Ricin A-chain is delivered into macrophages via receptor-mediated endocytosis. We have found that following uptake via the mannose receptor, ricin A-chain is rapidly cleaved by endosomal proteases. Inhibition of endosomal proteases such as cathepsin D and B leads to the accumulation of toxin inside the cell. Inhibition of cathepsin D reduces ricin A-chain cytotoxicity, while blocking cathepsin B enhances cytotoxicity. Similar results were obtained using fibroblasts transfected with the mannose receptor. Our data strongly suggest that the activation or membrane translocation of ricin A-chain is dependent upon the action of specific proteases.  相似文献

Capacitance computed tomography techniques were used to visualize particles movement in the draft tube of a spouted fluidized bed for the coating process of drug production. A total of 512 frames images of the particle concentration distribution were obtained at 10-millisecond intervals over a coating time of 5 min using a capacitance computed tomography system. The three-dimensional capacitance CT images (time and two-dimensional space images) were decomposed to wavelet time and space levels to extract the dominant particle distribution feature using three-dimensional discrete wavelet multiresolution at different coating times. As a result, the time and space dominant particle distribution with a specific frequency level can be visualized.  相似文献

We examined the issue of reliability of InP-based high-electron mobility transistors (HEMTs), focusing on the increase of drain resistance Rd. In investigations of the mechanism of Rd increase, we took note of contaminant incorporation and of the relations between the device lifetime and the strength of the channel electric field. In the fabrication process, reducing contaminants, especially fluorine, significantly suppressed the increase of source and drain resistances. Cross-sectional views of the gate of improved devices, which had a long lifetime, confirmed an almost contaminant-free surface around the gate. In acceleration tests, the most negative impact on drain resistance stability among several bias conditions was found when the current density was high and the channel electric field was large at the same time. The dependence of drain-gate electric field strength E showed that the device lifetimes of HEMTs determined from Rd increase obeyed exp(1/E), which means that impact ionization was the main cause of degradation. We elucidate that the interactions of hot carriers with contaminants around the gate are the main causes of the Rd increase in HEMTs. Suppression of device degradation was achieved by optimizing the fabrication process around the gate. In this way, device lifetime was remarkably enhanced.  相似文献

Both the neutral and acidic fractions of the acetone extract of yellow fieldcress (Kireha-inugarashi,Rorippa sylvestris Besser) inhibited lettuce seed germination. Salicylic,p-hydroxybenzoic, vanillic, and syringic acid were identified in the acidic fraction. In the neutral fraction, hirsutin (8-methylsulfinyloctyl isothiocyanate), 4-methoxyindole-3-acetonitrile, and pyrocatechol were identified. Bioassay using a root exudate recirculating system showedR. sylvestris during flowering inhibited the lettuce seedling growth. Hirsutin (13g/plant/day) and pyrocatechol (9.3g/plant/day) were the major compounds released into the rhizosphere. Several combinations of pyrocatechol,p-hydroxybenzoic acid, vanillic acid, and hirsutin reduced lettuce seedling growth. These compounds seemed to be allelochemicals.  相似文献

The alumina–carbon composite fibers were obtained from poly[(acyloxy)aloxane] (PAA) with 3-ethoxypropanoic (EPA) and m-anisic acids (m-AA) legands. This preceramic polymer can be dissolved in p-xylene-methanol-EPA mixed solvent, and the concentrated solution exhibited an excellent spinnability. During the pyrolysis and sintering processes, aliphatic carboxylate in the side groups was easily decomposed and eliminated. The aromatic carboxylate, however, seems to be converted and migrated to a carbon domain in the alumina matrix into which aloxane repetition was converted. The fibers pyrolyzed up to 800 and 1000°C have electrical conductivities that monotonically increase with increasing temperature. The fiber pyrolyzed up to 1200°C showed the electrical conductivity in a rather complicated manner.  相似文献

Experimental techniques for evaluating the interfacial properties between fiber bundles and the matrix of three-dimensionally reinforced carbon/carbon composites were examined. Specially arranged fiber bundle push-out and pull-out tests were conducted up to 2273 K in vacuum. In these tests, a fiber bundle in the specimens was extruded or pulled out by external compressive or tensile loads. Post-fracture observations revealed that a shear fracture was successfully induced within the carbon matrices at the loaded fiber bundle interface. The interfacial shear strength and initial sliding stress of the fiber bundle monotonically increased with the test temperature. The relief of residual thermal stress and increases in the frictional resistance and anchor effect at the fiber bundle interface were considered to be the major mechanisms that caused the enhancements. The study presented in this paper concerns the influence of the counter materials on wear and friction performance of polytetrafluoroethylene (PTFE) reservoirs arranged in distinct patterns on coated surfaces. Al-bronze and Mo coatings were deposited on a mild steel substrate using an atmospheric plasma spray process. Three patterns of PTFE reservoirs were used. Pins, which served as counter surface, were made of three different materials. Wear tests were carried out in a pin-on-disc test rig at room temperature and under dry contact conditions. The tests were carried out at a constant pressure of 10 MPa. An average linear speed of the test disc was 0.036 m/s. The importance of appropriately matched hardness of two surfaces in sliding contact is emphasised. Due to inadequate hardness of the counter material, performance of Mo coating was adversely affected and expected beneficial action of PTFE reservoirs severely hindered. Al-bronze coating proved to be performing far better than Mo coating. Serious deterioration of Mo coating occurred faster than that for Al-bronze coating.  相似文献

A new hair-conditioning agent, N-[3-(dimethylamino)propyl]docosanamide (APA-22) hydrochloric acid salt (APA-22 HCl), has excellent solubility in water with a solubility limit over 10-times larger than that of other APA hydrochloric salts with shorter alkyl chain lengths. The physicochemical characteristics of APA-22 HCl were studied at 25 degrees C by using equilibrium surface tension (gamma), solubilization of an oil-soluble dye, steady-state fluorescence, and chloride ion selective electrode techniques. The APA-22 HCl salt is considered to form an aggregate at a concentration, C(1), which is about twice the concentration of the solubility limit of APAs with shorter alkyl chains. At a higher concentration, C(2), the aggregates of APA-22 HCl start forming another kind of aggregate, which is able to solubilize oil-soluble materials. That is, C(1) is considered to represent the critical aggregation concentration (cac) and C(2) a morphology transition. In the concentration region between C(1) and C(2) the solutions are seemingly transparent whereas at above C(2) they appear bluish or translucent. Since the Krafft point of APA-22 HCl is 55 degrees C, all the solutions, including the aggregates, are thought to exist in metastable states. Notably, however, these metastable solutions do not change significantly over a few months. The excellent solubility of APA-22 HCl in water is considered to be a result of this unique stepwise aggregation with increasing concentration.  相似文献
