Traumatic stress due to conflict and war causes major mental health problems in many resource-poor countries. The objective of this study was to examine whether trained lay counselors can carry out effective treatment of posttraumatic stress disorder (PTSD) in a refugee settlement. In a randomized controlled dissemination trial in Uganda with 277 Rwandan and Somalian refugees who were diagnosed with PTSD the authors investigated the effectiveness of psychotherapy administered by lay counselors. Strictly manualized narrative exposure therapy (NET) was compared with more flexible trauma counseling (TC) and a no-treatment monitoring group (MG). Fewer participants (4%) dropped out of NET treatment than TC (21%). Both active treatment groups were statistically and clinically superior to MG on PTSD symptoms and physical health but did not differ from each other. At follow-up, a PTSD diagnosis could not be established anymore in 70% of NET and 65% TC participants, whereas only 37% in MG did not meet PTSD criteria anymore. Short-term psychotherapy carried out by lay counselors with limited training can be effective to treat war-related PTSD in a refugee settlement. This paper presents a stochastic modelling framework based on stochastic automata networks (SANs) for the analysis of complex biochemical reaction networks. Our approach takes into account the discrete character of quantities of components (i.e. the individual populations of the involved chemical species) and the inherent probabilistic nature of microscopic molecular collisions. Moreover, as for process calculi that have recently been applied to systems in biology, the SAN approach has the advantage of a modular design process being adequate for abstraction purposes. The associated composition operator leads to an elegant and compact representation of the underlying continuous-time Markov chain in form of a Kronecker product. SANs have been extensively used in performance analysis of computer systems and a large variety of numerical and simulative analysis algorithms exist. We illustrate that describing a biochemical reaction network by means of a SAN offers promising opportunities to get insight into the quantitative behaviour of systems in biology while taking advantage of the benefits of a compositional modelling approach. 相似文献

High-temperature steam electrolysis by solid oxide electrolysis cells (SOEC) is a method with great potential for transforming clean and renewable energy from non-fossil sources to synthetic fuels such as hydrogen, methane or dimethyl ether, which have been identified as promising alternative energy carriers. With the same technology, fuel gas can be used in a very efficient way to reconvert chemically stored energy into electrical energy, since SOECs also work in the reverse mode, operating as solid oxide fuel cells (SOFC). As solid oxide cells (SOC) perform at high-temperatures (700–900 °C), material degradation and evaporation can occur, e.g., from the cell-sealing material, leading to poisoning effects and aging mechanisms that decrease the cell efficiency and long-term durability. To investigate such cell degradation processes, thorough examination of SOCs often requires a chemical and structural characterisation at a microscopic and nanoscopic level. The combination of different microscopic techniques such as conventional scanning electron microscopy (SEM), electron probe microanalysis (EPMA) and the focused ion beam (FIB) preparation technique for transmission electron microscopy (TEM) allows for post-mortem analysis at a multi-scale level. These complementary techniques can be used to characterise structural and chemical changes over a large and representative sample area (micro-scale) as well as at the nano-scale level for selected sample details. This article presents a methodical approach for the structural and chemical characterisation of changes in aged cathode-supported electrolysis cells produced at Risø DTU, Denmark. Polymeric and porous ceramic micro- and nanospheres were prepared by a simple solidification process with a liquid preceramic polymer in an oil–water-emulsification process. The system was composed of a water-crosslinkable poly(methoxymethylsiloxane) as the preceramic polymer and a linear hydroxyl-terminated poly(dimethylsiloxane) as a source for in situ water generation. Emulsification followed by subsequent curing was carried out via an ultrasonic treatment in the presence of surfactants and a crosslinking active organometallic catalyst. The rheological behavior of the polymeric base system was characterized and the crosslinking behavior was investigated as a function of catalyst concentration and temperature. Effects of composition and processing parameters such as energy input and type and concentration of the surfactant were determined with respect to the microstructure and particle size distribution of the obtained polymeric spheres. The particle diameters of the as-synthesized spheres were mainly in the sub-micron range. Among mammals, serotonin is predominantly found in the gastrointestinal tract, where it has been shown to participate in pathway-regulating satiation. For the stomach, vascular serotonin release induced by gastric distension is thought to chiefly contribute to satiation after food intake. However, little information is available on the capability of gastric cells to synthesize, release and respond to serotonin by functional changes of mechanisms regulating gastric acid secretion. We investigated whether human gastric cells are capable of serotonin synthesis and release. First, HGT-1 cells, derived from a human adenocarcinoma of the stomach, and human stomach specimens were immunostained positive for serotonin. In HGT-1 cells, incubation with the tryptophan hydroxylase inhibitor p-chlorophenylalanine reduced the mean serotonin-induced fluorescence signal intensity by 27%. Serotonin release of 147 ± 18%, compared to control HGT-1 cells (set to 100%) was demonstrated after treatment with 30 mM of the satiating amino acid L-Arg. Granisetron, a 5-HT3 receptor antagonist, reduced this L-Arg-induced serotonin release, as well as L-Arg-induced proton secretion. Similarly to the in vitro experiment, human antrum samples released serotonin upon incubation with 10 mM L-Arg. Overall, our data suggest that human parietal cells in culture, as well as from the gastric antrum, synthesize serotonin and release it after treatment with L-Arg via an HTR3-related mechanism. Moreover, we suggest not only gastric distension but also gastric acid secretion to result in peripheral serotonin release. 相似文献

The widely used pESC vector series (Stratagene, La Jolla, CA, USA) with the bidirectional GAL1/GAL10 promoter provides the possibility of simultaneously expressing two different genes from a single vector in Saccharomyces cerevisiae. This system can be induced by galactose and is repressed by glucose. Since S. cerevisiae prefers glucose as a carbon source, and since its growth rate is higher in glucose than in galactose‐containing media, we compared and evaluated seven different promoters expressed during growth on glucose (pTEF1, pADH1, pTPI1, pHXT7, pTDH3, pPGK1 and pPYK1) with two strong galactose‐induced promoters (pGAL1 and pGAL10), using lacZ as a reporter gene and measuring LacZ activity in batch and continuous cultivation. TEF1 and PGK1 promoters showed the most constant activity pattern at different glucose concentrations. Based on these results, we designed and constructed two new expression vectors which contain the two constitutive promoters, TEF1 and PGK1, in opposite orientation to each other.
