Vinylidenebisphosphonic acid tetraethyl ester (1) and diazo ketones 7a-1 in ether at 22 degrees C yield pyrazoline bisphosphonate tetraethyl esters 8a-1 in moderate to good yield. These compounds were evaluated in animal models of arthritis: rat adjuvant-induced polyarthritis (AIP) and murine antigen-induced arthritis (AIA) and a murine model of chronic inflammation, the delayed type hypersensitivity granuloma reaction (DTH-GRA). (5-Benzoyl-2,4-dihydro-3H-pyrazol-3-ylidene)-bisphosphonic acid tetraethyl ester (8a), and [5-(3-fluorobenzoyl)-2,4-dihydro-3H-pyraxol-3-ylidene]- bisphosphonic acid tetraethyl ester (8d) significantly inhibited the arthritis models, AIP (15 mg/kg) and AIA (25 mg/kg), as well as the DTH-GRA (25 mg/kg). Conversion of 8a to the corresponding bisphosphonic acid, 10a, resulted in loss of activity. Compounds with alkyl substituents on the pyrazoline nitrogen, 9a-d, were inactive in the DTH-GRA. These results show that 8a and 8d have novel antiinflammatory activity and are capable of inhibiting chronic arthritis and inflammation in animals. Such compounds might be useful in man for treating chronic tissue injury associated with arthropathies such as inflammatory joint disease as well as other chronic inflammatory diseases.  相似文献

OBJECTIVE: To estimate more precisely the risk of fetal loss and congenital abnormalities after maternal parvovirus B19 infection, and to assess the long term outcome for surviving infants. DESIGN: Prospective cohort study of pregnant women with confirmed B19 infection with follow up of the surviving infants. The rate of fetal loss in the study cohort was compared with that in pregnant women with varicella. SETTING: Cases reported by laboratories in England and Wales between 1985-1988 and 1992-1995. SAMPLE: Four hundred and twenty-seven pregnant women with B19 infection and 367 surviving infants of whom 129 were followed up at 7-10 years of age. METHODS: Questionnaires to obstetricians and general practitioners on outcome of pregnancy and health of surviving infants. Maternal infection confirmed by B19-specific IgM assay and/or IgG seroconversion. RESULTS: The excess rate of fetal loss in women with B19 infection was confined to the first 20 weeks of gestation and averaged 9%. Seven cases of fetal hydrops followed maternal infections between 9 and 20 weeks of gestation (observed risk 2.9%, 95% CI 1.2-5.9). No abnormalities attributable to B19 infection were found at birth in surviving infants (observed risk 0%, upper 95% CI 0.86%). No late effects were found at 7-10 years. CONCLUSIONS: Around 1 in 10 women infected before 20 weeks of gestation will suffer a fetal loss due to B19. The risk of an adverse outcome of pregnancy after this stage is remote. Infected women can be reassured that the maximum possible risk of a congenital abnormality due to B19 is under 1% and that long term development will be normal.  相似文献

A novel technique is described which enables reliable fracture toughness measurements to be made in impact test on relatively small specimens of a tough polyethylene. Composite specimens have been made in which a tough polyethylene is sandwiched between two layers of a more brittle polyethylene. The overall fracture toughness is interpreted on the basis of simple additivity of the strain energy release rate associated with each of the component layers. Brittle plane strain failures were obtained for specimens in which the relative thickness of the layers was varied over a substantial range and the fracture toughness of each layer determined by suitable extrapolation. The fracture toughness of the brittle layer obtained in this way agreed well with direct measurements on that material.  相似文献

Yield and fracture of polyethylene have been studied in torsion tests under superposed hydrostatic pressures. Two ductile-to-brittle transitions have been observed. At high strain rates and pressures, a conventional ductile-to-brittle transition was found with increasing strain rate and pressure. A second ductile-to-brittle transition was observed at low strain rates with decreasing strain rate. The yield stress showed a region of low, relatively constant, rate dependence at low strain rates, high temperatures and low pressures and a second region of higher strain-rate dependence at high strain rates and pressures. In contrast, the fracture stress was found throughout to have a relatively constant strain-rate dependence of intermediate value between those obtained for the yield stress. These features confirmed that failure can be considered as competition between yield and fracture processes. The fracture stress became lower than the yield stress at both high and low strain rates where brittle fracture was observed, with fully ductile behaviour resulting in intermediate conditions where the fracture stress exceeded the yield stress. The pressure, strain rate and temperature dependence of the yield stress was well described by two Eyring processes acting in parallel, both processes being pressure dependent.  相似文献

The temporal changes in progesterone (delta 4P), 20 alpha-dihydroprogesterone (20 alpha-DHP) and luteinizing hormone (LH) concentrations in pseudopregnant (PSP) rats after treatment with a single subcutaneous Silastic-PVP tube containing 600 micrograms PGF2 alpha were correlated. Progesterone levels fell and LH levels rose significantly 2h after initiation of treatment, while 20 alpha-DHP levels were found to increase significantly 12h after treatment. Since the changes in delta 4P and LH concentrations occurred concurrently, it seems that the increase in LH levels could have been due to a direct effect of PGF2 alpha on the ovary causing a reduction in delta 4P and thus a negative feedback effect on LH release. Alternatively, PGF2 alpha might exert a direct effect on LH secretion at the hypothalamic-pituitary level.  相似文献
