We hypothesized that increasing ruminal pH would lead to enrichment of adipose tissue with conjugated linoleic acid (CLA). Twenty-four Korean native (Hanwoo) steers were used to investigate the additive effects of monensin (30 ppm, SO-BM) and/or fish oil (0.7%, SO-BMF) in the diets along with soybean oil (7%) and sodium bicarbonate (0.5%, SO-B) on cis-9, trans-11 and trans-10, cis-12 CLAs in adipose tissue. The steers were assigned to randomly four groups of six animals each based on body weight. The control group (CON) was fed a commercial concentrate for the late fattening stage. Supplementation of oil and sodium bicarbonate reduced feed intake and daily gain, and fish oil further decreased feed intake (P < 0.001) and daily gain (P < 0.087) compared to steers fed other diets. Total CLA and CLA isomers in M.longissimus dorsi were not affected when steers were fed SO-B and SO-BM diets compared with those of steers fed CON and SO-BMF diets. However, total poly unsaturated fatty acids were higher (P = 0.03) in steers fed SO than in CON steers.  相似文献

Conjugated linoleic acid (CLA) isomers can affect the lipid profile and signaling of cells and thereby alter their function. A total of 5,700 bovine oocytes were used in a structured series of experiments to test the effects of CLA cis-9,trans-11 and CLA trans-10,cis-12 in vitro. In experiment 1, high doses of each CLA isomer during in vitro maturation (IVM) were compared with high or low doses during the entire in vitro culture (IVC) of parthenogenetic embryos. High doses of the CLA isomers ranged from 50 to 200 μM and low doses were 15 and 25 μM. In experiment 2, the low doses of each CLA isomer were tested during IVM/IVC on embryos produced by in vitro fertilization (IVF). Experiment 3 compared the effects of 15 μM doses of each CLA isomer during IVM or IVC of IVF embryos. In experiment 4, post-rewarming survival rates and blastomere counts were assessed for embryos supplemented with each CLA isomer during IVM or for 36 h before vitrification. In experiment 1, when either CLA isomer was provided only during IVM, we observed no effects on overall rates of maturation, cleavage, or blastocysts (92.2 ± 1.6%, 78.3 ± 4.1%, and 28.9 ± 5.1%, respectively). However, high doses of each CLA isomer, but not low doses, during the entire embryo culture period decreased blastocyst rates (5–20%) in a dose-dependent manner. Cleavage rates improved with 15 or 50 μM CLA trans-10,cis-12. Progesterone concentrations in maturation media were significantly increased by high doses of each CLA isomer compared with control, but low doses of CLA isomers had no effect. In experiment 2 with IVF embryos, low doses of each CLA isomer did not alter cleavage rates (average 84.9 ± 1.9%) and only 25 μM CLA trans-10,cis-12 during IVC reduced blastocyst rates below those of controls (25.5 ± 2.1 vs. 38.2 ± 2.3%). The lipid content of embryos was increased and relative expression of the BIRC5 (baculoviral IAP repeat containing 5) gene was depressed by CLA trans-10,cis-12. In experiment 3, low doses (15 μM) of each CLA isomer during IVC significantly reduced blastocyst rates (20.6 ± 2.4% and 27.7 ± 1.2% vs. 34.18 ± 1.2% for CLA trans-10,cis-12 and CLA cis-9,trans-11 compared with control, respectively) with less effect of each CLA during IVM. In experiment 4, adding 100 μM CLA cis-9,trans-11 during the final 36 h of culture resulted in a high survival rate after rewarming and culture, and the higher embryo blastomere count was comparable to that of control embryos not undergoing vitrification. In conclusion, supplementation with either CLA isomer did not improve embryo production, but inclusion of CLA cis-9,trans-11 before vitrification improved the quality of bovine IVF embryos after rewarming and culture.  相似文献

The objective of this study was to examine the effect of ruminal pulse dose of free linoleic acid (LA) and free docosahexaenoic acid (DHA) on microbial populations in the rumen, duodenal fatty acid (FA) flow, milk composition, and milk FA profiles of Chinese Holstein dairy cows. Four rumen- and duodenal-fistulated Chinese Holstein cows in mid lactation (138.5 ± 10 d in milk) were randomly assigned to 3 treatment groups and 1 control group in a 4 × 4 Latin square design over 4 periods (3 wk per period). Diets contained either no LA or 2.7% LA and either no DHA or 0.5% DHA in a 2 × 2 factorial arrangement of treatments. Ruminal pulse dose with DHA increased counts of Megasphaera elsdenii, decreased Fibrobacter succinogenes, but did not affect Butyrivibrio fibrisolvens or Ruminococcus flavefaciens. The pulse dose of LA at 2.7% dry matter had no effect on the population sizes of the 3 major cellulolytic bacterial species or M. elsdenii, and no interaction was observed between LA and DHA. The pulse dose of LA or DHA, either alone or in combination, increased the duodenal flow of vaccenic acid (VA). The milk VA and cis-9,trans-11 conjugated linoleic acid (CLA) contents also increased in response to the fatty acid pulse dose, and the pulse dose of both LA and DHA together had the most profound stimulatory effect. This study indicated that ruminal pulse dose of LA or DHA could be used to increase duodenal flow of VA and the milk contents of potentially health-promoting FA, such as VA and cis-9,trans-11 CLA. These results might be useful in formulating dietary interventions to improve milk cis-9,trans-11 CLA contents.  相似文献

Duration of soybean oil (SBO) supplementation needed to enhance carcass conjugated linoleic acid (CLA) and trans-vaccenic (TVA) content was examined using 96 beef steers (293.6 ± 3.9 kg) fed a 78% corn-based diet supplemented with SBO for 0, 77, 137, or 189 days before slaughter. Duration of SBO supplementation had no effect (P ? 0.15) on animal performance or carcass traits, nor (P ? 0.15), total, total saturated, or total polyunsaturated fatty acids of Longissimus dorsi (LD). Concentrations of CLA in LD were not affected (P ? 0.18) by SBO supplementation. Concentrations of monounsaturated fatty acids (MUFA) decreased linearly (P = 0.03) in LD, whereas TVA increased (P = 0.04) in adipose tissue and tended (P = 0.07) to increase in LD with increasing duration of SBO supplementation. Supplementing SBO to a concentrate-based diet may enhance TVA without impacting CLA, while reducing the MUFA content of lean beef.  相似文献

Activity of stearoyl-CoA desaturase (SCD) in the mammary gland is important for determining the relative proportions of saturated and monounsaturated fatty acids in milk and the concentration of the conjugated linoleic acid isomer rumenic acid (RA; cis-9,trans-11 18:2). Previous studies identified a large degree of between-cow variation in SCD activity, which was consistent across diets and suggests a genetic influence. The objectives of this study were to quantify genetic and phenotypic variations in fatty acid concentrations and SCD indices in milk fat and to estimate their heritabilities in a population of United Kingdom dairy cows. Milk samples were collected from 2,408 daughters of 597 Holstein-Friesian sires on 325 commercial farms for determination of fatty acid profiles. Indices of SCD activity were calculated by expressing each SCD product (cis-9 14:1, cis-9 16:1, cis-9 18:1, and RA) as a proportion of the precursor plus product [e.g., SCDI14 = cis-9 14:1/(14:0 + cis-9 14:1)]. For individual fatty acids, phenotypic variance was considerably greater than additive genetic variance, resulting in small and nonsignificant heritability estimates (± standard error) for all except 6:0 (h2 = 0.27 ± 0.10), 8:0 (h2 = 0.27 ± 0.09), 12:0 (h2 = 0.13 ± 0.07), cis-9 14:1 (h2 = 0.28 ± 0.10), and cis-9 18:1 (h2 = 0.12 ± 0.07). Heritability estimates of desaturase indices were significant for SCDI14 (h2 = 0.38 ± 0.11), SCDI18 (h2 = 0.19 ± 0.09), and SCDIRA (h2 = 0.21 ± 0.09), but not for SCDI16 (h2 = 0.05 ± 0.06). This study provides evidence that additive effects are responsible for a significant proportion of the phenotypic variation in SCD activity in dairy cows. It is concluded that because heritability of desaturase indices is moderate and significant in many cases, these indices could be investigated further for use in future breeding programs to increase concentrations of monounsaturated fatty acids and RA while decreasing concentrations of saturated fatty acids in milk fat.  相似文献

Eight Holstein (189 ± 57 DIM) and 4 Brown Swiss (126 ± 49 DIM) multiparous cows were used in a replicated 4 × 4 Latin square with 28-d periods to determine the minimal dietary concentration of fish oil necessary to maximize milk conjugated linoleic acid (CLA) and vaccenic acid (VA). Treatments consisted of a control diet with a 50:50 ratio of forage to concentrate (dry matter basis), and 3 diets with 2% added fat consisting of 0.33% fish oil, 0.67% fish oil, and 1% fish oil with extruded soybeans providing the balance of added fat. Dry matter intake (23.1, 22.6, 22.8, and 22.9 kg/d, for control, low, medium, and high fish oil diets, respectively) was similar for all diets. Milk production (21.5, 23.7, 22.7, and 24.2 kg/d) was higher for cows fed the fat-supplemented diets vs. the control. Milk fat (4.42, 3.81, 3.80, and 4.03%) and true protein (3.71, 3.58, 3.54, and 3.55%) concentrations decreased when cows were fed diets containing supplemental fat. Concentration of milk cis-9,trans-11 CLA (0.55, 1.17, 1.03, and 1.19 g/100 g of fatty acids) was increased similarly by all diets containing supplemental fat. Milk VA (1.12, 2.47, 2.13, and 2.63 g/100 g of fatty acids) was increased most in milk from cows fed the low and high fish oil diets. Milk total n-3 fatty acids were increased (0.82, 0.96, 0.92, and 1.01 g/100 g of fatty acids) by all fat-supplemented diets. The low fish oil diet was as effective at increasing VA and CLA in milk as the high fish oil diet, showing that only low concentrations of dietary fish oil are necessary for increasing concentrations of VA and CLA in milk.  相似文献

Three levels (0%, 1% and 2%) of an enriched conjugated linoleic acid oil (CLA) were combined with two levels of monounsaturated fatty acids (MUFA) (low (19%) average and high (39%) average) for pig feeding. The profile of volatile compounds of fresh and dry-cured loin as affected by dietary CLA, MUFA and CLA × MUFA interaction was studied by headspace-solid phase microextraction coupled to gas chromatography–mass spectrometry. A total of 27 and 69 compounds were identified in fresh loin and dry-cured loin, respectively. Identified compounds were alcohols, aldehydes, aliphatic hydrocarbons, aromatic hydrocarbons, esters, furans, ketones, nitrogen compounds, sulphur compounds and terpenes. No qualitative differences in volatile compounds caused by the assayed treatments were found neither in fresh loin nor in dry-cured loin. Dietary CLA, MUFA and their interaction did not affect the level of most detected volatiles. Nevertheless, in fresh loin, the level of heptanal significantly increased due to dietary CLA (p = 0.001) and the area units (AU) for ethyl benzene and 1,3-dimethyl benzene increased at 1% CLA (p < 0.05). In the case of dry-cured loin, the AU of heptanal, nonanal and 2-nonenal contents increased due to dietary CLA (p = 0.016, 0.024 and 0.019, respectively).  相似文献

β-Lactoglobulin (β-LG) is a member of the lipocalin protein family and can bind a variety of hydrophobic molecules, such as fatty acids, in vitro. In this study, a potential colon-targeted antitumor drug was developed using bovine β-LG as a carrier loaded with cis-9, trans-11 conjugated linoleic acid (CLA). The intrinsic tryptophan fluorescence intensity of β-LG monitored by spectrofluorometer showed that 2.46 mol of CLA can be bound per mole of β-LG. Dynamic light scattering showed the formation of a β-LG-CLA self-assembled complex with particle size of 170 ± 0.08 nm. After treatment with gastrointestinal pH and digestive enzymes, β-LG-CLA complex showed very good stability in gastrointestinal conditions in vitro, measured by zeta potential analyzer and sodium dodecyl sulfate PAGE, respectively. In an intestinal model in vitro, the concentration of CLA in Caco-2 cells was detected by reverse-phase HPLC, and the level of CLA in cells after treatment with β-LG-CLA complex was significantly greater than after treatment with CLA, which means β-LG served as a capsular vehicle of CLA for intracellular transport. According to cell proliferation assay, β-LG-CLA complex can inhibit the viability of Caco-2 cells, and the inhibition rate is significantly greater than with the same concentration of CLA (100 μM). The study revealed that bovine β-LG as a carrier binding with CLA can potentially be used for colon cancer therapy.  相似文献

Two strains each of Propionibacterium freudenreichii ssp. shermanii and P. freudenreichii ssp. freudenreichii were tested for their ability to produce conjugated linoleic acid (CLA) in sodium lactate medium (SLM), De Man-Rogosa-Sharpe (MRS) medium and skim-milk. Data showed that both strains were able to produce CLA in three media supplemented with different concentrations of sunflower oil. Maximum production of CLA (78.8 μg/ml) was observed after 36 h of incubation in MRS containing 12 mg/ml of sunflower oil by P. freudenreichii ssp. shermanii. Moreover, the growths of both strains were inhibited by sunflower oil and a positive relationship between CLA production and ability to tolerate sunflower oil was observed. At the same time, it was also observed that the inhibitory effects on P. freudenreichii ssp. shermanii and P. freudenreichii ssp. freudenreichii in three media follow the order SLM > skim-milk > MRS and SLM > MRS > skim-milk, respectively. Micro aerobic conditions were in favour of increasing the amounts of CLA. The amounts of CLA increased from 0 to 36 h under micro aerobic conditions and no significant (p > 0.05) increases in total CLA levels were observed after 80 h of incubation. Results showed that P. freudenreichii may have potential for producing CLA.  相似文献

Effects of culture pH and corn oil (CO) concentration on biohydrogenation (BH) of unsaturated fatty acids and disappearance of neutral detergent fiber (NDF) in batch culture were evaluated in a 2 × 3 factorial design experiment. Culture vessels (100 mL; 4 replicates/treatment per time point) included ground alfalfa hay plus CO at 0, 1, or 2% dry matter inclusion rate and were incubated at pH 5.8 (low pH) or 6.2 (high pH) for 0, 6, 12, 18, or 24 h. Effects of culture pH, CO, time, and their interactions were determined. Adding CO increased total fatty acid concentration in substrates to 1.01, 2.31, and 3.58% dry matter for 0, 1, and 2% CO, respectively. Corn oil concentration interacted with culture pH and resulted in different effects on BH of cis-9,cis-12 18:2 at low or high culture pH. After 24 h of incubation, low pH, compared with high pH, reduced disappearance of NDF by 35% and BH extent of cis-9,cis-12 18:2 by 31%. Increasing CO increased disappearance of NDF across pH treatments and decreased BH extent of cis-9,cis-12 18:2 at low pH and increased it at high pH over 24 h. Compared with high pH, low pH reduced concentrations of 18:0 by 31% and increased concentrations of trans-10,cis-12 18:2 and trans-10 18:1 by 110 and 79% after 24 h, respectively. Adding CO at low pH had greater effect on BH intermediates of cis-9,cis-12 18:2 compared with adding oil at high pH. In particular, increasing CO to 1 and 2% DM at low pH, compared with at high pH, resulted in a 36 and 46% reduction in the concentration of 18:0, an 84 and 131% increase in the concentration of trans-10,cis-12 18:2, and an 81 and 129% increase in the concentration of trans-10 18:1, respectively. Despite the interactions between culture pH and CO concentration, main effects across time were also significant for the response variables of interest. In conclusion, culture pH interacted with CO concentration to affect BH of UFA and disappearance of NDF in batch culture, as the effects were greater at low culture pH than at high culture pH.  相似文献

Based on the potential benefits of cis-9, trans-11 conjugated linoleic acid (CLA) for human health, there is a need to develop effective strategies for enhancing milk fat CLA concentrations. Levels of cis-9, trans-11 CLA in milk can be increased by supplements of fish oil (FO) and sunflower oil (SO), but there is considerable variation in the response. Part of this variance may reflect time-dependent ruminal adaptations to high levels of lipid in the diet, which lead to alterations in the formation of specific biohydrogenation intermediates. To test this hypothesis, 16 late lactation Holstein-British Friesian cows were used in a repeated measures randomized block design to examine milk fatty acid composition responses to FO and SO in the diet over a 28-d period. Cows were allocated at random to corn silage-based rations (8 per treatment) containing 0 (control) or 45 g of oil supplement/kg of dry matter consisting (1:2; wt/wt) of FO and SO (FSO), and milk composition was determined on alternate days from d 1. Compared with the control, the FSO diet decreased mean dry matter intake (21.1 vs. 17.9 kg/d), milk fat (47.7 vs. 32.6 g/kg), and protein content (36.1 vs. 33.3 g/kg), but had no effect on milk yield (27.1 vs. 26.4 kg/d). Reductions in milk fat content relative to the FSO diet were associated with increases in milk trans-10 18:1, trans-10, cis-12 CLA, and trans-9, cis-11 CLA concentrations (r2 = 0.74, 0.57, and 0.80, respectively). Compared with the control, the FSO diet reduced milk 4:0 to 18:0 and cis 18:1 content and increased trans 18:1, trans 18:2, cis-9, trans-11 CLA, 20:5 n-3, and 22:6 n-3 concentrations. The FSO diet caused a rapid elevation in milk cis-9, trans-11 CLA content, reaching a maximum of 5.37 g/100 g of fatty acids on d 5, but these increases were transient, declining to 2.35 g/100 g of fatty acids by d 15. They remained relatively constant thereafter. Even though concentrations of trans-11 18:1 followed the same pattern of temporal changes as cis-9, trans-11 CLA, the total trans 18:1 content of FSO milk was unchanged because of the concomitant increases in the concentration of other isomers (Δ4-10 and Δ12-15), predominantely trans-10 18:1. In conclusion, supplementing diets with FSO enhances milk fat cis-9, trans-11 CLA content, but the high level of enrichment declines because of changes in ruminal biohydrogenation that result in trans-10 replacing trans-11 as the major 18:1 biohydrogenation intermediate formed in the rumen.  相似文献

The objective of the current study was first to characterize lipid raft microdomains isolated as detergent-resistant membranes (DRM) from mammary gland tissue, and second to determine how dietary fatty acids (FA) such as conjugated linoleic acid (CLA), 19:1 cyclo, and long-chain n-3 polyunsaturated FA affect lipid raft markers of mammary cells, and to finally establish relationships between these markers and lactation performance in dairy cows. Eight Holstein cows were used in a replicated 4 × 4 Latin square design with periods of 28 d. For the first 14 d, cows received daily an abomasal infusion of (1) 406 g of a saturated FA supplement (112 g of 16:0 + 230 g of 18:0) used as a control; (2) 36 g of a CLA supplement (13.9 g of trans-10,cis-12 18:2) + 370 g of saturated FA; (3) 7 g of Sterculia fetida oil (3.1 g of 19:1 cyclo, STO) + 399 g of saturated FA; or (4) 406 g of fish oil (55.2 g of cis-5,cis-8,cis-11,cis-14,cis-17 20:5 + 59.3 g of cis-4,cis-7,cis-10,cis-13,cis-16,cis-19 22:6, FO). Mammary biopsies were harvested on d 14 of each infusion period and were followed by a 14-d washout interval. Cholera toxin subunit B, which specifically binds to ganglioside M-1 (GM-1), a lipid raft marker, was used to assess its distribution in DRM. Infusions of CLA, STO, and FO were individually compared with the control, and significance was declared at P ≤ 0.05. Milk fat yield was decreased with CLA and FO, but was not affected by STO. Milk lactose yield was decreased with CLA and STO, but was not affected by FO. Mammary tissue shows a strong GM-1-signal enrichment in isolated DRM from mammary gland tissue. Caveolin (CAV) and flotillin (FLOT) are 2 proteins considered as lipid raft markers and they are present in DRM from mammary gland tissue. Distributions of GM-1, CAV-1, and FLOT-1 showed an effect of treatments determined by their subcellular distributions in sucrose gradient fractions. Regardless of treatments, data showed positive relationships between the yield of milk fat, protein, and lactose, and the abundance GM-1 in DRM fraction. Milk protein yield was positively correlated with relative proportion of FLOT-1 in the soluble fraction, whereas lactose yield was positively correlated with relative proportion of CAV-1 in the DRM fractions. Infusion of CLA decreased mRNA abundance of CAV-1, FLOT-1, and FLOT-2. Regardless of treatments, a positive relationship was observed between fat yield and mRNA abundance of FLOT-2. In conclusion, although limited to a few markers, results of the current experiment raised potential links between variation in specific biologically active component of raft microdomains in bovine mammary gland and lactation performances in dairy cows.  相似文献

The objective of the study was to determine the effect of fatty acid additions to the cells and enzyme extract of Lactobacillus delbrueckii ssp. bulgaricus (CCRC14009) on CLA production. Washed cells of L. delbrueckii ssp. bulgaricus, obtained by cultivation in a MRS broth, were mixed with BSA and each of the three fatty acids: linoleic, oleic, and linolenic acids in sodium phosphate buffer at pH 6.5. After incubation at 37 °C for 108 h, CLA concentration was analyzed by HPLC. Enzyme extract from the culture was also reacted with each fatty acid at 50 °C for 10 min at pH 5 to test for CLA production. Results showed that linoleic acid addition to the culture improved CLA production, indicating the presence of linoleic acid isomerase activity in the culture. The crude enzyme extract from the culture was observed to be capable of oleic and linolenic acid conversions into CLA, demonstrating the possible presence of desaturase activity in the enzyme extract.  相似文献

The present study was conducted to determinate the responsiveness of different levels of dietary conjugated linoleic acid (CLA) on intramuscular fat (IMF) and fatty acid composition of skeletal muscle and fat in pigs fed from 59.5 to 133.5 kg. Forty female Large White × (Large White × Landrace) pigs were used. Four levels (0%, 0.5%, 1% and 2%) of a commercial enriched CLA oil supplementation (60% of CLA isomers, 30% cis-9, trans-11 and 30% trans-10, cis-12) were fed to pigs. Carcass, ham, foreleg and loin weights were recorded. Dietary CLA enrichment increased the loin weight (P < 0.01) and the combined weights of hams + forelegs + loins (P < 0.02). IMF content in Longissimus dorsi was also increased by dietary CLA treatment (P < 0.001) and a linear response was observed. Dietary CLA increased saturated fatty acids (SFA) and decreased monounsaturated fatty acids (MUFA) in muscle and adipose tissue (P < 0.001). Feeding 1% CLA to finishing swine increases IMF in heavy pigs slaughtered at an average weight of 133.5 kg.  相似文献

Three levels (0%, 1% and 2%) of a conjugated linoleic acid enriched oil (CLA) were combined with two levels of monounsaturated fatty acids (MUFA) (19% and 39% average) for pig feeding. Changes in instrumental colour, lipid oxidation (thiobarbituric acid reactive substances, TBARS) and volatile profile of fresh loin chops, as affected by dietary CLA, MUFA and CLA × MUFA, were studied throughout 7 days of refrigerated storage. Lightness (L∗) evolution was conditioned by dietary CLA, whereas changes in redness (a∗) and yellowness (b∗) were unaffected by dietary supplements. Dietary CLA at 2% led to higher TBARS values of loin chops at day 7 of refrigerated storage (p < 0.05), while MUFA supplementation and CLA × MUFA interaction did not affect lipid oxidation. Dietary CLA, MUFA or CLA × MUFA did not affect most volatile compounds of loin chops after 7 days of storage.  相似文献

Effects of condensed tannins (CT), either via extract or plant-bound, and saponin extract on ruminal biohydrogenation of α-linolenic acid (ALA) were investigated in vitro. Grass-clover hay served as basal diet (control). The control hay was supplemented with extracts contributing either CT from Acacia mearnsii [7.9% of dietary dry matter (DM)] or saponins from Yucca schidigera (1.1% of DM). The fourth treatment consisted of dried sainfoin (Onobrychis viciifolia), a CT-containing forage legume, in an amount also providing 7.9% CT in dietary DM. All diets were supplemented with linseed oil at a level contributing 60% of total dietary ALA in all treatments. Diets were incubated for 10 d (n = 4) in the rumen simulation technique system, using the last 5 d for statistical evaluation. Fatty acids were analyzed in feed, feed residues, incubation fluid, and its effluent. Data were subjected to ANOVA considering diet and experimental run as main effects. Both CT treatments reduced ruminal fiber and crude protein degradation, and lowered incubation fluid ammonia concentration. Only the CT extract suppressed methane formation and shifted microbial populations toward bacteria at cost of protozoa. The saponin extract remained without clear effects on fermentation characteristics except for increased protozoal counts. The extent of ALA biohydrogenation was 20% less with the CT plant, but this probably resulted from reduced organic matter degradability rather than from an inhibition of biohydrogenation. After incubation analysis of incubation fluid effluent and feed residues showed a considerable proportion of the 3 biohydrogenation intermediates, cis-9, trans-11, cis-15 C18:3, trans-11, cis-15 C18:2, and trans-11 C18:1, which did not occur in the initial feeds. Only the CT-extract diet led to a different profile in the effluent compared with the control diet with trans-11 C18:1 being considerably increased at cost of C18:0. This could have been achieved by suppressing protozoa and enhancing the bacterial population, thus removing potential microbes involved in biohydrogenation and increasing competition between bacteria involved in biohydrogenation and others. The elevation of trans-11 C18:1 as the precursor of cis-9, trans-11 conjugated linoleic acid formed in body tissue and mammary gland is probably favorable from a human health point of view.  相似文献

During biohydrogenation-induced milk fat depression (MFD), nutrients are spared from milk fat synthesis and are available for other metabolic uses. Acetate is the major carbon source spared and it may increase lipid synthesis in adipose tissue during MFD. The objective of this study was to compare the effect of trans-10,cis-12 conjugated linoleic acid (CLA) and the amount of acetate spared during CLA-induced MFD on adipose tissue lipogenesis. Nine multiparous, lactating, ruminally cannulated Holstein cows (244 ± 107 d in milk; 25 ± 8.4 kg of milk/d; mean ± standard deviation) were randomly assigned to treatments in a 3 × 3 Latin square design. Experimental periods were 4 d followed by a 10-d washout. Treatments were control (CON), ruminal infusion of acetate (AC; continuous infusion of 7 mol/d adjusted to pH 6.1 with sodium hydroxide), or abomasal infusion of CLA (10 g/d of both trans-10,cis-12 CLA and cis-9,trans-11 CLA). Dry matter intake, milk yield, and milk protein yield and percentage were not affected by treatments. Compared with CON, milk fat yield decreased 23% and fat percent decreased 28% in CLA, and milk fat yield increased 20% in AC. Concentration and yield of milk de novo synthesized fatty acids (<C16) were reduced and concentration of preformed fatty acids (>C16) was increased by CLA, compared with CON. Yield of de novo synthesized fatty acids and palmitic acid was increased by AC, compared with CON. Lipogenesis capacity of adipose tissue explants was decreased 72% by CLA, but was not affected by AC. Acetate oxidation by adipose explants was not affected by treatments. Treatments had no effect on expression of key lipogenic factors, lipogenic enzymes, and leptin; however, expression of fatty acid binding protein 4 was reduced in CLA compared with CON. Additionally, hormone-sensitive lipase and perilipin 1 were decreased by CLA and acetate. Plasma glucose and glucagon concentrations were not affected by treatments; however, CLA increased nonesterified fatty acids 17.7%, β-hydroxybutyrate 16.1%, and insulin 27.8% compared with CON, and AC increased plasma β-hydroxybutyrate 18%. In conclusion, during CLA-induced MFD in low-producing cow adipose tissue was sensitive to the anti-lipogenic effects of CLA, while spared acetate did not stimulate adipose lipogenesis. However, acetate may play an important role in stimulating lipogenesis and improving energy status in the mammary gland under normal conditions.  相似文献

The objective of this study was to investigate the effect of dietary fiber level on milk fat concentration, yield, and fatty acid (FA) profile of cows fed diets low in polyunsaturated fatty acid (PUFA). Six rumen-fistulated Holstein dairy cows (639 ± 51 kg of body weight) were used in the study. Cows were randomly assigned to 1 of 2 dietary treatments, a high fiber (HF; % of dry matter, 40% corn silage, 27% alfalfa silage, 7% alfalfa hay, 18% protein supplement, 4% ground corn, and 4% wheat bran) or a low fiber (LF; % of dry matter, 31% corn silage, 20% alfalfa silage, 5% alfalfa hay, 15% protein supplement, 19% ground wheat, and 10% ground barley) total mixed ration. The diets contained similar levels of PUFA. The experiment was conducted over a period of 4 wk. Ruminal pH was continuously recorded and milk samples were collected 3 times a week. Milk yield and dry matter intake were recorded daily. The rumen fluid in cows receiving the LF diet was below pH 5.6 for a longer duration than in cows receiving the HF diet (357 vs. 103 min/d). Neither diet nor diet by week interaction had an effect on milk yield (kg/d), milk fat concentration and yield, or milk protein concentration and yield. During wk 4, milk fat concentration and milk fat yield were high and not different between treatments (4.30% and 1.36 kg/d for the HF treatment and 4.31% and 1.33 kg/d for the LF treatment, respectively). Cows receiving the LF diet had greater milk concentrations (g/100 g of FA) of 7:0; 9:0; 10:0; 11:0; 12:0; 12:1; 13:0; 15:0; linoleic acid; FA <C16; and PUFA; and lower concentrations of iso 15:0; 18:0; trans-9 18:1; cis-9, trans-11 conjugated linoleic acid (CLA); trans-9, cis-12 18:2; 20:0; and cis-9 20:1 compared with cows receiving the HF diet. Milk concentrations (g/100 g of FA) of total trans 18:1; trans-10 18:1; trans-11 18:1; trans-10, cis-12 CLA, and trans-9, cis-11 CLA were not different between treatments. The study demonstrated that cows fed a diet low in fiber and low in PUFA may exhibit subacute ruminal acidosis and moderate changes to milk fatty acid profile but without concomitant milk fat depression. The changes in FA profile may be useful for the diagnosis of SARA even in the absence of milk fat depression.  相似文献

The objective of this study was to evaluate the effect of solids dilution rate (SDR) and oil source [soybean oil (SBO) or linseed oil (LSO)] on the ruminal production of trans C18:1 and conjugated linoleic acid (CLA). A dual-flow continuous culture system consisting of 4 fermenters was used in a 4 × 4 Latin square design with a factorial arrangement of treatments over 4 consecutive periods of 10 d each. Treatment diets (50:50 forage to concentrate) were fed at 120 g/d of dry matter (DM) in 3 equal portions. The concentrate mix contained 1% fish oil and either 2% SBO or 2% LSO on a DM basis. Treatments were as follows: 1) SBO at 6%/h SDR, 2) SBO at 3%/h SDR, 3) LSO at 6%/h SDR, and 4) LSO at 3%/h SDR. The oil source by SDR interaction was not significant for trans C18:1 and CLA fatty acids. The concentrations of trans C18:1 and vaccenic acid were greater in effluents when diets were supplemented with SBO vs. LSO (37.11 vs. 34.09 and 32.71 vs. 29.70 mg/g of DM, respectively) and at high SDR than low SDR (37.60 vs. 33.61 and 32.72 vs. 29.61 mg/g of DM, respectively). The concentration of cis-9, trans-11 CLA in effluents was also greater with SBO than LSO (0.81 vs. 0.40 mg/g of DM) supplementation and at high SDR than low SDR (0.68 vs. 0.54 mg/g of DM). Biohydrogenation of linoleic acid and linolenic acid increased at higher SDR within each oil treatment. Based on these results, SBO supplementation at high SDR enhances ruminal production of vaccenic acid, and therefore could potentially enhance cis-9, trans-11 CLA in milk fat through synthesis by Δ9-desaturase.  相似文献
