rate and reliability. To reflect the current status with respect to implementation of protein/peptide biomarkers, an investigation of the most recent (last 6 years) clinical studies from clinicaltrials.gov is presented. Forty‐two clinical trials involving the direct use of protein or peptide biomarkers in patient stratification and/or disease diagnosis and prognosis are highlighted. Most of the clinical trials that include proteomics/protein assays are aiming toward implementation of non‐invasive diagnostic tools for early detection, while many of the clinical trials are targeting to correlate the protein abundance with the risk of a disease event. Less in number are the studies in which the protein biomarkers are applied to stratify the patients for intervention. All the above areas of application are considered of great importance for improving disease management, in an era where implementation toward precision medicine is the desired outcome of proteomics biomarker research. 相似文献

Kinase profiling is an important component of the drug discovery process. Profiling with IC50 provides simultaneous potency and selectivity information about compounds of interest. Profiling data assist in the detection of inhibitors that may have potential toxic effects due to off-target activity. Compounds that exhibit the strongest potency and specificity profiles can be identified as leads for further optimization. Miniaturizing this profiling process in uHTS format generates data from a wide range of compound concentrations, providing more meaningful and accurate information. The combination of the Kinase-Glo Plus Assay with low-volume liquid-dispensing equipment provides an excellent solution for assay optimization and kinase profiling. 相似文献

Excitation-contraction coupling, a process of linking electrical activity to contraction, is the basis of cardiac function at cellular level. Here we developed a multi-scale electrode array (MSEA) as an in vitro platform to synchronously monitor excitation and contraction of cultured cardiomyocytes. The electrodes of MSEA were with sizes ranging from several tens of micrometers to about 1 mm and were classified into type_1, type_2 and type_3 by their electrode area. The former two types of electrodes were electroplated with platinum black to reduce baseline noise and no extra treatment was done on type_3 electrodes. In experiment, the micrometer-scale electrodes recorded extracellular field potentials like generally used multielectrode arrays and especially, the millimeter-scale electrodes recorded a “hump” followed after extracellular field potential. By analyzing temporal behavior of hump and observing its change under treatment of microtubule inhibitor vinblastine, it was verified that the hump expressed mechanical contraction of cardiomyocytes. In addition, we analyzed that the pathway of contraction recording was through cell-electrode contractile coupling, in which charges at electrode-electrolyte interface were reestablished due to contractile behavior of cellular layer and then the hump was formed. Finally, the dependence of amplitude and time duration of hump on electrode area, electrode property and contractile strength was also explored. 相似文献

针对工程中对高压高阻检定精度不断提高的问题,设计了一套基于高压电桥法和ARM的数字式高压高阻鉴定系统。该系统硬件部分主要包括高压电源模块、高压电桥模块、信号采集模块、ARM cortex M4嵌入式控制模块。在控制模块设置测量电压,将高压电源模块输出电压加载到被测电阻两端,根据量程选择恒流恒压方式进行测量,经过A/D采集计算出所测量的电阻,同时可以通过LCD多级菜单设置测量系数并显示鉴定结果。本系统将电桥测量原理、数字处理技术结合在一起,实现了对目前国内市场上各种电压、电阻范围高阻箱的检定。经过试验表明鉴定结果符合JJG 166-93、DL/T 979-2005,可替代传统的高压高阻电桥。 相似文献
