Among various carrier materials for drug delivery, silica has been found to be significant for loading and sustained release of the drug. In the present study, the antibiotic gentamicin was incorporated in various amounts on to a silica matrix, using a sol–gel subcritical drying route. Crack free silica rods with a length of 1–1.5 cm, diameter of 0.5 cm and density of 1.35 g/cm3 incorporated with gentamicin were obtained. The incorporation of gentamicin into the silica matrix was revealed by FTIR. The adsorption–desorption isotherms and pore size distribution were studied in detail. The drug elution patterns were analysed and show a biphasic mode of release. After the initial burst release, a constant release pattern is observed. A release value above the minimum inhibitory concentration (MIC) is always maintained. The amount of drug released is higher than that of many other bioactive ceramic materials. An attempt has been made to correlate the functional properties with the porosity features.  相似文献

Biomedical implant devices made out of titanium and its alloys are benefited by a modified surface or a bioactive coating to enhance bone bonding ability and to function effectively in vivo for the intended period of time. In this respect hydroxyapatite coating developed through pulsed laser deposition is a promising approach. Since the success of the bioactive ceramic coated implant depends mainly on the substrate-coating strength; an attempt has been made to produce micro patterned surface structure on titanium substrate for adherent hydroxyapatite coating. A pulsed Nd-YAG laser beam (355 nm) with 10 Hz repetition rate was used for surface treatment of titanium as well as hydroxyapatite deposition. The unfocussed laser beam was used to modify the substrate surface with 500–18,000 laser pulses while keeping the polished substrate in water. Hydroxyapatite deposition was done in a vacuum deposition chamber at 400°C with the focused laser beam under 1 × 10−3 mbar oxygen pressure. Deposits were analyzed to understand the physico-chemical, morphological and mechanical characteristics. The obtained substrate and coating surface morphology indicates that laser treatment method can provide controlled micro-topography. Scratch test analysis and microindentation hardness values of coating on laser treated substrate indicate higher mechanical adhesion with respect to coatings on untreated substrates.  相似文献

Gold nanorod molecular probes (GNrMPs) were designed and fabricated for multiplex identification of cell surface markers in HBECs. Cells were probed directly using dark field microscopy integrated with a spectral imager for simultaneous detection of up to three surface markers. The immunophenotype composition of these cell lines indicative of their metastasis potential was assessed using the GNrMPs. The technique has the potential to become an important tool for diagnosis and prognosis of breast and other cancers.  相似文献

Graft copolymerization of acrylonitrile (AN) onto sodium alginate was studied in an aqueous medium using ferrous ammonium sulfate–hydrogen peroxide (Fenton's reagent) as the redox initiator. To optimize the reaction conditions for affording maximum percentage of grafting, the concentrations of sulfuric acid, initiator components, monomer, and also temperature and time were varied. Transparent hydroxyapatite (HAP) was prepared by sintering gel-cast powder compacts at 1000°C for 2 h; the resultant HAP material was studied using X-ray diffractometry, transmission electron microscopy, scanning electron microscopy, and microhardness measurement. Nanoscale HAP crystallites were prepared using a precipitation method that involved calcium nitrate and ammonium dihydrogen orthophosphate solutions; the preparation was conducted at a temperature of 0°C. The precipitate was gel-cast and sintered at 1000°C in the form of a transparent ceramic that had a uniform grain size of 250 μm. The maximum Vickers microhardness obtained for a sample sintered at 1000°C was 6.57 GPa. The sintering behavior of gel-cast samples prepared from high-temperature-precipitated HAP was compared with that of material prepared at 0°C.  相似文献

Breast cancers are classified into five intrinsic subtypes: Luminal subtype A, Luminal subtype B, HER2+, Basal, and Normal-like. In this study, we compared the plasma proteome of patients with Luminal A, Luminal B, HER2+, and Basal subtype with plasma from healthy individuals. Protein changes were considered significant if q-value (false discovery rate) was less than 5%. The highest number of changes in the plasma proteome was observed in patients with Luminal type B followed by Basal type breast cancers. The plasma proteome of Luminal A and HER2+ breast cancer patients did not differ significantly from healthy individuals. In Basal breast cancer, a significant number of plasma proteins were downregulated compared with healthy individuals. Acute phase-response proteins α-glycoprotein orosomucoid 1 and serum amyloid protein P were specifically upregulated in the plasma of Luminal B breast cancer patients, suggesting prevalence of low-grade inflammation. Proteins involved in immune response and free radical scavenging were downregulated in the plasma of Luminal B patients, which is in agreement with defective immune system observed in cancer patients. These results reveal intrinsic subtype specific changes in the plasma proteome that may influence tumor progression as well as the systemic effects of cancer.  相似文献

Many of the plant leucine rich repeat receptor-like kinases (LRR-RLKs) have been found to regulate signaling during plant defense processes. In this study, we selected and sequenced an LRR-RLK gene, designated as Oryza rufipogon receptor-like protein kinase 1 (OrufRPK1), located within yield QTL yld1.1 from the wild rice Oryza rufipogon (accession IRGC105491). A 2055 bp coding region and two exons were identified. Southern blotting determined OrufRPK1 to be a single copy gene. Sequence comparison with cultivated rice orthologs (OsI219RPK1, OsI9311RPK1 and OsJNipponRPK1, respectively derived from O. sativa ssp. indica cv. MR219, O. sativa ssp. indica cv. 9311 and O. sativa ssp. japonica cv. Nipponbare) revealed the presence of 12 single nucleotide polymorphisms (SNPs) with five non-synonymous substitutions, and 23 insertion/deletion sites. The biological role of the OrufRPK1 as a defense related LRR-RLK is proposed on the basis of cDNA sequence characterization, domain subfamily classification, structural prediction of extra cellular domains, cluster analysis and comparative gene expression.  相似文献

The present work deals with the photopolymerization of 2-phenylethyl (meth)acrylates and estimation of their kinetic parameters. Formulations were made by independently homogenizing the monomers with photoinitiators of two different classes. Two different compositions of photoinitiators were used to study the effect of concentration of photoinitiator on cure kinetics. These compositions obtained were tested for photo curing performance using differential photocalorimetry (DPC) or photo DSC under polychromatic radiation. The heat flow against time was recorded for all formulations under isothermal conditions and the rates of polymerization as well as the percentage conversions were estimated. It was observed that due to a longer timescale for reaction diffusion, the methacrylate formulations showed a higher conversion than acrylate formulations. Other parameters such as induction time, maximum rate and conversion attained as well as the time to attain peak maximum were noted. The photopolymerization and kinetic estimations of the formulations including evaluation of kinetic models are discussed.  相似文献
