When observers look down into a gap in the ground plane, their judgments of the widest gap they can step across (gapmax) decrease as gap depth increases (Y. Jiang & L. S. Mark, 1994). This study investigated the possibility that Jiang and Mark's viewing conditions did not afford observers a sufficient opportunity to perform exploratory movements needed to detect information about gap width. Experiment 1 showed that the gap depth by gaze interaction disappeared only when restrictions were not imposed on observers' exploratory activities (eye, head, and body movements). Experiment 2 showed that observers tended to see the vertical surface as slanted away from them, which made the bottom of the surface appear farther away from them than the top. Only when observers were able to view the gap binocularly under conditions that did not restrict exploratory activity did their slant perception improve and their gapmax judgments no longer covary with gap depth. The data indicate that the exploratory movements of prospective actors are essential for the pickup of information about their action capabilities. Matrix remodeling could be an important mode of action of multipotent mesenchymal stromal cells (MSC) in extracellular matrix (ECM) disease, but knowledge is limited in this respect. As MSC are well-known to adapt their behavior to their environment, we aimed to investigate if their mode of action would change in response to healthy versus pathologically altered ECM. Human MSC-derived ECM was produced under different culture conditions, including standard culture, culture on Matrigel-coated dishes, and stimulation with the pro-fibrotic transforming growth factor-β1 (TGFβ1). The MSC-ECM was decellularized, characterized by histochemistry, and used as MSC culture substrate reflecting different ECM conditions. MSC were cultured on the different ECM substrates or in control conditions for 2 days. Culture on ECM increased the presence of surface molecules with ECM receptor function in the MSC, demonstrating an interaction between MSC and ECM. In MSC cultured on Matrigel-ECM and TGFβ1-ECM, which displayed a fibrosis-like morphology, gene expression of collagens and decorin, as well as total matrix metalloproteinase (MMP) activity in the supernatant were decreased as compared with control conditions. These results demonstrated that MSC adapt to their ECM environment, which may include pathological adaptations that could compromise therapeutic efficacy.  相似文献

Thyroid hormone (TH) has diverse effects on mitochondria and energy expenditure (EE), generating great interest and research effort into understanding and harnessing these actions for the amelioration and treatment of metabolic disorders, such as obesity and diabetes. Direct effects on ATP utilization are a result of TH’s actions on metabolic cycles and increased cell membrane ion permeability. However, the majority of TH induced EE is thought to be a result of indirect effects, which, in turn, increase capacity for EE. This review discusses the direct actions of TH on EE, and places special emphasis on the indirect actions of TH, which include mitochondrial biogenesis and reduced metabolic efficiency through mitochondrial uncoupling mechanisms. TH analogs and the metabolic actions of T2 are also discussed in the context of targeted modulation of EE. Finally, clinical correlates of TH actions on metabolism are briefly presented.  相似文献

Three heteroaromatic thiols (2-mercaptobenzothiazole, 2-mercaptobenzoxazole and 2-mercaptobenzimidazole) were investigated in regard to their abilities as a co-initiator in free radical polymerization induced by cyanine dye/borate salt photoinitiating system. The kinetic results shown that the new three-component photoinitiating systems are more effective than two-component ones. The mechanism of photochemical and chemical processes occurring during the irradiation of the three-component initiators was proposed on the basis of the laser flash photolysis. The addition to the cyanine dye borate salt of a second co-initiator (thiol) results in the generation besides butyl radical the second free radical which can start the polymerization chain reaction.  相似文献

Three-dimensional (3D) imaging has a significant impact on many challenges of life sciences. Three-dimensional matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI-IMS) is an emerging label-free bioanalytical technique capturing the spatial distribution of hundreds of molecular compounds in 3D by providing a MALDI mass spectrum for each spatial point of a 3D sample. Currently, 3D MALDI-IMS cannot tap its full potential due to the lack efficient computational methods for constructing, processing, and visualizing large and complex 3D MALDI-IMS data. We present a new pipeline of efficient computational methods, which enables analysis and interpretation of a 3D MALDI-IMS data set. Construction of a MALDI-IMS data set was done according to the state-of-the-art protocols and involved sample preparation, spectra acquisition, spectra preprocessing, and registration of serial sections. For analysis and interpretation of 3D MALDI-IMS data, we applied the spatial segmentation approach which is well-accepted in analysis of two-dimensional (2D) MALDI-IMS data. In line with 2D data analysis, we used edge-preserving 3D image denoising prior to segmentation to reduce strong and chaotic spectrum-to-spectrum variation. For segmentation, we used an efficient clustering method, called bisecting k-means, which is optimized for hierarchical clustering of a large 3D MALDI-IMS data set. Using the proposed pipeline, we analyzed a central part of a mouse kidney using 33 serial sections of 3.5 μm thickness after the PAXgene tissue fixation and paraffin embedding. For each serial section, a 2D MALDI-IMS data set was acquired following the standard protocols with the high spatial resolution of 50 μm. Altogether, 512?495 mass spectra were acquired that corresponds to approximately 50 gigabytes of data. After registration of serial sections into a 3D data set, our computational pipeline allowed us to reveal the 3D kidney anatomical structure based on mass spectrometry data only. Finally, automated analysis discovered molecular masses colocalized with major anatomical regions. In the same way, the proposed pipeline can be used for analysis and interpretation of any 3D MALDI-IMS data set in particular of pathological cases.  相似文献

In this contribution the dissociative electron attachment to metabolites found in aerobic organisms, namely oxaloacetic and citric acids, was studied both experimentally by means of a crossed-beam setup and theoretically through density functional theory calculations. Prominent negative ion resonances from both compounds are observed peaking below 0.5 eV resulting in intense formation of fragment anions associated with a decomposition of the carboxyl groups. In addition, resonances at higher energies (3–9 eV) are observed exclusively from the decomposition of the oxaloacetic acid. These fragments are generated with considerably smaller intensities. The striking findings of our calculations indicate the different mechanism by which the near 0 eV electron is trapped by the precursor molecule to form the transitory negative ion prior to dissociation. For the oxaloacetic acid, the transitory anion arises from the capture of the electron directly into some valence states, while, for the citric acid, dipole- or multipole-bound states mediate the transition into the valence states. What is also of high importance is that both compounds while undergoing DEA reactions generate highly reactive neutral species that can lead to severe cell damage in a biological environment.  相似文献

The paper investigates the effect of PF resin modification on the possibility of shortening the time or reducing the temperature when pressing particleboards glued with phenolic resin modified with amide polymers, and on the physical and mechanical properties of the boards. The applied activators of PF resin were as follows: polycaprolactam and its N-hydroxymethylene derivative, polyacrylamide and acrylamide. The scope of the research included studies regarding the influence of these modifiers upon properties of fluid PF resin, the production of particleboards at various pressing conditions and the determination of their physical and mechanical properties. The obtained results show that the modification of PF resin with amide polymers makes it possible to increase the reactivity of the resin and to produce, in a shorter time or at a lower temperature, particleboards with comparable or better properties than those of the boards produced under industry-like conditions.  相似文献

We have precisely measured the surface resistance (R S) of high quality YBCO thin films, deposited on Lanthanum Aluminate and Silver doped YBCO thin films on Cesium buffered Sapphire substrates using the Hakki-Coleman dielectric resonator at frequency of 10 GHz as a function of temperature from 25 K to 85 K. We have also studied the microwave power dependence of YBCO films on both substrates and the frequency dependence of films on LAO. Precise microwave characterisation of HTS films is needed for selection of best films for mobile phone base station receivers.  相似文献
