In order to observe intracellular structures by scanning electron microscopy, excess cytoplasmic matrix must be removed from the fractured surface of cells. Previously we reported an Osmium-DMSO-Osmium method devised for this purpose. This method is very effective in revealing intracellular structures, but requires osmium tetroxide for initial fixation with some consequent disadvantages. In the present study, a revised Osmium-DMSO-Osmium method is reported, in which an aldehyde mixture is used as the initial fixative instead of osmium tetroxide. As fixation is carried out by perfusion in this revised method, better preservation of fine structures is achieved than by the original method, especially in the central nervous tissue which tends to suffer from post-mortem degeneration. Moreover this method can be applied to cytochemical studies of intracellular structures with a scanning electron microscope (SEM). The rainbow schlieren deflectometry has been combined with the computed tomography to obtain three-dimensional density fields of shock containing free jets and we call the method the schlieren CT. Experiments on the schlieren CT have been performed at a nozzle pressure ratio of 4.0 by using an axisymmetric convergent nozzle with an inner diameter of 10 mm at the exit where the nozzle was operated at an underexpanded condition. Multidirectional rainbow schlieren pictures of an underexpanded sonic jet can be acquired by rotating the nozzle about its longitudinal axis in equal angular intervals and the three-dimensional density fields are reconstructed by the schlieren CT. The validity of the schlieren CT is verified by a comparison with the density fields reconstructed by the Abel inversion method. As a result, it is found that excellent quantitative agreement is reached between the three-dimensional jet density fields reconstructed from both methods. 相似文献

Conventional and gradient CoNiCrAlYSi coatings were produced by using low vacuum plasma spray and an additional step of diffusional over aluminizing (pack cementation) techniques on an Inconel-738 substrate. Hot corrosion of these coatings was investigated using Na2SO4–20wt%NaVO3 molten salt at 880?°C for 800?h. Hot corrosion rate was determined by measuring the weight gain of the specimens at regular intervals for a duration of 20?h. X-ray diffraction, field emission scanning electron microscopy and electron probe micro analysis techniques were used to determine the nature of phases, investigation of the thermally grown oxide, examination of the surface attack and determination of the elemental distribution. The gradient coating showed better performance by re-healing alumina scale due to its possession of more β phase as Al reservoir. Results indicated that pack cementation process caused an increase in amount of aluminum-rich β phase and better hot corrosion properties of gradient coatings owing to the Al enrichment in the outer layer and rapid formation of protective oxide on the surface. 相似文献

The safe decommissioning as well as decontamination of the radioactive waste resulting from the nuclear accident in Fukushima Daiichi represents a huge task for the next decade. At present, research and development on long-term safe storage containers has become an urgent task with international cooperation in Japan. One challenge is the generation of hydrogen and oxygen in significant amounts by means of radiolysis inside the containers, as the nuclear waste contains a large portion of sea water. The generation of radiolysis gases may lead to a significant pressure build-up inside the containers and to the formation of flammable gases with the risk of ignition and the loss of integrity.In the framework of the project “R&D on technology for reducing concentration of flammable gases generated in long-term waste storage containers” funded by the Japanese Ministry of Education, Culture, Sports, Science and Technology of Japan (MEXT), the potential application of catalytic recombiner devices inside the storage containers is investigated. In this context, a suitable catalyst based on the so-called intelligent automotive catalyst for use in a recombiner is under consideration. The catalyst is originally developed and mass-produced for automotive exhaust gas purification, and is characterized by having a self-healing function of precious metals (Pd, Pt and Rh) dissolved as a solid solution in the perovskite type oxides. The basic features of this catalyst have been tested in an experimental program. The test series in the REKO-4 facility has revealed the basic characteristics of the catalyst required for designing the recombiner system. 相似文献

A microtubule gliding assay is a biological experiment observing the dynamics of microtubules driven by motor proteins fixed on a glass surface. When appropriate microtubule interactions are set up on gliding assay experiments, microtubules often organize and create higher-level dynamics such as ring and bundle structures. In order to reproduce such higher-level dynamics on computers, we have been focusing on making a real-time 3D microtubule simulation. This real-time 3D microtubule simulation enables us to gain more knowledge on microtubule dynamics and their swarm movements by means of adjusting simulation parameters in a real-time fashion. One of the technical challenges when creating a real-time 3D simulation is balancing the 3D rendering and the computing performance. Graphics processor unit (GPU) programming plays an essential role in balancing the millions of tasks, and makes this real-time 3D simulation possible. By the use of general-purpose computing on graphics processing units (GPGPU) programming we are able to run the simulation in a massively parallel fashion, even when dealing with more complex interactions between microtubules such as overriding and snuggling. Due to performance being an important factor, a performance model has also been constructed from the analysis of the microtubule simulation and it is consistent with the performance measurements on different GPGPU architectures with regards to the number of cores and clock cycles. 相似文献

Development of clothing in consideration of the shape and body function of a person with spinal cord injury is an important task. Then, a dummy robot with a deformation mechanism was developed in this study for evaluating the comfortable level of clothings. Specifically, a trunk joint mechanism and an abdominal mechanism that can realize various deformations of the abdominal area and various trunk poses were developed. The trunk joint mechanism was implemented in order to simulate the seated posture of persons with spinal cord injury. The abdominal deformation mechanism was implemented using linear actuators and rotating servomotors in order to simulate abdominal obesity of persons with spinal cord injury. This study clarifies the absorption behavior of micro-droplets of water on treated paper to support the design of functional
microstructures, such as electronics and micro-fluid channels, on paper. The period of time between when a micro-droplet of
water ejected from an ink-jet head lands on the paper’s surface and its complete disappearance by absorption was defined as
the micro-sizing degree (MSD), and an MSD measurement method was established. The MSD was evaluated using microscopic high-speed
video images of the side view recorded every millisecond. Several grades of commercially available ink-jet paper media and
laboratory sheets having different levels of water repellency prepared from a pulp and a sizing agent were examined. The MSD
of commercial ink-jet papers, which are known to absorb water very quickly, was 3–6 ms. Weakly sized laboratory sheets exhibited
a lower MSD of 2–3 ms. The absorption behavior was analyzed in terms of the capillary pressure with and without the Laplace
pressure; the theoretical and experimental results agreed moderately well. The results indicated that the Laplace pressure
cannot be neglected in the analysis. The MSD of a wet surface where a preceding micro-droplet had already landed was higher
than that on a dry or partially wet surface, presumably because water remains inside pores for an unexpectedly long time. 相似文献
