A single-blind randomized study was conducted on young (18–21 years, n = 16) and mature (25–35 years, n = 16) drivers to assess how age, combined with a modest dose of alcohol (0.7 g/kg for males and 0.6 g/kg for females), influenced performance on a driving simulator. The driving tasks included detecting the presence of a vehicle on the horizon as quickly as possible, estimating the point on the road that an approaching vehicle would have passed by the participants’ vehicle (time-to-collision) and overtaking another vehicle against a steady stream of oncoming traffic. The results of the vehicle detection task showed that detection times were significantly slower with maturity, alcohol consumption and lower approaching vehicle speeds (50 kph), particularly on curved sections of road. Approaching vehicle speed was also found to significantly influence time-to-collision (TTC) judgments, such that faster approach speeds led to less underestimated (and therefore riskier) judgments of TTC than slower speeds. In the overtaking task, mature participants demonstrated impaired discrimination skills with varying approaching vehicle speeds, while young participants recorded significantly slower speeds while overtaking a vehicle, thus increasing the time that they spent in the opposing lane. In conclusion, young and mature drivers demonstrated pivotal differences in behavior in this study. Young drivers showed a greater tendency to engage in risky driving, while experienced drivers appeared to be more susceptible to perceptual influences. Overall, alcohol consumption impaired a driver's ability to divide attention, but had little effect on decision-making processes.  相似文献

Fluorescence imaging of single molecules at room temperature is a powerful technique for studying complex condensed phase systems and revealing structure and dynamics hidden by ensemble measurements. Successful single-molecule spectroscopic experiments rely upon strong emitters that can be detected at the level of individual copies above the relevant background signals. This Account discusses a class of nonlinear optical chromophores that not only are well-suited for single-molecule imaging but also offer additional beneficial properties such as a significant ground-state dipole moment, moderate hyperpolarizability, and sensitivity to local environment. An overview of the photophysical properties of several members of this class of molecules as well as a mechanism to help understand the environmental sensitivity is presented. Some preliminary applications of the chromophores as single-molecule reporters in cellular and polymer systems are discussed, along with detection of the emitters by two-photon fluorescence.  相似文献

Recombinant Escherichia coli strains were constructed which simultaneously expressed the genes encoding the (S)‐oxynitrilase from cassava (Manihot esculenta) together with the wild‐type or a mutant variant of the arylacetonitrilase from Pseudomonas fluorescens EBC191 in a single organism under the control of a rhamnose‐inducible promoter. The whole cell catalysts obtained converted benzaldehyde and potassium cyanide in aqueous media at pH 5.2 mainly to (S)‐mandelic acid and/or (S)‐mandelic amide and synthesized only low amounts of the corresponding (R)‐enantiomers. The conversion of benzaldehyde and potassium cyanide (KCN) by a whole‐cell catalyst simultaneously expressing the (S)‐oxynitrilase and the wild‐type nitrilase resulted in a ratio of (S)‐mandelic acid to (S)‐mandelic amide of about 4:3. This could be explained by the strong nitrile hydratase activity of the wild‐type nitrilase with (S)‐mandelonitrile as substrate. The relative proportion of (S)‐mandelic amide formed in this system was significantly increased by coexpressing the (S)‐oxynitrilase with a carboxy‐terminally truncated variant of the nitrilase. This whole‐cell catalyst converted benzaldehyde and KCN to mandelic amide and mandelic acid in a ratio of about 9:1. The ee of the (S)‐mandelic amide formed was calculated to be >95%.  相似文献

Branched and dendrimeric cationic peptides have shown better transfection efficiency than linear peptides, owing to their superior capacity for inducing DNA condensation. We have designed and synthesized two analogously guanidinocarbonylpyrrole-substituted (GCP-substituted) branched cationic tripeptides that provide extremely strong electrostatic attraction towards DNA. Both ligands 1 and 2 can bind to DNA and form condensed complexes, owing to the branched structure and high positive charges, as demonstrated by isothermal titration calorimetry (ITC), ζ potential and atomic force microscopy (AFM). After the replacement of the carboxylate group by an amide group, binding of ligand 2 to DNA shows exothermic enthalpy and positive entropy changes relative to ligand 1 . Rational interpretation would suggest that ligand 2 might aid the translocation of plasmid pF143 to HEK 293T cells, showing high gene transfection efficiency. This work therefore provides a facile way, by modifying a branched cationic tripeptide with GCP, to turn a peptide even a tripeptide into an efficient gene transfection vector.  相似文献

Tissue engineering requires the precise positioning of mammalian cells and biomaterials on substrate surfaces or in preprocessed scaffolds. Although the development of 2D and 3D bioprinting technologies has made substantial progress in recent years, precise, cell-friendly, easy to use, and fast technologies for selecting and positioning mammalian cells with single cell precision are still in need. A new laser-based bioprinting approach is therefore presented, which allows the selection of individual cells from complex cell mixtures based on morphology or fluorescence and their transfer onto a 2D target substrate or a preprocessed 3D scaffold with single cell precision and high cell viability (93–99% cell survival, depending on cell type and substrate). In addition to precise cell positioning, this approach can also be used for the generation of 3D structures by transferring and depositing multiple hydrogel droplets. By further automating and combining this approach with other 3D printing technologies, such as two-photon stereolithography, it has a high potential of becoming a fast and versatile technology for the 2D and 3D bioprinting of mammalian cells with single cell resolution.  相似文献

The particle based Discrete Element Method (DEM) can be applied to examine comminution processes. In this study, a DEM framework has been extended to model particle breakage without mass loss. After a breakage event occurs, spherical particles, as often considered in the DEM, are replaced by size reduced spherical fragments. During the following time steps, the fragments grow to their desired sizes, so that the mass loss can be counterbalanced. Previously defined overlaps with adjacent unbroken and broken particles (fragments) as well as walls are allowed. The breakage model has been realized in a parallelized DEM framework because comminution processes are often attributed to large numbers of particles and by parallelization the computational time can be reduced efficiently. An oedometer (one-dimensional compression in axial direction of a confined particle bed) has been modelled to investigate the parallelization efficiency and the influence of the permitted overlaps during the growth process on the growth duration. A simplified roller mill has been considered to examine the applicability of the breakage procedure considering parallelization. The results show that parallelization reduces computational time considerably. The breakage procedure is suitable to model comminution processes involving even densely packed particle systems and is superior to existing approaches.  相似文献

Flooding induces low-oxygen environments (hypoxia or anoxia) that lead to energy disruption and an imbalance of reactive oxygen species (ROS) production and scavenging enzymes in plants. The influence of hypoxia on roots of hydroponically grown maize (Zea mays L.) plants was investigated. Gene expression (RNA Seq and RT-qPCR) and proteome (LC–MS/MS and 2D-PAGE) analyses were used to determine the alterations in soluble and membrane-bound class III peroxidases under hypoxia. Gel-free peroxidase analyses of plasma membrane-bound proteins showed an increased abundance of ZmPrx03, ZmPrx24, ZmPrx81, and ZmPr85 in stressed samples. Furthermore, RT-qPCR analyses of the corresponding peroxidase genes revealed an increased expression. These peroxidases could be separated with 2D-PAGE and identified by mass spectrometry. An increased abundance of ZmPrx03 and ZmPrx85 was determined. Further peroxidases were identified in detergent-insoluble membranes. Co-regulation with a respiratory burst oxidase homolog (Rboh) and key enzymes of the phenylpropanoid pathway indicates a function of the peroxidases in membrane protection, aerenchyma formation, and cell wall remodeling under hypoxia. This hypothesis was supported by the following: (i) an elevated level of hydrogen peroxide and aerenchyma formation; (ii) an increased guaiacol peroxidase activity in membrane fractions of stressed samples, whereas a decrease was observed in soluble fractions; and (iii) alterations in lignified cells, cellulose, and suberin in root cross-sections.  相似文献
