Enhancing effect of natural killer cell stimulatory factor (NKSF/interleukin-12) on cell-mediated cytotoxicity against tumor-derived and virus-infected cells

Natural killer cell stimulatory factor (NKSF) or interleukin-12 (IL-12) is a heterodimeric cytokine with pleiomorphic effects on T and NK cells, including induction of lymphokine production, mitogenesis, and enhancement of spontaneous cytotoxic activity. Similarly to IL-2, NKSF/IL-12 enhances NK cell-mediated cytotoxicity within a few hours and independently from induced proliferation. This effect is independent from other induced cytokines, because it is not prevented by antibodies neutralizing interferon (IFN)-alpha, IFN-beta, IFN-gamma, IL-2 or tumor necrosis factor (TNF)-alpha and, unlike the induction of IFN-gamma production by peripheral blood lymphocytes, it does not require HLA class II-positive accessory cells. Enhanced cytotoxicity is accompanied by morphologic changes in NK cells, including a significant increase in the number of cytoplasmic granules. In addition to the previously described ability to enhance the cytotoxic activity of NK cells against tumor-derived target cells, NKSF/IL-12 is also a potent stimulator of cytotoxicity against virus-infected cells, either fibroblasts acutely infected with herpes viruses or T cell lines chronically infected with human immunodeficiency virus-1. NK cell-mediated antibody-dependent cytotoxicity or anti-CD16 antibody-redirected lysis is not significantly enhanced by NKSF/IL-12. However, the ability of resting peripheral blood T cells to mediate anti-CD3 antibody-redirected lysis is enhanced by 18-h incubation with NKSF/IL-12, indicating that this lymphokine can modulate the cytotoxic capability of both NK and T cells.     

The Advance Toward Distinction in Engineering Technology

This paper traces the development of engineering technology from bifurcation to the present time. They assert that ET desires to be distinguished more than it seeks to be distinct. This analysis of the successes and failures of bifurcation lead to a differentiation between engineering technology's being distinct as a discipline and becoming distinguished in its own right. They offer some conjecture about the bright future of ET as a partner with science and engineering in an atmosphere of increased technological development but intense international competition.     

New long-chain hydroxy acids fromGrevillea decora

In addition to the ω-5 olefinic acids found in otherGrevillea species, about 10% of the acyl groups ofG. decora seed oil contain a hydroxy group and an ω-5 double bond. The chainlengths of these acids are from C₂₂ to C₃₀, with the largest concentration at the C₂₆ and C₂₈ chainlengths. The hydroxy group is located on odd carbons from carbon-5 through carbon-13. These acids previously were unknown in nature. The most abundant of these are 7-hydroxy-cis-17-docosenoic, 7-hydroxy-cis-19-tetracosenoic, 9-hydroxy-cis-19-tetracosenoic, 9-hydroxy-cis-21-hexacosenoic, 11-hydroxy-cis-21-hexacosenoic, and 13-hydroxy-cis-23-octacosenoic acids. The oil also contains the largest known concentration of the unoxygenated C₂₆ and C₂₈ ω-5 monoenes.     

Motion patterns of Nabesna Glacier (Alaska) revealed by interferometric SAR techniques

Nabesna Glacier is one of the longest land-terminus mountain glaciers in North America. However, its flow has never been studied. We derived detailed motion patterns of Nabesna Glacier in winter and spring 1994–1996 from the synthetic aperture radar (SAR) images acquired by the European Remote Sensing satellites (ERS-1 and ERS-2) using interferometric SAR (InSAR) techniques. Special effort was made to assess the accuracy of the motion estimates, and remove data points with high uncertainty from the motion profiles, enabling us to obtain reliable glacier flow patterns along the highly curved main course of Nabesna Glacier. The results, covering 80 km of the 87 km main course of the glacier, were used to delineate four distinctive sections in terms of spatial and temporal variability of the glacier speed: (1) the upper section where glacier flow was apparently random both temporally and spatially presumably due to development of crevasses; (2) the upper-middle section with relatively steady flow around 0.27 to 0.4 m/day; (3) the middle section with a stable pattern of acceleration from 0.27–0.3 m/day to the maximum about 0.67–0.73 m/day, followed by a general deceleration to 0.17–0.33 m/day before reaching (4) the lower section where the glacier motion was generally slow yet highly variable although uncertainty in the estimation is high. Occurrence of the flow maximum, as well as many local maxima and minima at consistent locations over different periods suggests that the valley geometry affect the overall flow pattern. On top of this general trend, many small-scale temporal/spatial variations in the glacier flow patterns were observed along the glacier, especially in the lower sections. On average, the flow speeds were in the range of 0.3 to 0.7 m/day; however this lacks any measurements of summer flow speeds which are expected to be significantly higher.     

Detection of failure sites by focused ion beam and nano-probing in the interconnect of three-dimensional stacked circuit structures

Focused ion beam (FIB) and nano-probing were applied for failure analysis of three-dimensional stacked circuits with copper through-silicon-vias between the stacked chips. The failure analysis was done after high temperature storage and thermal cycling tests. Passive voltage contrast in FIB allowed to pinpoint the open sites. FIB cross-sections showed the presence of opens at the bottom of the copper vias. The failure cause was suspected to be an interlayer particle, which was confirmed by optical profilometry. Nano-probing was used on another sample to pinpoint the failure location through the measurement of the local resistance within the daisy chains. The failure was traced out to be related with surface contamination.     

Behavioral and subjective effects of D-amphetamine and modafinil in healthy adults.

Modafinil is indicated for the management of excessive daytime sleepiness; however, recent studies have examined a broad range of potential uses. Given that clinical uses of modafinil may be expanding, this study compared modafinil and d-amphetamine effects on subjective and performance measures. Across 11 sessions, 11 healthy adults were tested after oral doses of placebo (5 sessions), modafinil (1.75 mg/kg, 3.50 mg/kg, or 7.00 mg/kg), and d-amphetamine (0.035 mg/kg, 0.070 mg/kg, 0.140 mg/kg) under double-blind, randomized conditions. Assessments of cognitive performance and subjective effects were completed before drug administration, 30 min after drug administration, and at hourly intervals after drug administration for 5 hr. Modafinil increased ratings on the Amphetamine and Morphine Benzedrine Group scales of the Addiction Research Center Inventory (ARCI) and increased ratings on the Vigor and Total Positive scales of the Profile of Mood States. d-Amphetamine increased visual analog ratings of feeling stimulated and liking the drug and increased ratings on the Morphine Benzedrine Group scale of the ARCI. Both medications significantly reduced visual analog scale ratings of feeling sleepy, and modafinil decreased ratings on the ARCI Pentobarbital-Chlorpromazine-Alcohol Group scale. Both medications sustained performance that deteriorated across time on the Sternberg Number Recognition Test. Modafinil also enhanced performance rate on the Digit-Symbol Substitution Task above baseline levels and increased response rate on the Repeated Acquisition of Response Sequences Task. These results suggest that modafinil engenders alerting effects and increases performance in healthy non-sleep-deprived individuals comparable with that of d-amphetamine. KATRIN NEG pumping concept investigation

The Karlsruhe Tritium Neutrino experiment (KATRIN) is going to use a very large electro-static tandem spectrometer to measure the electron spectrum from the tritium beta decay, where several kilometers of non-evaporable getter (NEG) strips (type ST707) are to be used to achieve the UHV requirements. Different geometrical configurations of the NEG strips have been studied by Monte Carlo simulations. It is shown that the resulting pumping speed of the getter pumps will allow to get a pressure below 10⁻¹¹ mbar in the huge vessel with a volume of 1400 m³. By systematic assessment of the statistics of the pumping surfaces, it could be demonstrated that the design is sound.     

An efficient implementation of SELF, a dynamically-typed object-oriented language based on prototypes

We have developed and implemented techniques that double the performance of dynamically-typed object-oriented languages. Our SELF implementation runs twice as fast as the fastest Smalltalk implementation, despite SELF's lack of classes and explicit variables.To compensate for the absence of classes, our system uses implementation-levelmaps to transparently group objects cloned from the same prototype, providing data type information and eliminating the apparent space overhead for prototype-based systems. To compensate for dynamic typing, user-defined control structures, and the lack of explicit variables, our system dynamically compilesmultiple versions of a source method, eachcustomized according to its receiver's map. Within each version the type of the receiver is fixed, and thus the compiler can statically bind andinline all messages sent toself.Message splitting andtype prediction extract and preserve even more static type information, allowing the compiler to inline many other messages. Inlining dramatically improves performance and eliminates the need to hard-wire low-level methods such as+, ==, andifTrue:.Despite inlining and other optimizations, our system still supports interactive programming environments. The system traverses internal dependency lists to invalidate all compiled methods affected by a programming change.
