Pharmacological characterization of the pseudopterosins: novel anti-inflammatory natural products isolated from the Caribbean soft coral, Pseudopterogorgia elisabethae

Pseudopterosin E (PSE), a C-10 linked fucose glycoside and pseudopterosin A (PSA), a C-9 xylose glycoside isolated from the marine gorgonian Pseudopterogorgia elisabethae were both effective in reducing PMA-induced mouse ear edema when administered topically (ED50 (microg/ear) PSE(38), PSA(8)) or systemically (ED50 (mg/kg, i.p.) PSE (14), PSA (32)). Both compounds exhibited in vivo analgesic activity in phenyl-p-benzoquinone-induced writhing (ED50 (mg/kg, i.p.) PSE(14), PSA(4). PSE inhibited zymosan-induced writhing (ED50 = 6 mg/kg, i.p.), with a concomitant dose-dependent inhibition of peritoneal exudate 6-keto-prostaglandin F1alpha (ED50 = 24 mg/kg) and leukotriene C4 (ED50 = 24 mg/kg). In vitro, the pseudopterosins were inactive as inhibitors of phospholipase A2, cyclooxygenase, cytokine release, or as regulators of adhesion molecule expression. PSA inhibited prostaglandin E2 and leukotriene C4 production in zymosan-stimulated murine peritoneal macrophages (IC50 = 4 microM and 1 microM, respectively); however, PSE was much less effective. These data suggest that the pseudopterosins may mediate their anti-inflammatory effects by inhibiting eicosanoid release from inflammatory cells in a concentration and dose-dependent manner.     

AN EXAMINATION OF THE PERCEIVED EFFECTIVENESS OF COMMUNITY DEVELOPMENT CORPORATIONS: A PILOT STUDY

This study reports the results of a nationwide evaluation of community development corporations. Collective models of neighborhood economic revitalization are compared with other strategic approaches to community development. Special attention is given to the policy implications of the findings. The future of community development corporations is discussed, especially in light of Reaganomics and the decline in funds for the urban poor.     

DRYING OF PULP AND PAPER SLUDGE IN A PULSED FLUID BED DRYER

Hydrodynamics and drying kinetics for the pulp and paper primary sludge dried in a pulsed fluid bed dryer with relocated air stream are presented. Batch experiments have indicated that drying of disintegrated sludge to the required 12% moisture content takes place during the first drying period at practically constant material temperature close to the wet bulb temperature with respect to the inlet air conditions. Equations were developed for pressure drop, minimum pulsed-fluidization velocity, dynamic bed height, and volumetric mass transfer coefficient. Continuous experiments under drying conditions determined from the average residence time concept have confirmed that transportation of disintegrated sludge along the dryer follows the plug flow model.     

Utility of technetium-99m-sestamibi to assess osseous tumor spread

We report the staging results and the surgical outcome of a male patient with squamous-cell carcinoma in the floor of the mouth and a bone SPECT scan suggestive of local tumor infiltration of the mandible. Additional 99mTc-sestamibi SPECT imaging of the primary tumor and superimposing of both studies excluded osseous tumor spread and less extensive surgery was performed. Pathohistological examination confirmed the scintigraphic results and indicated a nonspecific periostal reaction as the cause of the positive bone scan. Nevertheless, a high-resolution camera and careful superimposition of both studies is mandatory if the differential diagnosis of an osseous tumor spread of a malignant tumor in the floor of the mouth and possibly less extensive surgery is at stake.     

Characterization of crystalline low temperature GaAs layers annealed from an amorphous phase

The results from an in-depth characterization of as-grown and annealed low-temperature GaAs layers deposited at less than 260°C are presented. The layers, amorphous as grown, became crystalline after annealing. The crystallization was documented by several characterization techniques including photoreflectance, Raman spectroscopy, photoluminescence, transmission electron microscopy, and double-crystal x-ray diffraction. The n-type conductivity of the annealed films was exploited for the construction of a diode structure.     

Chronic stress, social support, and persistent alterations in the natural killer cell response to cytokines in older adults

To address the long-term physiological consequences of chronic stressors, 14 continuing or current family caregivers of Alzheimer's disease (AD) patients, 17 former AD caregivers, and 31 control subjects were compared. Continuing and former caregivers did not differ on depressive symptomatology or perceived stress; both groups were significantly more depressed and stressed than controls. Furthermore, continuing and former caregivers did not differ in the response of NK cells in vitro to recombinant interferon-gamma and recombinant interleukin-2, and both groups had a significantly poorer response to these cytokines than controls. The physiological and psychological consequences of chronic stressors may persist well beyond the cessation of the actual stressor.     

Psychosocial enhancement of immunocompetence in a geriatric population.

Assessed the enhancement of immunocompetence by relaxation and social contact in 45 60–88 yr old geriatric residents of independent-living facilities. Ss were randomly assigned to 1 of 3 protocols: (1) relaxation training, (2) social contact, or (3) no contact. Ss in the relaxation and social-contact conditions were seen individually 3 times/wk for 1 mo. Blood samples and self-report data were obtained at baseline, at the end of the intervention, and at a 1-mo follow-up. At the end of the intervention, Ss in the relaxation group, who had been taught progressive relaxation and guided imagery as an active coping skill, showed a significant increase in natural killer cell activity and significant decreases in antibody titers to Herpes simplex virus and self-rated distress. The other 2 groups showed nonsignificant changes. There was a general increase in the T-lymphocyte response to phytohemagglutinin stimulation at the end of the intervention, with greater change at lower mitogen concentrations.
