The purpose of this study was to increase the water solubility and potential bioavailability of quercetin by encapsulation in whey protein isolate (WPI) based on a green, efficient pH-driven method. According to the results, the water solubility of quercetin increased by 346.9: times after loading into WPI nanoparticles. When the initial quercetin concentration was 0.25 mg mL−1 and WPI was 2% w/v, the encapsulation efficiency reached 94.1%, the Z-average diameter was 36.63 nm, and the zeta potential was −36.4 mV at pH 7.0. The fluorescence spectroscopy assay suggested the molecular complexation of quercetin and WPI at pH 12.0. X-ray diffraction assay indicated the enclosure of amorphous quercetin in WPI. Correspondingly, the bioaccessibility increased from 2.76% to 31.23% and the Caco-2 cell monolayer uptake increased from 0% to 2.12% after nanoencapsulation. This work confirmed that the pH-driven method is an effective approach to prepare WPI–quercetin nanocapsules to improve physical and potentially biological properties of quercetin.  相似文献

Abstract

Different drying methods (spray drying (SD), vacuum drying (VD), microwave vacuum drying (MVD), and infrared vacuum drying (IFVD)) were applied in order to compare the hygroscopicity behavior of chicken powders. The hygroscopicity curves and glass transition temperature were used to evaluate the influence of ambient humidity and temperature on moisture absorption of powders. The results showed that the chicken powder dried by MVD had the lowest moisture absorption, followed by IFVD, VD, and SD. The hygroscopicity of SD chicken powders was different from other three kinds of chicken powders due to the physical properties of particles and the changes of protein secondary structure as detected by the Fourier transform-infrared spectrometer. For the three vacuum drying methods, the difference of protein secondary structure was the main reason of differences in hygroscopicity. Although MVD chicken powders were slightly inferior to SD chicken powders in taste, MVD chicken powders were the best in terms of smell and color as suggested by instrumental sensory parameter evaluations. It was found that MVD had a positive effect on reducing moisture absorption and maintaining sensory quality of chicken powders.  相似文献

Incorporating high level of potato flour into wheat flour enhances nutritional values of bread but induces a series of problems that lead to the decline of the bread quality. To overcome the barrier, wheat gluten and carboxymethylcellulose (CMC) were added into potato–wheat composite flour to improve dough machinability and bread quality. The rheological properties, thermo-mechanical properties and microstructures of dough were investigated. The results showed that the interaction between gluten and CMC mitigated the discontinuity of gluten matrix and gluten protein aggregation caused by the addition of potato flour, which yielded a more branched and compact gluten network. The compact three-dimensional viscoelastic structure induced improvements of gas retention capacity and dough stability, making it mimic the machinability properties of wheat flour dough. Bread qualities were apparently improved with the combined use of 4% gluten and 6% CMC, of which specific volume increased by 42.86%, and simultaneously, hardness reduced by 75.93%.  相似文献

In this work a multicommuted flow system employing copper–4,4′- dipyridyl coordination compound as the solid-phase reagent for the spectrophotometric determination of reducing sugar was developed. The coordination compound was synthesized through a reaction of the 4,4′-dipyridyl and copper (II) nitrate, under hydrothermal conditions. The complex was characterized by infrared spectroscopy (FTIR), power X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive spectrometer (EDS) and thermogravimetric analysis (TGA). Based on the characterization, a multicommuted spectrophotometric procedure for the determination of reducing sugar using copper (II) complex as solid reagent is proposed. The proposed method was based on the redox reaction between a monosaccharide, such as fructose and glucose (reducing sugar) and Cu(II). This reaction, mediated in an alkaline medium, produces a yellow compound that can be determined by absorption electronic spectroscopy (λABS = 420 nm). Under optimum experimental conditions, a linear response ranging from 1.0 to 20.0 g L−1 (R = 0.9978 and n = 5), a detection (3σ criterion) and quantification (10σ criterion) limit estimated at 0.23 and 0.75 g L−1, respectively, a standard deviation relative of 4.7% (n = 7), for a reference solution of 10.0 g L−1 reducing sugar, and a sampling rate of 75 determinations per hour were achieved. The proposed system was applied to the determination of reducing sugars in coconut water and juices. The analysis of ten samples and the application of the t-test to the results found, and those obtained using reference procedures (AOAC), provided no significant differences at a 95% confidence level. This system enabled the analysis of reducing sugar with ease and simplicity, providing a significant economy of the solid reagent (600 μg per determination) and reducing effluent generation.  相似文献

Opening catalytically active sites in metal organic frameworks is an issue of fundamental importance for the development of effective and efficient catalysts. In this work, we first reported two metal metalloporphyrin–organic frameworks (MMPFs) with unoccupied pyridine groups as base catalysts. The reaction of Mn(II) and Co(II) with 5,10,15,20-tetrapyridylporphyrin produces two different metal metalloporphyrin–organic frameworks, {[(MnTPyP)]·H2O}n (MMPF-Mn) and [(CoTPyP)]n (MMPF-Co) (TPyP = 5,10,15,20-tetrapyridylporphyrin) under hydrothermal conditions. These two MMPFs have been fully characterized by single-crystal X-ray diffraction, powder XRD, elemental analysis and thermogravimetry (TG). MMPF-Mn displays a 3D network with a nbo topology, large and open hexagonal channels, MMPF-Co reveals a 1D single zigzag chain architecture. Interestingly, both MMPFs have a high thermal stability and opening basic pyridine group, which have been tested for the base catalyzed Knoevenagel condensation reaction. The catalytic study has demonstrated that MMPF-Mn catalysts having exposed pyridine group within 1D channel displayed an excellent performance for Knoevenagel condensation reaction. When MMPF-Mn was recycled four times, its catalytic activity remained with an inconspicuous decrease. We attribute MMPF-Mn showing a better performance than MMPF-Co to its active sites being aligned in extra-large cavity with an interior diameter of 20 Å.  相似文献

Through a mechanism called quorum sensing, bacteria are able to express specific genes in response to population density. Cell-to-cell communication in bacteria is mediated by signal molecules such as acylated homoserine lactones (AHLs). This work aimed to detect AHL production in Gram-negative psychrotrophic bacteria isolated from raw milk. A total of 84.9% of the bacteria were identified as AHL producers eliciting a diversity of responses in the AHL-monitor systems. These results demonstrate that AHL-production is common among psychrotrophic bacteria isolated from milk, and indicate that quorum sensing may play an important role in the spoilage of this product.  相似文献

Metallic silver has been prepared by direct hydrogen reduction of Ag2S in basic slurry under hydrothermal conditions. The XRD pattern and SEM micrographs of the reaction product are given. Metallic silver was predominant in the reduction product obtained by using ZnO as additive and PdCl2 as catalyst at 220 °C, pH 12.0 and 2–3 MPa of partial hydrogen pressure for 4 h.  相似文献

Cucurbitaceae family seeds are mostly discarded as agro-industrial wastes. Gurum (Citrullus lanatus var. colocynthoide) is an underutilized wild cucurbit plant, closely related to desert watermelon, which is grown abundantly in some African countries. Gurum seeds can play a significant role in health and nutrition due to their high oil content. This review describes the nutritional composition of gurum seeds and their oil profile. Gurum seeds are a good source of oil (27–35.5%), fiber (26–31%), crude protein (15–18%), and carbohydrates (14–17%). Gurum seeds oil is extracted by supercritical CO2 (SFE), screw press, and solvent extraction techniques. The gurum seeds oil is composed of unsaturated fatty acids with a high proportion of linoleic acid (C18:2) and oleic acid (C18:1). Gurum seeds oil contains various bioactive compounds, such as tocopherols, phytosterols, and polyphenols. It is reported that solvent extraction gives a higher yield than the screw press and SFE, but the SFE is preferred due to safety issues. More studies are required for producing better quality gurum seeds oil by using novel extraction techniques that can increase oil yield.  相似文献

Aspartate ammonia lyase (Asp) is one of three types of ammonia lyases specific for aspartate or its derivatives as substrates, which catalyzes the reversible reaction of l-aspartate to yield fumarate and ammonia. In this paper, the catalytic mechanism of Asp has been studied by using combined quantum-mechanical/molecular-mechanical (QM/MM) approach. The calculation results indicate that the overall reaction only contains two elementary steps. The first step is the abstraction of Cβ proton of l-aspartate by Ser318, which is calculated to be rate limiting. The second step is the cleavage of CαN bond of l-aspartate to form fumarate and ammonia. Ser318 functions as the catalytic base, whereas His188 is a dispensable residue, but its protonation state can influence the active site structure and the existing form of leaving amino group, thereby influences the activity of the enzyme, which can well explain the pH dependence of enzymatic activity. Mutation of His188 to Ala only changes the active site structure and slightly elongates the distance of Cβ proton of substrate with Ser318, causing the enzyme to remain significant but reduced activity.  相似文献

Metamaterial absorbers have been widely studied and continuously concerned owing to their excellent resonance features of ultra-thin thickness, light-weight, and high absorbance. Their applications, however, are typically restricted by the intrinsic dispersion of materials and strong resonant features of patterned arrays (mainly referring to narrow absorption bandwidth). It is, therefore essential to reassert the principles of building broadband metamaterial absorbers (BMAs). Herein, the research progress of BMAs from principles, design strategies, tunable properties to functional applications are comprehensively and deeply summarized. Physical principles behind broadband absorption are briefly discussed, typical design strategies in realizing broadband absorption are further emphasized, such as top-down lithography, bottom-up self-assembly, and emerging 3D printing technology. Diversified active components choices, including optical response, temperature response, electrical response, magnetic response, mechanical response, and multi-parameter responses, are reviewed in achieving dynamically tuned broadband absorption. Following this, the achievements of various interdisciplinary applications for BMAs in energy-harvesting, photodetectors, radar-IR dual stealth, bolometers, noise absorbing, imaging, and fabric wearable are summarized. Finally, the challenges and perspectives for future development of BMAs are discussed.  相似文献
