为得到超声辅助提取芥菜总黄酮的最佳工艺,并评价其脂质抗氧化活性,在超声时间、液料比、乙醇浓度和超声温度4个单因素试验的基础上,对影响芥菜总黄酮提取率的工艺因素进行Box-Behnken设计并进行响应面优化,通过芥菜总黄酮对大豆油和猪油的作用来评价其脂质抗氧化活性。结果表明最佳的工艺条件为:超声时间25 min,液料比30∶1(mL/g),乙醇浓度76%和超声温度71℃,总黄酮提取率为26.82 mg/g,与理论最佳工艺的预测值(26.94 mg/g)相比,其相对误差为0.45%,说明该二次多项式回归模型准确率高、可靠性强,可用于芥菜总黄酮超声辅助提取工艺的优化与预测。芥菜总黄酮对大豆油和猪油均有抗氧化能力,并呈量效关系,说明芥菜总黄酮有一定的抗脂质氧化能力,可添加到油脂产品中以提高产品的货架期。  相似文献

研究蓖麻叶与花总黄酮的提取方法和含量测定.用亚硝酸钠-硝酸铝-氢氧化钠比色法测定总黄酮含量,比较索氏提取法和微波辅助提取法提取蓖麻叶与花中总黄酮的效果.结果2种提取方法中微波法提取效率最高,结果重现性好,并测得叶与花中总黄酮含量分别为2.0411%和1.8655%.总黄酮在蓖麻叶中的含量明显高于花中的含量.  相似文献

通过对槐米总黄酮提取,考察体积分数、提取时间、提取温度、料液比四因素对槐米总黄酮提取率的影响.用RAS软件程序对试验数据进行二次响应面分析,对3项工艺参数进行优化,得出槐米总黄酮提取的二次回归方程;通过分析得出槐米总黄酮提取的最佳条件为:60%乙醇(体积分数),提取2.2 h,料液比(g:mL)1:17.9,温度80.0℃.在此条件下测得的总黄酮提取率为18.46%.  相似文献

为了研究小米辣的绿色泡制方法,本文用中药提取液对小米辣进行护色.采用超声提取法提取中药,以总黄酮含量做指标,通过正交试验对总黄酮的提取工艺进行优化.结果显示:护色液总黄酮最佳提取工艺条件为:70%乙醇,提取温度80℃,固液比为20∶1,提取时问30 min,提取率达5.21%,护色效果最好.  相似文献

以广佛手为原料,用复合酶法(纤维素酶量:果胶酶量=3:1,质量比)提取总黄酮.通过单因素试验和L9(34)正交试验对佛手总黄酮的提取工艺进行优化,确定佛手总黄酮提取的最佳工艺务件:酶用量1.0%(E/S)、酶解时间120 min、酶解温度45℃、pH4.5,此条件下的佛手总黄酮得率为4.093%.  相似文献

目的 比较不同方法提取苦荞种子总黄酮的效果.方法 分别用乙醇回流提取法、微波辅助提取法、超声波提取法、碱水浸提法及热水浸提法提取苦荞种子总黄酮,用高效液相色谱法测定总黄酮含量,计算提取产率.结果 乙醇回流提取法提取总黄酮产率最高,微波和超声波提取法次之,碱水和热水浸提法最低.结论 微波提取法具有明显的高效性,适于大量提取苦荞种子总黄酮.  相似文献

研究了茴香中黄酮类化合物的提取,采用溶剂法,用正交实验法分析考察乙醇浓度、提取时间、提取温度及料液比等因素对黄酮提取率的影响,用NaNO2-Al(NO3)3-NaOH比色法测定茴香中总黄酮含量.结果表明:用80%乙醇水溶液为溶剂,料液比为1:20,在75℃回流2h的工艺务件下,总黄酮提取量较高,提取率为2.69%.  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
