Sex pheromone produced by Callosobruchus subinnotatus females stimulates conspecific males to walk upwind toward the source. Gas chromatographic and mass spectrometric analyses of sex pheromone components and their hydrogenated derivatives suggested that the sex pheromone consisted of two short-chain fatty acids, (E)-3-methyl-2-heptenoic acid (E32C7) and (Z)-3-methyl-2-heptenoic acid (Z32C7). The composition of the sex pheromone was confirmed by electrophysiological and behavioral bioassays with synthetic compounds. A mixture of 5 ng E32C7 and 5 ng Z32C7 elicited a 2.1-mV male EAG response, whereas the solvent control elicited 0.1 mV. Mixtures of the two compounds at various ratios were attractive to males in Y-tube bioassays. A pitfall-type trap equipped with a lure impregnated with 1 ng E32C7 and 1 ng Z32C7 was effective in trapping males. Traps baited with the lure caught 80% of males in a glass aquarium during nighttime and 50% during daytime, whereas control traps (solvent treated) in a separate aquarium caught 20% and 10%, respectively.  相似文献

To date, all known or suspected pheromones of click beetles (Coleoptera: Elateridae) have been identified solely from species native to Europe and Asia; reports of identifications from North American species dating from the 1970s have since proven to be incorrect. While conducting bioassays of pheromones of a longhorned beetle (Coleoptera: Cerambycidae), we serendipitously discovered that males of Cardiophorus tenebrosus L. and Cardiophorus edwardsi Horn were specifically attracted to the cerambycid pheromone fuscumol acetate, (E)-6,10-dimethylundeca-5,9-dien-2-yl acetate, suggesting that this compound might also be a sex pheromone for the two Cardiophorus species. Further field bioassays and electrophysiological assays with the enantiomers of fuscumol acetate determined that males were specifically attracted by the (R)-enantiomer. However, subsequent analyses of extracts of volatiles from female C. tenebrosus and C. edwardsi showed that the females actually produced a different compound, which was identified as (3R,6E)-3,7,11-trimethyl-6,10-dodecadienoic acid methyl ester (methyl (3R,6E)-2,3-dihydrofarnesoate). In field trials, both the racemate and the (R)-enantiomer of the pheromone attracted similar numbers of male beetles, suggesting that the (S)-enantiomer was not interfering with responses to the insect-produced (R)-enantiomer. This report constitutes the first conclusive identification of sex pheromones for any North American click beetle species. Possible reasons for the strong and specific attraction of males to fuscumol acetate, which is markedly different in structure to the actual pheromone, are discussed.  相似文献

Field experiments showed that males of the seed-feeding, Neotropical wasp Bephratelloides pomorum are strongly attracted to females. That this attraction was semiochemically mediated was demonstrated by both the response of male antennae to the whole-body extract of females and by behavioral bioassays. In the field, males were captured in traps baited with female thoraces, but not with other body parts (abdomen and head). Volatiles collected from the headspace of female wasps or whole-body extracts, concentrated and transferred to cotton plugs, elicited a full sequence of sexual behavior in male wasps. The attraction of males to female-baited traps showed a peak of activity at noon, but in indoor bioassays male responded to female extracts even at night.  相似文献

Antennal and behavioral responses of the rust-red grain beetle,Cryptolestes ferrugineus, and the flat grain beetle,C. pusillus, to synthetic samples of the macrocyclic lactones reported to comprise their aggregation pheromones were investigated. Electroantennogram (EAG) recordings were obtained successfully from both species for the first time. Females of both species showed larger EAGs than males. The EAGs ofC. ferrugineus showed a high degree of specificity for conspecific aggregation pheromone components;C. pusillus showed much less specificity. Behavioral tests were conducted using two-choice pitfall bioassays. Separation of the results into the two effects of activity stimulation and direction finding showed that both effects contributed to the overall response, although sometimes to different extents. The strain ofC. pusillus studied responded equally well to both components of its pheromone, whereas it had been reported previously that only one was active, the other acting as a Synergist and eliciting no response when tested alone. With both species, behavioral response was elicited with a single lactone, suggesting that it might not be necessary to use both components for field use. Particularly surprising was thatC. pusillus showed a greater response to the pheromone components ofC. ferrugineus than to its own. Aeration of the two species and thermal desorption of the collected volatiles confirmed production of the expected lactones, and aeration of authentic lactones showed that the response was not due to the C.ferrugineus pheromone components being markedly more volatile. This response, which seems to be an actual preference, is the first to be discovered among the cucujid beetles and encourages optimism that a practical lure for various species may not need to be as complex as originally feared.  相似文献

Cryptoblabes gnidiella Milliére (Lepidoptera: Pyralidae) is an economically important exotic pest of vineyards in Southern Brazil and Uruguay. The sex pheromone of C. gnidiella was identified as a mixture of (Z)-11-hexadecenal and (Z)-13-octadecenal, and has been used to monitor populations of this pest in Israel. The development of mating disruption for this species may be hampered by the chemical instability of the natural pheromone components. Therefore, studies on more stable pheromone analogs may provide tools for a control strategy based on behavior-modifying chemicals. We report here the electrophysiological and behavioral responses of C. gnidiella males to (Z)-9-tetradecenyl formate and (Z)-11-hexadecenyl formate, structural analogs of the pheromone components. In gas chromatography-electroantennogram detection (GC-EAD) studies, both analogs elicited responses from C. gnidiella male antennae. Pre-exposure to the formates did not affect the subsequent EAD responses to the natural pheromone components. The formates acted as pheromone antagonists in wind tunnel tests, inhibiting the responses of males toward both synthetic pheromone and calling females. In the field, captures of males in pheromone-baited traps decreased, in a dose-response pattern, when different amount of formates were added to the pheromone. These pheromone antagonists, thus, are potentially useful as mating disruptants for C. gnidiella in commercial vineyards.  相似文献

Volatiles from chocolate mediate upwind flight behavior in Ephestia cautella and Plodia interpunctella. We used gas chromatography with electroantennographic detection and found 12 active compounds derived from three different chocolate types, i.e., plain, nut-containing, and rum-flavored. Eight of the compounds were identified with mass spectrometry, and the activity of three compounds, ethyl vanillin, nonanal, and phenylacetaldehyde (PAA), was subsequently confirmed in both electrophysiological and behavioral assays. In the electroantennogram experiment, PAA and nonanal were consistently eliciting responses in both species and sexes. Ethyl vanillin was active in males of both species, and also in P. interpunctella females. E. cautella females showed no antennal activity in response to ethyl vanillin. All three volatiles were attractive to E. cautella males and P. interpunctella females in a flight tunnel. E. cautella females were significantly attracted only to ethyl vanillin. P. interpunctella males were attracted to PAA. Ethyl vanillin is a novel insect attractant, whereas both nonanal and phenylacetaldehyde mediate behavior in many insect species. A final experiment revealed that a blend of the three volatiles was required to induce landing in the flight tunnel bioassay, and that the landing rate was dependent on dose. The three-component blend attracted both sexes of P. interpunctella and females of E. cautella, whereas E. cautella males were not attracted.  相似文献

Stressed woody plants represent an ephemeral and unpredictable resource for larvae of some species of longhorned beetles (Coleoptera: Cerambycidae) because prime subcortical tissues are rapidly degraded by a guild of xylophagous competitors. Selection favors efficient mechanisms of host and mate location to expedite colonization of hosts by larvae. Based on previous research, we hypothesize that mate location in some species of the subfamily Cerambycinae involves three sequential behavioral stages: (1) both sexes are attracted to larval hosts by plant volatiles; (2) males attract females over shorter distances with pheromones; and (3) males recognize females by contact pheromones in their epicuticular wax layer. We already have evidence of second-stage and third-stage behaviors in three species in this subfamily whose xylophagous larvae feed in hardwood trees: Xylotrechus colonus, Megacyllene caryae, and Neoclytus mucronatus mucronatus. In this report, we evaluate the first behavioral stage of mate location behavior (i.e., independent response of both sexes to host plant volatiles) for the same three species. Supporting our hypothesis, both males and females responded to volatiles emanating from hickory logs in Y-tube olfactometer bioassays.  相似文献

In comparison to the large amount of study on the communication abilities of females in ant societies and their associated chemical ecology and sensory physiology, such study of male ants has been largely ignored; accordingly, little is known about their olfactory sensory capabilities. To address this, we explored peripheral odor sensitivities in male Harpegnathos saltator by measuring the electrophysiological activity of olfactory sensory neurons within antennal trichoid and coeloconic sensilla using an extracellular recording technique. In an initial trial of 46 compounds, sensilla trichodea responded strongly to two alarm pheromone components, while a limited number of non-hydrocarbon odorants elicited strong responses in sensilla coeloconica. Both sensillar types responded indifferently to 31 cuticular hydrocarbons (CHCs) and synthetic long-chain hydrocarbons (HCs) typically found on insect cuticle. In a search for sensilla responding to CHCs and other compounds, we found some sensilla that responded to synthetic HCs and CHCs from virgin queen postpharyngeal glands that are potentially used in close range mate recognition. Olfactometer bioassays of male ants to 15 non-HCs correlated sensory responsiveness to the respective behavioral responses. Comparing olfactory responses between H. saltator males and females, we found that sensilla coeloconica and basiconica of workers showed greater responses and broader selectivity to all compounds. The rarity of CHC-responding trichoid sensilla in Harpegnathos males suggests a more specific role in sexual communication compared to that in females, which use CHCs in a broader communication context.  相似文献

Electroantennograms from males and females ofDendroctonus frontalis andD. brevicomis in response to serial dilutions of the pheromonesexo brevicomin and frontalin and the host terpene hydrocarbons 3-carene and α-pinene show no significant differences in the threshold concentration for response to the compounds for either sex or species. The intensity of response was greater to higher concentrations of the pheromones (10 to >104 μg) than the terpene hydrocarbons, suggesting the presence of more receptors on the antennae for pheromones than terpene hydrocarbons. Antennal olfactory responses to the compounds did not correlate to published data on behavioral responses by the beetles to the compounds in both field and laboratory studies. Adaptation experiments indicated that forD. frontalis, both frontalin andexo brevicomin share the same receptors on the antennae. The results indicated that the terpene hydrocarbons share some, but not all, of the same receptors.  相似文献

Previous laboratory studies have shown that the mirid Lygus hesperus is attracted to volatiles emitted from alfalfa; feeding damage increases the amounts of several of these volatiles, and visual cues can enhance attraction further. The present study tested single plant volatiles in electrophysiological and behavioral trials with L. hesperus. Electroantennogram (EAG) analyses indicated that antennae responded to most plant volatiles included in the test, and that when gender differences were observed, males usually were more responsive than females. Antennal responses to the alcohols ((E)-3-hexenol, (Z)-3-hexenol, 1-hexanol), the acetate (E)-2-hexenyl acetate, and the aldehyde (E)-2-hexenal were among the strongest. Moderate responses were observed for (E)-β-ocimene, (E,E)-α-farnesene, (±)-linalool, and methyl salicylate. A dose dependent response was not observed for several terpenes (β-myrcene, β-caryophyllene, (+)-limonene, or both (R)-(+)- and (S)-(−)-α-pinenes). EAG responses, however, were not always consistent with behavioral assays. In Y-tube bioassays, males did not exhibit a positive behavioral response to any of the compounds tested. Instead, males were repelled by (E)-2-hexenyl acetate, (±)-linalool, (E,E)-α-farnesene, and methyl salicylate. In contrast, female L. hesperus moved upwind towards (R)-(+)-α-pinene, (E)-β-ocimene, and (E,E)-α-farnesene, and showed a negative response towards (Z)-3-hexen-1-ol, (S)-(−)-α-pinene, and methyl salicylate. This study emphasizes the use of multiple approaches to better understand host plant finding in the generalist herbivore L. hesperus.  相似文献

Volatile emissions from twigs of eastern white pine, Pinus strobus L., stimulated oviposition by Dioryctria abietivorella (Grote) in laboratory bioassays. Chemical identification and percentage composition of monoterpenes emitted by twigs were determined by capillary GC and GC/MS analysis of headspace and of volatiles entrained on Porapak Q. Enantiomeric excess of optically active monoterpenes was determined by capillary GC analysis with a β-cyclodextrin chiral column. Four of the six most abundant monoterpenes identified in twig volatiles elicited a significant oviposition response. Myrcene and car-3-ene [predominantly the (+)-enantiomer] were the most active compounds in both electroantennogram (EAG) and oviposition bioassays. In contrast, (?)-limonene elicited a significant oviposition response but was the least stimulating monoterpene in EAG tests. Female EAG responses to monoterpenes generally increased with age and mating and were larger than those from males of the same age.  相似文献

A series of lignans fromBupleurum salicifolium Soland (Umbelliferae) were tested for nematostatic activity on the cysts and freed secondstage juveniles of the potato cyst nematodesGlobodera rostochiensis andG. pallida. None of the six lignans tested—bursehernin, matairesinol, syringaresinol, the novel product buplerol, guayarol, and a derivative, nortrachelogenin triacetate—showed nematicidal activity in an in vitro analysis with second-stage juveniles, but significant differences were noted when the lignans were assayed for nematostatic activity as cyst hatching inhibitors. Bursehernin and matairesinol showed the greatest activity, at concentrations of 50 ppm. This is the first known instance of a natural product inhibiting the hatch of the nematodeG. pallida. The HID (hatching inhibiting dose) of bursehernin was estimated, and some conclusions were drawn about the structure-activity relationships of the lignans under study.  相似文献

Volatile chemicals produced by plants mediate host location, mate-finding and oviposition behavior in insects. State-dependent response to plant cues allows for timing of foraging, mating and oviposition on ephemeral host plants or plant parts. Caloptilia fraxinella is a herbivorous specialist on the foliage of ash trees (Fraxinus). Adults are long-lived and undergo a nine-month reproductive diapause over the fall and winter. Mating and oviposition occur in the spring when volatile chemicals released by ash leaves mediate host location. This study tested the plasticity of olfactory response of C. fraxinella to host plant volatiles using both electroantennogram and behavioral bioassays. The effect of moth physiological state on olfactory response was tested on male and female moths in different nutritional, mating, and diapause states. Antennal responses to host plant volatiles were plastic and depended on moth physiological state, and were highest when moths were reproductively active and would be seeking oviposition hosts. Moth sex and nutritional status also impacted antennal response to host plant volatiles. Oriented flight of females to ash seedlings varied with physiological state and nutritional status, with fed, reproductively active females having the highest response. Physiological state impacted oriented flight of males to female-produced sex pheromone signals whether or not a host plant was present, and there was no increase in behavioral response to sex pheromone in the presence of an ash host.  相似文献

Wasps of genus Asobara, a larval parasitoid of Drosophila, have become model organisms for the study of host-parasite interactions. However, little is known about the role of pheromones in locating mates and courtship behavior in this genus. In the present study, we aimed to identify the female courtship pheromone in Asobara tabida. The chemical compositions of solvent extracts from male and female wasps were analyzed by GC/MS. These extracts, fractions thereof, and synthetic pheromone candidates were tested for their activity in behavioral bioassays. The results demonstrate that the courtship pheromone of A. tabida is characterized by a remarkable chemical diversity. A multi-component blend of female-specific compounds including methyl 6-methylsalicylate (M6M), fatty alcohol acetates (FAAs), and cuticular hydrocarbons (CHCs) released male courtship behavior. Using a combinatory approach that included both purified natural products and synthetic analogs, it was shown that none of the three chemical classes alone was sufficient to release a full behavioral response in males. However, a blend of M6M and FAAs or combinations of one or both of these with female-derived CHCs resulted in wing-fanning responses by males comparable to those elicited by the crude extract of females. Thus, components from all three chemical classes contribute to the bioactivity of the pheromone, but none of the elements plays a key role or is irreplaceable. The fact that one of the FAAs, vaccenyl acetate, is also used as a kairomone by Asobara females to locate Drosophila hosts suggests that a pre-existing sensory responsiveness to vaccenyl acetate might have been involved in the evolution of the female sex pheromone in Asobara.  相似文献

Chemical communication between adults of the American warble fly, Dermatobia hominis (Diptera: Oestridae), was investigated by electroantennography and behavioral bioassays. Significant electroantennographic responses were recorded from both sexes to hexane-soluble cuticular lipids from either sex. Olfactometer tests indicated an attraction between males and females, and between females. Copulatory behavior of males with a white knotted string treated with female extract confirmed production of a sexual stimulant by females. Such behavior was not observed in tests with male extract, demonstrating that the pheromone acts also as a sex recognition factor. Cuticular hydrocarbons of sexually mature female and male D. hominis were identified by Gas chromatography–mass spectrometry and consist of a mixture of saturated n-, monomethyl-, and dimethylalkanes in both sexes. Sexual dimorphism was characterized by a higher relative concentration of dimethylalkanes in males and the presence of alkenes only in females.  相似文献

Abstract

The optical resolution of racemic tryptophan was performed by ultrafiltration using the BSA solution system. The pH of the feed solution had a strong influence on the complexation constants between BSA and tryptophan, especially for L-tryptophan. The complexation constant for L-tryptophan reached a maximum value at pH 9 (K L = 110,000), varying by 2 orders of magnitude in the range from pH 6 (K L = 1000) to pH 11 (K L = 21,000). Smaller variations of the complexation constant of D-tryptophan were observed. Based on these data, the recovery and the purity of the permeate were optimized by a proper control of the physicochemical parameters of the feed solution (essentially pH and initial concentrations). In one stage, 91% purity with a 89% recovery of D-tryptophan has been easily obtained with a high permeation rate (6.3 × 10?4 mol·m?2s?1 at 1.5 bar).  相似文献

The objective of the present investigation was to develop a technique to synthesize submicronic particles of Nb-doped strontium titanate with a homogeneous composition. This was achieved by a modified glycine-nitrate process, using Ti-lactate, Nb-oxalate, and Sr(NO3)2 as starting materials. A combination of both citric acid and glycine was needed in order to integrate the useful features of both complexation and combustion natures of citric acid and glycine, respectively. The amount of citric acid, glycine, and nitrates in the starting solution, as well as the source for extra nitrates, and the uniformity of heating during the thermal dehydration step were found to have significant influence on the final phase purity of the material. Calcination at 1100 °C in 7% H2 in N2 produced single phase Nb-doped strontium titanate with grain sizes of about 100 nm in diameter on average.  相似文献

In biological systems, it is expected that gene expression levels generally will correlate with temporally varying physiological and biological needs, and that gene expression levels could regulate biological capabilities. In moth species, male response to female sex pheromones often is affected by moth age and mating status. Odorant receptors (ORs) expressed in neurons within male antennae are critical for detecting the female pheromones. Therefore, we hypothesized that the expression level of these receptor proteins would be affected by age and mating status of male moths. We examined expression levels of two OR genes that are preferentially expressed in the male antennae of Heliothis virescens (HvOR13 and HvOR15) and Heliothis subflexa (HsOR13 and HsOR15). Antennae were dissected from virgin males at 2 h, 1 d, 2 d, 4 d, and 8 d. We also dissected antennae from 4-d-old mated males. We found that age had no effect on expression levels of either OR in either species, except for a small difference in HsOR15 expression between 2 h and 8-d-old virgin males. Furthermore, we found no effect of mating status on expression level of these ORs in either species. We discuss these findings in relationship to studies of age and mating status effects on male electrophysiological and behavioral response to female pheromones, and contrast our results to studies on the effects of age and mating status on gene expression of pheromone receptor proteins and pheromone binding proteins in other moths.  相似文献

Analysis of frass extracts and aerations from adult beetles confirmed earlier studies identifying 1-heptanol as a principal pheromone constituent produced by female Dendroctonus jeffreyi Hopkins. The aerations also indicated that males produced 33% (+) and 67% (–)-frontalin, previously unreported for this species, and >99% (+)-exo-brevicomin. Both males and females were responsive to these compounds in electrophysiological studies. Laboratory bioassays and field trapping experiments indicated that males and females were attracted to a mixture of 1-heptanol plus the principal host resin component heptane, but females were less attracted than males. Inclusion of racemic exo-brevicomin at 0.1% concentration in the traps increased the response of females. Inclusion of racemic frontalin above a 1% concentration in the normally attractive lures blocked arrival of beetles to the traps. While the female-produced pheromone components and host synergists are very different between the sibling species D. jeffreyi and Dendroctonus ponderosae Hopkins, the principal male-produced compounds are the same, and the response of the female beetles to those compounds appears to be similar.  相似文献

Many pollinators specialize on a few plants as food sources and rely on flower scents to recognize their hosts. However, the specific compounds mediating this recognition are mostly unknown. We investigated the chemical basis of host location/recognition in the Campanula-specialist bee Chelostoma rapunculi using chemical, electrophysiological, and behavioral approaches. Our findings show that Ca. trachelium flowers emit a weak scent consisting of both widespread and rare (i.e., spiroacetals) volatiles. In electroantennographic analyses, the antennae of bees responded to aliphatics, terpenes, aromatics, and spiroacetals; however, the bioassays revealed a more complex response picture. Spiroacetals attracted host-naive bees, whereas spiroacetals together with aliphatics and terpenes were used for host finding by host-experienced bees. On the intrafloral level, different flower parts of Ca. trachelium showed differences in the absolute and relative amounts of scent, including spiroacetals. Scent from pollen-presenting flower parts elicited more feeding responses in host-naive bees as compared to a scentless control, whereas host-experienced bees responded more to the nectar-presenting parts. Our study demonstrates the occurrence of learning (i.e., change in the bee’s innate chemical search-image) after bees gain foraging experience on host flowers. We conclude that highly specific floral volatiles play a key role in host-flower recognition by this pollen-specialist bee, and discuss our findings into the broader context of host-recognition in oligolectic bees.  相似文献
