The astigmatid mite Acarus farris is the main species of mite infesting Cabrales cheese, a traditional Spanish product. This species may reach population density levels of 260 mobile mites/cm2, resulting in high economic losses. Different chemical and physical methods that were able to control this pest without affecting the organoleptic quality of Cabrales cheese were tested. The efficacy of four short-chain fatty acids (caproic, caprylic, pelargonic and capric) and two sugar alcohols (ribitol and xylitol) were tested under laboratory conditions against mobile stages of A. farris. Only pelargonic acid significantly increased mortality in comparison with controls (63.4 vs. 26.1%, respectively). Additionally, the contact effect of the monoterpene eucalyptol was assessed in cheese maturing rooms at doses of 2.5 and 1.25 μl/cm2. The first dose effectively controlled A. farris, but its application negatively affected the organoleptic values of the treated cheeses. Finally, the application of the waxy food coating READOM CBR to Cabrales cheeses in maturing rooms, kept the mite population low but negatively modified the external appearance of the cheese, an important quality parameter.The physical approach was based on lowering the usual cheese maturing temperature (15 °C) to 6, 4 or 2 °C. As temperature decreased, A. farris density was drastically reduced from 174 ± 33 mites/cm2 at the control temperature (15 °C) to 14, 11 and 1 mites/cm2 for 6, 4 and 2 °C, respectively, though the maturing time was considerably extended. The feasibility of both chemical and physical methods for the control of A. farris is discussed.  相似文献

The synanthropic mould mite, Tyrophagus putrescentiae infests various stored products, plant seeds and mushroom beds. Its biology was studied under controlled laboratory conditions at 10, 15, 20, 25, and 30 °C, 70% r.h. and a photoperiod of 16:8 (L:D) h when reared on button mushroom, Agaricus bisporus and oyster mushroom, Pleurotus ostreatus. Tyrophagus putrescentiae had a significantly shorter developmental time at 25 °C when reared on button mushroom compared with oyster mushroom and this time decreased with increasing temperature. The pre-oviposition period was not influenced by the species of mushroom, but the other reproductive periods showed significant changes and female longevity was influenced by food type. The sex ratio of the offspring (♀/♂) and female percentage were 1.16% and 53.7%, respectively. When reared on button mushroom at 25 °C, the net reproductive rate (R0) for T. putrescentiae was 90.13 and the intrinsic rate of increase (rm) was 0.196 which were significantly different from the 18.97 and 0.122, respectively, when reared on oyster mushroom. The doubling times were 3.74 and 5.62 days for the two mushroom types, respectively. The developmental thresholds for eggs, larvae, protonymphs and tritonymphs were 8.38, 8.01, 7.51 and 10.12 °C, respectively, and the thermal unit (degree-days) for all immature stages was 250.62 °C. The spoilage potential of Brochothrix campestris and Brochothrix thermosphacta was investigated in vacuum-packed lamb. Striploins (n = 338) were inoculated and stored for twelve weeks at temperatures − 1.5, 0, 2 and 7 °C. Growth around 5–6 log10 CFU/cm2 was recorded after six weeks at 0, 2 and 7 °C, and ~ 3 log10 CFU/cm2 after nine weeks at − 1.5 °C. B. campestris was shown to cause spoilage by nine weeks at temperatures above 0 °C by the presence of green drip and unacceptable odours. Molecular based assays for the detection and differentiation of B. thermosphacta and B. campestris were developed and validated. A TaqMan assay was designed to target a unique single-nucleotide polymorphism in the Brochothrix 16 s rRNA gene with a sensitivity of < 7 CFU per reaction. Secondly a specific PCR was designed for B. campestris targeting the structural genes, brcA and brcB. These testing regimes offer a rapid and cost effective method for the detection and screening of Brochothrix species in meat products and processing environments.  相似文献

Late-instar larvae, pupae, and adults of Tribolium castaneum (Herbst), the red flour beetle, or Tribolium confusum (DuVal), the confused flour beetle, were exposed for variable durations at 36-54 °C. Beetles were placed in laboratory ovens set at a baseline of 27 °C, the temperature was increased by 0.1 °C per minute until the target temperature was achieved, and beetles were then held for specified exposure durations. There was no mortality after initial exposure or after a 1-week holding period of any life stage of T. castaneum or T. confusum exposed for 32 h to 36, 39, or 42 °C. At 45 °C, there was no initial mortality of either species exposed for different time intervals except for those exposed for 28 h. However, there was a significant increase in mortality after the 1-week holding period of those beetles exposed initially for at least 16 h to 45 °C. There was a sharp increase in mortality after the initial exposures of 4 h at 48 °C; mortality of T. confusum larvae was 90.0±5.7% but was only 10.0±10.0% for larvae of T. castaneum, and no pupae of either species were dead. All life stages of both species were killed after the initial exposure of 12 h, and 1-week mortality of beetles exposed for 4 and 8 h was generally greater than initial mortality. At 51 and 54 °C, 2- and 1-h exposures, respectively, killed all life stages of each species. Mortality in conditions of gradual temperature increase was less than previous studies with sudden temperature increases.  相似文献

The spoilage potential of Brochothrix thermosphacta, Serratia proteamaculans and Rahnella aquatilis was investigated in vacuum packaged high (5.9 to 6.4) and low (5.4 to 5.8) pH lamb. Vacuum packaged fore shank (m. extensor carpi radialis) and striploins (m. longissimus dorsi) (n = 306) inoculated with ~ 100 CFU of individual bacteria were stored for twelve weeks at temperatures − 1.5, 0, 2 and 7 °C. Spoilage characteristics and bacterial numbers were recorded and analysed in comparison to un-inoculated control samples. All three bacterial species were shown to grow in vacuum packaged lamb of pH values between 5.4 and 6.4, when stored at chilled temperatures (− 1.5 to 7 °C) for up to 84 days. B. thermosphacta and S. proteamaculans caused spoilage to the meat under these conditions whilst R. aquatilis spoiled high pH meat at 7 °C. These results go against previous beef models stipulating that Brochothrix and Enterobacteriacae species cannot grow on or cause spoilage of low pH meat in the absence of oxygen.  相似文献

Characterisation of esterase activities from the edible mushroom species, Amanita vaginata var. vaginata and Tricholoma terreum, were investigated. Native electrophoresis of the crude extracts prepared from both mushroom samples showed the presence of esterolytic activities. The extracts had the greatest activity in the presence of p-nitrophenyl butyrate (pNPB) as a substrate. pH and temperature optima were found to be 8.0 and 30 °C for both enzymes, respectively. Vmax and Km values were determined as 14.2 U/l and 71 μM for A. vaginata var. vaginata and 34.6 U/l and 9.6 μM for T. terreum, respectively. The pH-stability profile showed a stationary line between 3.0 and 10.0 for both enzymes. The esterolytic activities from the extracts were maintained between 10 and 40 °C for 4 h and started to decrease at 50 °C. The effects of EDTA, NaN3, DTT and PMSF on the enzyme activity were also investigated.  相似文献

Laboratory experiments were conducted in order to assess the insecticidal effect of a diatomaceous earth formulation (Silicosec®, Biofa GmbH, Germany) against Sitophilus oryzae and Tribolium confusum on stored wheat. Adults of the two species were exposed on wheat treated with diatomaceous earth at four dose rates: 0.25, 0.5, 1 and 1.5 g/kg of wheat, respectively. For each dose rate, the treated wheat was placed at 22°C, 25°C, 27°C, 30°C and 32°C. Dead adults were counted after 24 and 48 h, 7 and 14 d of exposure. After the 14-d interval, the live adults were removed and placed for 7 d in untreated wheat (in the case of S. oryzae) or untreated flour (in the case of T. confusum), and the production of F1 was recorded. For both species, dose rate, temperature and exposure interval significantly affected mortality (P<0.001). Mortality was higher at longer exposure intervals. The efficacy of SilicoSec against S. oryzae increased with temperature, but for T. confusum mortality was lower at 32°C, compared to 30°C, for 24 and 48 h exposure intervals. Tribolium confusum proved less susceptible to SilicoSec than S. oryzae. In general, the rates of 1 and 1.5 g/kg of wheat provided a satisfactory level of protection against the two species examined. For S. oryzae, F1 emerged only at 22°C, in wheat treated with 0.25 or 0.5 g/kg. However, for T. confusum, F1 were recorded at 22°C for 0.5 g/kg and at 22°C, 25°C, 27°C and 30°C for 0.25 g/kg.  相似文献

Kinetic parameters for the thermal inactivation of Geobacillus stearothermophilus ATCC 7953 and Bacillus flexus 1316 spores were determined for temperatures ranging from 100 to 130 °C and 100 to 125 °C, respectively. Ringer's solution (pH = 7.1) was used as the heating medium. A batch-heating system, in which the samples are kept in small tubes that are heated with steam, and a continuous-heating system, which is based upon a heat exchanger and enables high temperature short time heating, were used. Experiments were conducted in both systems and the heat resistances of the two species were determined. Additionally, a comparison of the two heating systems was carried out. The reaction rate constant at the reference temperature, kref, the activation energy, Ea, the D-value and the z-value were calculated for the two species. The D-values at 121 °C for G. stearothermophilus and for B. flexus in the batch-heating system were found to be 42 and 4.2 s, respectively. The z-values were calculated as 13 K for G. stearothermophilus and 16 K for B. flexus in the batch-heating system. The results from both systems differed significantly, wherein the continuous-heating system had been more lethal than the batch-heating system.  相似文献

In this study, the effects of cold storage, rearing temperature, parasitoid age, and irradiation on the performance of the egg parasitoid Trichogramma evanescens were investigated. Pupae of T. evanescens can be stored at 4 °C for up to 3 weeks without much loss of performance. The longevity and walking speed of adults emerging from chilled pupae significantly decreased after longer storage periods. The F1 generation of adults which emerged from pupae stored up to 3 weeks was able to parasitize as well as the control. The parasitization rate was similar at 24, 27, and 30 °C, but significantly decreased at 33 and 36 °C. Although T. evanescens developed to the pupal stage at 36 °C, no adult emergence was observed at this temperature. Developmental periods were longer at 24 °C than at higher temperatures. The optimum age for T. evanescens to successfully parasitize host eggs ranged from 24 to 90 h. The parasitization frequency of the 56-78 h aged females was higher than for the other age groups. The daily egg laying pattern of female T. evanescens adults was similar when they were reared on Ephestia kuehniella or Plodia interpunctella eggs. Gamma- or ultraviolet-irradiated and unirradiated host eggs were equally preferred by adult females.  相似文献

A crude extract was prepared from the fruiting body of Lepista flaccida, an edible mushroom and endoglucanase activity of the extract was increased 14-fold with ammonium sulphate precipitation. Maximum enzyme activity was seen at pH 4.0 and 50 °C when carboxymethylcellulose was used as a substrate. K0.5 and Vmax values of the partially purified endoglucanase were 7.7 mg/ml and 25 ± 0.9 U/mg protein, respectively. The enzyme was quite stable over a broad range of pH (2.0–9.0) at 4 °C. When it was incubated at temperatures between 20 °C and 60 °C for 12 h, it conserved much of its original activity (over 40%). The activity of the enzyme increased by 234 ± 3.6% in the presence of 1 mM Mn2+. The endoglucanase was inhibited by EDTA, PMSF, β-ME and DDT. In conclusion, pH and thermal stability of the L. flaccida endoglucanase could make it useful for industrial purposes.  相似文献

The aim of this study was to determine the effect of a natural carotenoid mixture (Capsantal FS-30-NT), containing capsanthin and capsorubin, on growth and mycotoxin production of ochratoxin A-producing A. ochraceus, A. westerdijkiae, and A. tubingensis isolates.One isolate of each species, previously isolated from paprika or chilli, was inoculated on Czapek Yeast extract Agar (CYA) medium supplemented with different amounts of capsantal (0 to 1%) and incubated at 10, 15 and 25 °C for 21 days. Growth rates and lag phases were obtained, and OTA production was determined at 7, 14 and 21 days.The taxonomically related A. ochraceus and A. westerdijkiae showed the same behavior at 15 °C, but A. ochraceus was able to grow at 10 °C and had higher growth rates at 25 °C. A. tubingensis had the highest growth rates and lowest OTA production capacity of the assayed isolates, and it was not able to grow at 10 °C.Capsantal addition resulted in increased lag phases at 15 °C for all the strains, while growth rates remained rather constant. At 25 °C capsantal reduced growth rates, with rather constant lag phases. However, the effect of capsantal on OTA production was inconclusive, because it depended on temperature or time, and mostly was not significant.Low temperature has been a crucial factor in OTA production, regardless of the capsantal concentration tested, especially for A. tubingensis and A. westerdijkiae. Industrial storage temperature for paprika and chilli is approximately 10 °C. If this temperature is maintained, mould growth and OTA production should be reduced.  相似文献

Cronobacter sakazakii and Salmonella species have been associated with human illnesses from consumption of contaminated nonfat dry milk (NDM), a key ingredient in powdered infant formula and many other foods. Cronobacter sakazakii and Salmonella spp. can survive the spray-drying process if milk is contaminated after pasteurization, and the dried product can be contaminated from environmental sources. Compared with conventional heating, radio-frequency dielectric heating (RFDH) is a faster and more uniform process for heating low-moisture foods. The objective of this study was to design an RFDH process to achieve target destruction (log reductions) of C. sakazakii and Salmonella spp. The thermal destruction (decimal reduction time; D-value) of C. sakazakii and Salmonella spp. in NDM (high-heat, HH; and low-heat, LH) was determined at 75, 80, 85, or 90°C using a thermal-death-time (TDT) disk method, and the z-values (the temperature increase required to obtain a decimal reduction of the D-value) were calculated. Time and temperature requirements to achieve specific destruction of the pathogens were calculated from the thermal destruction parameters, and the efficacy of the RFDH process was validated by heating NDM using RFDH to achieve the target temperatures and holding the product in a convection oven for the required period. Linear regression was used to determine the D-values and z-values. The D-values of C. sakazakii in HH- and LH-NDM were 24.86 and 23.0 min at 75°C, 13.75 and 7.52 min at 80°C, 8.0 and 6.03 min at 85°C, and 5.57 and 5.37 min at 90°C, respectively. The D-values of Salmonella spp. in HH- and LH-NDM were 23.02 and 24.94 min at 75°C, 10.45 and 12.54 min at 80°C, 8.63 and 8.68 min at 85°C, and 5.82 and 4.55 min at 90°C, respectively. The predicted and observed destruction of C. sakazakii and Salmonella spp. were in agreement, indicating that the behavior of the organisms was similar regardless of the heating system (conventional vs. RFDH). Radio-frequency dielectric heating can be used as a faster and more uniform heating method for NDM to achieve target temperatures for a postprocess lethality treatment of NDM before packaging.  相似文献

The combined effects of high pressure processing (HPP) and pH on the glycolytic and proteolytic activities of Lactococcus lactis subsp. lactis, a commonly used cheese starter culture and the outgrowth of spoilage yeasts of Candida species were investigated in a fermented milk test system. To prepare the test system, L. lactis subsp. lactis C10 was grown in UHT skim milk to a final pH of 4.30 and then additional samples for treatment were prepared by dilution of fermented milk with UHT skim milk to pH levels of 5.20 and 6.50. These milk samples (pH 4.30, 5.20 and 6.50) with or without an added mixture of two yeast cultures, Candida zeylanoides and Candida lipolytica (105 CFU mL−1 of each species), were treated at 300 and 600 MPa (≤20 °C, 5 min) and stored at 4 °C for up to 8 weeks. Continuing acidification by starter cultures, as monitored during storage, was substantially reduced in the milk pressurised at pH 5.20 where the initial titratable acidity (TA) of 0.40% increased by only 0.05% (600 MPa) and 0.10% (300 MPa) at week 8, compared to an increase of 0.30% in untreated controls. No substantial differences were observed in pH or TA between pressure-treated and untreated milk samples at pH 4.30 or 6.50. The rate of proteolysis in milk samples at pH values of 5.20 and 6.50 during storage was significantly reduced by treatment at 600 MPa. Treatment at 600 MPa also reduced the viable counts of both Candida yeast species to below the detection limit (1 CFU mL−1) at all pH levels for the entire storage period. However, samples treated at 300 MPa showed recovery of C. lipolytica from week 3 onwards, reaching 106–107 CFU mL−1 by week 8. In contrast, C. zeylanoides did not show any recovery in any of the pressure-treated samples during storage.  相似文献

This study determined the starvation tolerance of Tribolium castaneum (Herbst), Rhyzopertha dominica (F.) and Sitophilus oryzae (L.) in terms of both adult survival and reproduction, the impact of starvation on reproduction not having been studied before. Experiments were conducted at 30 °C and 55% or 70% r.h. using a laboratory strain and a field strain of each species. The number of progeny was a better indicator of the impact of starvation on a species than adult survival. Tribolium castaneum was the most tolerant species, requiring up to 35 d starvation before no progeny were produced. Rhyzopertha dominica and S. oryzae required up to 8 d starvation before no progeny were produced. The results suggest that hygiene will have a greater impact on populations of S. oryzae and R. dominica than T. castaneum.  相似文献

Illnesses from Escherichia coli O157:H7, Listeria monocytogenes, and Salmonella have been associated with the consumption of numerous produce items. Little is known about the effect of consumer handling practices on the fate of these pathogens on celery. The objective of this study was to determine pathogen behavior at different temperatures under different storage conditions. Commercial fresh-cut celery was inoculated at ca. 3 log CFU/g onto either freshly cut or outer uncut surfaces and stored in either sealed polyethylene bags or closed containers. Samples were enumerated following storage for 0, 1, 3, 5, and 7 days when held at 4 °C or 12 °C, and after 0, 8, and 17 h, and 1, and 2 days when held at 22 °C. At 4 °C, all populations declined by 0.5–1.0 log CFU/g over 7 days. At 12 °C, E. coli O157:H7 and Salmonella populations did not change, while L. monocytogenes populations increased by ca. 0.5 log CFU/g over 7 days. At 22 °C, E. coli O157:H7, Salmonella, and L. monocytogenes populations increased by ca. 1, 2, or 0.3 log CFU/g, respectively, with the majority of growth occurring during the first 17 h. On occasion, populations on cut surfaces were significantly higher than those on uncut surfaces. Results indicate that populations are reduced under refrigeration, but survive and may grow at elevated temperatures.  相似文献

Pseudomonas of pallet-packaged raw pork grown at 0, 5, 10, 15, 20 and 25 °C has been studied in this paper. The modified Gompertz, Baranyi and Huang models were used for data fitting. Statistical criteria such as residual sum of squares, mean square error, Akaike's information criterion, and pseudo-R2 were used to evaluate model performance. Results showed that there was an apparent decline in Pseudomonas growth at initial-storage phase at low temperatures. The modified Gompertz model outperformed the others at 5, 15, and 20 °C, while Baranyi model was appropriate for 0 and 25 °C. The Huang model was optimal at 10 °C. No single model can give a consistently preferable goodness-of-fit for all growth data. The Gompertz model, with the smallest average values of RSS, AIC, MSE and the biggest pseudo-R2 at all temperatures, is the most appropriate model to describe the growth of Pseudomonas of raw pork under pallet packaging.  相似文献

Sulfuryl fluoride (SF) is a fumigant used for the control of stored-product pest insects and is a feasible alternative to some uses of methyl bromide. SF was first registered in Germany in 2004 for disinfestation of emptied flourmills, emptied food storages and dried fruit. One- to 4-day-old eggs, larvae and pupae of the warehouse moth, Ephestia elutella (Hübner) (Lepidoptera) were investigated for their susceptibility to SF under different conditions. Each life stage was exposed for 18 h, 24 h or 48 h, to 11.6 g/m3 or 21.3 g/m3 at 15 °C, 20 °C or 25 °C and 65% relative humidity. A Fourier transform infrared (FTIR) spectrometer was used to determine the concentrations of SF. Within 18 h of exposure, all larvae and pupae of E. elutella died at 11.6 g/m3 at all three temperatures. The 1 and 2- day-old eggs were generally more susceptible, whereas the 3 and 4-day-old eggs were more tolerant to the SF treatment. All eggs of all ages were controlled within 48 h of exposure to the concentration of 21.3 ± 1.3 g/m3 at temperatures of 20 °C and 25 °C.  相似文献

The physicochemical properties, fatty acid, tocopherol, thermal properties, 1H NMR, FTIR and profiles of non-conventional oil extracted from Citrullus colocynthis (L.) Schrad seeds were evaluated and compared with conventional sunflower seed oil. In addition, the antioxidant properties of C. colocynthis seed oil were also evaluated. The oil content of the C. colocynthis seeds was 23.16%. The main fatty acids in the oil were linoleic acid (66.73%) followed by oleic acid (14.78%), palmitic acid (9.74%), and stearic acid (7.37%). The tocopherol content was 121.85 mg/100 g with γ-tocopherol as the major one (95.49%). The thermogravimetric analysis showed that the oil was thermally stable up to 286.57 °C, and then began to decompose in four stages namely at 377.4 °C, 408.4 °C, 434.9 °C and 559.2 °C. The present study showed that this non-conventional C. colocynthis seed oil can be used for food and non-food applications to supplement or replace some of the conventional oils.  相似文献

Heat resistance of Neosartorya fischeri was studied in three different juices (apple, pineapple and papaya). The optimum heat activation temperature and time for the ascospores of the N. fischeri (growth for 30 days at 30 °C) was 85 °C for 10 min. Of the three juices tested, apple juice exhibited maximum 1/k values at 80, 85 and 90 °C (208.3, 30.1 and 2.0 min, respectively). The 1/k values for papaya juice (129.9, 19.0 and 1.9 min) and pineapple juice (73.5, 13.2 and 1.5 min) decreased with acidity and °Brix/acidity (ratio) level. The Z* values for apple, papaya and pineapple juices were 5, 5.5 and 5.9 °C, respectively. The sterilization F values (4-log reduction) for apple, pineapple and papaya juices were 56.3, 38.0 and 7.2 s, respectively. Considering the thermal treatments commercially applied to pineapple (96 °C/30 s) and apple juices (95 °C/30 s), it is concluded that such treatments will not guarantee that less than 1 ascospore in each set of 103 packs survive. Only the treatment applied to papaya juice (100 °C/30 s) will be sufficient because the F value is less than 30 s.  相似文献

This work aimed to study the factors influencing the growth of Carnobacterium divergens strain M35 and its ability to produce a new class IIa bacteriocin (divergicin M35) in various synthetic media and in medium supplemented with snow crab hepatopancreas (SCH), a natural-grade by-product of crustacean processing. C. divergens M35 growth and bacteriocin production in SCH-supplemented medium was evaluated at different temperatures in batch fermentations and under controlled pH. C. divergens M35 was shown to grow well in SCH medium at tested temperatures between 4 and 30 °C, except at 37 °C. Maximum divergicin M35 production was obtained after 10 h of growth in SCH medium at 25 and 30 °C with a total activity of 3.7 × 104 AU mL−1. Less growth was observed at 37 °C, for which a total bacteriocin activity of only 256 AU mL−1 was obtained. The production of divergicin M35 was greatly influenced by the medium composition, especially by the type of added carbon source. The best production of divergicin M35 in SCH medium was observed at a controlled pH of 7.0. This study describes the optimal parameters for the growth of C. divergens M35 and production of divergicin M35 and demonstrates the effectiveness of the marine by-product as a medium supplement for this culture.  相似文献
