γ‐Stearidonic acid, 18:4n‐6, a potential product of β‐oxidation of arachidonic acid (20:4n‐6), was only recently positively identified in a living organism—a thermophilic cyanobacterium Tolypothrix sp., albeit at low levels, whilst some indirect evidence suggests its wider presence, e.g. in a unicellular marine alga. We have prepared 18:4n‐6 using an iodolactonisation chain‐shortening approach from 22:5n‐6 and obtained its 1H‐, 13C‐, COSY‐ and HSQC NMR spectra, with 18:5n‐3 spectra also recorded for a comparison. The GC and GC‐MS behaviour of its methyl ester was also studied. Like another Δ3 polyunsaturated acid, octadecapentaenoic (18:5n‐3), 18:4n‐6 rapidly yields 2‐trans isomer upon formation of dimethyloxazoline derivative. On a polar ionic liquid phase (SLB‐IL100, 200 °C) the methyl ester could be mistaken for 18:3n‐3, while on methylsilicone phase (BP1, 210 °C) it eluted ahead of 18:3n‐6 and 18:4n‐3, suggesting that when present it may be easily misidentified during GC analysis of fatty acids.  相似文献

Long‐chain polyunsaturated fatty acids (LC‐PUFA) now have a proven role in human health and nutrition, including the n‐3 forms normally found in fish oils. Unfortunately, global fish stocks are now more than ever subject to over‐fishing and environmental pollution, indicating the need for an alternative source of fish oils. Recent efforts have focussed on the production of LC‐PUFA in transgenic plants to provide a sustainable and clean source of fish oils. The current progress in this area is considered, as well as the bottlenecks that remain to be overcome.  相似文献

The splenocyte fatty acid profile and immune response of broiler chickens were investigated. One hundred and twenty day‐old broiler chicks were fed diets containing conjugated linoleic acid (CLA) (Diet I), sunflower oil (Diet II), flaxseed oil (Diet III) or fish oil (Diet IV). The total lipid content of the diets was 3.5%. Body weight and feed intake was higher (P <0.05) in Diet IV compared to Diets I, II and III. Birds fed Diet III and IV had a higher content of n‐3 fatty acids in splenocytes than those fed Diets I and II. Serum anti‐BSA immunoglobulin content was higher (P <0.05) in birds fed Diets III and IV, compared to those fed Diets I and II. Delayed type hypersensitivity response, measured as the wing web skin swelling reaction (thickness) to Mycobacterium butyricum injection (s.c.), increased (P <0.05) from 0.71 and 0.98 mm in Diets IV and III, respectively, to 1.19 and 1.41 mm in Diets I and II, respectively. The number of CD4+ and CD8+ blood lymphocytes and CD4+, CD8+ and IgM+ splenocytes did not differ (P >0.05) between treatment groups. N‐3 fatty acids increased production performances and antibody mediated responses, while n‐6 fatty acids and conjugated linoleic acid increased cell mediated responses in broiler birds.  相似文献

The effects of supplementing diets with n‐3 alpha‐linolenic acid (ALA) and docosahexaenoic acid (DHA) on plasma metabolites, carcass yield, muscle n‐3 fatty acids and liver messenger RNA (mRNA) in lambs were investigated. Lambs (n = 120) were stratified to 12 groups based on body weight (35 ± 3.1 kg), and within groups randomly allocated to four dietary treatments: basal diet (BAS), BAS with 10.7 % flaxseed supplement (Flax), BAS with 1.8 % algae supplement (DHA), BAS with Flax and DHA (FlaxDHA). Lambs were fed for 56 days. Blood samples were collected on day 0 and day 56, and plasma analysed for insulin and lipids. Lambs were slaughtered, and carcass traits measured. At 30 min and 24 h, liver and muscle samples, respectively, were collected for determination of mRNA (FADS1, FADS2, CPT1A, ACOX1) and fatty acid composition. Lambs fed Flax had higher plasma triacylglycerol, body weight, body fat and carcass yield compared with the BAS group (P < 0.001). DHA supplementation increased carcass yield and muscle DHA while lowering plasma insulin compared with the BAS diet (P < 0.01). Flax treatment increased (P < 0.001) muscle ALA concentration, while DHA treatment increased (P < 0.001) muscle DHA concentration. Liver mRNA FADS2 was higher and CPT1A lower in the DHA group (P < 0.05). The FlaxDHA diet had additive effects, including higher FADS1 and ACOX1 mRNA than for the Flax or DHA diet. In summary, supplementation with ALA or DHA modulated plasma metabolites, muscle DHA, body fat and liver gene expression differently.  相似文献

A plant source of omega-3 fatty acid (FA) that can raise tissue eicosapentaenoic acid (EPA) and/or docosahexaenoic acid (DHA) is needed. A soybean oil (SBO) containing approximately 20% stearidonic acid [SDA; the delta-6 desaturase product of alpha-linolenic acid (ALA)] derived from genetically modified soybeans is under development. This study compared the effects of EPA to SDA-SBO on erythrocyte EPA+DHA levels (the omega-3 index). Overweight healthy volunteers (n=45) were randomized to SDA-SBO (24 ml/day providing approximately 3.7 g SDA) or to regular SBO (control group) without or with EPA ethyl esters (approximately 1 g/day) for 16 weeks. Serum lipids, blood pressure, heart rate, platelet function and safety laboratory tests were measured along with the omega-3 index. A per-protocol analysis was conducted on 33 subjects (11 per group). Compared to baseline, average omega-3 index levels increased 19.5% in the SDA group and 25.4% in the EPA group (p<0.05 for both, vs. control). DHA did not change in any group. Relative to EPA, SDA increased RBC EPA with about 17% efficiency. No other clinical endpoints were affected by SDA or EPA treatment (vs. control). In conclusion, SDA-enriched SBO significantly raised the omega-3 index. Since EPA supplementation has been shown to raise the omega-3 index and to lower risk for cardiac events, SDA-SBO may be a viable plant-based alternative for providing meaningful intakes of cardioprotective omega-3 FAs.  相似文献

Omega-3 polyunsaturated fatty acids (omega-3 PUFAs) are essential for improving the health and performance of athletes. The present study aimed to evaluate the nutritional status of omega-3 PUFAs in Chinese elite athletes by both dietary intake analysis and serum biomarker detection. A cross-sectional analysis of data from 54 elite athletes (24 men and 30 women) from Shanghai professional sports teams was conducted. A food frequency questionnaire (FFQ) was employed to analyze dietary intake, and gas chromatography–mass spectrometry (GC–MS/MS) was conducted to measure serum biomarkers of PUFAs. Correlation analysis was performed to investigate the relationships of PUFA biomarkers with diet, inflammation and oxidative stress. The results showed that the median intake of EPA + DHA among athletes was 132 mg/d, which is lower than the minimum value recommended by dietary guidelines (250 mg/d). The average serum EPA + DHA was 4.0 ± 1.1%, and the ratio of omega-6/omega-3 was 7.7 ± 1.7. Most (96.3%) of the athletes were below the targeted value of serum EPA + DHA, which is associated with a reduction in cardiovascular risk. Correlation analysis showed that the serum EPA + DHA was positively correlated with the long-term dietary intake of EPA + DHA and negatively correlated with inflammatory markers. In conclusion, the serum circulating EPA + DHA and omega-6/omega-3 ratio are effective biomarkers reflecting the nutritional status of PUFAs in athletes. Omega-3 PUFAs have a potential effect on inhibiting inflammatory markers. Hence, it is necessary for Chinese athletes to improve their suboptimal nutritional status of PUFAs through dietary intervention.  相似文献

There is growing scientific evidence that consumption of n‐3 very long‐chain polyunsaturated fatty acids (n‐3 VLC‐PUFA) helps in brain and eye development, and protects against a range of common degenerative diseases. This has provided the impetus to the scientists to develop new and renewable sources for these important fatty acids so that the food industry is able to produce and market products fortified with n‐3 VLC‐PUFA. The bioactive efficacy and stability of food products containing n‐3 VLC‐PUFA may be determined not only by the amount of n‐3 VLC‐PUFA present but also by the positional distribution of these acids within the triacylglycerol (TAG) molecules (regiopurity). Studies of the effects of positional distribution on the functionality of n‐3 VLC‐PUFA containing oils have been hampered by a general lack of pure TAG regioisomers for experimentation. This paper reviews methods that have been used for the synthesis of TAG regioisomers containing n‐3 VLC‐PUFA, with special reference to those in which one n‐3 VLC‐PUFA occurs in combination with two long‐chain saturated acids.  相似文献

In the prevention and treatment of cardiovascular disease, in addition to the already proven effective treatment of dyslipidemia, hypertension and diabetes mellitus, omega-3 polyunsaturated fatty acids (n-3 PUFAs) are considered as substances with additive effects on cardiovascular health. N-3 PUFAs combine their indirect effects on metabolic, inflammatory and thrombogenic parameters with direct effects on the cellular level. Eicosapentaenoic acid (EPA) seems to be more efficient than docosahexaenoic acid (DHA) in the favorable mitigation of atherothrombosis due to its specific molecular properties. The inferred mechanism is a more favorable effect on the cell membrane. In addition, the anti-fibrotic effects of n-3 PUFA were described, with potential impacts on heart failure with a preserved ejection fraction. Furthermore, n-3 PUFA can modify ion channels, with a favorable impact on arrhythmias. However, despite recent evidence in the prevention of cardiovascular disease by a relatively high dose of icosapent ethyl (EPA derivative), there is still a paucity of data describing the exact mechanisms of n-3 PUFAs, including the role of their particular metabolites. The purpose of this review is to discuss the effects of n-3 PUFAs at several levels of the cardiovascular system, including controversies.  相似文献

Significant amounts of polyunsaturated fatty acids (PUFA) with a chain length of 26 carbon atoms were detected in lipids of five deep water species of Ophiuroidea besides common fatty acids with chain lengths between 14 and 24. By means of hydrogenation, GC–MS of the methyl esters, and 4,4‐dimethyloxazoline (DMOX) derivatives of these C26 acids were characterized as 5,8,11,14,17,20,23‐hexacosaheptaenoic [26:7(n‐3)]; 8,11,14,17,20,23‐hexacosahexaenoic [26:6(n‐3)]; 5,8,11,14,17,20‐ hexacosahexaenoic [26:6(n‐6)]; and 11,14,17,20,23‐hexacosapentaenoic [26:5(n‐3)]. Concentrations of these acids varied from 0.3 to 4.5 mol% of the total FA. In all the samples investigated, the main component of C26PUFA was hexacosaheptaenoic acid 26:7(n‐3). These C26PUFA are localized mainly in polar lipids. The presence of the possible biosynthesis precursors suggests that the C26PUFA are produced by the brittle stars, and are not accumulated from food sources. This finding can also explain the presence of small amounts of the 26:7(n‐3) acid detected earlier in flesh lipids of the roughscale sole Clidoderma asperrimum, which feeds on deep water brittle stars. We suggest a possible scheme of the biosynthesis of C26 PUFA.  相似文献

Iso‐oleic acid is a mixture of C18‐unsaturated branched‐chain fatty acid isomers with a methyl group on various positions of the alkyl chain, which is the product of the skeletal isomerization reaction of oleic acid and is the intermediate used to make isostearic acid (C18‐saturated branched‐chain fatty acid isomers). Methyl iso‐oleate, a mixture of C18‐unsaturated branched‐chain fatty acid methyl ester isomers, is obtained via acid catalyzed esterification of iso‐oleic acid with methanol. The branched‐chain materials are liquid at room temperature and their “oiliness” property makes them an attractive candidate for the lubricant industry. In this paper, we report characterization of these branched‐chain materials using comprehensive two‐dimensional GC with time‐of‐flight mass spectrometry (GC × GC/TOF‐MS) and their physical and lubricity properties using tribology measurements.  相似文献

Background – Since n‐3 fatty acids, abundant in fatty fish, may improve health, we raised the question whether self‐reported intake frequency of fatty fish (FF) might be related to the percentage of n‐3 fatty acids in serum phospholipids (PL‐n‐3), and also to self‐rated health (H). Design – The study followed a cross‐sectional design. Methods – In the cross‐sectional Oslo Health Study, PL‐n‐3 were determined in 121 middle‐aged ethnic Norwegians and 102 immigrants from the Indian subcontinent and correlated with FF and H. Logistic regression was used to study the relationship between PL‐n‐3 and H (dichotomized, i.e. Poor vs. Good health). Results – FF correlated positively with PL20:5n‐3 (PL‐EPA, r = 0.467, p <0.001) and PL22:6n‐3 (PL‐DHA, r = 0.499, p <0.001), and negatively with PL20:4n‐6 (PL‐AA, r = –0.350, p = 0.001). H was positively associated with PL‐EPA (r = 0.321, p <0.001) and PL‐DHA (r = 0.275; p <0.001), but negatively with PL‐AA (r = –0.220, p = 0.001). The odds ratio for reporting Poor vs. Good health was significantly higher in subjects with low levels of PL‐EPA (OR = 1.49; 95% confidence interval = 1.17–1.89, p = 0.001), persisting after adjusting for sex, physical activity, ethnicity and length of education. Conclusion – The intake frequency of fatty fish is related to n‐3 fatty acids in the serum phospholipids, and to self‐rated health.  相似文献

Antimicrobial activity of monoacylglycerols (MAG) with odd number of carbons prepared from undecanoic (MAG C11:0) and undecenoic (MAG C11:1) was investigated. Data showed that both studied substances successfully inhibited Gram‐positive cocci (Staphylococcus aureus) and spore‐forming rods (Bacillus cereus). Gram‐negative bacteria were highly resistant against MAG C11:0 and showed considerable tolerance against MAG C11:1. Minimum inhibitory concentration (MIC) of MAG C11:1 was possible to determine for Escherichia coli only. Complete inhibition of three fungi genera (Alternaria, Cladosporium, Trichothecium) was achieved using MAG C11:1 with a concentration higher than 750 µg/mL. The growth of Aspergillus niger, Mucor racemosus, Phoma, Scopulariopsis, Trichoderma was significantly reduced. Generally, MAG C11:1 was more efficient than MAG C11:0 against all tested microorganisms. Due to a rich production of degradation enzymes, some fungi species surprisingly adapted their metabolism during growth and were even able to utilise tested MAG as a source of carbon. Practical applications: MAG composed of fatty acids with odd number of carbons in molecule are a promising group of substances applicable in food industry and cosmetics. They represent materials with broad spectrum of antimicrobial activity, especially against Gram‐positive bacteria. More efficient MAG C11:1 with a double bond in molecule can be recommended for reduction of S. aureus that cause food enterotoxicosis.  相似文献

Epidemiological and clinical studies have suggested that intake of n-3 polyunsaturated fatty acids (PUFA) reduces the incidence of allergic airway diseases and improves pulmonary function in patients with allergic asthma. However, the pharmacological targets of PUFA have not been elucidated upon. We investigated whether free fatty acid receptor 4 (FFA4, also known as GPR120) is a molecular target for beneficial PUFA in asthma therapy. In an ovalbumin (OVA)-induced allergic asthma model, compound A (a selective agonist of FFA4) was administrated before OVA sensitization or OVA challenge in FFA4 wild-type (WT) and knock-out (KO) mice. Compound A treatment of RBL-2H3 cells suppressed mast cell degranulation in vitro in a concentration-dependent manner. Administration of compound A suppressed in vivo allergic characteristics in bronchoalveolar lavage fluid (BALF) and lungs, such as inflammatory cytokine levels and eosinophil accumulation in BALF, inflammation and mucin secretion in the lungs. Compound A-induced suppression was not only observed in mice treated with compound A before OVA challenge, but in mice treated before OVA sensitization as well, implying that compound A acts on mast cells as well as dendritic cells. Furthermore, this suppression by compound A was only observed in FFA4-WT mice and was absent in FFA4-KO mice, implying that compound A action is mediated through FFA4. Activation of FFA4 may be a therapeutic target of PUFA in allergic asthma by suppressing the activation of dendritic cells and mast cells, suggesting that highly potent specific agonists of FFA4 could be a novel therapy for allergic asthma.  相似文献

Phosphatidylglycerol (PG) is a highly functional phospholipid (PL), which has many physiological functions. However, naturally occurring PG binding n‐3 polyunsaturated fatty acid (n‐3 PUFA) is low in content, resulting in a scarcity of industrial bio‐resources of n‐3 PUFA enriched PG. The current study investigates the preparation of salmon roe PG (SRPG) from three types of salmon roe lipids and glycerol via phospholipase D (PLD)‐mediated transphosphatidylation. The yields of SRPG obtained from salmon roe total lipid (SRTL) and salmon roe PL (SRPL) are higher than those obtained from purified salmon roe phosphatidylcholine (SRPC) in aqueous system. Following a 24 h reaction with 0.75 U PLD, SRTL, and SRPL yield up to 96.4 mol% and 96.7 mol% SRPG, respectively. In addition, more fatty acids are released from synthesized SRPG via hydrolysis by pancreatic enzymes than from SRPC and soybean PC in in vitro digestion model. Fatty acids at the sn‐2 position of SRPG are completely liberated by 0.04 U of phospholipase A2 (PLA2) during a 6 h reaction, whereas fatty acids of SRPC are partially unhydrolyzed even after a 24 h reaction. Our results suggest that SRPG converted from salmon lipids by PLD is a functional PL with high bioavailability of n‐3 PUFAs. Practical Applications: Phosphatidylglycerol rich in n‐3 PUFAs is prepared from salmon roe lipids (SRPG) catalyzed by PLD. The SRPG yields reach 96.4 mol% and 96.7 mol% of phosphatidylcholine contained in SRTL and SRPL, respectively, in aqueous reaction system. Fatty acids rich in n‐3 PUFAs at sn‐2 position of prepared SRPG are rapidly liberated by PLA2 in an in vitro digestion model.  相似文献
