A 40‐wk experiment was conducted using Hy‐Line W‐36 laying hens (19‐wk old) to investigate the impact of feeding cholecalciferol‐enriched diets on egg yolk quality. Feeds were enriched with 4 cholecalciferol levels, 9700 (diet 2), 17200 (diet 3), 24700 (diet 4), and 102200 (diet 5) IU/kg feed. The control (diet 1) contained 2200 IU cholecalciferol/kg feed. Eggs from each replicate group of enriched diets were collected daily and the yolks were pooled into 2‐d period during the first 2 wk. During weeks 3, 4, 6, 8, 12, 16, 20, 24, 28, 32, 36, and 40, pooled samples were generated by daily collection of 3 consecutive days of egg production. The cholecalciferol content of egg yolk from the enriched diets increased rapidly during the first 3 wk. The peak cholecalciferol concentrations in egg yolk that occurred at week 3 were 865, 1641, 2411, and 34815 IU/100 g egg yolk (wet basis) from diet 2 to 5. The average cholecalciferol concentration in yolk during weeks 3 to 40 and the deposition rate of cholecalciferol during the first 3 wk were both linearly increased with the dietary cholecalciferol level when the feed contained no more than 24700 IU/kg cholecalciferol. Egg yolk lipid profile (total lipid content, fatty acid composition, phospholipid composition, and unsaponifiables), physical and functional properties (yolk viscosity and emulsifying property), and sensory quality of hard‐boiled egg yolk were not affected by the cholecalciferol enrichment in the feed. Practical Application: A linear dose‐response relationship between dietary vitamin D3 level and egg yolk vitamin D3 content was established at relatively low enrichment levels. Such relationship can be used to formulate feed to achieve a target egg vitamin D level. High vitamin D yolk showed no difference from the conventional yolk in other compositional, functional, and sensory properties. 相似文献

In this pilot study, ethosomes and transethosomes were investigated as potential delivery systems for cholecalciferol (vitamin D3), whose deficiency has been correlated to many disorders such as dermatological diseases, systemic infections, cancer and sarcopenia. A formulative study on the influence of pharmaceutically acceptable ionic and non-ionic surfactants allowed the preparation of different transethosomes. In vitro cytotoxicity was evaluated in different cell types representative of epithelial, connective and muscle tissue. Then, the selected nanocarriers were further investigated at light and transmission electron microscopy to evaluate their uptake and intracellular fate. Both ethosomes and transethosomes proven to have physicochemical properties optimal for transdermal penetration and efficient vitamin D3 loading; moreover, nanocarriers were easily internalized by all cell types, although they followed distinct intracellular fates: ethosomes persisted for long times inside the cytoplasm, without inducing subcellular alteration, while transethosomes underwent rapid degradation giving rise to an intracellular accumulation of lipids. These basic results provide a solid scientific background to in vivo investigations aimed at exploring the efficacy of vitamin D3 transdermal administration in different experimental and pathological conditions. 相似文献

Morphological, molecular, and physiological effects of vitamin D on skeletal muscle have been analyzed both in animals and humans. Vitamin D may be a potential therapeutic for increasing muscle mass and function. The presence of vitamin D receptors in skeletal muscle cells is already established. However, there is still need for more evidence about the effect of vitamin D on muscle. Some studies have associated vitamin D and skeletal muscle in chronic kidney disease (CKD) patients; most of these studies enrolled hemodialysis patients. FGF-23 and Klotho were recently described in mineral and bone disorders in CKD, resulting in reductions in calcitriol levels. Therefore, both Klotho and FGF-23 may play a role in muscle loss in CKD, which is related to morbidity and mortality risk. Therefore, this article presents a narrative review, aiming to discuss the available information associating skeletal muscle and vitamin D, highlighting the results in CKD and dialysis patients. 相似文献

Vitamin D3, known to regulate bone homeostasis, has recently been shown to have many pleiotropic effects in different tissues and organs due to the presence of its receptor in a wide range of cells. Our previous study demonstrated that vitamin D3 was able to increase the wound healing respect to the control sample, 24 h after cutting, without however leading to a complete repair. The aim of the study was to combine vitamin D3 with silver nanoparticles to possibly enable a faster reparative effect. The results showed that this association was capable of inducing a complete wound healing only after 18 h. Moreover, a treatment of vitamin D3 + silver nanoparticles yielded a small percentage of keratinocytes vimentin-positive, suggesting the possibility that the treatment was responsible for epithelial to mesenchymal transition of the cells, facilitating wound healing repair. Since vitamin D3 acts via sphingolipid metabolism, we studied the expression of gene encoding for the metabolic enzymes and protein level. We found an increase in neutral sphingomyelinase without involvement of neutral ceramidase or sphingosine kinase2. In support, an increase in ceramide level was identified by Ultrafast Liquid Chromatography–Tandem Mass Spectrometry, suggesting a possible involvement of ceramides in wound healing process. 相似文献

A novel simple method to detect vitamins in cosmetic products by gas chromatography-mass spectrometry (GC-MS) has been developed. Three vitamins (panthenol, cholecalciferol and tocopherol) were used for this study. Vitamins were prepared by dissolving in tetrahydrofuran (ThF), and silylated with bis-trimethylsilyltri-fluoroacetamide- trichloromethylsilane (BSTFA). Silylated vitamins were separated on a fused-silica capillary column coated with DB-5. The identification of each vitamin was accomplished by retention time and mass spectrum library search with a computer, and the quantitation was made in the selected-ion monitoring (SIM) mode of GC-MS. SIM mode had given sensitivity to determine 50 pg of panthenol, 285 pg of cholecalciferol and 130 pg of tocopherol. Linearity was maintained over the range 0.005-0.20% for each vitamin. Each cosmetic product (i.e. hair tonic and lotion) was found to contain amounts of the vitamins. This method was sensitive and gave 77.5-99.9% recovery of each vitamin from these cosmetic products. From these results, we concluded that silylation with BSTFA followed by GC-MS analysis allows the simple, convenient and exact determination of panthenol, cholecalciferol and tocopherol. 相似文献

There is emerging interest in linking vitamin D status to physiological health and disease states in the dog, as evidenced by the recent increase in publications in this area. This research has most likely been spurred by the studies exploring vitamin D and disease in humans. However, there are important differences in vitamin D intake and metabolism between humans and dogs that should be accounted for. The understanding of basic vitamin D metabolism and the relationship between vitamin D intake and vitamin D status in dogs remains even more limited than current knowledge in humans. This review will summarize current knowledge of vitamin D in the dog, including metabolism and dietary recommendations. Emphasis is placed on the limitations to current knowledge. Studies investigating links between vitamin D and disease will be discussed in light of this knowledge. Suggestions for future research, including the development of reference ranges to define blood vitamin D sufficiency, are provided. 相似文献
