Glycosaminoglycans (GAGs) and proteoglycans (PGs) are major components of the glycocalyx. The secreted GAG and CD44 ligand hyaluronic acid (HA), and the cell surface PG syndecan-1 (Sdc-1) modulate the expression and activity of cytokines, chemokines, growth factors, and adhesion molecules, acting as critical regulators of tumor cell behavior. Here, we studied the effect of Sdc-1 siRNA depletion and HA treatment on hallmark processes of cancer in breast cancer cell lines of different levels of aggressiveness. We analyzed HA synthesis, and parameters relevant to tumor progression, including the stem cell phenotype, Wnt signaling constituents, cell cycle progression and apoptosis, and angiogenic markers in luminal MCF-7 and triple-negative MDA-MB-231 cells. Sdc-1 knockdown enhanced HAS-2 synthesis and HA binding in MCF-7, but not in MDA-MB-231 cells. Sdc-1-depleted MDA-MB-231 cells showed a reduced CD24-/CD44+ population. Furthermore, Sdc-1 depletion was associated with survival signals in both cell lines, affecting cell cycle progression and apoptosis evasion. These changes were linked to the altered expression of KLF4, MSI2, and miR-10b and differential changes in Erk, Akt, and PTEN signaling. We conclude that Sdc-1 knockdown differentially affects HA metabolism in luminal and triple-negative breast cancer model cell lines and impacts the stem phenotype, cell survival, and angiogenic factors. 相似文献

Number of blends were prepared by intergrinding clinker, gypsum, fly ash, calcined clay, microsilica and limestone in laboratory ball mill in varying percentages, and their physical properties such as fineness, consistency, setting time and compressive strength have been determined. The durability tests on selected compositions were also conducted by exposing the mortar cubes separately in 5% Na2SO4 and 5% NaCl solutions till the age of 90 and 180 days. The performance was observed by compressive strength development criteria after various length of exposure. We consider the classic problem of pole placement by state feedback. We offer an eigenstructure assignment algorithm to obtain a novel parametric form for the pole-placing feedback matrix that can deliver any set of desired closed-loop eigenvalues, with any desired multiplicities. This parametric formula is then exploited to introduce an unconstrained nonlinear optimisation algorithm to obtain a feedback matrix that delivers the desired pole placement with optimal robustness and minimum gain. Lakes play a vital role in regulating water storage, flow of river water, and ultimately maintaining a balanced ecosystem. Spatial and temporal variations in physicochemical parameters of water in Harike Wetland, a Ramsar site in the northwestern state of Punjab, India, were studied. This study was conducted on a monthly basis from January to December 2015. The water quality was studied at ten locations from sites 1 to 10 upstream, central and downstream from Harike Lake for ten physicochemical parameters, including temperature, pH, electrical conductivity, turbidity, dissolved oxygen concentration biological oxygen demand, nitrate and phosphate concentrations and salinity. The findings of this study revealed that, except for temperature and pH, all parameters exhibited relatively higher values for the Sutlej River, compared with the Beas River, with sampling sites 5 to site 7 exhibiting intermediate results. The mean seasonal temperature variations ranged from 16.9 to 26.6 °C, the pH from 7.7 to 8.2, electrical conductivity from 223 to 303 μS cm?1 and TDS concentration from 148.7 to 180.4 ppm. Correlation analysis was conducted to assess the relations between the variables. The electrical conductivity exhibited a high positive correlation with salinity and biological oxygen demand, whereas it correlated negatively with the dissolved oxygen concentration. The Harike Wetland situated in Punjab is a Ramsar site and a wetland of national importance. The present study was undertaken to assess the spatiotemporal dynamics of the wetland on the basis of geospatial technology and ground‐based studies. Landsat images for the years 2002 and 2014 were acquired from the United States Geological Survey and classified digitally to generate landuse/land cover maps involving four classes (water, grassland (including water hyacinth), agriculture, built‐up (settlement), barren land). The total area of the Harike Wetland was found to be 8023.68 ha. Water sampling at eleven sites was carried out and evaluated for physicochemical parameters. The water quality at several sampling points was found to be severely degraded. Change detection analysis revealed the submerged area (area under water) and grassland (including water hyacinth) had decreased over the past 12 years, whereas that area under agriculture and built‐up land has increased, indicating a shrinkage in the total wetland area. The present study also indicated that the near‐infrared band is a good indicator of water quality parameters, as indicated by the significant positive correlation between the near‐infrared band and relevant water parameters. Because the wetland is important from both an ecological perspective and economic perspective, regular monitoring is recommended, for which geospatial technology has proven to be very useful. 相似文献

Developing countries diets are based on a variety of plant foods that often are the main suppliers of important amounts of iron (Fe) and zinc (Zn). The objectives of this study were 1) to measure the total and dialyzable amounts of Fe and Zn in foods from Northern Mexico (Sonora) and from Southern Mexico (Oaxaca) and 2) to evaluate the effect of meat content of diets on the dialyzable amount of Fe and Zn. Methods to calculate the total dialyzable amount of Fe and Zn, were those of the AOAC and of Shen et al. Total Fe in e northern Mexican foods went from 0.78 +/- 0.0 to 11.59 +/- to 0.03 mg/ 100g (dry weight, DW); in southern Mexican foods the same micronutrient amounts were 0.86 +/- 0.18 to 8.8 +/- 0.57 mg/100 g (BS). Total Zn values were 0.91 +/- 0.00 to 13.58 +/- 0.05 mg/100 g (DW) in Sonora, and 0.64 +/- 0.18 to 20.80 +/- 0.33 mg/100 g (DW) in Oaxaca. In northern Mexico, foods dialyzable Fe had values from 0.1 +/- 0.04% to 10.6 +/- 0.36% and for Zn from 4.0 +/- 0.21% to 55.32 +/- 0.14%. Meanwhile, the range of values of dialyzable Fe for foods from Oaxaca were from 0.22 +/- 0.06% to 9.40 +/- 0.14% for and from 2.41 +/- 0.26% to 54.27 +/- 1.49% for dialyzable Zn. Background and objective
The degeneration of the balance control system in the elderly and in many pathologies requires measuring the equilibrium conditions very often. In clinical practice, equilibrium control is commonly evaluated by using a force platform (stabilometric platform) in a clinical environment.In this paper, we demonstrate how a simple movement analysis system, based on a 3D video camera and a 3D real time model reconstruction of the human body, can be used to collect information usually recorded by a physical stabilometric platform.Methods
The algorithm used to reconstruct the human body model as a set of spheres is described and discussed. Moreover, experimental measurements and comparisons with data collected by a physical stabilometric platform are also reported. The measurements were collected on a set of 6 healthy subjects to whom a change in equilibrium condition was stimulated by performing an equilibrium task.Results
The experimental results showed that more than 95% of data collected by the proposed method were not significantly different from those collected by the classic platform, thus confirming the usefulness of the proposed system.Conclusions
The proposed virtual balance assessment system can be implemented at low cost (about 500$) and, for this reason, can be considered a home use medical device. On the contrary, astabilometric platform has a cost of about 10,000$ and requires periodical calibration. The proposed system does not require periodical calibration, as is necessary for stabilometric force platforms, and it is easy to use. In future, the proposed system with little integration can be used, besides being an emulator of a stabilometric platform, also to recognize and track, in real time, head, legs, arms and trunk, that is to collect information actually obtained by sophisticated optoelectronic systems. 相似文献38.
In this paper, a tailored blended image adaptive watermarking scheme has been presented, which is based on DWT and SVD. Through this paper an attempt has been made to solve the problem of false positive while maintaining the robustness and imperceptibility with the help of principal component and perceptual tuning of the image. Perceptual tuning is a non-blind technique and based on the objective quality of image. The embedding strength is made dependent on watermark features as well as of host in wavelet domain by using tuning parameter which is user specific. The idea of embedding the principal component of intermediate frequency sub-bands of watermark image into singular values of perceptually tuned intermediate frequency sub-bands of host image have been exploited. The proposed algorithm is providing the adaptive behavior towards the image content for perceptual transparency and at the same time avoiding the possibility of false watermark extraction well supported by a private key, which is necessary at the time of extraction. Thus the proposed watermarking algorithm is a kind of non-blind, image adaptive and suitable for rightful ownership. This article describes the development and use of broken probes: prompted processes of degradation that produce unique identifiers with which to associate and retrieve digitally recorded histories. We offer our design and deployment of Broken Probes as a methodology for eliciting insights into how broken objects and acts of breakage may be given new life through their integration with ubiquitous computing technologies. Based on these developments, we introduce the genre of worn media—a variety of computational material with which to frame and critically examine the manifestation of wear among digital things. We end by discussing how the genre of worn media sensitizes designers and Ubicomp researchers to issues of incompleteness, impermanence, and imperfection to help account for the ethical, material, and historical terms of endurance in a digital age. 相似文献

Rich Internet applications have removed most of the constraints of Web 1.0 while giving users more responsiveness and advanced browsing and interaction experiences. These new horizons, however, raise many challenges for people with disabilities or using limited hardware and software technologies, whose risk to be excluded from the benefits deriving from advanced web applications. To address this problem, WCAG 2.0 guidelines have been released as the newest World Wide Web Consortium recommendation for accessible web content, and WAI-ARIA is a candidate recommendation which provides reference specifications for accessible rich Internet applications. However, both specifications contain a huge amount of information that often discourages most web designers from dealing with accessibility issues. Moreover, guidelines are suitable and usually adopted to judge a design solution a posteriori, but they do not suggest how to face a design problem constructively. This paper proposes a design pattern language for accessibility. The language can be regarded as a universal design resource for helping web designers create accessible rich Internet applications compliant with the most recent standards. Knowledge representation through design patterns reflects the problem-solving approach usually followed by software and web designers, while pattern organization in a structured language aims to guide web designers throughout the design process. The language has been implemented as an accessible rich Internet application itself, thus allowing designers with disabilities to participate in web design. In order to evaluate the design pattern language, a three-step process was carried out including: (1) a heuristic analysis with a group of human–computer interaction experts, (2) a survey study with a group of web designers, and (3) a validation on the field with two designers who have been requested to apply the language in real design cases. 相似文献
