Antioxidants, preferentially those of dietary origin, have for a long time been considered to help against diseases that are presumably aggravated by oxidative stress, such as cardiovascular diseases, cancer, and neurodegenerative disorders. The outcome of clinical trials undertaken to corroborate this hypothesis, however, remained largely inconclusive. Evidence is now emerging that some dietary "antioxidants" influence signaling pathways and the expression of genes relevant in atherosclerosis by mechanisms other than antioxidative ones. By concrete examples we show that (1) vitamin E has gene regulatory functions which might be more important than acting as an antioxidant in vivo, (2) selenium itself is not an antioxidant at all, and even not in general when incorporated into glutathione peroxidases, and (3) a moderate oxidative stress is beneficial rather than detrimental since it can induce defense mechanisms counteracting xenobiotic and oxidative stress. Thus, there is only a future for antioxidants in the prevention of any disease if their real mechanism of action is considered and suitable read-outs and biomarkers are established.  相似文献

It is currently assumed that benzene contributes only negligibly to secondary organic aerosol formation in the atmosphere. Our understanding of the capacity of benzene to generate secondary aerosols is based on the work of Izumi and Fukuyama (Atmos. Environ. 1990, 24A, 1433) in which two photosmog experiments with benzene in the presence of NOx were performed and no particle formation was observed. In contrast to the observations of Izumi and Fukuyama, experiments performed in the EUPHORE large outdoor simulation chamber have clearly shown aerosol formation during the photochemical oxidation of benzene in various NOx regimes. The maximum aerosol yields of 8-25% on a mass basis are comparable to yields obtained during the photochemical oxidation of other aromatic compounds under similar conditions. In addition, a density of 1.35+/-0.04 g/cm3 for the secondary organic aerosol from the benzene photochemical oxidation in the presence of NOx has been determined through the simultaneous measurement of aerosol volume and aerosol mass using two independent measurement techniques. Comparing the results in the present work with previous findings underscores the strong influence that the NOx content in the system has on aerosol formation during the photochemical oxidation of aromatic hydrocarbons and the importance of performing experiments with NOx concentrations relevant to the atmosphere.  相似文献

A thorough understanding of the factors that regulate the secretion of FSH is critical for the development of efficient methods for fertility regulation. The purpose of this review is to evaluate what is currently known about the existence of FSH-releasing factor(s). It considers the obstacles encountered in understanding the control of FSH secretion, current knowledge of the nature of FSH secretion and the mechanisms involved in controlling FSH secretion, the arguments that have been posed against the need for an FSH-releasing factor and finally the evidence available to date supporting the existence of FSH-releasing factor(s).  相似文献

BackgroundThe gut microbiota is a putative target for dietary interventions for cardio-metabolic health (CMH), including prevention of obesity, type 2 diabetes, and cardiovascular disease. This has generated considerable interest, but the actual feasibility for diet or specific foods to induce measurable, sustained and meaningful benefits for CMH risk by this route remains uncertain.Scope and approachThis report summarises an expert workshop assessing the gut microbiota as a relevant, feasible and competitive target for CMH benefits by dietary interventions. It summarises the expert presentations and overall view of participants on the current status and outlook, considering also implications for the food industry.Key findings and conclusionsChanging the gut microbiota by diet is possible, but an assessment of the impact on CMH risk is still needed, including clarifying advantages above other known dietary routes. The individual gut microbiota composition may in part determine the impact of diet and its effects on health. Therefore, future developments may identify individuals at risk and thus possible modification of the microbiota to achieve benefits in susceptible (sub) populations depending on their initial microbiota composition. Prebiotics currently appear to be the most promising ingredients; however, required doses may be relatively high and the actual role of gut microbiota needs further assessment. Overall, causal evidence linking gut microbiota interventions with CMH benefits are developing in preclinical models but are still lacking in humans. A significant research effort is needed and ongoing to determine whether potential effects can be reliably substantiated.  相似文献

OBJECTIVE: To describe the extent of the tobacco industry involvement in establishing international standards for tobacco and tobacco products and the industry influence on the International Organization for Standardization (ISO). METHODS: Analysis of tobacco industry documents made public as part of the settlement of the Minnesota Tobacco Trial and the Master Settlement Agreement. Search words included "ISO", "CORESTA", "Barclay", "compensation and machine smoking", "tar and nicotine deliveries", and the name of key players, in different combinations. RESULTS: It is clear that the tobacco industry, through the Cooperation Centre for Scientific Research Relative to Tobacco (CORESTA), play a major role in determining the scientific evidence and suggesting the standards that are eventually adopted as international standards for tobacco and tobacco products in several areas, including the measurement of cigarette tar and nicotine yield. CONCLUSIONS: ISO's tobacco and tobacco products standards are not adequate to guide tobacco products regulatory policies, and no health claims can be made based on ISO's tobacco products standards. There is an urgent need for tobacco control advocates and groups worldwide to be more involved with the work of the ISO, both directly and through their national standardisation organisations.  相似文献
