Covering the soil with plastic film is an important method to modify soil temperature and improve the conditions for plant growth. For the optimal use of films in horticulture, it is necessary to quantify this effect, which depends particularly on the optical properties of the film. Five films, used mainly for soil covering in asparagus cultivation, were tested by measuring the soil temperature as well as the transmittance and reflectance in the short‐ and long‐wavelength bands: ‘Taschenfolie schwarz/weiß’, a film with black and white surfaces and pockets for fastening; ‘Antitaufolie’, a film with anti‐condensation coating, ‘Solartherm plus Folie’, a film with black top and transparent sides; ‘MaterBi Folie’, a biodegradable film; and ‘Thermoplus Folie’, a black polyethylene with transparent sides. All films were produced in Germany. The temperature measurements were carried out under controlled irradiation (short‐wavelength band) and ambient temperature conditions in the laboratory: first, 182 W m?2 and 11 °C at a depth of 0.4 m, and second, 340 W m?2 at 20 °C. A significant relationship between the temperature increase or decrease by covering the soil with the film and the calculated apparent quantities of the reflectance in the short and long‐wavelength band was derived (r2 = 0.85, n = 12). Furthermore, first tests with a physical model were carried out to obtain basic information, which should be used in the development of strategies for film management in the field. The heat-induced formation of lysinoalanine (LAL) was studied in raw skim milk that had been subjected to heat treatments as is usual in milk technology. Preheat treatment of milk at temperatures below 100 degrees C up to 20 min resulted in neglectable LAL amounts below 10 ppm i.p. (i.p.=in the protein) if at all. Tests with an UHT pilot plant showed that there was no proven formation of LAL with direct UHT-treatment at 110-130 degrees C for 10-25 min. In indirect UHT-treated milk vert small LAL amouints up to 50 ppm i.p. were detected only in those milk samples that were treated at temperatures high than 145 degrees C and holding times longer than 10 s. Autoclave sterilization in the range of 110-129 degrees C/10-25 min induced LAL amounts of 110 to 710 ppm i.p.. LAL formation in autoclave sterilized milk was almost directly dependent on heating temperature and time. Pre-treatment at temperatures below 100 degrees C induced no further LAL formation in any sterilization processes subsequent to preheating. In the pH range 6,50-6,90 LAL amounts in autoclave-sterilized milk increased directly with higher pH values at all temperatures. The varying degree of LAL formation with pH value was substantially more noticeable at higher than at lower temperatures. Increasing of lactose concentration caused only an insignificant decrease in LAL formation in autoclave-sterilized milk.  相似文献

In this paper we address the extension of a recently proposed reduced integration eight‐node solid‐shell finite element to large deformations. The element requires only one integration point within the shell plane and at least two integration points over the thickness. The possibility to choose arbitrarily many Gauss points over the shell thickness enables a realistic and efficient modeling of the non‐linear material behavior. Only one enhanced degree‐of‐freedom is needed to avoid volumetric and Poisson thickness locking. One key point of the formulation is the Taylor expansion of the inverse Jacobian matrix with respect to the element center leading to a very accurate modeling of arbitrary element shapes. The transverse shear and curvature thickness locking are cured by means of the assumed natural strain concept. Further crucial points are the Taylor expansion of the compatible cartesian strain with respect to the center of the element as well as the Taylor expansion of the second Piola–Kirchhoff stress tensor with respect to the normal through the center of the element. Autophagy is a self-degradative process that involves turnover and recycling of cytoplasmic components in healthy and diseased tissue. Autophagy has been shown to be protective at the early stages of programmed cell death but it can also promote apoptosis under certain conditions. Earlier we demonstrated that oxygen contributes to the pathogenesis of neonatal brain damage, which can be ameliorated by intervention with recombinant human erythropoietin (rhEpo). Extrinsic- and intrinsic apoptotic pathways are involved in oxygen induced neurotoxicity but the role of autophagy in this model is unclear. We analyzed the expression of autophagy activity markers in the immature rodent brain after exposure to elevated oxygen concentrations. We observed a hyperoxia-exposure dependent regulation of autophagy-related gene (Atg) proteins Atg3, 5, 12, Beclin-1, microtubule-associated protein 1 light chain 3 (LC3), LC3A-II, and LC3B-II which are all key autophagy activity proteins. Interestingly, a single injection with rhEpo at the onset of hyperoxia counteracted these oxygen-mediated effects. Our results indicate that rhEpo generates its protective effect by modifying the key autophagy activity proteins.  相似文献

A thought translation device (TTD) for brain-computer communication is described. Three patients diagnosed with amyotrophic lateral sclerosis (ALS), with total motor paralysis, were trained for several months. In order to enable such patients to communicate without any motor activity, a technique was developed where subjects learn to control their slow cortical potentials (SCP) in a 2-s rhythm, producing either cortical negativity or positivity according to the task requirement. SCP differences between a baseline interval and an active control interval are transformed into vertical or horizontal cursor movements on a computer screen. Learning SCP self regulation followed an operant-conditioning paradigm with individualized shaping procedures. After prolonged training over more than 100 sessions, all patients achieved self-control, leading to a 70-80% accuracy for two patients. The learned cortical skill enabled the patients to select letters or words in a language-supporting program (LSP) developed for inter-personal communication. The results demonstrate that the fast and stable SCP self-control can be achieved with operant training and without mediation of any muscle activity. The GTPases Rac and Cdc42Hs control diverse cellular functions. In addition to being mediators of intracellular signaling cascades, they have important roles in cell morphogenesis and mitogenesis. We have identified a novel PAK-related kinase, PAK4, as a new effector molecule for Cdc42Hs. PAK4 interacts only with the activated form of Cdc42Hs through its GTPase-binding domain (GBD). Co-expression of PAK4 and the constitutively active Cdc42HsV12 causes the redistribution of PAK4 to the brefeldin A-sensitive compartment of the Golgi membrane and the subsequent induction of filopodia and actin polymerization. Importantly, the reorganization of the actin cytoskeleton is dependent on PAK4 kinase activity and on its interaction with Cdc42Hs. Thus, unlike other members of the PAK family, PAK4 provides a novel link between Cdc42Hs and the actin cytoskeleton. The cellular locations of PAK4 and Cdc42Hs suggest a role for the Golgi in cell morphogenesis.  相似文献

Echinocandins are cyclic nonribosomal hexapeptides based mostly on nonproteinogenic amino acids and displaying strong antifungal activity. Despite previous studies on their biosynthesis by fungi, the origin of three amino acids, trans‐4‐ and trans‐3‐hydroxyproline, as well as trans‐3‐hydroxy‐4‐methylproline, is still unknown. Here we describe the identification, overexpression, and characterization of GloF, the first eukaryotic α‐ketoglutarate/FeII‐dependent proline hydroxylase from the pneumocandin biosynthesis cluster of the fungus Glarea lozoyensis ATCC 74030. In in vitro transformations with L ‐proline, GloF generates trans‐4‐ and trans‐3‐hydroxyproline simultaneously in a ratio of 8:1; the latter reaction was previously unknown for proline hydroxylase catalysis. trans‐4‐Methyl‐L ‐proline is converted into the corresponding trans‐3‐hydroxyproline. All three hydroxyprolines required for the biosynthesis of the echinocandins pneumocandins A0 and B0 in G. lozoyensis are thus provided by GloF. Sequence analyses revealed that GloF is not related to bacterial proline hydroxylases, and none of the putative proteins with high sequence similarity in the databases has been characterized so far.  相似文献
