A large metabolomics study was performed on 600 plasma samples taken at four time points before and after a single intake of a high fat test meal by obese and lean subjects. All samples were analyzed by a liquid chromatography-mass spectrometry (LC-MS) lipidomic method for metabolic profiling. A pragmatic approach combining several well-established statistical methods was developed for processing this large data set in order to detect small differences in metabolic profiles in combination with a large biological variation. Such metabolomics studies require a careful analytical and statistical protocol. The strategy included data preprocessing, data analysis, and validation of statistical models. After several data preprocessing steps, partial least-squares discriminant analysis (PLS-DA) was used for finding biomarkers. To validate the found biomarkers statistically, the PLS-DA models were validated by means of a permutation test, biomarker models, and noninformative models. Univariate plots of potential biomarkers were used to obtain insight in up- or downregulation. The influence of the bacteriocinogenic culture Lactobacillus sakei (105/g) and semi-purified bacteriocin mesenterocin Y (2560 AU/kg) on the safety and quality of traditional Croatian fermented sausages was investigated. The addition of Lb. sakei and/or mesenterocin Y reduced microbial counts (P < 0.05) in the final products. After 28 days of ripening, coagulase-negative cocci decreased 1.5–2.0 log, yeasts 1.2–1.4 log and enterococci 1.7–2.7 log. In the case of the addition of Lb. sakei, the lactic acid bacteria count was significantly (P < 0.05) higher at day 7 of ripening, and was accompanied by a lower pH and a higher amount of lactic acid (P < 0.05). In the final product the amount of acetic acid was significantly lower. More intensive proteolysis and an increase in ammonia content were found at the beginning of fermentation, and in the second phase of ripening in the control samples, respectively. The free fatty acid concentration was significantly lower during the entire ripening process compared to the control (P < 0.05). Semi-purified mesenterocin Y did not affect the sensory properties of the sausages, whilst the addition of Lb. sakei enhanced them. 相似文献

This study analyses the efficiency of fungal pectinolytic preparations produced in laboratory and commercial products used in the clarification process of apple, butia palm fruit, blueberry, and grape juices. Two crude enzymatic extracts, produced by Aspergillus niger T0005007-2 (TE1) and Aspergillus oryzae IPT 301 (TE2), were tested in solid-state and submerse cultures, respectively, in comparison to the commercial preparations Pectinex®Clear and Pectinex®BE Colour, used for the clarification of clear (apple, butia palm fruit) and dark (blueberry and grape) juices, respectively. With pectinases total activity at 1 U/mL of fruit juice, reactions were conducted at 30 and 50 °C, for 30 and 60 min. Time increase resulted in an improved clarification, whereas temperature increase could not be clearly linked to a greater clarification. When compared to the commercial products, the use of the crude preparation TE1 resulted in similar clarification of apple and blueberry juices. For butia palm and grape juice, however, TE1 presented the highest clarification results, revealing the potential of A. niger T0005007-2 enzymes for commercial application. 相似文献

The present study was performed to evaluate the antioxidant capacity, the polyphenol and metal content of conventionally and organically produced wines, which underwent similar winemaking processes. The wine grapes were grown under well-defined organic and conventional conditions from the two different wine-growing sub-regions of Croatia, Prigorje and Southern Dalmatia. The values of antioxidant activity, as evaluated by two free radical methods, ABTS and DPPH, was found systematically higher in organic wines compared to conventional ones. In a reversed phase HPLC analysis, used in order to characterise the phenol fraction of wines, higher concentrations of chlorogenic acid, ferulic acid, catechin, trans-resveratrol, all studied hydroxybenzoic acids and flavonols were found in the organically produced wines. No apparent trend was found in the metal contents of the wines, but ICP-MS analysis confirmed that both wine productions, organic and conventional, were well within the toxicological safety limits. 相似文献

Magnesium, as a biodegradable metal, offers great potential for use as a temporary implant material, which dissolves in the course of bone tissue healing. It can sufficiently support the bone and promote the bone healing process. However, the corrosion resistance of magnesium implants must be enhanced before its application in clinical practice. A promising approach of enhancing the corrosion resistance is deposition of bioactive coating, which can reduce the corrosion rate of the implants and promote bone healing. Therefore, a well-designed substrate-coating system allowing a good control of the degradation behavior is highly desirable for tailored implants for specific groups of patients with particular needs. In this contribution, the influence of coating formation conditions on the characteristics of potentiostatically electrodeposited CaP coatings on magnesium substrate was evaluated. Results showed that potential variation led to formation of coatings with the same chemical composition, but very different morphologies. Parameters that mostly influence the coating performance, such as the thickness, uniformity, deposits size, and orientation, varied from produced coating to coating. These characteristics of CaP coatings on magnesium were controlled by coating formation potential, and it was demonstrated that the electrodeposition could be a promising coating technique for production of tailored magnesium-CaP implants. 相似文献

Results of a study devoted to provide the pure enantiomers of isoflurane and desflurane from racemic mixtures using gas chromatography are presented. For that purpose, a cyclodextrin‐based selector described in earlier work was immobilized on porous glass beads. The adsorption isotherms were determined and applied to predict operating parameters which provide the highest possible productivity of the separation. The analysis included evaluation of the performance of larger columns applying simplifying scale‐up considerations. Using repetitive batches, the method can provide per day with a laboratory scale column approximately 1 g pure enantiomer. Selected model predictions were validated experimentally. 相似文献
