Depolymerisation of mangrove Rhizophora apiculata bark extracts in the presence of phloroglucinol nucleophiles in ethanol was carried out. The flavan-3-ols and their phloroglucinol adducts were separated using reversed phase liquid chromatography (HPLC). The HPLC analysis of mangrove R. apiculata showed that catechin was the most common component of the flavanoid monomers. The antioxidant activities of these mangrove tannins were evaluated and compared with several commercial tannins by using reducing power, DPPH and ABTS assays with butylated hydroxytoluene, BHT and l-(+)-ascorbic acid as standards. All tannins had reducing power and percentage scavenging activities similar to the (+)-catechin and l-(+)-ascorbic acid standards. In the DPPH assay, >90% of the maximum scavenging activity was attained at 30 μg ml−1. Mangrove tannins had stronger antioxidant activity than the BHT standard in the DPPH assay. The results of the ABTS assay were correlated with the DPPH assay. Scavenging activity in the ABTS assay increased as the tannin concentration increased, up to a plateau at 50 μg ml−1.  相似文献

The pigments from molasses alcohol wastewater were extracted by the macroporous resin adsorption method. The antioxidant and free radical scavenging activities of these pigments were also investigated. The adsorptive characteristics of five macroporous resins including HPD-600, HPD-500, D301-R, NKA-II and D296-R were studied and the results showed that the macroporous resin HPD-600 was most appropriate for extracting the pigments from molasses alcohol wastewater. The antioxidant and free radical scavenging activities of pigments extracted from alcohol wastewater were evaluated using nitrate, hydroxyl radical, superoxide anion radical and 1,1-diphenyl-2-picrylhydrazyl (DPPH) in vitro model systems. The pigment extract exhibited a concentration-dependent radical scavenging activity in all the systems. Meanwhile, scavenging activity of pigment extract in the DPPH system was found to be significantly (P < 0.05) higher than that in other systems and the 50% inhibitory concentration (IC50 value) was about 0.07 mg/ml. The scavenging effect of pigment extract on superoxide anion radical was very weak with IC50 value greater than 10 mg/ml.  相似文献

The ethanol extract and its solvent subfractions, partitioned by n-hexane (HX), chloroform (CF) and ethylacetate (EA), from Enteromorpha prolifera were measured for antioxidant activities, and a structural identification of the active compound was performed using spectroscopic techniques. The CF fraction showed the most potent 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl (OH) radical scavenging activities with strong reducing ability. The DPPH and hydroxyl radical scavenging capacities of the CF fraction were comparable to the capacities of the positive controls, BHA and α-tocopherol, at concentrations ranging from 0.25 to 1.0 mg/mL. However, little correlation (r2 = 0.03–0.48) was observed between antioxidant activities and total phenolic contents of the extracts. Further fractionation and spectroscopic analysis of the CF fraction suggested that the strong antioxidant activity of the extracts from E. prolifera was because of a chlorophyll compound, pheophorbide a, rather than phenolic compounds.  相似文献

Screening of 91 native plants from central Argentina was carried out with the aim of finding new sources of anti-tyrosinase compounds. Extracts obtained from Achyrocline satureioides, Artemisia verlotiorum, Cotoneaster glaucophylla, Dalea elegans, Flourensia campestris, Jodina rhombifolia, Kageneckia lanceolata, Lepechinia floribunda, Lepechinia meyenii, Lithrea molleoides, Porlieria microphylla, Pterocaulon alopecuroides, Ruprechtia apetala, Senna aphylla, Sida rhombifolia, Solanum argentinum, Tagetes minuta and Thalictrum decipiens exhibited more than 90% inhibition of tyrosinase monophenolase activity at 1000 μg ml−1. D. elegans,L. meyenii and L. molleoides were the most potent with IC50 values of 0.48, 10.43 and 3.77 μg ml−1, respectively. D. elegans, L. molleoides and T. decipiens also showed more than 90% inhibition of diphenolase activity at 1000 μg ml−1, with the first of these being the most effective (IC50 = 49.27 μg ml−1). (Z,Z)-5-(trideca-4,7-dienyl)-resorcinol (1) was isolated from L. molleoides as an effective tyrosinase inhibitor with l-tyrosine or l-DOPA as substrates (IC50 = 0.49 and 14.94 μg ml−1, respectively). Compound 1 was 37 times more active in monophenolase inhibitory activity than kojic acid used as a reference. Effective extracts as well as (Z,Z)-5-(trideca-4,7-dienyl)-resorcinol could prove to be promising preservative agents for use in the food industry.  相似文献

Agrocybe cylindracea (DC: Fr.) Mre. was available in the form of fruit bodies, mycelia and fermentation filtrate. From these three forms, hot-water extracts were prepared and their antioxidant properties were studied. Antioxidant activities of hot-water extracts from fruit bodies, mycelia and filtrate were 63.6%, 81.6% and 56.8% at 20 mg ml−1, respectively. EC50 values in reducing power were 2.72, 3.97 and 3.09 mg ml−1 whereas those in scavenging abilities of 1,1-diphenyl-2-picrylhydrazyl radicals were 0.62, 1.66 and 0.82 mg ml−1 for fruit bodies, mycelia and filtrate, respectively. At 20 mg ml−1, the scavenging abilities of hydroxyl radicals were 80.1%, 57.0% and 54.3% for fruit bodies, mycelia and filtrate, respectively. With regard to EC50 values in chelating abilities on ferrous ions, the hot-water extract from filtrate was better than that from mycelia. Total phenols were the major naturally occurring antioxidant components found in hot-water extracts and in the range of 23.74–30.16 mg g−1. From EC50 values obtained, it can be concluded that hot-water extracts from three forms of A. cylindracea were good in antioxidant properties.  相似文献

The ethyl acetate and methanol extracts from 16 Salvia L. species were screened for their inhibitory activity against acetylcholinesterase, butyrylcholinesterase, lipoxygenase, and tyrosinase; the enzymes linked to neurodegeneration. Their antioxidant activity was also tested using DPPH radical scavenging, metal-chelation, and ferric-reducing antioxidant power (FRAP) assays. Total flavonoid content of the extracts was determined by AlCl3 reagent, while HPLC technique was applied for analysis of various phenolic acids in the extracts. The extracts exerted weak cholinesterase and tyrosinase inhibition, and remarkable inhibition against lipoxygenase (13.07 ± 2.73-74.21 ± 5.61%) at 100 μg ml−1. The methanol extracts showed higher antioxidant activity in DPPH radical scavenging and FRAP assays. The extracts were analyzed for their gallic, protocateuchic, p-hydroxy-benzoic, vanillic, caffeic, chlorogenic, syringic, o- and p-coumaric, ferulic, rosmarinic, and tr-cinnamic acid contents and the methanol extract of Salvia ekimiana (153.50 mg 100 g−1) was revealed to be the richest in terms of rosmarinic acid.  相似文献

Various solvent extracts of Kappaphycus alvarezii, an edible red seaweed (family Solieriaceae) were screened for total phenol content and antioxidant activity using 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferrous ion chelating activity, reducing power and antioxidant activity assays in a linoleic acid system with ferrothiocyanate reagent (FTC). The total phenol content of different extracts of K. alvarezii varied from 0.683 ± 0.040% to 2.05 ± 0.038%. The radical-scavenging activity of ethanol extract was, as IC50 3.03 mg ml−1, whereas that of the water extract was IC50 4.76 mg ml−1. Good chelating activity was recorded for methanol extract (IC50 3.08 mg ml−1) wherein 67.0 ± 0.924% chelation was obtained using 5.0 mg ml−1 of extract. The reducing power of the samples was in the following order: BHT > methanol > ethanol > ethyl acetate > water > hexane. But, in the linoleic acid system, the ethanol extract proved superior to the synthetic antioxidants butylated hydroxytoluene (BHT). Hence, these extracts could be considered as natural antioxidants and may be useful for curing diseases arising from oxidative deterioration.  相似文献

Knipholone anthrone (KA), isolated from an Ethiopian medicinal plant, Kniphofia foliosa, Hochst (Asphodelaceae) has been shown to display a potent antiprotozoal activity and a relatively little cytotoxic effect on mammalin cells. In the present study, the antioxidant potential of KA was assessed using a variety of in vitro assay models. KA displayed a concentration-dependant scavenging effect against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals. The IC50 values obtained for KA and the positive control (−)-epicatechin (EC) in the DPPH assay were 22 ± 1.5 and 8.7 ± 0.9 μM (mean ± SEM; n = 3 separate experiments), respectively. KA displayed a better activity than EC in scavenging superoxide anions and preventing deoxyribose degradation by hydroxyl radicals. KA appeared to form a complex with Fe2+, displayed a concentration-dependant reducing power, and also protected (at concentrations of 4.4 μM and above) isolated DNA from damage induced by Fenton reaction-generated hydroxyl radicals.  相似文献

This study was designed to examine the in vitro antioxidant activities and rosmarinic acid levels of the methanol extracts of Salvia verticillata subsp. verticillata and S. verticillata subsp. amasiaca. The extracts were screened for their possible antioxidant activity by two complementary test systems, namely DPPH free radical-scavenging and β-carotene/linoleic acid systems. In the first case, S. verticillata subsp. verticillata was superior to the subsp. amasiaca with an IC50 value of 14.5 ± 1.21 μg mg−1. In the β-carotene/linoleic acid test system, inhibition capacity of S. verticillata subsp. verticillata was 74.4 ± 1.29%. Antioxidant activities of BHT, ascorbic acid, curcumin and α-tocopherol were determined in parallel experiments. Activity of rosmarinic acid was also screened for better establishing the relationship between rosmarinic acid level and antioxidant activity for the plant extracts. S. verticillata subsp. verticillata had the highest rosmarinic acid level with a value of 28.7 ± 0.89 μg mg−1. There is a strong correlation between the rosmarinic acid level and antioxidant activity potential. Our results showed that rosmarinic acid and its derivatives are more likely to be responsible for most of the observed antioxidant activities of Salvia species.  相似文献

The potential of using Rhodotorula glutinis alone or in combination with salicylic acid (SA) for the control of postharvest Rhizopus rot of strawberries, and their effects on enzyme activities of fruits were investigated. The combination of R. glutinis (1 × 108 CFU ml−1) with SA (100 μg ml−1) resulted in a significant reduction in the disease incidence and lesion diameter of Rhizopus rot on the strawberry fruits at 20 °C and 4 °C, and more so than with SA or yeast alone. SA at the concentration of 100–1000 μg ml−1 significantly inhibited spore germination of Rhizopus stolonifer. About 100 μg ml−1 of SA did not inhibit the growth of the antagonistic yeast, and could significantly increase the population growth of R. glutinis in strawberry wounds at 20 °C. SA, combined with R. glutinis, increased the activity of strawberry host defence enzymes (POD) and cell wall lytic enzymes (β-1,3-glucanase).  相似文献

A novel red pigment (RP) was isolated from Osmanthus fragrans  ’ seeds. The optimised experimental parameters of extraction obtained with a four-factor at three-level orthogonal array experimental design L9(34)L9(34) were ethanol concentration, temperature, pH and extraction time as 90%, 78 °C, 2.5 and 40 min, respectively. A yield of 34.6 ± 2.2 g/100 g was obtained under optimised conditions. The red pigment directly from O. fragrans’ seeds can be dissolved in alkaline, acidic waters solutions and hydrophilic organic solvents in common use. The colour of a water solution of RP changed with pH. RP was stable to heat in the temperature range of 25–100 °C. Physical and chemical properties of RP revealed that the red pigment was also stable in the presence of Na2SO3, NaCl, amino acid, organic acid, sugar, starch or metal-ions (such as Ca2+, Cu2+, Fe3+, Zn2+, Al3+, Mg2+ and Na+), but was bleached by strong oxidants (KMnO4, K2Cr2O7 and NaOCl). Subsequently, free radical scavenging activities of RP were assessed using 2,2′-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radicals using a new resonance scattering spectral method. RP showed an excellent DPPH radical scavenging activity and was superior to butylated hydroxytoluene (BHT), and exhibited quite a strong concentration-dependent inhibition of hydroxyl radical at low concentrations compared with ascorbic acid and quercetin. When the concentration of RP was 0.03 μg/ml, the scavenging percentage of hydroxyl radical reached 92.3%. Salidroside was isolated as an active principle.  相似文献

Six genotypes of sweet potato commercially available in Taiwan, including TNG57, TNG66, TNG68, TYY1, RP and WP, were used as samples in this study of the effects of steaming and kneading with pre-steaming treatments on the antioxidant components and antioxidant properties of methanolic extracts. Steam treatment increased the total phenols contents of all genotypes (2–13 times), flavonoids content of RP (1.3 times) and anthocyanins contents of RP and WP (5–6 times). Steam treatment also increased the reducing power and scavenging DPPH radical effect of sweet potato flours. For the methanolic extracts of steamed and kneaded flours, reducing powers were 0.02–1.70 at 5.0 mg ml−1 and the scavenging effects on DPPH radicals were 19–92% at 2.5 mg ml−1. Both showed the order of RP > WP > TYY1 and TNG66 > TNG57 and TNG68. However, the chelating effect of the six genotypes at 1.0 mg ml−1 ranged from 50% to 73%. Contents of total phenols, flavonoids, and anthocyanins of sweet potato flours were significantly positively correlated with the reducing power and scavenging DPPH radical effects. After steaming and kneading treatments, RP showed the highest increase in the contents of total phenols, flavonoids and anthocyanins among the six genotypes studied.  相似文献

In the present work, we report the results of a study aimed at evaluating the antiradical activity, the antioxidant activity and the acetylcholinesterase (E.C. 3.1.1.7.) inhibitory capacity of essential oils, ethanol and boiling water extracts from five aromatic herbs growing wild in Portugal and used in traditional food preparations: fennel (Foeniculum vulgare), mint (Mentha spicata), pennyroyal (Mentha pulegium), rosemary (Rosmarinus officinalis) and wild thyme (Thymus serpyllum). The water extracts of M. spicata and M. pulegium showed the highest radical-scavenging activity (DPPH test) values (IC50 = 5.7 ± 0.4 and 8.9 ± 0.2 μg ml−1 respectively). This activity was higher than that found with the standard antioxidant BHT. The ethanol extracts of M. spicata, T. serpyllum and F. vulgare showed the highest antioxidant activities measured by the β-carotene/linoleic acid assay, IC50 = 36.9 ± 0.1, 41.2 ± 0.1 and 68.7 ± 0.1 μg ml−1, respectively. The inhibition of AChE was higher in the essential oil fraction. The highest activity was found for R. officinalis with an IC50 = 69.8 ± 0.1 μg ml−1.  相似文献

The methanol extract from mature fruits of Aristotelia chilensis (Mol) Stuntz (Elaeocarpaceae) showed antioxidant activities and cardioprotective effects on acute ischemia/reperfusion performed in rat heart in vivo. This extract protected animals from heart damage by the incidence of reperfusion dysrythmias, and the no-recovery of sinus rhythm. On the other hand, the MeOH extract of the fruit was able to prevent these harmful events in the animal’s heart by diminishing lipid oxidation and reducing the concentration of thiobarbituric acid reactive substances (TBARS), a lipid peroxidation index. In addition, MeOH extract of A. chilensis was evaluated for DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging, crocin radical scavenging, oxygen radical absorption capacity (ORAC), ferric reducing antioxidant power (FRAP), an estimation of lipid peroxidation in liposomes through the inhibition of formation of TBARS. MeOH extract was found to have IC50 of 1.62 ppm against DPPH and 2.51 ppm against TBARS, compared with the juice, whose IC50 was 12.1 ppm and 9.58 ppm against DPPH and TBARS formation, respectively. Antioxidant activities of MeOH extract were strongly correlated with total polyphenol content. Consistent with this finding, MeOH had the greatest ORAC and FRAP values as percentage of activity. These results show that these fruits could be useful as antioxidant, cardioprotective and nutraceutical sources.  相似文献

Davallia solida rhizome has long been used as an herb tonic to treat osteoporosis, arthralgia, and arthritis. The aqueous extract of D. solida rhizome contains a high content of phenolic compounds [210.8 ± 4.6 mg catechin equivalents (CE)/g dry weight] and shows a strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity (IC50 = 15.93 ± 1.21 μg dry weight/ml). Further solvent partition of the aqueous extract yielded chloroform, n-butanol, and water layers. Among them, n-butanol layer has the highest phenol content (806.3 ± 12.3 mg CE/g dry weight) and DPPH scavenging potential (IC50 = 3.93 ± 0.31 μg dry weight/ml). Isolation and purification from the n-butanol layer identified 12 compounds. They included four new compounds: 3′-O-p-hydroxybenzoylmangiferin (1), 4′-O-p-hydroxybenzoylmangiferin (2), 6′-O-p-hydroxybenzoylmangiferin (3), and 3-O-p-hydroxybenzoylmangiferin (4); as well as eight known compounds: mangiferin (5), 2-C-β-d-xylopyranosyl-1,3,6,7-tetrahydroxyxanthone (6), 4β-carboxymethyl-(−)-epicatechin (7), 4β-carboxymethyl-(−)-epicatechin methyl ester (8), eriodictyol (9), eriodictyol-8-C-β-d-glucopyranoside (10), icariside E5 (11), and icariside E3 (12). DPPH scavenging and Trolox equivalent antioxidant capacity (TEAC) analyses revealed that the most potent antioxidants are 1, 2, and 3, which exerted more than triple activity as compared with the positive controls, α-tocopherol and Trolox.  相似文献

Major anthocyanins, ascorbic acid content, total phenolic content, and the radical scavenging activity against ABTS and DPPH radicals in petals of orange Nasturtium flowers (Tropaeolum majus), were investigated. Anthocyanin (ACN) content in the petals was 72 mg/100 g FW and pelargonidin 3-sophoroside represented 91% of the total ACN content. The ascorbic acid content was 71.5 mg/100 g and the total phenolic content as determined by the Folin–Ciocalteau method was 406 mg GAE/100 g FW. The radical scavenging activities against ABTS and DPPH radicals were 458 and 91.87 μm trolox eq/g FW, respectively. The excellent free radical scavenging activities along with high phenolic and ascorbic acid content of Nasturtium flowers suggest that they could be source of natural pigments and antioxidants for applications in functional foods.  相似文献

The antioxidative and hepatoprotective potential of Solidago microglossa D.C, a widely used medicinal plant from Brazil was investigated. The leaf extract showed inhibition against thiobarbituric acid reactive species (TBARS) induced by different prooxidants (10 μM FeSO4 and 5 μM sodium nitroprusside SNP) in rat liver, brain and phospholipid homogenates from egg yolk. Moreover, the free radical scavenging activities of the extract was evaluated by the scavenging of 2,2-diphenyl-1-picrylhydrazyl (DPPH) (IC50, 3.8 ± 0.5 μg/ml) and hydroxyl radical on benzoic acid hydroxylation (IC50, 32.3 ± 1.3 μg/ml) and deoxyribose (IC50, 39.1 ± 2.4 μg/ml) assays. The ethanolic extract showed significant hepatoprotective activity against paracetamol (250 mg/kg) induced liver damage in mice in a dose dependent manner. The phenolic composition and their quantification by high performance liquid chromatography (HPLC) resulted in the identification of gallic acid and flavonoids: quercetrin (quercetin-3-O-rhamnoside), rutin (quercetin-3-O-rutinoside) and quercetin.  相似文献

Antioxidant capacity and antimicrobial activities of Laetiporus sulphureus (Bull.) Murrill. extracts obtained with ethanol were investigated in this study. The study was aimed at determining the antioxidant activity (DPPH free radical-scavenging, β-carotene/linoleic acid systems), total phenolic content and total flavonoid concentration of L. sulphureus. Inhibition values both of L. sulphureus ethanol and the standards increased parallel with the elevation of concentration in the linoleic acid system. Inhibition values of L. sulphureus (LS) extract, BHA and α-tocopherol standards were found to be 82.2%, 96.4% and 98.6%, respectively, at a concentration of 160 μg/ml. DPPH free radical-scavenging activity was found to exhibit 14%, 26%, 55% and 86% inhibition, respectively, at concentrations of 100, 200, 400 and 800 μg/ml. Total flavanoids were 14.2 ± 0.12 μg mg−1 (quercetin equivalent) while the phenolics were 63.8 ± 0.25 μg mg−1 (pyrocatechol equivalent) in the extract. Positive correlations were found between total phenolic content in the mushroom extracts and their antioxidant activities. Edible mushrooms may have potential as natural antioxidants. L. sulphureus showed narrow antibacterial activity against Gram-negative bacteria and strongly inhibited the growth of the Gram-positive bacteria tested. The crude extract exhibited high anticandidal activity on Candida albicans. Therefore, the extracts could be suitable as antimicrobial and antioxidative agents in the food industry.  相似文献

Dorystoechas hastata (D. hastata) is a monotypic plant endemic to Antalya province of Turkey. D. hastata leaves are used to make a tea locally called “çalba tea”. Diethyl ether (E), ethanol (A), and water (W) were used for the sequential preparation of extracts from dried D. hastata leaves. A hot water extract (S) was also prepared by directly boiling the powdered plant in water. The antioxidant activities of the extracts were tested by ferric thiocyanate (FTC) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging methods. E extract exhibited the greatest antioxidant activity with FTC method, whereas S extract exhibited the lowest IC50 value (6.17 ± 0.53 μg/ml) for DPPH radical scavenging activity. Total phenolic contents of the extracts were estimated by Folin–Ciocalteu method and S extract was found to contain the highest amount (554.17 ± 20.83 mg GAE/g extract) of phenolics. Extract A contained highest flavonoid content and there was a inverse linear correlation (R2 = 0.926) between IC50 values for DPPH radical scavenging activity and flavonoid contents of all extracts. Reducing power of extracts increased in a concentration-dependent manner. S extract was found to possess higher reducing power than equivalent amount of ascorbic acid at 20 and 25 μg/ml concentrations. Linear correlation between reducing power and concentration of E, A, and W extracts (R2 > 0.95) was observed. A, W, and S extracts contained relatively high levels of proline. The results presented suggest that D. hastata may provide a natural source of antioxidants and proline.  相似文献

Enzymatic hydrolysis of hemp protein isolate (HPI) by six proteases (alcalase, flavourzyme, neutrase, protamex, pepsin and trypsin) and antioxidant activities of the resulting hydrolysates, obtained for 2 and 4 h were investigated. The yield of trichloroacetic acid (TCA)-soluble peptides (Ysp), protein composition and surface hydrophobicity (Ho) of the hydrolysates were evaluated. The results showed that the hydrolysates exhibited varying DPPH radical scavenging (with lowest IC50, ∼2.3 mg/mL) and Fe2++ chelating (with lowest IC50 of 1.6–1.7 mg/mL) abilities and reducing power (with highest absorbance at 700 nm of 0.31–0.35), depending on their Ysp and Ho values. The DPPH radical scavenging and Fe2++ chelating abilities of the hydrolysates were positively correlated with their Ysp or Ho values. The results suggest that enzymatic hydrolysis can be used as an effective technique to produce high value-added products of hemp proteins.  相似文献
