The effect of initial head spaces of air, 4.9% CO2/2.1% O2/93% N2 and 5% CO2/95% N2 on the microbial flora of shredded carrots was studied at 4 and 10°C. The microbial flora of shredded carrots comprised lactic acid bacteria, pseudomonads and yeasts. Lactic acid bacteria were the predominant organisms in all samples. The pH dropped during the storage of carrots and this was more pronounced at 10°C. The concentration of different organic acids such as lactic, acetic, tartaric, citric and succinic increased in all samples stored under modified atmosphere packaging conditions at both temperatures. The spoilage of carrots stored under 5% CO2/95% N2 was delayed, as indicated by the changes in their texture, colour and odour, compared with those samples stored under air or 4.9% CO2/2.1% O2/93% N2.  相似文献

ABSTRACT:  We investigated the inhibition of Clostridium perfringens spore germination and outgrowth by the biopolymer chitosan during abusive chilling of cooked ground beef (25% fat) and turkey (7% fat) obtained from a retail store. Chitosan was mixed into the thawed beef or turkey at concentrations of 0.5%, 1.0%, 2.0%, or 3.0% (w/w) along with a heat-activated 3-strain spore cocktail to obtain a final spore concentration of 2 to 3 log10 CFU/g. Samples (5 g) of the ground beef or turkey mixtures were then vacuum-packaged and cooked to 60 °C in 1 h in a temperature-controlled water bath. Thereafter, the products were cooled from 54.4 to 7.2 °C in 12, 15, 18, or 21 h, resulting in 4.21, 4.51, 5.03, and 4.70 log10 CFU/g increases, respectively, in C. perfringens populations in the ground beef control samples without chitosan. The corresponding increases for ground turkey were 5.27, 4.52, 5.11, and 5.38 log10 CFU/g. Addition of chitosan to beef or turkey resulted in concentration- and time-dependent inhibition in the C. perfringens spore germination and outgrowth. At 3%, chitosan reduced by 4 to 5 log10 CFU/g C. perfringens spore germination and outgrowth ( P ≤ 0.05) during exponential cooling of the cooked beef or turkey in 12, 15, or 18 h. The reduction was significantly lower ( P < 0.05) at a chilling time of 21 h, about 2 log10 CFU/g, that is, 7.56 log10 CFU/g (unsupplemented) compared with 5.59 log10 CFU/g (3% chitosan). The results suggest that incorporation of 3% chitosan into ground beef or turkey may reduce the potential risk of C. perfringens spore germination and outgrowth during abusive cooling from 54.4 to 7.2 °C in 12, 15, or 18 h.  相似文献

ABSTRACT: Fresh-cut'Carabao'and'Nam Dokmai'mango cubes were stored in air or in high CO2 atmospheres (3%, 5%, and 10%) at 5 °C and 13 °C. Freshly sliced'Carabao'mango cubes had a lower respiration rate and total bacterial count and higher L-ascorbic acid content and firmness than'Nam Dokmai'mango cubes. The shelf life of fresh-cut mango, based on browning discoloration and water-soaked appearance, was 6 d at 5 °C and 4 d at 13 °C for'Carabao'and 2 d at 5 °C and less than 1 d at 13 °C for'Nam Dokmai'. High CO2 atmospheres retarded the development of water-soaked'Carabao'cubes at 5 °C and 13 °C and'Nam Dokmai'cubes at 5 °C. Texture of'Carabao'cubes was enhanced by high CO2, but ethanol and L-ascorbic acid contents were not affected at 5 °C and 13 °C. Total bacterial count was lower in'Carabao'cubes than in'Nam Dokmai'cubes during storage at both temperatures, and a 10% CO2 only reduced the bacterial count on'Carabao'and'Nam Dokmai'cubes stored at 13 °C. Bacterial flora in'Nam Dokmai'mango cubes consisted mostly of Gram-negative rods assigned primarily to phytopathogenic bacteria such as Pantoea agglomerans and Burkholderia cepacia . The genera of bacteria isolated from cubes stored in 10% CO2 were similar to those from cubes on the initial day.  相似文献

We studied the microbial changes on fresh (still warm) pork carcasses immediately after electrostatic spraying of lactic acid solutions and during chilled storage. Lactic acid decontamination (LAD) included electrostatic spraying of hot (55C) solutions of 1% lactic acid for 90 s and 2%- and 5% lactic acid for 30 and 90 s. The treatments increased the carcass surface temperature from 22C to about 32–43C. The bactericidal activity of lactic acid killed mainly Gram-negative bacteria. Reductions in "total" psychrotrophic Gram-negative and Entero-bacteriaceae counts were found reliable indicators for the efficacy of LAD. Hot 2–5% LAD treatments achieved overall reductions in Gram-negative psychrotrophs ranging from 0.8 to more than 1.4 log10 cfu per cm2. Overall reductions in mesophilic Enterobacteriaceae counts ranged from approximately 1.2 to more than 1.8 log10 cfu per cm2. Treatments with 1% lactic acid achieved marginal 0.5–0.6 overall log10 reductions in these groups of organisms. Bactericidal effects after LAD occurred during a lag period of whose duration increased with the lactic acid concentration of the spray solution. The lag lasted less than 1 day on 1% LAD, 2 days on 2% LAD and 4 days on 5% LAD meat. During these lags the meat surface pH rose from pH 2.7 to 3.2 immediately after LAD to pH 4.8 to 5.2. During aerobic chilled storage after LAD the growth of Gram-negative psychrotrophs was controlled only temporarily. These organisms became the dominant group of organisms. The practical utility of electrostatic spraying of fresh pork carcasses with hot 1–5% LAD to improve the keeping quality is questionable .  相似文献

A mixture of 2% acetic, 1% lactic, 0.25% citric and 0.1% ascorbic acid was applied to beef core samples of muscle inoculated with bacteria. Experimental variables were concentration and temperature of mixed acids and type of microorganism. Overall, an increase in either mixed acids concentration (0 to 3%) or temperature (20 to 70°C) resulted in reductions in counts of about one logi0 for aerobic bacteria and S. ryphimurium, less than one log10 for Enterobacteriaceae, and about one-half log10 for E. coli. Initial meat surface pH dropped to 4.3 after treatment with 3% mixed acids solution but rose to 5.2 after 24 hr. Overall, this mixture of acids performed about the same as acetic acid or lactic acid under the same conditions.  相似文献

ABSTRACT:  The ability of a portable hand-held electronic nose (EN) in detecting spoilage of whole Alaska pink salmon ( Oncorhynchus gorbuscha ) stored at 14 °C and in slush ice (1 °C) was investigated. Fish were sampled daily at 14 °C for up to 3 d, while fish stored in slush ice were sampled at various intervals up to 16 d. Sensory evaluations indicated that fish were rejected at day 3 when stored at 14 °C and at day 12 when stored in slush ice. Aerobic bacteria counts for fish skin at 14 °C ranged from 3.4 log10 colony-forming units (CFU)/cm2 (day 0) to 4.8 log10 CFU/cm2 (day 3) and for fish stored in slush ice ranged from 3.4 log10 CFU/cm2 (day 0) to 5.5 log10 CFU/cm2 (day 16). The correct classification rate using forward stepwise general discriminate analysis was 85% and 92% for EN analysis of belly cavity volatiles for fish held at 14 °C and in slush ice, respectively. A predictive model may be developed for spoilage of whole Alaska pink salmon by analyzing belly cavity odors using the EN.  相似文献

Longissimus muscle obtained from beef carcasses was used in this research. Initially, 0.596, 1.0% and 1.5% lactic and citric acid solutions were prepared. Meat was marinated in these solutions (1:4 w/v) in polyethylene bags at 4C for 72h. Bound water, pH, weight gain, cooking loss and Warner Bratzler shear (WBS) were evaluated. Differential Scanning Calorimetry (DSC) was used to determine the bound water content in meat samples. The latent heat of melting (ΔHm) and bound water were found to be functions of moisture content of marinated meat. There was a significant decrease in pH due to marination. Compared to lactic acid, the samples marinated with citric acid held less water. The WS values in control samples were higher than in marinated samples. Cooking loss was lower in samples marinated with lactic acid compared to citric acid marinated samples.  相似文献

ABSTRACT: The efficacy of 2% molecular weight 240, 2% molecular weight 360 polylactic acid (PLA), and an equal mix of both at reducing numbers of Escherichia coli O157:H7 and Lactobacillus plantarum on raw beef was determined. Fresh beef cubes inoculated with either organism were dipped in PLA solutions or wrapped in PLA-sprayed films. Samples were vacuum packaged and stored at 4°C for 42 d. Treated samples maintained a significantly lower pH than controls. Growth of E. coli O157:H7 was totally inhibited by both PLA treatments by up to 7.29 log10 CFU/cm2 when the spray method was used. However, PLA treatments against L. plantarum were not very effective.  相似文献

ABSTRACT:  Effect of organic acids (acetic, citric, and lactic; 100 mM, pH 5) on spore inactivation by pressure-assisted thermal processing (PATP; 700 MPa and 105 °C), high pressure processing (HPP; 700 MPa, 35 °C), and thermal processing (TP; 105 °C, 0.1 MPa) was investigated.  Bacillus amyloliquefaciens  spores were inoculated into sterile organic acid solutions to obtain a final concentration of approximately 1.3 × 108 CFU/mL.  B. amyloliquefaciens  spores were inactivated to undetectable levels with or without organic acids after 3 min PATP holding time. At a shorter PATP treatment time (approximately 2 min), the inactivation was greater when spores were suspended in citric and acetic acids than in lactic acid or deionized water. Presence of organic acids during PATP resulted in 33% to 80% germination in the population of spores that survived the treatment. In contrast to PATP, neither HPP nor TP, for up to 5 min holding time with or without addition of organic acids, was sporicidal. In a separate set of experiments, carrot puree was tested, as a low-acid food matrix, to study spore recovery during extended storage following PATP. Results showed that organic acids were effective in inhibiting spore recovery in treated carrot puree during extended storage (up to 28 d) at 32 °C. In conclusion, addition of some organic acids provided significant lethality enhancement ( P  < 0.05) during PATP treatments and suppressed spore recovery in the treated carrot puree.  相似文献

ABSTRACT:  This study investigated 2 sanitizer formulations and compared them with hydrogen peroxide (H2O2). Formulation number 1 contained citric acid and sodium dodecylbenzene sulfonate (SDBS). Formulation number 2 contained SDBS, citric, lactic, phosphoric acids, and benzoic acid. Low concentration levels of the sanitizers (1.0% for formulation 1 and 0.5% for formulation 2) were compared with 35% H2O2 for their efficacies on Escherichia coli , Listeria innocua, and Saccharomyces cerevisiae inoculated onto low-density polyethylene (LDPE) films and metal cans at room temperature (23 ± 1 °C) and 40 °C. The results showed that both formulations 1 and 2 required >120 s to sanitize both materials from microbial populations at room temperature, while <15 s was needed for the H2O2. Except for formulation 1 on the E. coli inoculated LDPE film surface, the sanitizers completely eliminated the bacterial populations on both materials in 60 s at 40 °C. In general, the formulations were more effective for reduction of the microbial numbers on the can material when compared with the LDPE film. The E. coli showed greater tolerance for the sanitizers when exposed to the process conditions in this study. All sanitizers completely eliminated the test organisms in ≤36 s at 40 °C when tested on a commercial Benco Aseptic packaging machine.  相似文献

ABSTRACT:  The effect of high-intensity pulsed electric field (HIPEF) treatment (35kV/cm, 4 μs pulse length in bipolar mode without exceeding 38 °C) as influenced by treatment time (200, 600, and 1000 μs) and pulse frequency (100, 150, and 200 Hz) for inactivating Salmonella enterica ser. Enteritidis inoculated in tomato juice was evaluated. Similarly, the effect of combining HIPEF treatment with citric acid (0.5%, 1.0%, 1.5%, and 2.0%[wt/vol]) or cinnamon bark oil (0.05%, 0.10%, 0.2%, and 0.3%[vol/vol]) as natural antimicrobials against S. Enteritidis in tomato juice was also studied. Higher treatment time and lower pulse frequency produced the greater microbial inactivation. Maximum inactivation of S. Enteritidis (4.184 log10 units) in tomato juice by HIPEF was achieved when 1000 μs and 100 Hz of treatment time and pulse frequency, respectively, were applied. However, a greater microbial inactivation was found when S. Enteritidis was previously exposed to citric acid or cinnamon bark oil for 1 h in tomato juice. Synergistic effects were observed in HIPEF and natural antimicrobials. Nevertheless, combinations of HIPEF treatment with 2.0% of citric acid or 0.1% of cinnamon bark oil were needed for inactivating S. Enteritidis by more than 5.0 log10 units (5.08 and 6.04 log10 reductions, respectively). Therefore, combinations of HIPEF with organic acids or essential oils seem to be a promising method to achieve the pasteurization in these kinds of products.  相似文献

Beef strip loins were decontaminated by spraying with solutions of various food grade acids (1.0% lactic acid, 1.0% acetic acid and an acid mixture containing 1.0% lactic acid, 2.0% acetic acid, 0.25% citric acid and 0.1% ascorbic acid) followed by vacuum packaging and storing at 4 ± 1°C. Initially and at days 3 and 6 of display in polyvinyl chloride (PVC) film, aerobic plate counts (APCs) of steaks fabricated from the acid treated loins that were stored for 0, 14, 28, 42, 56, 70 and 84 days were not significantly different (P > 0.05) from the APCs of steaks fabricated from control loins.  相似文献

The microbiological status of beef carcass meat was investigated at a traditional and a semi-modern abattoir in Indonesia. Carcasses from the traditional abattoir were generally less contaminated than those from the semi-modern abattoir. The average total aerobic viable bacterial count of carcasses produced at the traditional abattoir, (log10 3.62/cm2) was found to be significantly lower (p = 0.02) at the semi-modern abattoir (log10 4.02/cm2). More than 70% of carcasses at both abattoirs were found to be positive for Escherichia coli (> 10 organisms/cm2), while Salmonellae were found on approximately 15% of carcasses at both locations.  相似文献

Growth, sporulation, and enterotoxin formation by Clostridium perfringens NCTC 8239 were determined in chicken thigh meat incubated at 45°C for 1.5 h and 37°C for up to 12.5 h. With an inoculum of 106 vegetative cells per g, the cell counts reached mean log 10 7.32/g after 6 h of incubation and remained in that range through 14 h. Heat-resistant spores (log10 2.48/g) were first detected at 4 h, and the number increased to log10 5.19/g at 14 h. Enterotoxin (0.19 μg/g) was first detected after 2 h of incubation (1.5 h at 45°C and 0.5 h at 37°C) in the absence of detectable sporulation, and the enterotoxin concentration increased to 0.76 μg/g after 14 h. Significant differences (p < 0.01) in the odor, color, and texture scores for inoculated versus uninoculated cooked chicken following 2 h incubation correlated with the production of enterotoxin and suggested that these parameters could be used as indices of chicken spoilage by C. perfringens.  相似文献

Fresh cows' milk samples were treated with CO2 to give a calculated CO2 content of 77mM. The treated samples as well as the control were stored at 7° and 20 ± 5°C for three days and analysed periodically. The increase in the total count as well as the counts of psychrotrophic, lactic acid and coliform bacteria during storage at 7°C was found to be inhibited by the addition of CO2. The treated samples had higher titratable acidity and lower pH values than the controls but satisfied the clot-on-boiling test. The keeping quality of cooled milk can therefore be improved by the addition of CO2.  相似文献

This study was designed to investigate the microbiological and chemical characteristics of kurut, a dairy product obtained by drying yogurt under the sun in rural areas of Turkey. For this purpose, 50 randomly collected kurut samples from the Kars Region were analysed for determination of their microbiological and chemical characteristics. Analysis of kurut samples showed that kurut contained on average 4.52 log10 cfu/g of aerobic mesophilic bacteria, 2.78 log10 cfu/g of aerobic mesophyl spores, 3.60 log10 cfu/g of lactic acid bacteria, 3.60 log10 cfu/g of Lactococcus , 3.94 log10 cfu/g of mould and yeast, 2.13 log10 cfu/g of Enterobacteriaceae, l.51 log10 cfu/g of sulphite-reducing clostridia, and 1.81 log10 cfu/g of coagulase-positive s taphylococci . Coliform bacteria and enterococci bacteria were not detected in any of the kurut samples. Furthermore, chemical characteristics of the kurut were as follows: pH 4.2, acidity (lactic acid) 2.9, moisture 12.1%, fat 45.9% protein 25.5%, salt 6.7% and ash 10.0%. The quality characteristics of kurut presented here may help create a different perspective for new products to be produced in the dairy sector.  相似文献

The impact of high O2+ high CO2 modified atmospheres (MA), on the preservation of minimally processed carrots was studied. A combination of 50% O2+ 30% CO2 prolonged the shelf life of sliced carrots compared to storage in air by 2 to 3 d. When the carrots received a pre-treatment with a 0.1% citric acid dip and a sodium alginate edible coating prior to packaging, shelf life was extended by 5 to 7 d. Advantages and disadvantages of the proposed MA over previously recommended MA (1% O2+ 10% CO2), related to a range of physicochemical and microbiological characteristics of carrots are discussed.  相似文献

The effects of reduced water activity and 0.1% sorbate on Salmonella typhimurium ATCC 7136 in a soy-starch product were examined. The formulated product (final pH 6.7), was inoculated with cells to give a final concentration of approximately 2 × 106 cells/g. Ten gram samples were placed in a series of desiccators over saturated salt solutions and stored at either 22°C or 40°C. Water activities ranged from 0.79 to 0.97. Samples were withdrawn at prescribed intervals and appropriate dilutions made. A dual plating procedure was used to monitor growth (tryptic soy agar) and development of injury (Levine's Eosin Methylene Blue Agar supplemented with 2% NaCl and 0.05% sodium desoxycholate). Cells stored at 22°C remained viable longer at an aw of 0.79 than at aw values of 0.96 to 0.97. As storage time increased, cells were stressed at a faster rate at higher aw values. This effect was accelerated at 40°C and as storage time increased.  相似文献

Cylindrical samples of beef semitendinosus muscle were dipped in inocula of diluted (1:8) fresh bovine manure or suspensions of Escherichia coli or Salmonella typhimurium. After air drying 15 min, samples were sanitized by dipping in lactic acid(0, 1, 2 or 3%)for 15 s at 25, 40, 55 or 70°C. The most effective treatment was dipping the lean beef muscle in 3% lactic acid solution at 70°C. In general, as the temperature of acid at each concentration was increased from 25 to 70°C, numbers of surviving test organisms were increasingly reduced. Overall, concentration was an insignificant variable at the higher temperatures but became significant as temperature was lowered. Treatment tended to affect aerobes most and E. coli least. Bacterial injury was shown to be a factor to be considered when conducting sanitizing studies.  相似文献

ABSTRACT: The effect of nisin and nisin/EDTA followed by vacuum packaging on the reduction of Brochothrix thermosphacta and Salmonella typhimurium counts on beef was investigated. Beef cubes, inoculated with either bacteria, were dipped in nisin, or nisin combined with EDTA solutions, vacuum packaged, and stored at 4 °C for up to 25 d. Growth of B. thermosphacta was completely inhibited by nisin and nisin/EDTA by up to 6.76 log10 CFU/ml. However, neither treatment showed significant effects against S. Typhimurium. Thus, nisin or nisin/EDTA treatment, followed by vacuum packaging under refrigerated conditions could increase the shelf life of fresh beef by inhibiting the growth of B. thermosphacta , an important spoilage organism in beef.  相似文献
