采用电聚合的方法将普鲁士蓝(PBl聚合在玻碳电极表面,再将石墨烯修饰在PB上面,然后再采用电沉积的方法将HAuCL直接还原成纳米金粒子,沉积在石墨烯表面,最后将羊抗人IgG抗体直接固定于该修饰的玻碳电极表面,制备了用于人IgG抗原检测的非标记电化学免疫传感器。利用循环伏安法和交流阻抗研究了修饰电极表面的电化学特性,用差分脉冲伏安法对人IgG抗原进行了测定。实验表明,此免疫传感器在含不同浓度人IgG的PBS溶液(pH6.98)中测定,响应电流与人IgG浓度在5.55~455.5ng/mL范围内有良好的线性关系,其相关系数r=0.9926,检测限为0.015ng/mL(S/N=3)。该免疫传感器具有制备简单、响应时间快(5min)、稳定性好等特点。  相似文献

提出了一种基于纳米金结合银增强的质量放大方法以提高压电免疫传感器的灵敏度.通过夹心法将纳米金引入传感器表面后滴加银增强试剂,在纳米金的催化下可使银沉积在传感器上,从而对免疫反应引起的质量变化进行放大.以检测正常人IgG为例,对该方法的可行性作了研究,设计的传感器可在0.3~5μg/mL范围内实现对正常人IgG的定量检测.  相似文献

本工作采用电聚合手段在玻碳电极表面制备聚硫堇基质膜,借硫堇膜上富含的氨基基团实现对碳纳米管与纳米金颗粒二元纳米复合材料的化学组装,制备稳定的碳纳米管/纳米金/聚硫堇传感界面, 以此传感界面固定免疫活性物质,发展了一种无电子媒介的酶免疫传感器.结果表明,电极采用碳纳米管/纳米金颗粒界面固定免疫活性物质,其传感响应性能明显优于单独采用碳纳米管的情况.以人IgG免疫体系为例,电极以此界面固定人IgG抗体,并通过夹心方式直接响应酶催化H2O2的还原电流信号,实现了对人IgG浓度的灵敏测定.此外,采用甘氨酸-HCl缓冲液洗涤使用后的传感器,可有效洗脱其表面的免疫复合物,实现传感器的重复使用.  相似文献

快速检测大肠杆菌O157:H7的电化学阻抗免疫生物传感器   总被引:2,自引:0,他引:2  
建立了一种采用电化学阻抗谱技术快速检测大肠杆菌O157∶H7的生物传感器,它是通过石英晶体金电极表面附着一层蛋白A膜来固定抗体的。该生物传感器采用了三电极系统-工作电极石英晶体金电极、Ag/AgCl/Cl-SAT参考电极和铂对电极。和以往的普通金电极不同,第一次采用了石英晶体金电极。试验结果说明抗体的固定以及大肠杆菌O157∶H7与抗体的结合都增加了石英晶体金电极表面的电子传递阻抗,在[Fe(CN)6]3-/4-氧化还原对存在的情况下,用电化学阻抗谱测量该阻抗。该免疫生物传感器的检测限是103cfu/mL,石英晶体金电极的电子传递阻抗变化值和大肠杆菌O157∶H7的浓度在一定范围内呈线性关系,检测时间少于10 min。  相似文献

该文提出了一种新的、高灵敏的基于银增强放大和免疫胶体金技术的血吸虫抗体的免疫分析方法.免疫反应在聚苯乙烯微孔板中以间接分析模式(血吸虫抗原-兔抗血吸虫抗体-纳米金标记的羊抗兔IgG二抗)进行,通过反应上去的纳米金催化银离子的还原,使大量的银沉积在纳米金的周围,用酸将银溶解后,通过阳极溶出伏安法(ASV)对银离子进行检测,溶出峰电量的大小与待分析物血吸虫抗体(一抗)的浓度成正比.对免疫分析的一些实验条件如抗原与待测抗体的反应时间、纳米金标记二抗与待测抗体的反应时间、纳米金标记抗体稀释度以及银增强时间进行了优化.阳极溶出峰电量与日本血吸虫抗体的浓度在6.4μg/L~100mg/L范围内呈良好的线性关系,检测下限为3.0 μg/L.将该方法应用于兔血清中日本血吸虫抗体的测定,取得了满意的结果.  相似文献

近年来,随着交流阻抗谱实验技术的广泛应用,我们在APPLE—Ⅱ机上发展了阻抗谱的解析与拟合程序(ZRES)。该程序已成功地用于多种离子导电性材料的实验数据处理。本文在介绍ZRES程序主要特点的同时,通过一组模拟计算说明该程序在忽略测量误差情况下,计算精度至少可达10~(-3)Ω。例举了若干不同化学传感器敏感膜的阻抗谱及其拟合参数。  相似文献

本文提出了一种结合酪胺聚合膜和纳米金为界面的甲状腺激素压电免疫传感器.先在晶振表面电聚合一层酪胺聚合膜,然后通过聚酪胺一端的氨基和纳米金之间的作用力在聚合膜表面自组装一层纳米金,再在纳米金上固定T4抗体,实现对T4的检测.考察了一些实验条件,如免疫反应时间,抗体浓度等对传感器响应性能的影响,该传感器对T4在1.0~97.7 μg/dL范围具有很好的线性响应,并将此传感器与采用戊二醛交联法和半胱氨吸附界面的传感器进行了比较.  相似文献

发展了一种利用金纳米颗粒免疫凝集的压电传感技术用于人血清免疫球蛋白IgG的简单、快速、高灵敏检测.以金纳米颗粒替代传统胶乳标记羊抗人IgG诊断血清(抗IgG),利用石英晶体微天平(QCM)直接灵敏响虚因金纳米颗粒免疫凝集而引起溶液的非质量参数(密度、粘度等)的变化.考察了pH值、离子强度和抗IgG-金纳米颗粒浓度对免疫凝集反应的影响,并进行了质控实验.结果表明,该传感技术毋需固定活性组分,可快速榆测浓度下限0.38μg/mL的免疫球蛋白IgC.定量能力与经典ELISA法相接近,可基本满足临床疾病诊断的生化检测要求.  相似文献

利用滴涂于铂盘表面的Nation膜中负电性的磺酸基与anti—IgG分子中的氨基阳离子之间的静电作用实现抗体的结合,同时通过负电性的纳米金增加抗体的固定量,最后在修饰电极的表面涂敷一层明胶(Gelatin)薄膜进行固定,制成Gelatin/anti—IgG/Au/Nafion/Pt膜,制得非标记免疫传感器.用循环伏安法(CV)和交流阻抗法(Nyquist)对电极逐层修饰过程进行了表征,并对免疫传感器的性能进行了研究.该传感器测定IgG的最低浓度为2.0μg/L,标准曲线的线性范围在5—960μg/L,回归方程为:△E=2.2+31.7log[IgG],响应时间为5min.此传感器制备过程简单、稳定性好、灵敏度高、具有良好的选择性,用于生物样品分析,结果令人满意。  相似文献

构建一种基于氧化石墨烯/聚吡咯-铟锡氧化物GO/PPy-ITO(Graphene Oxide/Polypyrrole-Indium Tin Oxide)微电极的细胞阻抗生物传感器并用于细胞粘附增殖行为学检测。ITO微电极采用光刻技术对感光干膜绝缘层蚀刻而成,通过一步法电聚合技术在ITO微电极表面沉积GO/PPy纳米复合膜制备GO/PPy-ITO微电极;形状测量激光显微镜和扫描电子显微镜分别对GO/PPy表面粗糙度和拓扑形貌进行表征;电化学循环伏安法及阻抗谱表征GO/PPy-ITO微电极的电化学性质;人肺癌细胞株A549粘附、铺展和增殖实验考察GO/PPy界面的生物相容性;以GO/PPy-ITO微电极作为传感电极,利用电化学阻抗谱技术对A549细胞的粘附增殖行为进行检测。结果显示,ITO微电极表面上电沉积的GO/PPy纳米复合物表面平整,分布大量的微孔结构;电化————————————学实验结果显示GO/PPy-ITO微电极比裸ITO微电极具有更低的阻抗特征和更高的电化学活性;GO/PPy比纯PPy膜更能促进A549细胞粘附、铺展和增殖;GO/PPy-ITO微电极表面A549细胞的粘附增殖行为改变电极系统的阻抗谱特征,通过对阻抗谱数据进行等效电路拟合分析获得细胞粘附增殖行为学信息。本文发展的GO/PPy-ITO微电极兼具优良的电化学性质和细胞生物相容性,基于该电极系统构建的细胞阻抗生物传感器可用于细胞病理生理学行为、药物筛选等研究领域。  相似文献

The developmental process is seen as a dialogue between technologists and teachers. This dialogue is often problematic because of the differences in training, experience and culture between them. Abstract This paper starts with some reflections on the policy considerations and priorities which are shaping European Commission (EC) research programmes. Then it attempts to position the current projects which seek to capitalise on information and communications technologies for learning in relation to these priorities and the apparent realities of the marketplace. It concludes that while there are grounds to be optimistic about the contribution EC programmes can make to the efficiency and standard of education and training, they are still too technology driven.  相似文献

融合集成方法已经广泛应用在模式识别领域,然而一些基分类器实时性能稳定性较差,导致多分类器融合性能差,针对上述问题本文提出了一种新的基于多分类器的子融合集成分类器系统。该方法考虑在度量层融合层次之上通过对各类基多分类器进行动态选择,票数最多的类别作为融合系统中对特征向量识别的类别,构成一种新的自适应子融合集成分类器方法。实验表明,该方法比传统的分类器以及分类融合方法识别准确率明显更高,具有更好的鲁棒性。  相似文献

Development of software intensive systems (systems) in practice involves a series of self-contained phases for the lifecycle of a system. Semantic and temporal gaps, which occur among phases and among developer disciplines within and across phases, hinder the ongoing development of a system because of the interdependencies among phases and among disciplines. Such gaps are magnified among systems that are developed at different times by different development teams, which may limit reuse of artifacts of systems development and interoperability among the systems. This paper presents control charts models and the necessary simulation software for the location of economic values of the control parameters. The simulation program is written in FORTRAN, requires only 10K of main storage, and can run on most mini and micro computers. Two models are presented - one describes the process when it is operating at full capacity and the other when the process is operating under capacity. The models allow the product quality to deteriorate to a further level before an existing out-of-control state is detected, and they can also be used in situations where no prior knowledge exists of the out-of-control causes and the resulting proportion defectives.  相似文献

Going through a few examples of robot artists who are recognized worldwide, we try to analyze the deepest meaning of what is called “robot art” and the related art field definition. We also try to highlight its well-marked borders, such as kinetic sculptures, kinetic art, cyber art, and cyberpunk. A brief excursion into the importance of the context, the message, and its semiotics is also provided, case by case, together with a few hints on the history of this discipline in the light of an artistic perspective. Therefore, the aim of this article is to try to summarize the main characteristics that might classify robot art as a unique and innovative discipline, and to track down some of the principles by which a robotic artifact can or cannot be considered an art piece in terms of social, cultural, and strictly artistic interest. Although there are many arguments that logic is an appropriate tool for artificial intelligence, there has been a perceived problem with the monotonicity of classical logic. This paper elaborates on the idea that reasoning should be viewed as theory formation where logic tells us the consequences of our assumptions. The two activities of predicting what is expected to be true and explaining observations are considered in a simple theory formation framework. Properties of each activity are discussed, along with a number of proposals as to what should be predicted or accepted as reasonable explanations. An architecture is proposed to combine explanation and prediction into one coherent framework. Algorithms used to implement the system as well as examples from a running implementation are given.  相似文献

为了设计一种具有低成本、低功耗、易操作、功能强且可靠性高的煤矿井下安全分站,针对煤矿安全生产实际,文章提出了采用MCS-51系列单片机为核心、具有CAN总线通信接口的煤矿井下安全监控分站的设计方案;首先给出煤矿井下安全监控分站的整体构架设计,然后着重阐述模拟量输入信号处理系统的设计过程,最后说明单片机最小系统及其键盘、显示、报警、通信等各个组成部分的设计;为验证设计方案的可行性与有效性,使用Proteus软件对设计内容进行仿真验证,设计的煤矿井下安全监控分站具有瓦斯、温度等模拟量参数超标报警功能和电机开停、风门开闭等开关量指示功能;仿真结果表明:设计的煤矿井下安全监控分站具有一定的实际应用价值.  相似文献

This paper provides the author's personal views and perspectives on software process improvement. Starting with his first work on technology assessment in IBM over 20 years ago, Watts Humphrey describes the process improvement work he has been directly involved in. This includes the development of the early process assessment methods, the original design of the CMM, and the introduction of the Personal Software Process (PSP)SM and Team Software Process (TSP){SM}. In addition to describing the original motivation for this work, the author also reviews many of the problems he and his associates encountered and why they solved them the way they did. He also comments on the outstanding issues and likely directions for future work. Finally, this work has built on the experiences and contributions of many people. Mr. Humphrey only describes work that he was personally involved in and he names many of the key contributors. However, so many people have been involved in this work that a full list of the important participants would be impractical.  相似文献
