A technique to study nanowire growth processes on locally heated microcantilevers in situ in a transmission electron microscope has been developed. The in situ observations allow the characterization of the nucleation process of silicon wires, as well as the measurement of growth rates of individual nanowires and the ability to observe the formation of nanowire bridges between separate cantilevers to form a complete nanowire device. How well the nanowires can be nucleated controllably on typical cantilever sidewalls is examined, and the measurements of nanowire growth rates are used to calibrate the cantilever‐heater parameters used in finite‐element models of cantilever heating profiles, useful for optimization of the design of devices requiring local growth.  相似文献

Chronic otitis media is a common disease often accompanied by recurrent bacterial infections. These may lead to the destruction of the middle ear bones such that prostheses have to be implanted to restore sound transmission. Surface coatings with layered double hydroxides (LDHs) are evaluated here as a possibility for drug delivery systems with convenient advantages such as low cytotoxicity and easy synthesis. Male New Zealand White rabbits were implanted with Bioverit® II middle ear prostheses coated with the LDH Mg4Al2(OH)12(SO4)2·6H2O impregnated with ciprofloxacin. 12 (group 1) were directly infected with Pseudomonas aeruginosa and another 12 (group 2) 1 week after the implantation. Clinical outcome, blood counts, histological analyses and microbiological examination showed an excellent antimicrobial activity for group 1, whereas this effect was attenuated in animals where infection was performed 1 week after implantation. This is the first study to demonstrate an efficient drug delivery system with an LDH coating on prostheses in the middle ear.  相似文献

Nanowires are commonly used as tools for interfacing living cells, acting as biomolecule‐delivery vectors or electrodes. It is generally assumed that the small size of the nanowires ensures a minimal cellular perturbation, yet the effects of nanowires on cell migration and proliferation remain largely unknown. Fibroblast behaviour on vertical nanowire arrays is investigated, and it is shown that cell motility and proliferation rate are reduced on nanowires. Fibroblasts cultured on long nanowires exhibit failed cell division, DNA damage, increased ROS content and respiration. Using focused ion beam milling and scanning electron microscopy, highly curved but intact nuclear membranes are observed, showing no direct contact between the nanowires and the DNA. The nanowires possibly induce cellular stress and high respiration rates, which trigger the formation of ROS, which in turn results in DNA damage. These results are important guidelines to the design and interpretation of experiments involving nanowire‐based transfection and electrical characterization of living cells.  相似文献

A drainage/filtration apparatus was developed for automatically determining sedimentation velocity and dewatering rate. Pressure-step testing was used to study filter cake compressibility, resistance, and swelling. Activated sludge was analysed, and the data indicate that the sludge is highly compressible even at low pressures (10 kPa). Furthermore, compressed sludge cakes swell if the pressure is released. Hence, the average specific cake resistance decreases if the pressure is released, though the resistance is higher after the compression cycle than before. Sludge must be dewatered under low pressure, because higher pressure only compresses the cake and does not improve the dewatering rate.  相似文献

The formation of biogenic amines (histamine, cadaverine, putrescine and spermidine) was followed during vacuum packed storage at 2 °C or 10 °C in the scombroid fish mackerel and in the non-scombroid fish herring. Also the changes in the content of free amino acids and in the organoleptic and microbiological qualities were followed.At 10 °C the amine contents were 2–20 times higher at the time of rejection as compared with samples stored at 2 °C. In herring and mackerel similar amounts of histamine were accumulated, whilst cadaverine was formed at much higher levels in mackerel compared with herring. The high contents of cadaverine in mackerel can possibly explain why mackerel and not herring are often implicated in incidents of scombortoxic poisoning.

Zusammenfassung Die Bildung von biogenen Aminen (Histamin, Cadaverin, Putrescin und Spermidin) wurde während der Lagerung bei 2 °C oder 10 °C, vacuumverpackt, in Makrelen und Hering verfolgt, ebenso die Veränderungen im Gehalt an freien Aminosäuren und der sensorischen bzw. mikrobiologischen Eigenschaften.Bei 10 °C war der Amingehalt 2- bis 20mal höher als bei den Proben bei 2 °C. Beim Hering und der Makrele haben sich die Histaminmengen gleichermaßen angehäuft, wahrend sich Cadaverin in der Makrele viel starker gebildet hatte als im Hering. Der hohe Gehalt an Cadaverin in der Makrele kann möglicherwerise erklären, warum Makrelen und nicht die Heringe an Vergiftungen beitragen.

This paper covers a combined experimental and computational effort carried out at Vattenfall Research & Development AB in order to study the thermal mixing in the annular region between a top tube and a control-rod stem. The low frequency thermal fluctuations in this region can result in problems with thermal fatigue and have caused cracks in the control-rod stems of several nuclear reactors ( [Kobayashi et al., 2009] and [0070] ).The flow in the vertical annular region formed by the top tube and the control-rod stem is characterized by the mixing of hot bypass flow with cold crud-removal flow. The crud-removal flow is flowing upwards along the control-rod stem, and the warmer bypass flow is entering through eight horizontal holes positioned in the lower part of the guide tube and four holes in the upper part of the top tube, forming jets.Two full-scale models of a control rod, including the control-rod stem and the guide tube, were constructed. The first model, designed to work at atmospheric conditions, was made of Plexiglass, in order to be able to visualize the mixing process, whereas the second one was made of steel to allow for a higher temperature difference between the two flows, and the heating of the top tube.CFD simulations of the case at atmospheric conditions were also carried out.Both the experiments and the simulations showed that the mixing region between the cold crud-removal flow and the warm bypass flow is dominated by large flow structures coming from above. The process is characterized by low frequency, high amplitude temperature fluctuations. The process is basically hydrodynamic, caused by the downward transport of flow structures originated at the upper bypass inlets. The damping thermal effects through buoyancy is of secondary importance, as also the scaling analysis shows, however a slight damping of the temperature fluctuations can be seen due to natural convection due to a pre-heating of the cold crud-removal flow. The comparison between numerical and experimental results shows a rather good agreement, indicating that experiments with plant conditions are not necessary since, through the existing scaling laws and CFD calculations, the obtained results may be extrapolated to plant conditions.The problem of conjugate heat transfer has not yet been addressed experimentally since complex and difficult measurements of the heat transfer have to be carried out. These types of measurements, along with corresponding CFD calculations, constitute one of the main challenges to be dealt with in ongoing work.  相似文献

Active power converters are flexible and highly dynamic. Many power converters, as battery charging devices, use only a small part of their technical capabilities. This paper develops a new control method that combines the primary converter function with grid frequency control. Doing this, the frequency stability can be influenced positively, using the power converters control potential, without penetrating its primary functions under normal power system conditions. In case of severe frequency deviations, priority is given to this grid support functionality, which can significantly improve the security of supply. If the converter’s control algorithm does not take frequency stability aspects into consideration, the impact of thousands of these devices on the grid frequency stability will be negative. Electric and hybrid cars with battery storage and plug-in capabilities are used as an example for power conversion units. The impact of many cars, with the referred control method, on the UCTE (Union for the Co-ordination of Transmission of Electricity) grid frequency can be observed by simulations. Even a small percentage of electric cars can significantly influence the grid frequency after a power plant disconnection fault. No communication between network operator and converter is necessary for the basic grid supporting functions. Dispersed frequency support as suggested by the developed control method could be adopted as a requirement to future grid codes.  相似文献

The oxidative stability of mixtures of edible oils containing polyunsaturated fatty acids (PUFA) and microcrystalline cellulose (MCC) was investigated. The mixtures studied consisted of oils of either camelina (CAM), cod liver (CLO), or salmon (SO) mixed with either colloidal or powdered MCC. A 50:50 (w/w) ratio of oil:MCC resulted in an applicable mixture containing high levels of PUFA edible oil and dietary fiber. The oxidative stability of the formulated mixtures and the pure oils was investigated over a period of 28 days. The peroxide value (PV) was assessed as a parameter for primary oxidation products and dynamic headspace gas chromatography mass spectrometry (GC/MS) was used to analyze secondary volatile organic compounds (VOC). CAM and the respective mixtures were oxidatively stable at both 4 and 22 °C during the storage period. The marine oils and the respective mixtures were stable at 4 °C. At 22 °C, an increase in hydroperoxides was found, but no increase in VOC was detected during the time-frame investigated. At 42 °C, prominent increases in PV and VOC were found for all oils and mixtures. Hexanal, a common marker for the degradation of n-6 fatty acids, propanal and 2,4-heptadienal (E,E), common indicators for the degradation of n-3 fatty acids, were among the volatiles detected in the headspace of oils and mixtures. This study showed that a mixture containing a 50:50 ratio of oil:MCC can be obtained by a low-tech procedure that does not induce oxidation when stored at low temperatures during a period of 1 month.  相似文献

Phage display with filamentous phages is widely applied and well developed, yet proteins requiring a cytoplasmic environment for correct folding still defy attempts at functional display. To extend applicability of phage display, we employed the twin-arginine translocation (TAT) pathway to incorporate proteins fused to the C-terminal domain of the geneIII protein into phage particles. We investigated functionality and display level of fluorescent proteins depending on the translocation pathway, which was the TAT, general secretory (SEC) or signal recognition particle (SRP) pathway mediated by the TorA, PelB or DsbA signal sequences, respectively. Importantly, for green fluorescent protein, yellow fluorescent protein and cyan fluorescent protein, only TAT, but not SEC or SRP, translocation led to fluorescence of purified phage particles, although all three proteins could be displayed regardless of the translocation pathway. In contrast, the monomeric red fluorescent protein mCherry was functionally displayed regardless of the translocation pathway. Hence, correct folding and fluorophor formation of mCherry is not limited to the cytosol. Furthermore, we successfully displayed firefly luciferase as well as an 83 kDa argonaute protein, both containing free cysteines. This demonstrates broad applicability of the TAT-mediated phagemid system for the display of proteins requiring cytoplasmic factors for correct folding and should prove useful for the display of proteins requiring incorporation of co-factors or oligomerization to gain function.  相似文献

The curing of an epoxy consisting of the solid hardener dicyandiamide (DICY) and the resin diglycidyl ether of bisphenol A (DGEBA) is studied in a system consisting of a tablet of DICY embedded in liquid DGEBA. Dissolution of DICY within the liquid DGEBA in combination with the transport of dissolved DICY from the tablet border into DGEBA and the chemical reaction of both reactants is studied by scanning Brillouin microscopy and infrared spectroscopy. Scanning Brillouin microscopy demonstrates the spatial and temporal evolution of the static and dynamic hypersonic properties in the course of curing in the vicinity of the DICY tablet. Infrared spectroscopy performed on epoxy pieces extracted from the final sample at different distances from the tablet surface give information about the spatial evolution of the curing process. The results achieved by both techniques are finally combined to yield a better understanding of the curing of DICY-based epoxies, which transform upon curing from strongly heterogeneous systems towards increasingly homogeneous systems.  相似文献
