Fungi are distributed worldwide and can be found in various foods and feedstuffs from almost every part of the world. Mycotoxins are secondary metabolites produced by some fungal species and may impose food safety risks to human health. Among all mycotoxins, aflatoxins (AFs), ochratoxin A (OTA), trichothecenes, deoxynivalenol (DON and T‐2 toxin), zearalenone (ZEN), and fumonisins (FMN) have received much attention due to high frequency and severe health effects in humans and animals. Malaysia has heavy rainfall throughout the year, high temperatures (28 to 31 °C), and high relative humidity (70% to 80% during wet seasons). Stored crops under such conditions can easily be contaminated by mycotoxin‐producing fungi. The most important mycotoxins in Malaysian foods are AFs, OTA, DON, ZEN, and FMN that can be found in peanuts, cereal grains, cocoa beans, and spices. AFs have been reported to occur in several cereal grains, feeds, nuts, and nut products consumed in Malaysia. Spices, oilseeds, milk, eggs, and herbal medicines have been reported to be contaminated with AFs (lower than the Malaysian acceptable level of 35 ng/g for total AFs). OTA, a possible human carcinogen, was reported in cereal grains, nuts, and spices in Malaysian market. ZEN was detected in Malaysian rice, oat, barley, maize meal, and wheat at different levels. DON contamination, although at low levels, was reported in rice, maize, barley, oat, wheat, and wheat‐based products in Malaysia. FMN was reported in feed and some cereal grains consumed in Malaysia. Since some food commodities are more susceptible than others to fungal growth and mycotoxin contamination, more stringent prevention and control methods are required.  相似文献

为提高毕赤酵母重组菌产伏马毒素B1(fumonisin B1,FB1)羧酸酯酶的摇瓶发酵水平,以酶活力为指标,对发酵条件和培养基进行优化。通过单因素试验对发酵条件进行优化。通过Plackett-Burman试验筛选出关键培养基成分,再进行正交试验建立试验数据样本,最后建立误差反向传播神经网络模型进行预测并寻找最优发酵培养基组成。经优化得到的发酵条件:诱导温度28℃,初始p H 6.0,每24 h补加体积分数为1%的甲醇,诱导96 h;优化后发酵培养基组成:蛋白胨23 g/L、KH2PO444 g/L、PTM4 (Pichia trace minerals 4)微量元素溶液1.5 m L/L、K2SO47.15 g/L、Mg SO4·7H2O 5.85 g/L和酵母粉5 g/L。FB1羧酸酯酶酶活力达到402 U/m L以上,较优化前提高了1.19倍。优化后显著提高了重组菌株的产酶能力,研究结果为FB1羧酸酯酶发酵罐优化提供了基础数据。  相似文献

Based on fungal and fumonisin contamination of 870 freshly harvested samples, the quality of corn used by processing industries in the Northern region of Paraná State, Brazil (2003 and 2004 crop-year) was evaluated. Sampling was carried out for each crop at two points in the production chain, i.e. at reception by the processors and at the pre-drying step. Corn samples were more frequently contaminated with Fusarium sp. (100%) and Penicillium sp. (84.1–95.3%) than Aspergillus sp. (5.6–19.8%). Fumonisin B1 (FB1) was detected in all samples from the two points in both crop-years. FB1 levels ranged 0.02–11.83 µg g?1 in the reception and 0.02–10.98 µg g?1 in the pre-drying samples of the 2003 crop. Samples from the 2004 crop showed FB1 levels ranging 0.03–12.04 µg g?1 in the reception and 0.06–7.74 µg g?1 in the pre-drying samples. FB2 levels ranged 0.02–5.25 µg g?1 in the reception and 0.01–7.89 µg g?1 in the pre-drying samples (2003 crop-year). In samples from the 2004 crop, FB2 levels ranged 0.02–6.12 µg g?1 in the reception and 0.05–3.47 µg g?1 in the pre-drying samples. Low fumonisin levels were detected in most corn samples used by processors in the Northern region of Paraná State, showing a decreasing trend in fumonisin contamination over the years.  相似文献

We analysed a total of 2258 grain samples over a 3-year period (2006–2008) from 93 storage centers in the principal maize cultivation area of Italy to establish the levels of fumonisin contamination. Fumonisin concentrations were measured using ELISA (RIDASCREEN) fumonisin test kits. Mean levels of contamination were remarkably high in each year, with the highest value in 2006 (10.9?mg/kg) and the lowest in 2008 (4.8?mg/kg). Similarly, for each year, variations were quite large: from <LOQ to 77.0?mg/kg, <?LOQ to 26.3, and 0.1 to 19.0?mg/kg, respectively, in 2006, 2007 and 2008. The highest fumonisin concentration was found in samples harvested in Veneto (11.1?mg/kg) and Friuli Venezia Giulia (8.9?mg/kg), while the lowest were recorded in Emilia Romagna and Piemonte (4.6?mg/kg).  相似文献

ABSTRACT

Fresh and frozen pamonhas, a native Brazilian food made with sweet corn (Zea mays L.), similar to the Mexican tamal, which has been incorporated into the country’s diet, were investigated for fumonisins (FBs: FB1 and FB2) contamination and product characteristics. The laboratory conditions of the applied fumonisins method with liquid chromatography and fluorescence detection were validated. Five out of the total samples showed FBs contamination above the national maximum level of 1000–1500 µg/kg. Despite this, in 59.6% (31) of the samples FB1 and in 23.1% (12) FB2 could be detected. The pamonhas characteristics did not show uniformity among the brands surveyed. The moisture content of 59.4% and water activity of 0.97 for both (fresh and frozen) samples showed to be suitable conditions for fungal growth. One sample had a quite high pH. It is essential to establish guidance criteria for pamonha production by the regulatory agencies.  相似文献

Finger millet (Eleusine coracana) is a subsistence crop grown in Sub-Saharan Africa and the Indian Sub-continent. Fusarium species occurring on this crop have not been reported. Approximately 13% of the Fusarium isolates recovered from finger millet growing at three different locations in eastern Uganda belong to Fusarium verticillioides, and could produce up to 18,600?µg/g of total fumonisins when cultured under laboratory conditions. These strains are all genetically unique, based on AFLP analyses, and form fertile perithecia when crossed with the standard mating type tester strains for this species. All but one of the strains is female-fertile and mating-type segregates 13:20 Mat-1:Mat-2. Three new sequences of the gene encoding translation elongation factor 1??α were found within the population. These results indicate a potential health risk for infants who consume finger millet gruel as a weaning food, and are consistent with the hypothesis that F. verticillioides originated in Africa and not in the Americas, despite its widespread association with maize grown almost anywhere worldwide.  相似文献

BACKGROUND: The genus Fusarium comprises a diverse group of fungi including several species that produce mycotoxins in food commodities. In the present study, a multiplex PCR was standardised for the group‐specific detection of fumonisin‐producing and trichothecene‐producing strains of Fusarium species. Primers for genus‐level recognition of Fusarium spp. were designed from the internal transcribed spacer regions 1 and 2 of rDNA. Primers for group‐specific detection were designed from the tri5 and tri6 genes involved in trichothecene biosynthesis and the fum1 and fum13 genes involved in fumonisin biosynthesis. RESULTS: Among the various genera and their strains tested, all the 85 confirmed Fusarium strains were positive for rDNA gene and the rest stayed negative. From among the Fusarium strains, 15 had amplification for trichothecene‐ and 20 for fumonisin‐encoding genes. All PCR positive trichothecene chemotypes of Fusarium species tested were positive for chemical analysis but in the case of fumonisins, of the 20 PCR positive cultures, only 13 showed positive for chemical analysis by HPTLC. CONCLUSION: The assay described here provided a rapid and reliable detection of trichothecene‐ and fumonisin‐producing Fusarium directly from natural food grains and the results were always comparable with a conventional HPTLC detection method. The quality and safety of maize (Zea mays L.) from different grain storage units (GSUs), located in the main producing region of Rondônia State, Northern Brazil, were evaluated. Maize grains (n = 76) stored in four GSUs were collected from July to November 2014 and evaluated for grain damages, humidity, fungi and fumonisins (FBs) content. The climate conditions data were also obtained from plant growing to storage periods. Regarding the moisture content and water activity (aw), these varied from 10.0% to 16.1% and 0.5 to 0.8, respectively. As expected, fungi spores were present in 94.8% of the samples, prevailing Fusarium genera, with a fungi colony maximum of 2.2 × 104 CFU g?1. Regarding FBs, 60.5% of the samples were contaminated, below Brazilian and United States maximum limits, but 9.2% had levels higher than the European legislation.  相似文献

Mycotoxins are secondary metabolites produced by moulds in food that are considered a substantial issue in the context of food safety, due to their acute and chronic toxic effects on animals and humans. Therefore, new accurate methods for their identification and quantification are constantly developed in order to increase the performance of extraction, improve the accuracy of identification and reduce the limit of detection. At the same time, several industrial practices have shown the ability to reduce the level of mycotoxin contamination in food. In particular, a decrease in the amount of mycotoxins could result from standard processes naturally used for food processing or by procedures strategically introduced during processing, with the specific aim of reducing the amount of mycotoxins. In this review, the current methods adopted for accurate analyses of mycotoxins in cereals (aflatoxins, ochratoxins, trichothecenes, fumonisins) are discussed. In addition, both conventional and innovative strategies adopted to obtain safer finished products from common cereals intended for human consumption will be explored and analysed. © 2018 Society of Chemical Industry  相似文献
