1. To determine whether nitric oxide (NO) was involved in tolerance and sensitization to the effects of phencyclidine (PCP), we examined NO synthase activity and the number of NADPH-diaphorase (NADPH-d)-positive cells in discrete brain regions of saline-, acute PCP- and repeated PCP-treated mice. We also investigated the effects of a NO synthase inhibitor, NG-nitro-L- arginine methyl ester (L-NAME), on the behavioural changes induced by repeated PCP treatment in mice. 2. Acute PCP (1, 3, and 10 mg kg-1, s.c.) treatment induced dose-dependent hyperlocomotion, motor incoordination and stereotyped behaviours, consisting of sniffing, head movement and ataxia in mice. 3. In mice treated repeatedly with PCP (1, 3, and 10 mg kg-1 day-1), s.c., once a day for 14 days), the sniffing, head movement, ataxia and motor incoordination induced by PCP were attenuated (indicating the development of tolerance to these behaviours), whereas the hyperlocomotion induced by PCP was potentiated (indicating the development of sensitization to hyperlocomotion). The development of tolerance and sensitization to PCP-induced behaviours in the repeated PCP-treated mice was more marked at the dose of 10 mg kg-1 day-1) than at other doses. 4. NO synthase activity in the cerebral cortex and cerebellum, but not in the striatum and hippocampus, was significantly decreased by acute PCP (10 mg kg-1) treatment in comparison with saline treatment, and such changes in activity in the cerebral cortex and cerebellum were reversed by repeated PCP treatment (10 mg kg-1 day-1). 5. The number of neurones containing NADPH-d reactivity in the cerebral cortex, nucleus accumbens, and striatum of acute and repeated PCP-treated mice showed no change in comparison with saline-treated mice. 6. Tolerance to PCP (10 mg kg-1 day-1)-induced ataxia and motor incoordination was significantly attenuated by combined treatment with L-NAME (50 mg kg-1 day-1 i.p.). 7. Sensitization to PCP-induced hyperlocomotion was further enhanced by combined treatment with L-NAME (50 mg kg-1 day-1). However, NG-nitro-D-arginine methyl ester (D-NAME, 50 mg kg-1 day-1, i.p.), a less active enantiomer of L-NAME, had no effect, suggesting a stereospecific mechanism. 8. The PCP-induced behaviours in animals that had exhibited tolerance and sensitization to PCP (10 mg kg-1 day-1) were not influenced by acute L-NAME (5 and 50 mg kg-1, i.p.) or D-NAME (50 mg kg-1, i.p.) treatment. 9. These results suggest that NO may play an important role in the development, but not in the maintenance, of tolerance and sensitization to the effect of PCP in mice.  相似文献

The homomorphic filtering method is described for the detection and quantitation of left-to-right shunts from radionuclide angiocardiography. First, the method is investigated theoreticatly and numerically using a realistic model with systemic recirculation. It is demonstrated that even in a noisy situation the shunt flow fraction can be relatively well estimated, if only the characteristics of the fiters used are suitable. Second, the method is applied to real data, i.e., the pulmonary time-activity curves from radionucUde anglocardiography. The presence or absence of a left-to-right shunt is determined using the ratio AT/MTT. The pulmonary-to-systemic flow ratios calculated from the areas of the two decomposed curves are compared with those from oximetry at cardiac catheterization. Good agreement between the Qp/Qs ratios calculated by oximetry and radionucUide angiocardiography is obtained.  相似文献

The role of structure–activity relationships in the ability of catechins to inhibit P-glycoprotein (P-gp) function was investigated with respect to gallate and pyrogallol moieties. Experiments using octyl derivatives of gallic acid indicated that the gallate moiety required the catechol group and a neighboring carbonyl group to inhibit P-gp. On the other hand, the pyrogallol moiety appeared to require three hydroxyl groups to inhibit P-gp, according to comparisons between (−)-epicatechin gallate (ECG) and (−)-epigallocatechin gallate (EGCG). The difference in the number of hydroxyl groups that gallate or pyrogallol moieties required for P-gp inhibition, was due to the presence of a carbonyl group. The P-gp inhibition by catechins was restricted by their hydrophobicity. The pyrogallol moiety of EGC did not appear to inhibit P-gp because of its low hydrophobicity. The P-gp inhibitory activity of EGCG was speculated to be increased by the addition of long carbon chains to the C3″of gallate moieties.  相似文献

The ultrafiltration and dialysis characteristics of the semipermeable polymer blend membranes obtained from cellulose nitrate, poly(vinyl pyrrolidone), and N,N-dimethyl formamide were investigated under various conditions. The water content fraction and the ultrafiltration rate were dependent on the poly(vinyl pyrrolidone) content in the membranes, and the strengths were governed by the cellulose nitrate content in the polymer blend. If the pore radius in the membranes, calculated according to the Hagen-Poiseuilli equation for capillary model, was identical, the water content in each membrane was not identical. It was found that urea molecules broke very weak hydrogen bonds of the bound water in the membrane, but sodium chloride did not, also, the diffusion of urea through the membrane was more rapid than that of sodium chloride. The ratio of the membrane diffusion coefficient to the ultrafiltration rate was explained qualitatively by the capillary model, however, when the bound water in the water content fraction was considered, this ratio showed better agreement with the model.  相似文献

Three lipases, isolated previously in our laboratory, and a known lipase fromCandida cylindracea were screened for the enrichment of arachidonic acid (AA). The enzyme fromC. cylindracea was the most effective for the production of oil with high concentration of AA. When a single-cell oil fromMortierella alpina, containing 25% AA, was hydrolyzed with this lipase for 16 h at 35°C, the resulting glycerides contained 50% AA at 52% hydrolysis. After this, no further hydrolysis occurred, even with additional lipase. However, when the glycerides were extracted from the hydrolyzate and were hydrolyzed again with new lipase, the resulting oil contained 60% AA, with a recovery of 75% of its initial AA content. Triglycerides were the main components of the resulting oil. The release of each fatty acid from the oil depended on the hydrolysis rate of its ester. The fatty acid, whose ester is the poorest substrate for the enzyme, is concentrated in the glycerides.  相似文献

A recent physiological finding of neural coding for border ownership (BO) that defines the direction of a figure with respect to the border has provided a possible basis for figure-ground segregation. To explore the underlying neural mechanisms of BO, we investigated stimulus configurations that activate BO circuitry through psychophysical investigation of the BO-dependent tilt aftereffect (BO-TAE). Specifically, we examined robustness of the border ownership signal by determining whether the BO-TAE is observed when gestalt factors are broken. The results showed significant BO-TAEs even when a global shape was not explicitly given due to the ambiguity of the contour, suggesting a contour-independent mechanism for BO coding.  相似文献

Summary Poly(phenylene oxide) (PPO) — aromatic polyester multiblock copolymers were synthesized by polycondensation of bisphenol-A / isophthalic acid or m- and p-hydroxybenzoic acids in the presence of PPO having a carboxylic acid at one end and a phenolic hydroxy group at the other using triphenylphosphine / hexachloroethane as coupling agent.
