采用索氏提取法,以丙酮作为有机溶剂制备掌叶苹婆种子提取物,并利用滤纸药膜法研究了掌叶苹婆种子提取物对玉米象、谷蠹和锈赤扁谷盗3种试虫成虫的触杀作用。结果表明,掌叶苹婆种子丙酮提取物对玉米象、谷蠹和锈赤扁谷盗3种试虫成虫具有很强的触杀作用,且触杀作用随着处理时间延长和处理剂量增加而显著增强。在浓度3.923 mg/cm2处理剂量下处理96 h后提取物对3种试虫的触杀作用校正死亡率均达到100%。掌叶苹婆种子丙酮提取物对玉米象、谷蠹和锈赤扁谷盗成虫的触杀毒力LD50值分别为:0.875、0.716和1.389 mg/cm2。3种试虫成虫对提取物的敏感性为谷蠹>玉米象>锈赤扁谷盗。  相似文献

掌握温度对氮气气调中害虫不同虫态致死时间的影响程度对科学控制气调时间和成功杀虫具有重要意义。测定了18℃、23℃和28℃温度时锈赤扁谷盗的卵、幼虫、蛹和成虫在98%氮气气调过程中不同时间的死亡率及完全致死时间。温度18℃时锈赤扁谷盗卵、幼虫、蛹和成虫的完全致死时间分别为28、18、28和14 d,23℃时完全致死时间分别为22、14、24和8 d,28℃时完全致死时间分别为16、9、18和5 d。温度18℃时卵、幼虫、蛹和成虫的完全致死时间比23℃时相应延迟6、4、4和6 d,比28℃时相应延迟12、9、10和9 d。实验温度下98%氮气气调时锈赤扁谷盗耐受力最强的虫为蛹期,其后依次为卵、幼虫和成虫期。锈赤扁谷盗蛹在18、23和28℃温度下的死亡率-时间回归方程分别为y = 3.85x - 3.85、y = 4.01x - 3.53和y = 4.72x - 3.71。温度降低显著延迟98%氮气气调杀虫时间,氮气气调锈赤扁谷盗时应以杀死其耐受力最强的蛹为目标。  相似文献

采用电子加速器对长角扁谷盗、锈赤扁谷盗和土耳其扁谷盗成虫及小麦进行辐照处理,研究了0~1 000 Gy的电子束辐照对这3种试虫的致死效应,分析了它们对辐照的敏感性,以及0~5 000 Gy剂量辐照下小麦的加工品质。结果表明,辐照剂量越高,3种试虫的死亡速度越快;100 Gy的剂量辐照后,试虫在18 d内全部死亡;3种试虫对辐照的敏感性差异不显著,辐照后第12天,长角扁谷盗、锈赤扁谷盗和土耳其扁谷盗成虫的LD_(99)分别为280.2、260.4、281.3 Gy。电子束辐照对小麦的出粉率、湿面筋含量影响不显著(P0.05),但经2 000 Gy以上剂量辐照的小麦粉的面团形成时间和稳定时间显著下降(P0.05)。因此,100~2 000 Gy辐照剂量可以作为辐照防治扁甲科储粮害虫的适宜剂量。  相似文献

采用高效液相色谱法对871家蚕不同发育阶段、不同虫态胆固醇的含量进行了分析.测定结果表明,雌蚕胆固醇含量在五龄幼虫期的第6d最低(37.188mg/100g),第8d最高(117.170mg/100g),雄蚕胆固醇含量在五龄幼虫期的第8d最低(30.077mg/100g),成蛾后第1d最高(91.698mg/100g);雌蚕胆固醇的含量在幼虫期、成虫期均高于雄蚕;蚕卵、幼虫、蛹、成虫胆固醇含量分别为:132.583、55.105、80.428、100.376mg/100g,方差分析表明,不同虫态胆固醇含量差异显著.871家蚕不同虫态中胆固醇含量均较低,胆固醇含量排序:蚕卵>成虫>蛹期>幼虫.  相似文献

以新鲜大豆为原料,通过标准液添加获得黄曲霉毒素B1(AFB1)含量分别为0.1、0.5和1.0 mg/kg的染毒大豆,考察了紫外光照射、γ射线辐照及其联合技术对大豆中AFB1的降解效果。结果表明,利用紫外光照射降解大豆中AFB1时,紫外光照射强度对大豆中AFB1的降解具有显著影响(P0.05),而大豆中水分含量和AFB1含量对AFB1降解率的影响并不显著(P0.05),紫外光照射前10 min内,大豆中AFB1的降解率随光照时间的增加而增加,但是当照射时间超过10 min后,紫外光照射对大豆中AFB1降解率的影响并不显著(P0.05)。利用γ射线辐照降解大豆中AFB1时,大豆中AFB1降解率随γ射线辐照剂量和大豆水分含量的增加而增加,随大豆中AFB1含量的增加而降低,γ射线辐照剂量、水分含量和AFB1含量对大豆中AFB1的降解均具有显著影响(P0.05)。同时,研究还发现紫外光照射和γ射线辐照对大豆中AFB1的降解具有耦合增效作用。  相似文献

潜在危害的储粮害虫大黑粉盗Cynaeus angustus研究缺乏,掌握其磷化氢耐受力有助于科学治理。采用快速击倒和FAO推荐方法测定了磷化氢对大黑粉盗的KT50值和毒力方程,并与赤拟谷盗Tribolium castaneum和锈赤扁谷盗Cryptolestes ferrugineus进行了比较,测定了100、200、300、400、500 mL/m3磷化氢模拟熏蒸中3种害虫卵、幼虫、蛹和成虫不同时间的死亡率。磷化氢对大黑粉盗、赤拟谷盗和锈赤扁谷盗的KT50值分别为7、127和3 736 min,相应毒力方程斜率值b为3.67、8.28和9.94,相应LC50值为0.008、1.34和5.88 mg/L。害虫不同虫态在100~500 mL/m3浓度下的半数致死时间LT50值于大黑粉盗卵为4~2 h、幼虫4~1 h、蛹4~1 h、成虫3~1 h,于赤拟谷盗为卵12~5 d、幼虫11~5 d、蛹13~7 d、成虫10~4 d,于锈赤扁谷盗为卵28~13 d、幼虫为18~9 d,蛹26~11 d,成虫17~9 d。相应的完全致死时间(LT100)于大黑粉盗卵为21~6 h、幼虫8~3 h、蛹9~6 h、成虫5~3 h,于赤拟谷盗卵为21~18 d、幼虫21~15 d、蛹27~18 d、成虫21~12 d,于锈赤扁谷盗卵为54~30 d、幼虫42~30 d、蛹48~30 d、成虫36~25 d。所测大黑粉盗为磷化氢敏感品系,其各虫态对磷化氢的耐受力为卵>蛹>幼虫>成虫,其耐受力远小于赤拟谷盗和锈赤扁谷盗磷化氢抗性品系。  相似文献

玉米是我国重要的食品和饲料原料,当收获、加工和储藏等措施不当时,可能会造成黄曲霉毒素B1(aflatoxin B1,AFB1)污染玉米这一突出问题,AFB1已被国际癌症机构定为1级致癌物。尽管目前已建立了一些物理、化学和生物降解AFB1的方法,但高效、安全、经济的绿色降解方法仍很少。本研究以AFB1污染的玉米为试样,研究臭氧对玉米中AFB1的降解效果。结果表明:AFB1降解率随着臭氧质量浓度的增加和处理时间的延长而显著提高;当水分质量分数为20.37%的玉米经90 mg/L的臭氧处理40 min后,AFB1含量由77.6 μg/kg降低到21.42 μg/kg,降解率达72.4%。臭氧降解AFB1的动力学模拟结果表明,臭氧降解AFB1符合一级动力学模型。玉米中AFB1降解速率常数按以下次序递减:k90 mg/L>k65 mg/L>k40 mg/L。实验得到臭氧降解AFB1的动力学方程、反应速率常数、决定系数和半衰期,为最优地控制臭氧降解AFB1的反应条件奠定了理论和实践基础,也为臭氧降解AFB1污染玉米的应用提供了技术保障。  相似文献

将不同初始水分(5%、9%、12%、18%)的玉米胚在25℃、45%相对湿度的条件下进行储存,取不同储存时间的玉米胚样品检测其粗脂肪、粗蛋白含量及黄曲霉毒素B1(AFB1)含量,并提取玉米毛油检测其酸值、过氧化值、色泽及AFB1含量,研究储存条件对玉米胚及其毛油中AFB1含量和品质的影响。结果表明,在30 d的储存时间内,初始水分为5%和9%的玉米胚及其毛油的主要质量指标和AFB1含量均无明显变化,但毛油酸值(KOH)分别增加了1.31 mg/g和3.60 mg/g;初始水分为12%和18%的玉米胚,粗脂肪含量分别下降了5.05个百分点和6.09个百分点,毛油酸值(KOH)分别增加了25.89 mg/g和61.69 mg/g,色泽明显加深,玉米胚及其毛油中AFB1含量随储存时间的延长逐渐增加,至30 d时,玉米胚中AFB1含量分别达到3.192μg/kg和7.528μg/kg,对应的毛油中AFB1含量分别达到1.134μg/kg和2.128μg/kg。但所有玉米胚样品及其毛油样品中AFB1含量均未超过国家限量标准(20μg/kg)。  相似文献

以黄曲霉毒素B1(AFB1)含量超标的玉米毛油为原料,AFB1脱除率及含量为考察指标,研究碱炼法对玉米毛油中AFB1的脱除效果以及碱液质量分数、超量碱、碱炼时间及碱炼温度对AFB1脱除效果的影响,并通过单因素试验和正交试验设计确定脱除玉米毛油中AFB1的最佳工艺条件。结果表明:碱炼法对玉米毛油中AFB1的脱除率在90%以上,经碱炼玉米毛油中AFB1含量从73.35μg/kg降至5μg/kg以下,明显优于GB 2716—2005《食用植物油卫生标准》中规定的小于等于20μg/kg的限量指标;碱炼条件对AFB1脱除效果的影响程度为碱液质量分数超量碱碱炼温度碱炼时间;碱炼脱除玉米毛油中AFB1的最佳条件为碱液质量分数6.58%、超量碱0.4%、碱炼时间20min、碱炼温度55℃,此条件下可将玉米毛油中AFB1脱除至0.62μg/kg,脱除率99.15%。研究结果对改进和优化玉米油精炼中的碱炼脱酸过程从而实现对AFB1的高效脱除有很好的指导作用。  相似文献

为完善辐照技术防治储粮虫霉的理论,研究了电子束辐照对绿豆象种群发育及对绿豆品质的影响。绿豆象的卵、幼虫、蛹和成虫经0.1~1.0 kGy剂量的电子束辐照后,卵和幼虫在辐照的虫态死亡;蛹发育至成虫和成虫的死亡率显著增加,其种群存活个体的产卵总量显著减少;辐照处理种群的F1代不能孵化为幼虫或在1~2龄幼虫时全部死亡,而0.2 kGy及以上的辐照剂量则能完全抑制F1代孵化为幼虫。尽管0.2kGy的辐照剂量使绿豆的苗高下降了17%,脂肪酸值升高了12%,但对其发芽率、发芽势、可溶性蛋白质含量、降落数值和过氧化氢酶活性无显著影响,并且提高了绿豆的糊化液黏度。因此,推荐0.2 kGy的电子束辐照作为有效防治绿豆中绿豆象的参考剂量。  相似文献

为提高氨气对玉米中黄曲霉毒素B1(AFB1)的降解效率,首先通过单因素试验研究氨熏温度、氨气浓度、氨熏时间及玉米含水量对黄曲霉毒素B1 降解率的影响,然后通过二次回归正交试验设计及响应面分析确定主要影响因素的最优参数组合:温度37℃、玉米含水量20%、氨气体积分数7.05%、氨熏时间96h,在此条件下玉米中AFB1 降解率的模型预测值为90.4%,实测值为92%,两者较接近。  相似文献

应用高光谱技术研究和构建霉变玉米黄曲霉毒素B1(aflatoxin B1,AFB1)和玉米赤霉烯酮(zearalenone,ZEN)含量的检测方法,通过建立霉变玉米中这2?种毒素含量的预测模型,实现对玉米霉变程度的快速、无损、准确判别。首先,通过对比5?种预处理方法,确定标准正态变量校正法对原始光谱数据进行预处理;然后,采用光谱-理化值共生距离算法结合偏最小二乘回归(partial least squares regression,PLSR)法分析不同校正集样本预测AFB1和ZEN含量的差异,并分别优选出130?个和140?个校正集样本;在采用均匀光谱间隔法对原始光谱变量进行初降维的基础上,对比连续投影算法(successive projections algorithm,SPA)和竞争性自适应重加权算法2?种变量提取法。结果表明:经SPA分别筛选出17?个特征波段且基于较少校正集样本和特征波长光谱信息建立的PLSR模型能够获得较优的预测结果,对应AFB1和ZEN含量预测集的相关系数和均方根误差(root mean square error of prediction,RMSEP)(R2pre,RMSEP)由最初的(0.994?4,0.984?6)和(0.991?6,2.320?9)分别变为(0.997?3,0.681?5)和(0.997?7,1.144?1),在降低模型复杂度的情况下提高了预测精度,表明该模型对这2?种毒素含量能够实现较强的预测能力。因此,利用高光谱技术对玉米AFB1和ZEN含量实施无损检测具有可行性。  相似文献

本试验以2013年收获的“源玉3”和“先玉335”玉米原粮为试验对象,测定了玉米原粮中脂肪含量、脂肪酸组成及脂肪酸值随储藏时间的变化。并对完整籽粒和胚芽中的脂肪酸含量进行了比较分析。试验结果表明,在储藏270 d时间中,脂肪含量分别下降14.25%和12.19%。棕榈酸和硬脂酸含量呈先上升后下降趋势。油酸、亚油酸和亚麻酸含量呈下降趋势。五种脂肪酸含量在胚芽中所占比例略高于在完整籽粒中所占的百分比,但是脂肪酸含量变化规律保持一致,无显著差异。2014年4月1日后,在本试验模拟粮仓储藏条件下,应用玉米胚芽提取玉米油的玉米原粮的适宜储藏期不宜超过210 d,但并不影响玉米原粮的宜存品质。  相似文献

To evaluate the effectiveness of maize detoxification achieved with a modified tortilla-making process (MTMP), maize contaminated with aflatoxin B(1) (AFB(1)) and aflatoxin B(2) (AFB(2)) at levels of 22.46, 69.62, and 141.48 ng/g (AFB(1)+ AFB(2)) was processed into tortillas. Aflatoxin content was determined according to the 991.31 AOAC official method. Based on the results obtained with spiked samples (0.78 to 25 ng/g), the mean recovery was 92%, with a standard error of 1.2, and a coefficient variation value of 4.4%. The MTMP caused 68, 80, and an 84% decrease in aflatoxin content, respectively. Extract acidification (as occurs during digestion) prior to mycotoxin quantification caused some reformation of the aflatoxin structure in tortillas (up to 3%). According to these results, the MTMP seems to be safe for decontamination since a low percentage of the initial aflatoxin concentration can be reverted to the original fluorescent form upon acidification. PRACTICAL APPLICATION: The potential presence of aflatoxins in maize destined for human consumption is a serious problem to the Mexican food supply, as these toxic compounds may persist during the traditional alkaline-process for tortilla elaboration. Consequently, new detoxification procedures are needed that eliminate or at least minimize the aflatoxin risk, through lowering aflatoxin concentration in maize-based products. Under these considerations, the use of MTMP is recommended, since it has definite advantages including non-production of wastewater and reduced energy/time consumption.  相似文献

以普通玉米(苏玉24号,SY-24)、糯玉米(紫花糯玉米,ZY)和甜玉米(京晶3号,TY)为试材,研究了不同玉米品种的组分差异及不同发芽阶段生理生化和碳水化合物组成的变化.结果表明:不同品种玉米之间存在组分差异,SY-24的千粒重和直链淀粉含量最高,ZY中未检测到直链淀粉;ZY的水分、还原糖、可溶性糖、Mg、Na和Ca的含量最高;TY中的类胡萝卜素、可溶性蛋白、游离氨基酸、灰分、粗脂肪、Zn和K含量最高.SY-24中未检出Ca;TY和SY-24中Fe含量没有显著差异(P>0.05),但均显著高于ZY(P<0.05),分别是ZY中铁含量的4.69和1.04倍.在发芽过程中,芽长、发芽率、呼吸强度逐渐升高,干物质含量逐渐降低;淀粉酶活力而逐渐增加,β-淀粉酶活力显著高于α-淀粉酶活力(P<0.05);在淀粉酶作用下,淀粉含量逐渐降低,淀粉颗粒逐渐变小,ZY中没有检测到直链淀粉,SY-24和TY直链淀粉含量随发芽时间延长呈先增加后降低,并分别在第24h和第36h达到最大值,分别是未发芽的3.10倍和1.46倍;可溶性糖在发芽过程中呈现波动变化;ZY和SY-24中还原糖含量逐渐增加;TY中还原糖含量呈现先降低后增加的趋势,在发芽地12h达到最低,与未发芽相比降低了15.44%.  相似文献

Aflatoxin B1 (AFB1) is an unavoidable food contaminant. To evaluate the potential health risk of AFB1 to Koreans posed by food consumption, we determined the natural occurrence of AFB1 in food and estimated the excess risk for liver cancer through dietary exposure to AFB1. A total of 694 food samples collected from six different regions of South Korea were analyzed for their AFB, content. One hundred four of the 694 samples were found to give positive enzyme-linked immunosorbent assay (ELISA) readings for AFB1 and were further investigated with high-performance liquid chromatography. Thirty-two samples, including 2 maize samples, 3 soybean products, 20 peanut samples, nut samples, and their products, and 7 spices, were found to be contaminated with AFB1 (4.6% incidence), up to 48.6 microg kg(-1). The level of AFB1 contamination in 28 of the 32 food products was below 10 microg kg(-1), which is the legal tolerance limit in Korea. From data on daily food consumption, the exposure dose of AFB1 was estimated to be 6.42 x 10(-7) mg kg(-1) body weight (bw) day(-1). The major contributors to the dietary intake of AFB1 were soybean paste and soy sauce, which composed 91% of the total exposure to AFB1. The excess risk of liver cancer for those exposed to AFB1 through food intake was estimated to be 5.78 x 10(-6) for hepatitis B-negative individuals and 1.48 x 10(-4) for hepatitis B-positive individuals. These results suggest that special consideration is required to reduce the intake of AFB1 in hepatitis B-positive individuals.  相似文献

The feedstuffs used in dairy animals must be able to give consumers confidence about the wholesomeness of milk with regard to aflatoxin contamination. The aim of this study was to determine the excretion patterns of aflatoxin M(1) (AFM1) in the milk of dairy goats fed a single dose of pure aflatoxin B(1) (AFB1), which can occasionally occur if feeds are infected by hot-spot growth of molds that produce aflatoxins. Five dairy goats in midlactation were administered 0.8 mg of AFB1 orally. Individual milk samples were collected for 84 h after AFB1 dosage. Aflatoxin M(1) was found in milk in the highest concentration. In all goats, AFM1 was not detected in milk before AFB1 administration, but was detected in the first milking following AFB1 administration. The excretion pattern of AFM1 concentration in milk was very similar in all goats even if the values of the concentration differed between animals. The peak values for AFM1 concentration in milk was observed in milk collected during the milking at 3 and 6h. After the peak, the AFM1 in milk disappeared with a trend that fitted well a monoexponential decreasing function, and the toxin was not detected after 84 h. Only about 0.17% of the amount of AFB1 administered was detected as AFM1 in milk, and about 50% of this was excreted in the first liter of milk yielded after AFB1 intake. Correct procedures to prevent growth of molds, and consequent AFB1 contamination, on the feedstuffs for lactating goats represent the key to providing consumers a guarantee that milk is not contaminated by AFM1.  相似文献

Occurrence of aflatoxins (AFs) AFB1, AFB2, AFG1, AFG2 and ochra toxin A (OTA) was studied in 65 samples of stored and freshly harvested wheat, barley and maize collected in Tunisia. The mycotoxins were simultaneously extracted and quantified by high performance liquid chromatography. Determination of AF-producing (section Flavi) and OTA-producing Aspergillus species (sections Nigri and Circumdati) was conducted in these samples by species-specific polymerase chain reaction (PCR). Results showed that most of maize samples were contaminated with AFs, data after storage showing lower values than those collected at harvest. All contaminated maize samples contained AFG1 and AFG2, among which 27.78% also had AFB1 and AFB2. This AFs pattern was consistent with the A. parasiticus toxin profile. A. flavus however showed the highest frequency in maize but was also found in barley and wheat where no AFs were detected. In contrast, OTA was neither found in maize nor in barley and only one wheat sample contained OTA. A. niger was the only OTA-producing species detected.  相似文献

Weekly sampling of the Indian meal moth, Plodia interpunctella, was conducted over 15 consecutive weeks at 19 locations within a maize storage facility. Direct sampling consisted of maize samples from two depths from the maize surface, and was compared with three indirect sampling techniques: (1) cardboard traps intended for trapping of late instar larvae and pupae; (2) unbaited sticky traps at two heights above the maize intended for trapping adults; and (3) probe traps inserted into the maize intended for trapping both larvae and adults. Temperature was recorded weekly and maize moisture content every 2 weeks at each sampled location. Total catches for the entire sampling period were highest with cardboard traps followed by maize samples, sticky traps, and probe traps. Spatial statistics were used to examine the distribution pattern of weekly catches for each sampling technique: (1) larvae in both top and bottom maize samples indicated similar aggregated distribution patterns; (2) catches with cardboard traps and sticky traps at both heights suggested a random distribution pattern; and (3) catches with probe traps were considered too low for analysis. For each weekly sampling event, the level of spatial association between sampling techniques was investigated in which the spatial distribution of larvae in top samples was compared with: (1) larvae in bottom maize samples; (2) cardboard trap catches; and (3) adults sticky traps. Catches of adults in low sticky traps were compared with: (4) adults in high sticky traps; (5) cardboard trap catches; and (6) larvae in bottom maize samples. Of the six spatial associations, only larvae in the top and bottom maize samples were significantly associated, while there was no significant spatial association involving catches with indirect sampling techniques. We used response surface regression analysis to evaluate the relative contribution of six explanatory variables in a model fitted to the abundance of larvae in maize samples (order of importance): y-coordinate of sampling locations, vertical position of samples, temperature, number of weekly sampling event, moisture content, and x-coordinate of sampling locations. Using additional response surface regression analyses with the same explanatory variables, we showed that abundance of larvae in maize samples was a poor predictor for the occurrence of P. interpunctella individuals in indirect samples. The implications of different spatial distribution patterns for different sampling techniques of P. interpunctella are discussed.  相似文献

采用4 种方法脱除花生油中黄曲霉毒素B1(aflatoxin B1,AFB1),对比研究4 种方法的脱除效果以及对花生油酸价、过氧化值、VE含量、植物甾醇含量的影响。探究碱法脱除花生油中黄曲霉毒素的作用机理、安全性、油中游离碱残留及花生油风味的影响。花生(三级)经压榨后油中AFB1含量为21.50 μg/kg,经碱液处理(碱液pH 13.44、碱液占花生质量10%、30 min)后花生油AFB1残留量降至2.00 μg/kg,脱除率90.7%;膨润土一次吸附(膨润土用量质量分数2.0%、吸附时间30 min)花生油AFB1残留量为0.13 μg/kg,脱除率99.4%;膨润土复吸(首次吸附用量质量分数1.0%、复吸用量质量分数0.5%、每次吸附时间30 min)花生油AFB1残留量为0.10 μg/kg,脱除率为99.5%;碱炼脱酸(温度50 ℃、超碱用量0.8%、30 min)花生油AFB1残留量为0.10 μg/kg,脱除率为99.5%。碱液处理法比其他方法花生油中VE保留率最高,经感官评定比较,该方法花生油风味损失最低。  相似文献
