The main dietary sources of trans fatty acids are partially hydrogenated vegetable oils (PHVO), and products derived from polyunsaturated fatty acid biohydrogenation (PUFA‐BHP) in ruminants. Trans fatty acid intake has historically been associated with negative effects on health, generating an anti‐trans fat campaign to reduce their consumption. The profiles and effects on health of PHVO and PUFA‐BHP can, however, be quite different. Dairy products naturally enriched with vaccenic and rumenic acids have many purported health benefits, but the putative benefits of beef fat naturally enriched with PUFA‐BHP have not been investigated. The objective of the present experiment was to determine the effects of beef peri‐renal fat (PRF) with differing enrichments of PUFA‐BHP on lipid and insulin metabolism in a rodent model of dyslipidemia and insulin resistance (JCR:LA‐cp rat). The results showed that 6 weeks of diet supplementation with beef PRF naturally enriched due to flaxseed (FS‐PRF) or sunflower‐seed (SS‐PRF) feeding to cattle significantly improved plasma fasting insulin levels and insulin sensitivity, postprandial insulin levels (only in the FS‐PRF) without altering dyslipidemia. Moreover, FS‐PRF but not SS‐PRF attenuated adipose tissue accumulation. Therefore, enhancing levels of PUFA‐BHP in beef PRF with FS feeding may be a useful approach to maximize the health‐conferring value of beef‐derived fats.  相似文献

The current study evaluated the composition and relationships of polyunsaturated fatty acid biohydrogenation products (PUFA-BHP) from the perirenal (PRF) and subcutaneous fat (SCF) of yearling steers fed a 70 % grass hay diet with concentrates containing either sunflower-seed (SS) or flaxseed (FS). Analysis of variance indicated several groups or families of structurally related FA, and individual FA within these were affected by a number of novel oilseed by fat depot interactions (P < 0.05). Feeding diets containing SS increased the proportions of non-conjugated 18:2 BHP (i.e., atypical dienes, AD) and conjugated linoleic acids (CLA) with the first double bond from carbon 7 to 9, trans-18:1 isomers with double bonds from carbon 6 to 12, and these PUFA-BHP had greater proportions in SCF compared to PRF (P < 0.05). Enrichment of conjugated linolenic acids, AD and CLA isomers with the first double bond in position 11 or 12, and t-18:1 isomers with double bonds from carbon 13 to 16 were achieved by feeding diets containing FS, with PRF having greater proportions than SCF (P < 0.05). Principal component analysis visually confirmed interaction effects on these groups/families of FA, and further confirmed or suggested a number of relationships between PUFA-BHP. Feeding SS or FS in a grass hay diet and exploiting adipose tissue differences, therefore, present unique opportunities to differentially enrich a number of PUFA-BHP which seem to have positive health potential in humans (i.e., t11-18:1, c9,t11-18:2 and c9,t11,c15-18:3).  相似文献

Trans-10, cis-12 conjugated linoleic acid (CLA) is a potent inhibitor of milk fat synthesis in the cow and similarly reduces milk fat in rodents. The objective of this study was to determine whether dietary fat can overcome CLA inhibition of milk fat concentration in lactating mice. Wild type C57Bl/6J mice (n = 31) were fed semipurified diets containing either low fat (LF; 4% fat) or high fat (HF; 23.6% fat) starting 4–6 days postpartum. Dietary fat was increased by inclusion of high oleic sunflower oil. After 2 days on the experimental diets, lactating dams were orally dosed with either water (control) or trans-10, cis-12 CLA (20 mg/day) for 5 days. CLA treatment decreased pup growth similarly in both HF and LF diets. Milk fat percent was increased over 16% by the HF diet and decreased over 12% by CLA, but there was no interaction of dietary fat and CLA. Both CLA and the HF diet reduced the proportion of short- and medium-chain fatty acids that originate from de novo synthesis, and there was no interaction of diet and CLA. CLA had no effect on the percent of preformed fatty acids, but the HF diet increased their abundance. Dietary fat and CLA both modified mammary expression of lipogenic enzymes and regulators, but no interactions were observed. In conclusion, CLA reduced milk fat concentration and litter growth, but these effects were not overcome by increased dietary fat from high oleic sunflower oil. CLA inhibition of milk fat in the mammary gland is not substrate dependent, and the mechanism is independent from dietary supply of oleic acid.  相似文献

Bioactive and biocompatible conjugated linoleic acid (CLA) has been only considered as a food or medicine ingredient due to its rare natural occurrence. In this work, the surface activities and pH‐induced self‐assembling behaviors of the semi‐synthetic CLA molecules into micelles or vesicles were systematically investigated. First, the self‐assembling of CLA was studied in detail, and it was found that aside from temperature and ionic strength, pH is the prominent factor affecting the self‐assembling of CLA. Moreover, stable CLA ufasomes (unsaturated fatty acid liposomes) in uniform size were obtained by self‐crosslinking of the CLA ufasomes, and the morphologies of the crosslinked CLA assemblies were recorded by transmission electron microscopy, which made known the pH‐induced formation of the CLA ufasomes or the CLA micelles. The crosslinked CLA assemblies presented improved properties such as a higher calcium stability, a lower lime soap dispersing requirement and a better solubilization ability than that of the CLA molecules themselves or the pre‐crosslinked linoleic acids. These investigations could be helpful for comprehensively understanding effects of environment factors on self‐assembling behaviors of conjugated fatty acids and responsive polymerization of polymerizable surfactants.  相似文献

The incorporation of vaccenic acid (VA, 0.5 and 1.2%), conjugated linoleic acid (CLA, mixture of primarily c9,t11‐ and t10,c12‐CLA, 1.2%), linoleic acid (LA, 1.2%) and oleic acid (OA, 1.2%) into different tissues of mice was examined. The effects on the fatty acid composition of triacylglycerols (TAG) and phospholipids (PL) in kidney, spleen, liver and adipose tissue were investigated. VA and CLA (c9,t11‐ and t10,c12‐CLA) were primarily found in TAG, especially in kidney and adipose tissue, respectively. Conversion of VA to c9,t11‐CLA was indicated by our results, as both fatty acids were incorporated into all the analyzed tissues when a diet containing VA but not c9,t11‐CLA was fed. Most of the observed effects on the fatty acid profiles were seen in the CLA group, whereas only minor effects were observed in the VA groups compared with the OA group. Thus, CLA increased n‐3 polyunsaturated fatty acids (PUFA) in PL from kidney and spleen and lowered the ratio of n‐6/n‐3 PUFA in these tissues. Furthermore, CLA increased C22 PUFA in the PL fraction of kidney, spleen and liver, but reduced the level of arachidonic acid in PL of liver and spleen and lowered the Δ9‐desaturation indexes in all analyzed tissue TAG.  相似文献

The influence of the distribution of polyunsaturated fatty acids on the glycerol backbone of dietary triacylglycerols on the fatty acid profile of adipose tissue and muscle phospholipids was investigated in growing‐finishing pigs (48) and broiler chicken (84). The animals were fattened on barley/soybean meal diets supplemented with a blend of soybean oil and beef tallow, either in the ratio 3:1 w/w (high‐PUFA) or 1:3 w/w (low‐ PUFA). Part of the high‐ and low‐PUFA blends was chemically interesterified to randomly distribute all fatty acids over the three positions of the glycerol. Thus, two sets of diets of identical overall fatty acid composition, but differing in the distribution of fatty acids in the triacylglycerols, were fed. Growth performance and carcass composition were neither affected by fatty acid composition nor by randomisation of dietary fats in either animal species. Apparent digestibility of energy was slightly lower in pigs fed the low‐PUFA blends. Fatty acid profile of subcutaneous fat of pigs and broilers as well as of internal body fat (lamina subserosa) and muscle phospholipids of pigs varied according to the dietary fatty acid composition but was not affected by randomisation of dietary fats. These findings are explained in terms of the hydrolysis of TAG during transport of lipids from enterocytes to adipose tissue cells and the continuous lipolysis and re‐esterification of fatty acids that take place in adipose tissue cells.  相似文献

Liver fatty acid binding protein (Fabp1) and sterol carrier protein‐2/sterol carrier protein‐x (SCP2/SCPx) genes encode proteins that enhance hepatic uptake, cytosolic transport, and peroxisomal oxidation of toxic branched‐chain fatty acids derived from dietary phytol. Since male wild‐type (WT) mice express markedly higher levels of these proteins than females, the impact of ablating both genes (TKO) was examined in phytol‐fed males. In WT males, high phytol diet alone had little impact on whole body weight and did not alter the proportion of lean tissue mass (LTM) versus fat tissue mass (FTM). TKO conferred on dietary phytol the ability to induce weight loss as well as reduce liver weight, FTM, and even more so LTM. Concomitantly TKO induced hepatic lipid accumulation, preferentially threefold increased phospholipid (PL) at the expense of decreased triacylglycerol (TG) and total cholesterol. Increased PL was associated with upregulation of membrane fatty acid transport/translocase proteins (FATP 2,4), cytosolic fatty acid/fatty acyl‐CoA binding proteins (FABP2, ACBP), and the rate limiting enzyme in PL synthesis (Gpam). Decreased TG and cholesterol levels were not attributable to altered levels in respective synthetic enzymes or nuclear receptors. These data suggest that the higher level of Fabp1 and Scp2/Scpx gene products in WT males was protective against deleterious effects of dietary phytol, but TKO significantly exacerbated phytol effects in males.  相似文献

Mixed‐isomer conjugated linoleic acid (CLA) and the individual isomers, trans‐10, cis‐12 (CLAt10c12) and cis‐9, trans‐11 (CLAc9t11), decrease severity of collagen‐induced arthritis (CA) when consumed after disease onset. Few studies have been conducted exploring the role of CLA in the prevention of autoimmune diseases. These studies suggest that isomer‐specific effects may be occurring; however, a direct comparison of CLAt10c12 and CLAc9t11 has yet to be conducted. A study to compare the ability of CLAt10c12 and CLAc9t11 to prevent CA and assess their effects on early inflammation was performed. DBA/1 mice were fed a semipurified diet containing 6% corn oil (CO), 5.5% CO and 0.5% CLAt10c12, or 5.5% CO and 0.5% CLAc9t11 (n = 27 per diet) starting three weeks before CA primary immunization. Effects on disease incidence and severity, anticollagen antibodies, plasma and paw cytokines, and hepatic fatty acids were measured. Arthritis incidence was reduced by a minimum of 34% in mice fed either CLA isomer compared to those fed CO diet (p = 0.06). In mice that did develop arthritis (n = 9–12 mice per treatment), CLAt10c12 reduced arthritic severity to a greater extent than CLAc9t11 and CO (p = 0.03). CLA isomer treatment attenuated the increased hepatic arachidonic acid (ARA; 20:4n‐6) observed with arthritis at one‐week postonset (p = 0.03), while no differences in anticollagen antibodies or cytokines were observed between dietary treatments. These results suggest that CLA isomers may be effective at preventing specific immune‐mediated inflammatory diseases, in part, through modulation of the ARA cascade.  相似文献

In a performance trial, broiler chickens received 29 g per kg feed of a preparation containing 70% linoleic acid (LA) in the control treatment and another preparation containing approximately the same amount of conjugated linoleic acids (CLA) in the experimental treatment. Diets of CLA treatment contained 18 g CLA per kg feed. The CLA preparation contained the isomers cis‐9,trans‐11 and trans‐10,cis‐12 at a proportion 1:1, other CLA isomers were quantitively negligible. Performance parameters (weight gain and feed conversion ratio over a 42 day period) were not significantly influenced by CLA intake. However, fat content of liver, breast, and leg muscles was reduced and protein contents in liver and leg muscles were elevated significantly. Fat to protein ratios in the main edible parts were shifted in favour of protein in CLA treated animals. In all analysed tissue lipids the content of saturated fatty acids was increased and that of monounsaturated fatty acids was decreased significantly. At the same time CLA was incorporated in tissue lipids effectively reaching more than 10 g per 100 g of total fatty acids. With regard to isomers the cis‐9,trans‐11 isomer was found in higher concentrations in tissue lipid fractions compared to the trans‐10,cis‐12 isomer. It was concluded that nutrient repartitioning due to CLA intake described for other species is also valid for broilers. Using appropriate feeding strategies it is possible to produce CLA enriched food from broilers.  相似文献

Conjugated isomers of linoleic acid have beneficial effects on human health, with demonstrated anticarcinogenic, antiatherogenic, antioxidant, and anti‐inflammatory properties. Previous studies have shown that the loss of sex hormones induced by orchidectomy affects vascular function and serum lipids. The objective of this work is to evaluate the effect of conjugated linoleic acid (CLA) on serum lipids, cardiac, renal, and hepatic markers in orchidectomized Sprague‐Dawley rats. Male sex hormone deprivation is induced by orchidectomy to 24‐week‐old rats. The rats are fed a diet containing 1.8% w/w CLA, or a control diet for 8 weeks. At the end of the treatment, serum markers are measured using commercially available kits. The concentrations found in the orchidectomized‐CLA (ORX‐CLA) group of total cholesterol, low‐density lipoproteins (LDL), as well as the atherogenic index, concentrations of creatine kinase, lactate dehydrogenase, C‐reactive protein, glucose, urea, uric acid, creatinine, albumin, calcium, total protein, and bilirubin are significantly lower compared to the orchidectomized‐control (ORX‐C) group. The concentrations of high‐density lipoproteins (HDL), calcium, total protein, albumin, total bilirubin, and indirect bilirubin are significantly higher in the ORX‐CLA group than in the ORX‐C group. The CLA‐supplemented diet exerts a beneficial effect on different biochemical parameters negatively affected by orchidectomy, which may result in vascular protection. Practical Applications: The relationship between testosterone deficiency and the alteration of serum lipids is demonstrated; however, it is demonstrated in this study that the markers of renal damage, cardiac damage, and oxidative stress are also negatively affected by the lack of this hormone. This information has enriched the understanding about the relationship between testosterone deficiency and the development of different diseases such as metabolic syndrome, type 2 diabetes, kidney failure, osteoporosis, cardiovascular diseases, and cancer. On the other hand, CLA has been studied mainly as a nutraceutical and sports supplement; the information in this study will help to expand the knowledge about the benefits of these isomers of linoleic acid, providing guidelines for its use as a protection against negative effects triggered by orchidectomy.  相似文献

The present study was designed to determine if feeding steers extruded flaxseed and hay (25 and 75%; DM basis) together as a total mixed ration (TMR), or sequentially (non-TMR) would result in different enrichments of polyunsaturated fatty acids (PUFA) and their biohydrogenation intermediates (BHI) in beef adipose tissues [subcutaneous (SC) vs perirenal (PR) fat]. Forty-eight Angus cross steers (325 ± 16 kg) were stratified by weight to six pens, and pens were randomized to either TMR or non-TMR and fed ad libitum for an average of 242 days. The concentrations of α-linolenic acid increased by 18 mol% in both SC and PR in non-TMR steers compared to TMR steers (P < 0.01). trans 18:1 isomers were more concentrated in PR than SC (14.4 vs 9.5 mol%; P < 0.01) and increased by 10 mol% in both fat depots for non-TMR (P < 0.01). Other BHI including non-methylene-interrupted 18:2 (atypical dienes), conjugated linoleic acids and conjugated linolenic acids (CLnA) were affected by diet × tissue interactions (P < 0.01). The CLnA and CLA contents were higher in both fat depots when feeding the non-TMR, but the effect of diet was more pronounced in PR than in SC (P < 0.01). Atypical dienes were highest in PR from non-TMR and lowest in TMR fed steers (4.3 and 3.6 mol%) with SC contents being intermediate. The sequential feeding of lipid supplement can thus profoundly affect the enrichment of PUFA and their BHI in beef fat and their differentially enrichment is also fat depot dependant.  相似文献

The consumption of omega‐3 polyunsaturated fatty acids (n‐3 PUFA) is associated with a reduced risk of breast cancer. Studies in animals and in vitro have demonstrated mechanisms that could explain this apparent effect, but clinical and epidemiological studies have returned conflicting results on the practical benefits of dietary n‐3 PUFA for prevention of breast cancer. Effects are often only significant within a population when comparing the highest n‐3 PUFA consumption group to the lowest n‐3 group or highest n‐6 group. The beneficial effects of n‐3 PUFA eicosapentaenoic and docosahexaenoic on the risk of breast cancer are dose dependent and are negatively affected by total n‐6 consumption. The majority of the world population, including the most highly developed regions, consumes insufficient n‐3 PUFA to significantly reduce breast cancer risk. This review discusses the physiological and dietary context in which reduction of breast cancer risk may occur, some proposed mechanisms of action and meaningful recommendations for consumption of n‐3 PUFA in the diet of developed regions.  相似文献

trans 10,cis 12‐CLA has been reported to alter fatty acid composition in several non‐neurological tissues, but its effects are less known in neurological tissues. Therefore, the purpose of this study was to determine if CLA supplementation would alter brain and eye fatty acid composition and if those changes could be prevented by concomitant supplementation with docosahexaenoic acid (DHA; 22:6n3) or eicosapentaenoic acid (EPA; 20:5n3). Eight‐week‐old, pathogen‐free C57BL/6N female mice (n = 6/group) were fed either the control diet or diets containing 0.5% (w/w) t10,c12‐CLA in the presence or absence of either 1.5% DHA or 1.5% EPA for 8 weeks. CLA concentration was significantly (P < 0.05) greater in the eye but not in the brain lipids of the CLA group when compared with the control group. The sums of saturated, monounsaturated, polyunsaturated fatty acids, and n3:n6 ratio did not differ between these two groups for both tissues. The n3:n6 ratio and concentrations of 20:5n3 and 22:5n3 were significantly greater, and those of 20:4n6, 22:4n6, and 22:5n6 were lesser in the CLA + DHA and CLA + EPA groups than in the control and CLA groups for either tissue. DHA concentration was higher in the CLA + DHA group only but not in the CLA + EPA group when compared with the CLA group for both tissues. The dietary fatty acids generally induced similar changes in brain and eye fatty acid concentration and at the concentrations used both DHA and EPA fed individually with CLA were more potent than CLA alone in altering the tissue fatty acid concentration.  相似文献

A study was carried out to determine the profiles of fatty acids in fermented milks and dairy derivatives made with milk fat substituted by polyunsaturated fatty acid (PUFA)‐enriched fat. In order to improve the organoleptic properties of those products, whey protein concentrates (WPC) were added during the manufacturing process. Interest was focused during manufacturing and storage period on the contents of “healthy” fatty acids, mainly conjugated linoleic acid and n‐3 PUFA. Contents of these fatty acids were not affected by the manufacture practices and neither did addition of WPC during manufacturing nor cold storage cause their decrease. Percentages of total n‐3 fatty acids in fat from dairy derivatives enriched in PUFA after 21 d of storage (1.45%) were very close to those obtained before processing (1.39%). Contents did not differ either substantially when WPC were added during manufacturing (1.46%). The increase of volatile compounds was also examined. Although a slight decrease in the total volatile content was observed, percentages of different compounds were not modified when milk fat was substituted by PUFA enriched fat.  相似文献

Institute of Nutrition, Friedrich Schiller University of Jena, Jena, Germany In the last decade, conjugated linoleic acids (CLA) have been shown to have some beneficial (but also unfavourable) effects: anticarcinogenic properties, immune modulation, reduction of body fat and increase of lean body mass, normalisation of impaired glucose tolerance, promotion of fatty streak formation, and isomer‐specific effects. The research base on CLA has been derived almost exclusively from animal models, while some of the biological properties have been fairly well‐documented, others are still open to question. For about 5 years a lot of commercial CLA mixtures have been offered. These mixtures produced from linoleic acid‐rich oil like sunflower or safflower oil by alkali isomerization contained, besides cis‐9,trans‐11 and trans‐10,cis‐12 CLA isomers (about 20—40% of each), parts of cis,cis and trans,trans isomers as well. The quality of the recent products is significantly improved and they contain only two CLA isomers: cis‐9,trans‐11 and trans‐10,cis‐12. CLA play apparently a key role in regulating body composition. Several studies showed a reduction in body fat mass and a slight increase in lean body mass depending on the species. A possible explanation for the decrease of body fat may be a stimulation of lipolysis and a reduction of lipoprotein lipase activity in adipocytes. In adipose and muscle tissue a CLA‐stimulated increase of carnitine palmitoyltransferase activity resulting in an enhanced fatty acid oxidation was shown. There is evidence that CLA provide protection against cytokine‐induced (Tumour necrosis factor‐α, interleukin‐1) skeletal‐muscle catabolism (anabolic effect). The body composition modulating effects are most impressive in rodents and seem to become smaller in pigs and in humans. Data on humans are insufficient. Further research is essential to characterize the multifunctionality of CLA in humans, in order to identify the specific physiological mechanism of the biologically active isomers and to determine the optimal level of these isomers for beneficial effects.  相似文献

Structured phospholipids were synthesized with the functional lipid conjugated linoleic acid (CLA). The lipase‐ and phospholipase A2‐catalyzed enzymatic acidolysis reaction between phospholipids (PL) and CLA was used for fatty acid modification. Enzymatic processes were an effective way to produce structured PL. Screening of four lipases and immobilized phospholipase A2 and a combination of lipase and phospholipase showed that only Lipozyme RM IM and Lipozyme TL IM were effective in incorporation of CLA into PL. The maximum incorporation achieved by the latter enzyme was 16% with soy PL in 72 h.  相似文献

The effect of dietary C18:1 trans fatty acids on back fat composition in pigs was investigated with special emphasis on conjugated linoleic acids (CLA). A total of 12 × 4 siblings of Large White and Swiss Landrace breed were housed in groups and fattened from 22 to 103 kg live weight. Pigs were fed a control diet (barley, wheat, soybean meal) or experimental diets which consisted of the control diet with a 5% replacement of olein or stearin fractions of pork fat, or partially hydrogenated fat. The hydrogenated fat was rich in C18:1 trans fatty acids but contained only negligible amounts of CLA. In contrast olein and stearin fractions contained far less C18:1 trans fatty acids but some CLA. In the control diet no C18:1 trans fatty acids and only traces of CLA were detected. The partially hydrogenated fat led to the highest CLA content in back fat (0.44%). Intermediate amounts of CLA were measured in pigs fed the fractionated pork fat (0.22/0.23%). In pigs fed the control diet, also small amounts of CLA were detected. The results indicate that CLA may be produced by endogenous Δ9‐desaturation out of dietary trans vaccenic acid in pigs.  相似文献

The scope of this study was to assess the impact of calcium and conjugated linoleic acid (CLA) supplementation on plasma fatty acid profiles and to evaluate potential synergistic effects of both compounds against dietary obesity. Mice separated into five experimental groups were followed: control (C), high-fat diet (HF), HF with calcium (Ca), HF plus CLA and HF with both Ca and CLA. Plasma metabolites and fatty acids were determined by commercial kits and gas chromatography, respectively. Both dietary calcium and CLA supplementation contributed to lower body fat gain under a HF diet. Maximum efficacy was seen with calcium; no additional effect was associated with the combined treatment with CLA. Plasma leptin, adiponectin and HOMA index were in accordance with an altered glucose/insulin homeostasis in the HF and HF + CLA groups, whereas control levels were attained under Ca-enriched diets. Plasma fatty acids showed minor changes associated to CLA treatment, but a high impact on PUFA was observed under Ca-enriched diets. Our results show that the mechanism underlying the anti-obesity effects of calcium supplementation is mediated mainly by changes in PUFA plasma profile. In addition, the lack of synergy on body weight reduction in combination with associated lipid profiles of calcium and CLA suggests that calcium may interfere with absorption and/or bioactivity of CLA, which can be of relevance when using CLA-fortified dairy products against human obesity.  相似文献
