The electrical properties of carbon nanotube thin-film transistors (CNT-FETs) fabricated using plasma-enhanced chemical vapor deposition (PECVD) were studied by scanning probe microscopy. The measured results suggest the formation of an island structure in the subthreshold regime and the disappearance of the island structure at the ON state. These results were explained by the change in the effective number of CNTs that contributed to the electrical conduction due to the gate-bias-dependent resistance of the semiconducting CNTs. The results obtained by Monte Carlo simulation revealed similar results. The effects of metallic CNTs with defects and the scatter of the drain current in the subthreshold regime were also examined.  相似文献

Adsorption characteristics of three clay adsorbents – sepiolite, kaolin and synthetic talc – were investigated to determine their applicability to remove an anionic reactive dye – Reactive Yellow 138:1 – from a water stream. Results showed that synthetic talc had the highest maximum monolayer adsorption capacity of 10.07 mg/g, followed by kaolin (3.73 mg/g) and sepiolite (3.23 mg/g), for the removal of Reactive Yellow 138:1. Synthetic talc and sepiolite removed dyes from aqueous solutions mainly through physical adsorption depending on the surface area. By contrast, the adsorption mechanism of kaolin was strongly influenced by the anion exchange mechanism. Zeta potential analysis revealed that the adsorption capacity of clay adsorbents increased under acidic rather than alkaline conditions, owing to protonation of the clay surface. Moreover, synthetic talc was successfully regenerated by heating at more than 400°C, effectively restoring its original adsorption capacity.  相似文献

In the present study, we investigated a microplate-based culture (MBC) of Escherichia coli for the high-throughput, high-level production of PS-tag-fused scFvs (scFv-PS) in insoluble form. The Overnight Express™ Autoinduction System (OE system) was adopted to skip the laborious induction step of the addition of IPTG. ScFv and scFv-PS began to be expressed after 6 h by conventional flask culture (FLC) and by MBC when utilizing the OE system, and similar specific productivity levels were attained during cultivation. In MBC, an important factor that directly affected the production levels was rotational speed during cultivation, suggesting that the mass transfer of oxygen was rate-limiting. In a comparison of the productivity of flask cultures utilizing the 2YT-IPTG and OE systems, MBC utilizing the OE system was the highest, with approximately 1 mg of insoluble scFv-PS obtained from each well under optimal conditions (1400 rpm). The results of SDS-PAGE and a cross-contamination check indicated that very similar cultivation conditions were attained in each well, without cross-contamination. Thus, MBC using the OE system is very useful for the high-throughput, high-level production of scFv-PS, which can be activated on the surface of hydrophilic PS plates by solid-phase refolding. Therefore, the production of a variety of specific scFv-PSs for cytokines and biomarkers will make possible the construction of sensitive and low-cost antibody microarrays, which will be very useful in clinical diagnosis and biochemical research.  相似文献

Phosphatidylinositol (PI) 3-kinase (encoded by the pik3(+) gene) in Schizosaccharomyces pombe has been identified as a homologue of VPS34p, a protein required for proper vesicular protein sorting. The clone defective in this protein carries enlarged vacuoles and exhibits sensitivity to high temperature or high ion concentration. The effect of disruption of pik3(+) on sporulation of Sz. pombe was examined. The diploid cells underwent G(1) arrest and meiosis. However, the spores formed by the deltapik3 cells were not viable. Electron-microscopic analysis revealed that the growth of the forespore membrane of deltapik3 cells was not correctly orientated, failing to engulf the nucleus or forming extremely small spores, as was confirmed by the use of Spo3p-GFP and GFP-Psy1p, which are markers of the forespore membrane. The coating materials found along the forespore membrane of the wild-type were greatly reduced in these cells. PI 3-P, the product of Pik3p, was detected on the forespore membrane, suggesting that PI 3-P-dependent vesicle transport may take place in formation of the forespore membrane. Misshaped forespore membrane, accumulation of vesicles, formation of small non-viable spores, and suppression by over expression of Psy1p were the phenotypes commonly seen in deltapik3 and deltaspo3 cells, suggesting a relationship between the functions of Pik3p and Spo3p in formation of the forespore membrane in Sz. pombe.  相似文献

In the yeast two-hybrid system, the N-terminal region of Rap1p was shown to interact with Gcr1p and Gcr2p. Disruption of gcr1 and/or gcr2 in the two-hybrid reporter strain demonstrated that the interaction with Gcr1p does not require Gcr2p, whereas the interaction with Gcr2p is mediated through Gcr1p. Deletion of the N-terminal region of Rap1p alone did not show a growth phenotype, but a growth defect was observed when this mutation was combined with a gcr2 deletion. The poor growth of the gcr1 null mutant was not affected further by the N-terminal deletion of Rap1p, but the growth of gcr1 strains with mutations in the DNA binding region of Gcr1p was affected by the removal of the N-terminal region of Rap1p. These results suggest that one function of the N-terminal region of Rap1p, presumably the BRCT domain, is to facilitate the binding of Gcr1p to the promoter by a protein-protein interaction.  相似文献

Biosynthetic genes are not only responsible for the formation of bioactive substances but also suited for other applications including gene therapy. To test the feasibility of human cells producing antibiotics in situ when provided with a heterologous biosynthetic gene, we focused on cytochrome P450, the class of enzymes important in conferring bioactivity to natural product precursors. We selected Fma-P450 that plays a central role in the fumagillin antimicrobial biosynthesis in Aspergillus fumigatus to examine fungal metabolite production by HeLa cells that express fma-P450 heterologously. Here we show that HeLa cells harboring fma-P450 can biosynthesize 5-hydroxyl-β-trans-bergamoten and cytotoxic 5-epi-demethoxyfumagillol when supplemented with the nontoxic precursor β-trans-bergamotene. While the production level was insufficient to effect cell death, we demonstrate that programming human cells to autogenerate antibiotics by introducing a heterologous biosynthetic gene is feasible.  相似文献

The tritium breeder and structural materials are necessary components in the blanket to realize tritium (T) self-sustainment of nuclear fusion. The long-term exposure between tritium breeders and structural materials will cause surface corrosion in irradiation environments and then further affect the tritium release behavior. In this study, chemical compatibility between Li2TiO3 ceramic pebbles and advanced structural materials was studied systematically at 700 °C for 300 h under He+0.1 % H2 environment, respectively. The color of the Li2TiO3 ceramic pebbles changes from white to dark grey and black. Moreover, the grain size of Li2TiO3 ceramic pebbles increases to more than 5 μm, and the crushing load decreased slightly. For the structural materials, the Al-rich oxide layer with about 188.7 nm of 14Cr-5Al oxide dispersion strengthened (ODS) steel and Cr-Fe rich oxide layer with about 1.04 μm of 14Cr-ODS steel were observed on the cross-section. The effective diffusion coefficient of O element in Li2TiO3 ceramic moved into ODS steel at 700 °C was calculated to be 3.3 × 10−16cm2/s and 1.02 × 10−14 cm2/s. Unfortunately, when SiC ceramics were contacted with the pebbles, the crystal phase transformed into SiO2, which severely limits its application. Therefore, these results will provide guidance for the selection of structural materials in the future.  相似文献
