Theknowledge transfer problem in artificial intelligence consists of finding effective ways to elicit information from a human expert and represent it in a form suitable for use by an expert system. One approach to formalizing and guiding this knowledge transfer process for certain types of expert systems is to use psychometric scaling methods to analyze data on how the human expert compares or groups solutions. For example, Butler and Corter [1] obtained judgments of thesubstitutability of solutions from an expert, then analyzed the resulting data via techniques for fitting trees and extended trees [2]. The expert's interpretation of certain aspects of the solutions were directly encoded as production rules, allowing rapid prototyping. In this paper we consider the problem of combining information from multiple experts. We propose the use of three-way or individual differences multidimensional scaling, tree-fitting, and unfolding models to analyze two types of data obtainable from the multiple experts: judgments of the substitutability of pairs of solutions, and judgments of the appropriateness of specific solutions to specific problems. An application is described in which substitutability data were obtained from three experts and analyzed using the SINDSCAL program [3] for three-way multidimensional scaling [4].  相似文献

Evolutionarily elderly proteins commonly feature greater catalytic promiscuity. Cytochrome c is among the first set of proteins in evolution to have known prospects in electron transport and peroxidative properties. Here, we report that cyt c is also a proficient proton-transfer catalyst and enhances the Kemp elimination (KE; model reaction to show proton transfer catalytic property) by ∼750-fold on self-organized systems like micelles and vesicles. The self-organized systems mimic the mitochondrial environment in vitro for cyt c. Using an array of biophysical and biochemical mutational assays, both acid–base and redox mechanistic pathways have been explored. The histidine moiety close to hemin group (His18) is mainly responsible for proton abstraction to promote the concerted E2 pathway for KE catalysis when cyt c is in its oxidized form; this has also been confirmed by a H18A mutant of cyt c. However, the redox pathway is predominant under reducing conditions in the presence of dithiothreitol over the pH range 6–7.4. Interestingly, we found almost 750-fold enhanced KE catalysis by cyt c compared to aqueous buffer. Overall, in addition to providing mechanistic insights, the data reveal an unprecedented catalytic property of cyt c that could be of high importance in an evolutionary perspective considering its role in delineating the phylogenic tree and also towards generating programmable designer biocatalysts.  相似文献

针对核脉冲全谱数据采集技术中采用的深度加权滤波法在数据量少时平滑效果差、滞后大的缺点,设计了卡尔曼滤波算法替代传统滤波算法进行解谱的方案。滤波算法首先根据信号与噪声的状态空间模型,建立状态方程和测量方程,然后根据广义卡尔曼滤波对测量方程进行更新,最后根据现场的标准刻度井测量数据确定测量矩阵,对测量数据进行滤波。实测结果表明,该滤波算法相对于传统滤波算法,能够消除谱图的统计涨落,提高光滑度,遏制滞后,并保证滤波后曲线不失真。  相似文献

The stability and unfolding of an immunoglobulin (Ig) G bindingprotein based upon the B domain of protein A (SpAB) from Staphylococcusaureus were studied by substituting tryptophan residues at strategiclocations within each of the three a-helical regions (al-a3)of the domain. The role of the C-terminal helix, a3, was investigatedby generating two protein constructs, one corresponding to thecomplete SpAB, the other lacking a part of ct3; the Trp substitutionswere made in both one-and two-domain versions of each of theseconstructs. The fluorescence properties of each of the single-tryptophanmutants were studied in the native state and as a function ofguanidine-HCl-mediated unfolding, and their IgG binding activitieswere determined by a competitive enzyme-linked immunosorbentassay. The free energies of folding and of binding to IgG foreach mutant were compared with those for the native domains.The effect of each substitution upon the overall structure andupon the IgG binding interface was modelled by molecular graphicsand energy minimization. These studies indicate that (i) 3 contributesto the overall stability of the domain and to the formationof the IgG binding site in l and 2, and (ii) al unfolds first,followed by 2 and 3 together.  相似文献

针对镜面钢球缺陷检测装置中的球体全表面展开问题,建立展开机构接触模型并对球体运动进行分析,证明展开机构结构合理可行。首先由坐标变换建立数学模型,基于Matlab对模型空间接触位置进行计算,将展开运动合理简化,得到钢球和展开轮之间转动的同步关系;其次,对展开运动过程进行等效变换,寻找传动过程中能使钢球稳定展开的最优参数关系;最后,依据迭代法对球面上点的运动轨迹进行数值仿真,获得其具体规律为偏角固定的螺旋线。最终结果表明展开机构能够实现球体的全表面展开。  相似文献

CT辅助消化道胶囊内窥镜图像方法的主要目的在于更为直观地提高胶囊内窥镜的小病灶检出率,减少胶囊内窥镜的重复检查次数,减少患者检查费用,提高医生的工作效率.现有的虚拟外翻技术主要有两种,一种是展平法,一种是镜像法.CT辅助消化道胶囊内窥镜虚拟外翻是以镜像法为指导思想对三维肠道模型进行外翻,实验结果证明了这一方法的有效性.三维图像的显示采用了经典的移动立方体的算法.验证完成后,算法通过TI DaVinciDSP异构双核处理器(DM6467)和接口进行加速实现.通过合理分配双核的任务,算法的运算效率得到了很大提高.  相似文献

在流形学习的谱方法中, 流形展开被表述为优化问题. 这些优化问题的解是退化的, 即所有的样本将被嵌入到同一个点. 为了避免退化解, 谱方法对嵌入坐标人为地强加了一个单位协方差矩阵约束. 然而, 该约束往往导致流形展开的失真非常明显. 本文提出一种新的流形学习方法, 彻底抛弃了人为的单位协方差矩阵约束. 主要思路是先对流形边界进行嵌入, 然后再求流形内部的嵌入; 流形边界的嵌入位置被确定后, 流形内部样本的嵌入位置将被边界拉开, 使得它们不会都收缩到一个点上, 从而避免了退化解的出现. 将流形边界的嵌入位置作为边界条件, 求解一个线性方程组来得到内部样本的嵌入; 该线性方程组反映了尽量保持邻近样本间距离不变的要求. 流形边界的嵌入由简化流形的嵌入求出; 为此, 本文还设计了一种流形边界检测算法以及一种流形简化算法. 与目前代表性的几种流形学习方法进行了比较实验, 结果表明了本文方法的有效性, 其展开失真比谱方法明显要小.  相似文献

Satellite remote sensing has the potential to contribute to plant phenology monitoring at spatial and temporal scales relevant for regional and global scale studies. Historically, temporal composites of satellite data, ranging from 8 days to 16 days, have been used as a starting point for satellite-derived phenology data sets. In this study we assess how the temporal resolution of such composites affects the estimation of the start of season (SOS) by: 1) calibrating a relationship between satellite derived SOS with in situ leaf unfolding (LU) of trembling aspen (Populus tremuloides) across Canada and 2) quantifying the sensitivity of calibrated satellite SOS estimates and trends, over Canadian broadleaf forests, to the temporal resolution of NDVI data. SOS estimates and trends derived from daily NDVI data were compared to SOS estimates and trends derived from multiday NDVI composites that retain the exact date of the maximum NDVI value or that assume the midpoint of the multiday interval as the observation date. In situ observations of LU dates were acquired from the PlantWatch Canada network. A new Canadian database of cloud and snow screened daily 1-km resolution National Oceanic and Atmospheric Administration advanced very high resolution radiometer surface reflectance images was used as input satellite data. The mean absolute errors of SOS dates with respect to in situ LU dates ranged between 13 and 40 days. SOS estimates from NDVI composites that retain the exact date of the maximum NDVI value had smaller errors (~ 13 to 20 days). The sensitivity analysis reinforced these findings: SOS estimates from NDVI composites that use the exact date had smaller absolute deviations from the LU date (0 to − 5 days) than the SOS estimates from NDVI composites that use the midpoint (− 2 to − 27 days). The SOS trends between 1985 and 2007 were not sensitive to the temporal resolution or compositing methods. However, SOS trends at individual ecozones showed significant differences with the SOS trends from daily NDVI data (Taiga plains and the Pacific maritime zones). Overall, our results suggest that satellite based estimates of vegetation green-up dates should preferably use sub-sampled NDVI composites that include the exact observation date of the maximum NDVI to minimize errors in both, SOS estimates and SOS trend analyses. For trend analyses alone, any of the compositing methods could be used, preferably with composite intervals of less than 28 days. This is an important finding, as it suggests that existing long-term 10-day or 15-day NDVI composites could be used for SOS trend analyses over broadleaf forests in Canada or similar areas. Future studies will take advantage of the growing in situ phenology networks to improve the validation of satellite derived green-up dates.  相似文献
