用褐藻酸钙对小新月菱形藻进行固定化培养实验,测定不同胶粒大小、胶珠密度、CaCl2浓度及不同接种量对该藻的影响,比较了自由化生长与固定化细胞的生长曲线。小新月菱形藻在褐藻酸钙凝胶中仍具有呼吸和光合作用的能力。球径为3.5mm,CaCl2浓度为2%时,细胞生长快,每50mL培养液中加入200个胶珠时细胞生长量大,接种量不能低于104个细胞/mL。与游离的小新月菱形藻相比,固定化小新月菱形藻生长慢,但生长周期长。  相似文献

为探究栽植密度对樟树幼林生长的影响,设计了不同栽植密度下樟树生长量及生物量积累情况试验。结果表明:栽植密度会对樟树幼林的生长造成直接影响,在栽植密度为1 333~1 600株/hm2时,樟树幼林个体植株及林分整体生长均较为理想,是最适宜福州樟树的栽植密度。  相似文献

针对ELID磨削砂轮表面氧化膜的厚度测量及生长特性,采用激光和涡流传感器进行氧化膜的厚度模拟测试,以验证测量系统的准确性,并在预修锐阶段对氧化膜的生长特性进行观察。研究结果表明:在前期,氧化膜沿砂轮半径向内生长的速度大于向外生长的速度,且基本呈线性生长,氧化膜生长速度最快;在后期,向内生长速度减缓,向外生长速度稍快,但氧化膜总体生长速度变慢。    相似文献

为解决工业机器人运动规划中的冲击、振动、快速响应等问题,提出了一种源于细菌生长曲线的新算法。介绍了细菌生长曲线,证明了其与运动规划中速度变化情况的相关性;导出了理想速度曲线及公式,给出了应用实例。仿真实验及在真实机器人上实验证明算法可行而有效,可解决关节驱动器运动规划中的冲击振动等问题,同时可调节算法参数以满足其他规划要求如快速到达目标位置等。算法虽简单但新颖实用。  相似文献

磷酸二氢钾(KDP)晶体是一种经典的电光材料,是目前唯一可用于惯性约束核聚变的非线性光学材料。鉴于籽晶和生长方法对KDP晶体的生长和质量具有重要的影响,本文采用传统法生长法和点籽晶快速生长法生长KDP晶体,通过改变籽晶的种类及切向,不仅减少了晶体的恢复区,同时缩短了晶体的生长周期,生长出高透明度的单晶。对晶体的透过、结晶完整性及损伤等性能进行测试,结果表明,晶体在红外和可见光波段具有较高的透过率;晶体(001)面的衍射强度较强,峰形比较尖锐;相比于1-on-1方式,R-on-1处理使KDP的损伤阈值提高了1.7~2.1倍,晶体的抗损伤均匀性很高。生长的晶体具有优良的光学性能,籽晶对晶体的生长具有重要的影响。  相似文献

分析了以NURBS为代表的参数曲线表达方式在数控加工应用中的不足,借鉴了生物基于基因控制的生长模式,提出了基于规则控制的平面曲线描述方法,即以步长和转角来描述曲线。针对平面曲线的几种描述方式,给出了相应的提取曲线生长规则的方法,给出了平面曲线的自生长造型实例。并通过设计规则来设计新的曲线,最后将自生长造型应用于数控加工中。  相似文献

细胞培养是进行细胞研究的基础,为了在细胞体外培养时提供一种近似于体内的微环境,设计了一种可供细胞三维动态培养的微器件。首先设计了用于输运流体的微通道网络,培养池对称布置于微通道网络中,通过一系列"多进多出"型微通道分别与进样口和出样口相连。利用Comsol软件中的层流物理场和多孔介质物理场耦合对培养池内的流场进行仿真,通过比较流场的均一性和稳定性优化微通道网络结构。然后,采用静电直写技术在培养池内集成聚己内酯(PCL)三维支架,构建细胞三维培养空间。最后,封合微器件,检测微器件培养池内的流体流动情况,并进行细胞实验。实验结果表明,"2×2"型微器件培养池内的流体稳定性和均一性较好;PCL三维支架的纤维间距400μm,纤维直径80μm,孔隙率64%,细胞存活率达到90%以上。该细胞三维动态培养微器件更好地模拟了生物体内细胞生存所需的微环境,培养池内的细胞生长良好,满足设计要求。  相似文献

为解决企业文化落地评价难以准确量化、客观评价的问题。提出了基于熵权法与TOPSIS法的企业文化落地测评体系及评价方法。首先,从企业文化全生命周期发展的角度定义了企业文化落地的基本过程,构建了企业文化落地的评价指标体系;其次,应用熵权法对专家的经验打分进行了修正,确定了更加客观、准确的指标权重;对传统的TOPSIS进行了改进,简化了TOPSIS法综合评价的计算过程。最后,通过其县级供电公司企业文化落地评价的实例验证了本体系及计算方法的可信度及实用性。  相似文献

Devices for the rotational culture of cells and the study of biological reactions have been widely applied in tissue engineering. However, there are few reports exploring the effects of rotational culture on cell morphology, nitric oxide (NO) production, and cell cycle of the endothelial cells from human umbilical vein on the stent surface. This study focuses on these parameters after the cells are seeded on the stents. Results showed that covering of stents by endothelial cells was improved by rotational culture. NO production decreased within 24 h in both rotational and static culture groups. In addition, rotational culture significantly increased NO production by 37.9% at 36 h and 28.9% at 48 h compared with static culture. Flow cytometry showed that the cell cycle was not obviously influenced by rotational culture. Results indicate that rotational culture may be helpful for preparation of cell-seeded vascular grafts and intravascular stents, which are expected to be the most frequently implanted materials in the future.  相似文献

Ex vivo expansion of limbal stem cells from a small biopsy and its subsequent transplantation is the golden choice of treatment for limbal stem cell deficiency. Use of murine 3T3 feeder layer is a prerequisite for this ex vivo expansion. There is an ever‐increasing demand for feeder free cultures to avoid xenotoxicity and transmission of xeno‐diseases to human system. This study was aimed to establish an efficient xeno‐feeder free limbal culture system towards ocular surface regeneration. To study the effect of initial dispase treatment and culture system used, migratory distance of cells from explants was analyzed from phase contrast images using “interactive measurements” of Qwin software (Leica). Expression of p63 in different culture systems was studied by immunofluorescent staining, followed by quantitative confocal microscopy (Carl Zeiss). Results showed dispase treatment was not necessary for establishing limbal explant culture. A combination of Iscove's modified Dulbecco's medium and Panserin 801 resulted in formation of autofeeder layer with maintenance of progenitor characteristics, thus mimicking natural tissue architecture. Further analysis of this culture system showed that cells could be cultured till confluency. Immunofluorescent staining of ABCG2 revealed presence of stem cell marker in the confluent cell layer. Scanning Electron Micrographs demonstrated homogenous population of tightly packed cells in this culture system. Replacement of bovine serum with autologous serum did not affect morphology or growth of cells in this culture system. The subcommissural organ (SCO) is a brain circumventricular organ formed by ependymal and hypendymal secretory cells. It secretes glycoproteins into the cerebrospinal fluid of the third ventricle where they condense into a thread-like structure known as Reissner's fiber (RF). The present study was designed to investigate whether or not the bovine SCO continues to synthesize and release glycoproteins after a long-term culture. Cultured explants of SCO survive for several months. The content of the secretory granules present in the cultured ependymocytes displayed immunoreactive and lectin-binding properties similar to those of the core glycosylated glycoproteins found in the bovine SCO. The explants actively incorporated (35)S-cysteine. In the cultured ependymocytes, the pattern of distribution of the radioactive label and that of the immunoreactive secretory material was similar, thus indicating that this material has been synthesized during culture. At the ultrastructural level, the cultured tissue exhibited a high degree of differentiation comparable to that of the bovine SCO in situ. A striking finding was the observation of similar results when cerebrospinal fluid was used as a culture medium. The addition of antibodies against RF-glycoproteins into the culture medium allowed visualization, by means of different immunocytochemistry protocols, deposits of extracellular immunoreactive secretory material on the free surface of the cultured ependymocytes, indicating that release of secretory glycoproteins into the culture medium does occur. Primary culture of dispersed SCO ependymocytes, obtained either from fresh or organ cultured bovine SCO, showed that these cells release RF-glycoproteins that aggregate in the vicinity of each cell. The present investigation has shown that: (1) two types of secretory ependymocytes become evident in the cultured SCO; (2) under culture conditions, the SCO cells increase their secretory activity; (3) explants of bovine SCO synthesize RF-glycoproteins and release them to the culture medium; (4) after release these proteins aggregate but do not form a RF; (5) a pulse of anti-RF antibodies into the culture medium blocks the secretion of RF-glycoproteins for several days.  相似文献

Microalgae offer several advantages as renewable energy sources. To commercialize microalgae biofuel, the cost of microalgae product processing must be reduced. Mass culture and economical harvesting of microalgae are methods that can reduce the process cost. This paper proposes a separation method using ultrasonic waves for effective harvesting of microalgae. Experiments were conducted by considering four variables by ultrasonic separation process using the Taguchi method. Continuous culture of microalgae was performed based on the experimental results of the ultrasonic separation process. To harvest automatically microalgae in the process, an on-off control was implemented using the continuous culture and ultrasonic separation device to compose one combined system. Experiments were conducted on the combined system to increase production efficiency while simultaneously economically harvesting and performing mass culture of microalgae.  相似文献

Collagen often acts as an extracellular and intracellular marker for in vitro experiments, and its quality defines tissue constructs. To validate collagen detection techniques, cardiac valve interstitial cells were isolated from pigs and cultured under two different conditions; with and without ascorbic acid. The culture with ascorbic acid reached higher cell growth and collagen deposition, although the expression levels of collagen gene stayed similar to the culture without ascorbic acid. The fluorescent microscopy was positive for collagen fibers in both the cultures. Visualization of only extracellular collagen returned a higher correlation coefficient when comparing the immunolabeling and second harmonic generation microscopy images in the culture with ascorbic acid. Lastly, it was proved that the hydroxyproline strongly contributes to the second‐order susceptibility tensor of collagen molecules, and therefore the second harmonic generation signal is impaired in the culture without ascorbic acid.  相似文献

水质参数监测是工厂化水产养殖的重要组成部分,是工厂化养殖向自动化、大规模、高产量和高质量发展的关键环节。针对我国工厂化养殖水质监测手段落后、自动化水平低的现状,研制了一套适合国情的在线多参数水质监测系统,可对水温、水位、酸碱度(PH值)、溶解氧含量4个水质参数进行连续在线监测。以一台PC工业控制计算机作为现场上位机,以多台单片机作为现场鱼池的下位机,一旦发生故障,养殖池发生环境变化,能够及时报警,从而实现了对养殖现场中鱼类生长的实时监控。实际应用结果表明,系统运行稳定、安全可靠,测量数据的精确度完全可以达到工厂化渔业生产的要求。  相似文献

1992年以前,中国大学生文化与社会政治文化高度同质,建立在计划经济基础上的集体主义是大学生文化的当然价值取向。90年代以来,随着独特的大学生文化的形成,传统集体主义观受到大学生的不断质疑。中国大学生文化正在走向强调个人和集体共同发展的“后集体主义”,各类学生组织正是沟通个人自由与公共规范的中介以及形成新的集体主义精神的关键。  相似文献
