Two ethanol-producing yeast strains, CHY1011 and CHFY0901 were isolated from soil in South Korea using an enrichment technique in a yeast peptone dextrose medium supplemented with 5% (w v?1) ethanol at 30 °C. The phenotypic and physiological characteristics, as well as molecular phylogenetic analysis based on the D1/D2 domains of the large subunit (26S) rRNA gene and the internally transcribed spacer (ITS) 1 + 2 regions suggested that they were novel strains of Saccharomyces cerevisiae. During shaking flask cultivation, the highest ethanol productivity and theoretical yield of S. cerevisiae CHY1011 in YPD media containing 9.5% total sugars was 1.06 ± 0.02 g l?1 h?1 and 95.5 ± 1.2%, respectively, while those for S. cerevisiae CHFY0901 were 0.97 ± 0.03 g l?1 h?1 and 91.81 ± 2.2%, respectively. Simultaneous saccharification and fermentation for ethanol production was carried out using liquefied cassava (Manihot esculenta) starch in a 5 l lab-scale jar fermenter at 32 °C for 66 h with an agitation speed of 2 Hz. Under these conditions, S. cerevisiae CHY1011 and CHFY0901 yielded a final ethanol concentration of 89.1 ± 0.87 g l?1 and 83.8 ± 1.11 g l?1, a maximum ethanol productivity of 2.10 ± 0.02 g l?1 h?1 and 1.88 ± 0.01 g l?1 h?1, and a theoretical yield of 93.5 ± 1.4% and 91.3 ± 1.1%, respectively. These results suggest that S. cerevisiae CHY1011 and CHFY0901 have potential use in industrial bioethanol fermentation processes.  相似文献

Potato starch residue stream produced during chips manufacturing was used as an economical source for biomass and bioethanol production by Saccharomyces cerevisiae. Results demonstrated that 1% H2SO4 at 100 °C for 1 h was enough to hydrolyze all starch contained in the residue stream. Two strains of S. cerevisiae (y-1646 and commercial one) were able to utilize and ferment the acid-treated residue stream under both aerobic and semi-anaerobic conditions. The maximum yield of ethanol (5.52 g L?1) was achieved at 35 °C by S. cerevisiae y-1646 after 36 h when ZnCl2 (0.4 g L?1) was added. Addition of NH4NO3 as a source of nitrogen did not significantly affect either growth or ethanol production by S. cerevisiae y-1646. Some secondary by-products including alcohol derivatives and medical active compound were found to be associated with the ethanol production process.  相似文献

Sweet sorghum juice was fermented into ethanol using Saccharomyces cerevisiae (ATCC 24858). Factorial experimental design, regression analysis and response surface method were used to analyze the effects of the process parameters including juice solid concentration from 6.5 to 26% (by mass), yeast load from 0.5 g L−1 to 2 g L−1 and fermentation temperature from 30 °C to 40 °C on the ethanol yield, final ethanol concentration and fermentation kinetics. The fermentation temperature, which had no significant effect on the ethanol yield and final ethanol concentration, could be set at 35 °C to achieve the maximum fermentation rate. The yeast load, which had no significant effect on the final ethanol concentration and fermentation rate, could be set at 1 g L−1 to achieve the maximum ethanol yield. The juice solid concentration had significant inverse effects on the ethanol yield and final ethanol concentration but a slight effect on the fermentation rate. The raw juice at a solid concentration of 13% (by mass) could be directly used during fermentation. At the fermentation temperature of 35 °C, yeast solid concentration of 1 g L−1 and juice solid concentration of 13%, the predicted ethanol yield was 101.1% and the predicted final ethanol concentration was 49.48 g L−1 after 72 h fermentation. Under this fermentation condition, the modified Gompertz's equation could be used to predict the fermentation kinetics. The predicted maximum ethanol generation rate was 2.37 g L−1 h−1.  相似文献

A strain of Saccharomyces cerevisiae, which showed marked fermentation activity, ethanol and temperature tolerance and good flocculation ability, was selected for ethanol production. A stuck fermentation occurred at sucrose concentration of 25%. Increasing the yeast inoculum volume from 3% to 6% showed positive effects on fermentation from 25% sucrose. The ratio of added nitrogen to sucrose, which gave the best results (for the selected yeast strain), was determined. It was concluded that this ratio (nitrogen as ammonium sulphate at a rate of 5 mg g?1 of consumed sucrose) is constant at various sugar concentrations. Addition of nitrogen at this ratio produced 11.55% ethanol with complete consumption of 25% sucrose after 48 h of fermentation. However fermentation of 30% sucrose at the above optimum conditions was not complete. Addition of yeast extract at a level of 6 g l?1 together with thiamine at a level of 0.2 g l?1 led to complete utilization of 30% sucrose with resultant 14% ethanol production. However the selected yeast strain was not able to ferment 35% sucrose at the same optimum conditions. Addition of air at a rate of 150 dm3 min?1 m3 of reactor volume during the first 12 h of fermentation led to complete consumption of 35% sucrose and 16% ethanol was produced. Simultaneous saccharification and fermentation (SSF) of very high gravity (VHG) potato mash, containing 304 g L?1 of dissolved carbohydrates, was carried out for ethanol production. Potato tubers were ground into a mash, which was highly viscous. Mash viscosity was reduced by the pretreatment with mixed enzyme preparations of pectinase, cellulase and hemicellulase. The enzymatic pretreatment established the use of VHG mash with a suitable viscosity. Starch in the pretreated mash was liquefied to maltodextrins by the action of thermo-stable α-amylase at 85 °C. SSF of liquefied mash was performed at 30 °C with the simultaneous addition of glucoamylase, yeast (Saccharomyces cerevisiae) and ammonium sulfate as a nitrogen source for the yeast. The optimal glucoamylase loading, ammonium sulfate concentration and fermentation time were 1.65 AGU g?1, 30.2 mM and 61.5 h, respectively, obtained using the response surface methodology (RSM). Ammonium sulfate supplementation was necessary to avoid stuck fermentation under VHG condition. Using the optimized condition, ethanol yield of 16.61% (v/v) was achieved, which was equivalent to 89.7% of the theoretical yield.  相似文献

Volatile fatty acids (VFAs) are important mid-products in the production of methane, and their concentrations affect the efficiency of fermentation. However, their effects on methane yield and methanogenic bacteria growth have been less extensively studied. To address these effects, acetic acid, propionic acid, butyric acid and ethanol were used as substrates and an L9(34) orthogonal table was adopted to design anaerobic digestion tests. When the highest concentrations of ethanol, acetic acid and butyric acid were 2400, 2400 and 1800 mg L?1, respectively, there was no significant inhibition of the activity of methanogenic bacteria. However, when the propionic acid concentration was increased to 900 mg L?1, significant inhibition appeared, the bacteria concentration decreased from 6 × 107 to 0.6–1 × 107 ml?1 and their activity would not reconvert. These effects resulted in the accumulation of ethanol and VFAs, and the total methane yield consequently became very low (<321 ml). The original propionic acid concentration had a significant inhibitory effect on methanogenic bacteria growth (P < 0.01). An optimization analysis showed that ethanol, acetic acid, propionic acid and butyric acid at concentrations of 1600, 1600, 300 and 1800 mg L?1, respectively, led to the maximum accumulative methane yield of 1620 ml and the maximum methanogenic bacteria concentration of 7.3 × 108 ml?1.  相似文献

Fermentation of dilute sulfuric acid barley straw hydrolysate (BSH; undiluted/untreated) by Clostridium beijerinckii P260 resulted in the production of 7.09 gL?1 ABE (acetone butanol ethanol), an ABE yield of 0.33, and productivity of 0.10 gL?1 h?1. This level of ABE is much less than that observed in a control experiment (21.06 gL?1) where glucose (initial concentration 60 gL?1) was used as a substrate. In the control experiment, an ABE yield of 0.41 and productivity of 0.31 gL?1 h?1 were observed. This comparison suggested that BSH is toxic to the culture. To reduce this potential toxicity effect, BSH was treated with lime [Ca(OH)2] followed by fermentation. The treated BSH resulted in a successful fermentation and ABE concentration of 26.64 gL?1 was achieved. This was superior to both glucose and untreated BSH (initial sugar 60 gL?1) fermentations. In this fermentation, an ABE yield of 0.43 and productivity of 0.39 gL?1 h?1 (390% of untreated/undiluted BSH) was obtained. It should be noted that using lime treated BSH, a specific productivity of 0.55 h?1 was obtained as compared to 0.12 h?1 in the control fermentation suggesting that more carbon was directed to product formation.  相似文献

In order to obtain high ethanol yield and fermentation rate, response surface methodology (RSM) was employed to study the effect of culture medium on the ethanol productivity from stalk juice of sweet sorghum by immobilized yeast. A 23 central composite design (CCD) was chosen to explain the combined effects of the medium constituents, viz. nitrogen (adjusted by adding (NH4)2SO4), phosphorus (adjusted by adding KH2PO4), and pH. A mathematical correlation about the influence of the nitrogen, phosphorus, and pH on the ethanol productivity was established. It predicted that the maximum ethanol production rate (119.12 g/l h) was observed for a medium consisting of 0.77 g/l phosphorus, 2.15 g/l nitrogen, and pH = 6.39. Under this condition, the ethanol fermentation rate was 122.85 g/l h.  相似文献

Hydrothermal pretreatment on raw corn stover (RCS) with a loop reactor was investigated at 195 °C for different times varying between 10 min and 30 min. After pretreatment, the slurry was separated into water-insoluble solid (WIS) and liquid phase. Glucan and xylan were found in the both phases. The pretreatment condition showed a significant impact on xylan recovery. As the pretreatment time prolonged from 10 min to 30 min, the xylan recovery from liquid phase changed between 39.5% and 45.6% and the total xylan recoveries decreased from 84.7% to 61.6%. While the glucan recovery seemed not sensitive to the different pretreatment times. The glucan recovered from liquid was from 4.9% to 5.6% and the total glucan recoveries from all the pretreatments were higher than 98%. Besides HMF and furfural, acetic, lactic, formic and glycolic acids were also found in the liquid phase. All the concentrations of these potential inhibitors were lower enough not to affect the activity of the Saccharomyces cerevisiae (S. cerevisiae). Compared with the ethanol production of 32.4% from the RCS with S. cerevisiae, all the WISs gave higher ethanol productions ranging between 61.2% and 71.2%. When the xylan was taken into consideration, the best pretreatment condition would be 195 °C, 15 min and the estimated total ethanol production was 201 g kg?1 RCS by assuming the fermentation of both C-6 and C-5 with the ethanol yield of 0.51 g g?1 and 0.47 g g?1, respectively.  相似文献

Bio-hydrogen production by combined dark and light fermentation of ground wheat starch was investigated using fed-batch operation. Serum bottles containing heat-treated anaerobic sludge and a mixture of Rhodobacter sp. was fed with a medium containing 20 g dm?3 wheat powder (WP) at a constant flow rate. The system was operated at different initial dark/light biomass ratios (D/L). The optimum D/L ratio was 1/2 yielding the highest cumulative hydrogen (1548 cm3), yield (65.2 cm3 g?1 starch), and specific hydrogen production rate (5.18 cm3 g?1 h?1). Light fermentation alone yielded higher hydrogen production than dark fermentation due to fermentation of volatile fatty acids (VFAs) to H2 and CO2. The lowest hydrogen formation was obtained with D/L ratio of 1/1 due to accumulation of VFAs in the medium.  相似文献

The effects of five different biomass species and their chemical composition on the overall process efficiency and economic performance considering feedstock availability and feedstock costs to manufacture ethanol from lignocellulose were studied. First is a comparison of ethanol production and excess electricity generated between different biomass species. Results show that, at the same feedstock rate of 2000 Mg day?1, aspen wood has larger ethanol production than switchgrass, hybrid poplar and corn stover, while the excess electricity generated is as follows in increasing order: aspen < corn stover < hybrid poplar/switchgrass. Second, our results show that the ethanol production is largely linear with holocellulose (cellulose plus hemicellulose) composition of the various biomass species. However, the relationship between excess electricity generated and non-holocellulose combustible component is nonlinear. Last, on environmental performance, it is found that the water losses per unit ethanol production are in the following order: aspen wood < corn stover < hybrid poplar < switchgrass. While corn stover is a potential feedstock to produce cellulosic ethanol with the lowest ethanol production cost at the present time, hybrid poplar and switchgrass are the two promising future energy crops.The effects of plant size analysis showed that the estimated feedstock delivered costs, ethanol production, excess electricity generated and solid and gaseous waste emissions all increase with plant size for the various biomass species. The ethanol production costs decrease with the increase in plant size with optimal plant sizes for corn stover in the range from 2000 dry Mg day?1 to 4000 dry Mg day?1.  相似文献

Natural convection in a two-dimensional square cavity containing hydrodynamically and thermally anisotropic porous medium with internal heat generation is analyzed numerically by generalized non-Darcy approach. The properties considered for the study are permeability ratio (K1), inclination of the principal axes (θ), ratio of Forchheimer constants (F1) and thermal conductivity ratio (k1). Results are presented in terms of isotherms, streamlines and maximum temperature in the cavity to understand the flow physics. It is observed that the anisotropic properties have significant influence on the flow behaviour and heat transfer. A correlation for maximum temperature in the cavity for a wide range of parameters (107 ? Ra ? 108, 10?6 ? Da ? 10?3, 0° ? θ ? 90°, 1 ? F1 ? 100, 0.1 ? K1 ? 10 and 0.1 ? k1 ? 10) is developed.  相似文献

The structure, the thermal expansion coefficient, electrical conductivities of Ce0.8Gd0.2?xMxO2?δ (for M: Bi, x = 0–0.1, and for M: Sm, La, and Nd, x = 0.02) solid solutions, prepared for the first time hydrothermally, are investigated. The uniformly small particle size (28–59 nm) of the materials allows sintering of the samples into highly dense ceramic pellets at 1300–1400 °C. The maximum conductivity, σ700 °C around 4.46 × 10?2 S cm?1 with Ea = 0.52 eV, is found at x = 0.1 for Bi-co-doping. Among various metal-co-dopings, for x = 0.02, the maximum conductivity, σ700 °C around 2.88 × 10?2 S cm?1 with Ea = 0.67 eV, is found for Sm-co-doping. The electrolytic domain boundary (EDB) of Ce0.8Gd0.1Bi0.1O2?δ is found to be 1.2 × 10?19 atm, which is relatively lower than that of the singly doped samples. The thermal expansion coefficients, determined from high-temperature X-ray data are 11.6 × 10?6 K?1 for the CeO2, 12.1 × 10?6 K?1 for Ce0.8Gd0.2O2?δ, and increase with co-doping to 14.2 × 10?6 K?1 for Ce0.8Gd0.18Bi0.02O2?δ. The maximum power densities for the single cell based on the codoped samples are higher than that of the singly doped sample. These results suggest that co-doping can further improve the electrical performance of ceria-based electrolytes.  相似文献

Boron-doped hydrogenated microcrystalline silicon oxide (p-μc-Si:Ox:H) films have been deposited using catalytic chemical vapor deposition (Cat-CVD). The single-coiled tungsten catalyst temperature (Tfil) was varied from 1850 to 2100 °C and films were deposited on glass substrates at the temperatures (Tsub) of 100–300 °C. Different catalyst-to-substrate distances of 3–5 cm and deposition pressures from 0.1 to 0.6 Torr were considered.Optical and electrical characterizations have been made for the deposited samples. The sample transmittance measurement shows an optical-bandgap (Egopt) variation from 1.74 to 2.10 eV as a function of the catalyst and substrate temperatures. One of the best window materials was obtained at Tsub=100 °C and Tfil=2050 °C, with Egopt=2.10 eV, dark conductivity of 3.0×10?3 S cm?1 and 0.3 nm s?1 deposition rate.  相似文献

Smooth cordgrass (Spartina alterniflora), a saltmarsh plant with high production, was characterized for its potential for use as feedstock for anaerobic digestion processes. The anaerobic digestibility and biogas yield of S. alterniflora were evaluated by anaerobic batch digestion experiments performed at 35 ± 1 °C at initial volatile solids (VS) of 6%. The nutrient content analysis indicated that S. alterniflora contained the required nutrition for anaerobic microorganisms, but its high C/N of 58.8, high K and Na contents of 8.1, 22.7 g kg?1, respectively, may be disadvantageous to its anaerobic digestion. The cumulative biogas yield was determined to be 358 L kg?1 VS and the biodegradation efficiency was 45% after 60 days of digestion. The methane content of biogas increased from 53% on day 3 to around 62% after 13 days of digestion. The changes of volatile fatty acids (VFAs) indicated that the acidification of S. alterniflora was propionate-type fermentation with proportion of acetate and propionate ranging from 54.8% to 98.4%, and the hydrolysis of lignocellulose was the rate-limiting step for its anaerobic digestion. The analysis of cations suggested that K+ and Mg2+, with the maximum concentration of 1.35 and 0.43 g L?1 in fermentation liquor, respectively, could be inhibitory to the anaerobic digestion of S. alterniflora. It is concluded that S. alterniflora can be transformed into clean energy by anaerobic digestion and the high contents of K, Na, Ca and Mg may be the inhibitory factors when S. alterniflora is digested by continuous or semi-continuous anaerobic process.  相似文献

Mathematical simulation of unsteady natural convection modes in a square cavity filled with a porous medium having finite thickness heat-conducting walls with local heat source in conditions of heterogeneous heat exchange with an environment at one of the external boundaries has been carried out. Numerical analysis was based on Darcy–Forchheimer model in dimensionless variables such as a stream function, a vorticity vector and a temperature. The special attention was given to analysis of Rayleigh number effect Ra = 104, 105, 106, of Darcy number effect Da = 10?5, 10?4, 10?3, ∞, of the transient factor effect 0 < τ < 1000 and of the heat conductivity ratio k2,1 = 3.7 × 10?2, 5.7 × 10?4, 6.8 × 10?5 on the velocity and temperature fields. The influence scales of the defining parameters on the average Nusselt number have been detected.  相似文献

The phenomena of natural convection in an inclined square enclosure heated via corner heater have been studied numerically. Finite difference method is used for solving momentum and energy equations in the form of stream function–vorticity. One wall of the enclosure is isothermal but its temperature is colder than that of heaters while the remaining walls are adiabatic. The numerical procedure adopted in this analysis yields consistent performance over a wide range of parameters; Rayleigh number, Ra (103 ? Ra ? 106); Prandtl number, Pr (0.07 ? Pr ? 70); dimensionless lengths of heater in x and y directions (0.25 ? hx ? 0.75, 0.25 ? hy ? 0.75); and inclination angle, ? (0° ? ? ? 270°). It is observed that heat transfer is maximum or minimum depending on the inclination angle and depending on the length of the corner heaters. The effect of Prandtl number on mean Nusselt number is more significant for Pr < 1.  相似文献

The use of congress grass (Parthenium sp.) and water hyacinth (Eichhornia crassipes) as low cost raw materials for xylanase production from mutant Penicillium oxalicum SAUE-3.510 in submerged fermentation was investigated. For development of mutant from wild type P. oxalicum SA-8 ITCC 6024, a strategy of mixed mutagenesis was followed using UV-irradiation and ethidium bromide, which had resulted into 1.87 fold increases in the activity of the enzyme. For enzyme production, the fungus was cultivated in mineral medium containing congress grass as carbon source. Considerably higher levels of production (475.2 ± 6.0 IU ml?1) were achieved in media containing congress grass, although it was slightly less than that was obtained (488.5 ± 6.5 IU ml?1) in presence of commercial oat spelt xylan. This fact confirms the feasibility of using this low cost non-food resource as an alternative carbon source to save costs of the enzyme production process. Maximum xylanase activity was reported at 55 °C with its stability at 80 °C for 2 h. The highest activity of xylanase at pH 9.0 and its stability at similar pH for 24 h denote the alkalitolerant nature of enzyme.  相似文献

The biofixation of carbon dioxide (CO2) by microalgae has been proven to be an efficient and economical method, mainly due to the photosynthetic ability of these microorganisms to use this gas as a source of nutrients for their development. The aim of this work was to study the growth of Spirulina LEB18 and Chlorella kessleri microalgae, exposed to controlled and non-controlled conditions, with the injection of different concentrations of CO2. The cultures was carried out in 6 L open raceway ponds, under controlled conditions at 30 °C and 39 μE m?2 s?1 and under non-controlled conditions, protected by a tunnel of transparent film. The experiments were subjected to CO2 injections at concentrations of 0.038, 6, 12 and 18% (v/v). The highest concentration of biomass (4.95 g L?1) and maximum daily fixation (0.21 g g?1 d?1) were obtained for Spirulina LEB18 in culture that was prepared in non-controlled conditions with an injection of 6% (v/v) of CO2. C. kessleri had maximum (p < 0.0008) specific growth rate (0.84 d?1) when grown with 18% (v/v) of CO2 in non-controlled conditions of cultivation.  相似文献

This article analyzes the detailed heat transfer phenomena during natural convection within tilted square cavities with isothermally cooled walls (BC and DA) and hot wall AB is parallel to the insulated wall CD. A penalty finite element analysis with bi-quadratic elements has been used to investigate the results in terms of streamlines, isotherms and heatlines. The present numerical procedure is performed over a wide range of parameters (103 ? Ra ? 105,0.015 ? Pr ? 1000,0° ? φ ? 90°). Secondary circulations cells are observed near corner regions of cavity for all φ’s at Pr = 0.015 with Ra = 105. Two asymmetric flow circulation cells are found to occupy the entire cavity for φ = 15° at Pr = 0.7 and Pr = 1000 with Ra = 105. Heatlines indicate that the cavity with inclination angle φ = 15° corresponds to large convective heat transfer from the wall AB to wall DA whereas the heat transfer to wall BC is maximum for φ = 75°. Heat transfer rates along the walls are obtained in terms of local and average Nusselt numbers and they are explained based on gradients of heatfunctions. Average Nusselt number distributions show that heat transfer rate along wall DA is larger for lower inclination angle (φ = 15°) whereas maximum heat transfer rate along wall BC occur for higher inclination angle (φ = 75°).  相似文献
