Dewaka's banana grows well and abundant bananas production in Merauke area, but with a little bit sour taste when they’re ripe made this bananas often the second choice compared to another kind of bananas. The research has been conducted to determine concentration of bioethanol produced from dewaka's banana flour. Banana flour was obtained from crused the slices of dried bananas. Then concentration of ethanol can then be identified through a series of tests in the laboratory. The results of gas chromatographic analysis show that dewaka banana can be used as an alternative energy source with bioethanol content of 83.43%. The concentration was result of 5 (five) days fermentation with yeast. The hydrolysis which is used was acid hydrolysis, using hydrochloric acid. The optimum concentration of hydrolysis was 1.5 N HCl, the optimum temperature was 900C, whereas the optimum hydrolysis time was 70 minutes.  相似文献

本研究以库拉索芦荟为原料,滇皂荚多糖胶为复配稳定剂配料,通过正交试验、感官评定、稳定性试验等对芦荟悬浮饮料的生产工艺和产品配方进行了研究。试验结果表明:在芦荟悬浮饮料加工中复合稳定悬浮剂的最佳配方为:琼脂:0.05%,滇皂荚多糖胶:0.03%,氯化钾:0.03%。芦荟悬浮饮料中主要成分的最佳配方为:柠檬酸:0.20%,复合稳定剂:0.10%,白砂糖:11.0%.芦荟果肉:6.0%。  相似文献

采用PCR技术扩增了运动发酵单孢菌Zymomonasmobilis的两个产乙醇的关键酶基因pdc和adhⅡ ,分别克隆到载体 pUC19和pUC18中 ,形成重组质粒 pUC19::pdc和 pUC18::adh ,从中分离出两基因并串联到经EcoRⅠ酶切的 pUC18中 ,转化大肠杆菌E .coliDH5α获得重组质粒 pPA .携带pPA的重组菌Pp +a被证明具有丙酮酸脱羧酶酶活且提高了乙醇脱氢酶酶活 .通过代谢工程在Escherichiacoli中成功地构建了乙醇合成途径  相似文献

An anionic peroxidase POIII, molecular weight 56 kDa, was purified from the roots of horseradish cv. Balady. The enzyme exhibited high activity towards o-phenylenediamine and guaiacol, while o-dianisidine had moderate peroxidase activity. Pyrogallol and p-aminoantipyrine had low affinity toward POIII. POIII was found to have a temperature optimum at 40 °C; the enzyme activity remained stable up to 40 °C and retained 87%, 51% and 29% of its activity at 50, 60 and 70 °C, respectively. The enzyme exhibited more than 50% of activity in the pH range between 4.0 and 8.0 with its pH optimum at 5.5. Several metal cations had partial inhibitory effects toward POIII. Fe3+ enhanced the activity of the enzyme by 160% at 5 mM. All the metal chelators caused partial inhibitory effects toward POIII, except for EDTA at 1 mM, which had no effect on the enzyme.  相似文献

The effects of various sanitizers on the viability and cellular injury to structures of Escherichia coli and Listeria innocua were investigated. A food grade organic acidic formulation (pH 2.5) and acidic, neutral, and basic electrolyzed water [AEW (pH 2.7, oxidation reduction potential; ORP: 1100 mV, free available chlorine; FAC: 150 ppm), NEW (pH 6.9, ORP: 840 mV, FAC: 150 ppm), BEW (pH 11.6, ORP: -810 mV)] were used to treat E. coli and L. innocua cells. After 10 min of exposure to the sanitizers, changes to the bacterial numbers and cell structures were evaluated by plate counting and transmission electron microscopy (TEM), respectively. It was concluded from the results that the sanitizers reduced the E. coli cells between 2 and 3 log CFU/mL. Except for the BEW treatment, reductions in L. innocua population were greater (>1 log CFU/mL) than that of E. coli for all treatments. Data from the TEM showed that all sanitizers caused changes to the cell envelope and cytoplasm of both organisms. However, smaller changes were observed for L. innocua cells. Decrease in the integrity of the cell envelope and aggregation of the cytoplasmic components appeared to be mainly because of exposure to the sanitizers. The organic acid formulation and AEW were the most effective sanitizers against bacterial cells, indicating that penetration of acidic substances effectively caused the cell inactivation. PRACTICAL APPLICATION: An understanding of the method in which E-water and an acidic sanitizer cause injury to E. coli and L. innocua would be helpful in selecting an effective chemical agent as a food safety tool. This will allow a scientist to target similar microorganisms such as food borne bacteria with structures that are vulnerable to the sanitizer.  相似文献

The aim of this study was the evaluation of the use of selected lactic acid bacteria (LAB) starter culture of dairy origin in the production of low-acid fermented sausages (Salame nostrano) produced in a small-scale plant in Umbria (Italy), and their effect on microbiological, physico-chemical and sensorial properties of the products. Salame nostrano was obtained with two different technological processes: with and without the addition of selected LAB starter cultures. Microbial counts of safety indicators were lower in salami made with the addition of starter cultures. Pathogens after the first week of ripening were only detected from salami made without the addition of starter cultures. Control salami were rated as paler and harder, whereas those made with the addition of starter cultures as slightly saltier, juicier and in general more acceptable. Selected dairy-origin starter (SDS) cultures did prevent the growth of safety indicators, greatly reduced the rate of isolation of pathogens and increased the acceptability of full-ripened salami.  相似文献

以酵母AS2.1190为出发菌株,把含有木糖还原酶基因(XYL1)、木糖醇脱氢酶基因(XYL2)以及木酮糖激酶基因(XKS1)的质粒载体pYMIKP-xy127线性化后多拷贝整合进入其基因组,筛选得到基因工程菌株GZ4-127,并对此工程菌株进行葡萄糖、木糖共发酵试验.结果显示GZ4-127比出发菌株的菌体密度提高5%,木糖消耗提高2倍,酒精产率提高12%,说明工程菌已能够有效地利用木糖生产乙醇.  相似文献

人工合成的仿生鲶鱼抗菌肽DNA序列,经聚合酶链式反应(PCR)扩增后得到带有不同限制性酶切位点的序列XH1、XH2、XH3.将此基因克隆至载体pGEX-5X-3,成功构建了三串联的重组质粒.测序验证后转化大肠杆菌BL21(DE3),37℃IPTG诱导,获得高效表达,SDS—PAGE扫描分析表明,融合蛋白可达细菌全蛋白总量的30%,融合蛋白以包含体的形式存在于大肠杆菌细胞中.  相似文献
