An algorithm is presented for computing degrees of sequence conservation found among aligned amino acid sequences. Sequence identities are calculated for each position of an alignment and average identity values of neighboring positions are figured. The average identity value of the whole alignment is chosen as a limit to discriminate between well and less conserved sequence sections. A second algorithm is given to calculate the degree of divergence of individual sequences compared to the other sequences of the alignment. The approach is easy to use on microcomputers and gives an exact picture of sequence identities and differences in order to determine, first, protein regions of high functional or structural importance among homologous proteins, and, second, significant differences of single sequences that may contribute to individual properties of the analysed protein. The method is illustrated by an example analysing a sequence alignment of higher plant nitrate reductases.  相似文献

In a double-blind, double-dummy, randomized, multi-centre study, the effects of bendroflumethiazide vs. enalapril on blood pressure, glycaemic control, lipoprotein concentrations and albuminuria were compared in non-proteinuric, hypertensive type 2 diabetic patients; they were treated for 20 weeks with either bendroflumethiazide 2.5-5.0 mg (n = 59) or enalapril 10-20 mg (n = 55). Age, fasting plasma glucose, HbA1c and BMI were similar in the groups. Systolic and diastolic blood pressure were reduced in both groups. Bendroflumethiazide was accompanied by minor but significant elevations in fasting plasma glucose and serum C-peptide. HbA1c was increased during both treatments. Lipoproteins and urinary albumin/creatinine ratio were stable. Bendroflumethiazide caused a decrease in serum potassium and an increase in serum urate. No significant correlations were observed between the decline in blood pressure and changes in the metabolic risk factors. Baseline levels of age, sex, BMI, blood pressure or urinary albumin/creatinine ratio were not related to changes in blood pressure, metabolic parameters or urinary albumin/creatinine ratio.  相似文献

Polymethylmethacrylate (PMMA) bone cements present various problems with respect to biocompatibility and stability. In order to study the histological changes at the bone-cement interface following total hip replacement, a small animal model was created by implanting hip prostheses using two different bone cements (PMMA and glass-ionomer cement, GIC). Two problems with the use of GIC for fixation of the prosthesis became evident. One year following implantation, histomorphometric analysis of femurs containing the GIC demonstrated significantly higher amounts of osteoid at the bone-cement interface. This disturbance of mineralization is comparable with osteomalacia and probably due to leaching of aluminum ions. In addition the mechanical properties of GIC proved to be inadequate for the loads placed upon it in this hip replacement model.  相似文献

1. Diadenosine hexaphosphate (AP6A) exerts vasoconstrictive effects. The purpose of this study was to investigate whether AP6A has any effect on cardiac function. 2. The effects of AP6A (0.1-100 microM) on cardiac contractility and frequency were studied in guinea-pig and human isolated cardiac preparations. Furthermore, the effects of AP6A on the amplitude of the L-type calcium current, on the adenosine 3':5'-cyclic monophosphate (cyclic AMP) content and on the phosphorylation of regulatory phosphoproteins, i.e. phospholamban and troponin inhibitor, were investigated in guinea-pig isolated ventricular myocytes. 3. In isolated spontaneously beating right atria of the guinea-pig AP6A exerted a negative chronotropic effect and reduced the rate of contraction maximally by 35% (IC20 = 35 microM). 4. In isolated electrically driven left atria of the guinea-pig AP6A exerted a negative inotropic effect and reduced force of contraction maximally by 23% (IC20 = 70 microM). 5. In isolated electrically driven papillary muscles of the guinea-pig AP6A alone was ineffective, but attenuated isoprenaline-stimulated force of contraction maximally by 23% (IC20 = 60 microM). Furthermore, AP6A attenuated the relaxant effect of isoprenaline. 6. In human isolated electrically driven ventricular preparations AP6A alone was ineffective, but attenuated isoprenaline-stimulated force of contraction by maximally 42% (IC20 = 18 microM). Moreover, AP6A attenuated the relaxant effect of isoprenaline. 7. All these effects of AP6A were abolished by the selective A1-adenosine receptor antagonist 1,3-dipropyl-cyclopentyl-xanthine (DPCPX, 0.3 microM), whereas the M-cholinoceptor antagonist atropine (10 microM) and the P2-purinoceptor antagonist suramin (300 microM) failed to abolish the effects of AP6A. 8. AP6A 100 microM had no effect on the amplitude of the L-type calcium current, but attenuated isoprenaline-stimulated L-type calcium current. The maximum of the current-voltage relationship (I-V curve) was shifted to the left by isoprenaline and additional application of AP6A shifted the I-V curve back to the right to the control value. The phosphorylation state of phospholamban and the troponin inhibitor was unchanged by AP6A alone, but was markedly attenuated by AP6A in the presence of isoprenaline. Cyclic AMP levels remained unchanged by AP6A, even after stimulation with isoprenaline. 9. In summary, AP6A exerts negative chronotropic and inotropic effects in guinea-pig and human cardiac preparations. These effects are mediated via A1-adenosine receptors as all effects were sensitive to the selective A1-adenosine receptor antagonist DPCPX. Furthermore, the effects of AP6A on cyclic AMP levels, protein phosphorylation and the L-type calcium current are in accordance with stimulation of A1-adenosine receptors.  相似文献

Mycobacterium simiae was the third most common mycobacterium identified over a 2-year period from a single clinical laboratory in southern Arizona. Thirty-three isolates from 25 patients were identified over 1 year. The isolation of M. simiae was considered clinically significant for only two of 23 evaluable patients. None of five patients with human immunodeficiency virus infection had clinical disease associated with M. simiae. Twenty isolates were available for detailed study. All but one of the 20 isolates were niacin-negative, and 11 were nonphotochromogenic. All 20 isolates had a triple-cluster pattern consistent with M. simiae by high-performance liquid chromatography, and restriction fragment patterns were identical for 16 isolates. Analysis of 16S rDNA confirmed the identity of all the tested isolates as M. simiae. In this study, M. simiae was a frequent clinical isolate but was rarely associated with disease. The organisms isolated were confirmed to be M. simiae but appeared to be phenotypically distinct strains of low virulence.  相似文献

Environmental protection and cost effectiveness lead to the demand to increase the efficiency of energy conversion. In the past decade gas turbines have become an important factor in efficient power generation. Gas turbine inlet temperatures are being increased continuously to decrease the specific fuel consumption. The development of new base materials and sophisticated processes like single crystal solidification allows higher metal temperatures. This causes higher oxidative stress to the MCrAlY overlay coatings. As shown in an earlier paper (I), an improvement of the high temperature properties is achievable by the addition of Rhenium. In this paper we report on properl ies of MCrAlY coatings containing 8 to 12% Al and 10% Rhenium by weight. The coatings were applied on Inco 738 LC by LPPS (Low Pressure Plasma Spraying). Data for static oxidation and cyclic oxidation data at 950°C, I000°C up to 10,000 hours are presented. However, the main aim of this report is to discuss the thermal-mechanical properties of LPPS-coatings with Re. As known from earlier work, the variation or addition of different elements to overlay coatings lead to a change in the ductile-brittle transition. Ductile-Brittle-Transition-Temperature (DBTT) measurements on 10% Re containing systems under the variation of Co, Cr, Al and Si have been done. In order to get more information about the thermal-mechanical properties of the coating under service like conditions, TCF (Thermal Cycle Fatigue) and TMF (Thermal Mechanical Fatigue) tests were carried out. On discus shaped samples, the TCF behavior of an MCrAlYRe coating in comparison to an MCrAlY coating has been evaluated up to 1,500 cycles. The samples were heated in a radiation oven up to 1000°C and cooled down by compressed air. By this method a differentiation in the thermal fatigue behavior is possible. However, this kind of test does not cover all possible relaxation processes. A more realistic investigation of the thermal mechanical fatigue behavior is given by TMF tests. A service life cycle was taken to study the thermal mechanical behavior. It is shown that Re improves the thermal mechanical properties of MCrAlY coating systems considerably.  相似文献

The effect of a cellular prion protein (PrPc) deficiency on neuroenergetics was primarily analyzed via surveying the expression of genes specifically involved in lactate/pyruvate metabolism, such as monocarboxylate transporters (MCT1, MCT2, MCT4). The aim of the present study was to elucidate a potential involvement of PrPc in the regulation of energy metabolism in different brain regions. By using quantitative real-time polymerase chain reaction (qRT-PCR), we observed a marked reduction in MCT1 mRNA expression in the cortex of symptomatic Zürich I Prnp−/− mice, as compared to their wild-type (WT) counterparts. MCT1 downregulation in the cortex was accompanied with significantly decreased expression of the MCT1 functional interplayer, the Na+/K+ ATPase α2 subunit. Conversely, the MCT1 mRNA level was significantly raised in the cerebellum of Prnp−/− vs. WT control group, without a substantial change in the Na+/K+ ATPase α2 subunit expression. To validate the observed mRNA findings, we confirmed the observed change in MCT1 mRNA expression level in the cortex at the protein level. MCT4, highly expressed in tissues that rely on glycolysis as an energy source, exhibited a significant reduction in the hippocampus of Prnp−/− vs. WT mice. The present study demonstrates that a lack of PrPc leads to altered MCT1 and MCT4 mRNA/protein expression in different brain regions of Prnp−/− vs. WT mice. Our findings provide evidence that PrPc might affect the monocarboxylate intercellular transport, which needs to be confirmed in further studies.  相似文献

The structure-activity relationship of eclosion hormone from the silkworm, Bombyx mori, was analyzed. First, the probable active residues in silkworm eclosion hormone and also tobacco hornworm eclosion hormone were predicted by the average distance map method. To examine the contributions of those residues to the activity of silkworm eclosion hormone, Gly-substituted mutants for those predicted residues were produced by site-directed mutagenesis and their activities were evaluated by a bioassay. Finally, Glu12, Met24 and Phe25 were estimated to be the crucial residues for the eclosion hormone activity. The possibility of the development of a blocker of an eclosion hormone receptor on the basis of the present work is also discussed.   相似文献

The 3-D crystal structure of interleukin-1ß(IL-1ß)has been used to define its receptor binding surface by mutationalanalysis. The surface of IL-1ß was probed by site-directedmutagenesis. A total of 27 different IL-1ß muteinswere constructed, purified and analyzed. Receptor binding measurementson mouse and human cell lines were performed to identify receptoraffinities. IL-1ß muteins with modified receptor affinitywere evaluated for structural integrity by CD spectroscopy orX-ray crystallography. Changes in six surface loops, as wellas in the C- and N-termini, yielded muteins with lower bindingaffinities. Two muteins with intact binding affinities showed10- to 100-fold reduced biological activity. The surface regioninvolved in receptor binding constitutes a discontinuous areaof 1000 Å2 formed by discontinuous polypeptide chain stretches.Based on these results, a subdivision into two distinct localareas is proposed. Differences in receptor binding affinitiesfor human and mouse receptors have been observed for some muteins,but not for wild-type IL-1ß. This is the first timea difference in binding affinity of IL-1ß muteinsto human and mouse receptors has been demonstrated  相似文献

Experimental results for the electrodeposition of Ni-Zn alloys from chloride solutions, with addition of H3BO3 and without other additives, in a laboratory cell with a perforated nickel sheet cathode and with recirculating electrolyte are presented. The dependence of the zinc content in the alloy on the following operating conditions was investigated: cathodic current density, 1.0–20.0 A dm–2; temperature, 35–65°C; pH 1.5–5.5; total molarity,M tot=M Ni 2+ +M Zn 2+ =1.1–2.8 M; and, molar ratio,P=M Ni 2+ /M Zn 2+ =1.0–15. Depending on the operating conditions the Zn content in the alloy varied over the range 22–88 mol%. In separate experiments galvanostatic polarization curves were measured in the direction of increasing and then decreasing cathodic current density in the range 0.1–20.0 A dm–2 with all other operating conditions as used for electroplating experiments. In all cases significant hysteresis effects were observed. It was found that the current efficiency for the electrodeposition of Ni-Zn alloys from chloride solutions as a function of the zinc content in the alloy showed a sharp minimum of about 55% atX Zn=55 mol % irrespective of other operating conditions.  相似文献
