Endonuclease-mediated DNA fragmentation is both an immediate cause and a result of apoptosis and of all other types of irreversible cell death after injury. It is produced by nine enzymes including DNase I, DNase 2, their homologs, caspase-activated DNase (CAD) and endonuclease G (EndoG). The endonucleases act simultaneously during cell death; however, regulatory links between these enzymes have not been established. We hypothesized that DNase I, the most abundant of endonucleases, may regulate other endonucleases. To test this hypothesis, rat kidney tubular epithelial NRK-52E cells were transfected with the DNase I gene or its inactive mutant in a pECFP expression vector, while control cells were transfected with the empty vector. mRNA expression of all nine endonucleases was studied using real-time RT-PCR; DNA strand breaks in endonuclease genes were determined by PCR and protein expression of the enzymes was measured by Western blotting and quantitative immunocytochemistry. Our data showed that DNase I, but not its inactive mutant, induces all other endonucleases at varying time periods after transfection, causes DNA breaks in endonuclease genes, and elevates protein expression of several endonucleases. This is the first evidence that endonucleases seem to be induced by the DNA-degrading activity of DNase I.  相似文献

A coating system for polyester fabrics is developed containing metal effect pigments and a polyurethane binder. The metal pigment is made of copper, is of microscaled size and anisotropic shape. It is originally used for optical modification but can also be used to introduce other functional properties. The polyurethane binder is needed to fix the pigments on the textile surface. The aim of the current investigation is to present that the binder has not only the task to fix the pigment but can be also used to structure the pigment surface. The binder application leads to formation of microstructures on top of the pigment surface, probably as a result of a dewetting process. Based on this microstructured pigment surface, in future, textile coatings for improvement of water-repellent, antimicrobial, and antistatic properties could be developed.  相似文献

Summary: The creation of nanotemplates under dynamic shear flow conditions is described. The structures used are based on the self‐organization of comb‐coil‐like diblock copolymer systems with two intrinsic length scales, resulting from combining diblock copolymers with hydrogen‐bonding amphiphiles. Rheological data of polymeric supramolecular order formation are studied under oscillating shear flow conditions and it was observed that the order of the polymeric nanodomains can be extended to macroscopic distances. It is proposed that the key success factor to obtain highly ordered samples is to perform the orientation process using appropriate rheological conditions (temperature, frequency and strain) selected such that the ratio of the elastic component G′ to the viscous component G″ is larger than 1. This observation was used to select the rheological parameters for the three different samples investigated, where three different types of nanotemplates are observed after extracting the hydrogen‐bonding amphiphile; nanosheets, hollow cylinders and nanofibres. SAXS is primarily used to inspect the resulting structures. Additionally, AFM and TEM were utilized to image the morphologies and to further act as visual proof for the efficiency of the rheological orientation process.

Follicular fluid from bovine follicles at various stages of the estrous cycle and cysts were tested in vitro for immunosuppressive activity. Mitogen-stimulated lymphocyte assays and the mixed lymphocyte reaction were used as test procedures and thymidine uptake was monitored. Follicular fluid and cyst fluid inhibited the deoxyribonucleic acid synthesis response of bovine lymphocytes to mitogenic and antigenic stimulation. The nature of the suppression varied with the mitogen or antigen assay used, with the estrual stage, with size of follicle within the diestrual stage, and with the type of ovarian cyst. Blood sera from estrual or diestrual cows were not immunosuppressive. Heating (56 degrees C for 30 min) follicular fluid from normal follicles and cysts destroyed most of its suppressive activity toward Concanavalin A-stimulated lymphocytes but did not suppress that in luteal cyst fluid. The suppressive activity in follicular fluid toward phytohemagglutinin-stimulated lymphocytes was more stable to heat than that against Concanavalin A-stimulated lymphocytes. Because the immunosuppressive activity in follicular fluid was heat labile, estrogen, progesterone, and other steroids were not the active suppressive molecules. Proposed roles for these immunosuppressive substances are hypothesized in preventing autoantibody formation to the zona pellucida and in the inflammatory reaction leading to ovulation.  相似文献

The Internet provides a universal platform for large-scale distribution of information and supports inter-organizational services, system integration, and collaboration. Use of multimedia documents for dissemination and sharing of massive amounts of information is becoming a common practice for Internet-based applications and enterprises. With the rapid proliferation of multimedia data management technologies over the Internet, there is growing concern about security and privacy of information. Composing multimedia documents in a distributed heterogeneous environment involves integrating media objects from multiple security domains that may employ different access control policies for media objects. In this paper, we present a security model for distributed document management system that allows creation, storage, indexing, and presentation of secure multimedia documents. The model is based on a time augmented Petri-net and provides a flexible, multilevel access control mechanism that allows clearance-based access to different levels of information in a document. In addition, the model provides detailed multimedia synchronization requirements including deterministic and non-deterministic temporal relations and incomplete timing information among media objects.  相似文献

Topology optimization of foundation structures is conducted using a direct limit analysis based optimization approach. Originally developed for plastic topology design of 2D superstructures and industrial structures, which are traditionally designed based on elasticity, the direct limit analysis based design method demonstrated strong intrinsic properties that make of it a promising alternative for topology design. Unlike existing methods the one proposed here is fundamentally conceived and developed in the framework of direct plastic limit analysis which is the natural and usual known framework for the analysis and design of geotechnical structures. The method is extended to handle foundation design by providing for the simultaneous presence of two materials: the soil and the reinforcement, governed by Coulomb failure law. Two types of designs are considered, namely, continuous and discrete. The first allows a range of graded densities of reinforcement that may in practice represent the types of mixtures used in soil improvement, such as cementation. In the discrete design the reinforcement consists exclusively of solid structural elements. A series of two dimensional example problems demonstrate the capabilities of the method to generate rational classical designs or uncommon but intuitively sound foundation configurations that could inspire designers to develop novel solutions.  相似文献

Central Java province plays the main role in the production of food for national stocks in Indonesia, in which Demak is one of the 35 Regencies. Demak Regency had the sixth highest rice productivity in 2013, but at the same time, the seventh highest percentage of child malnutrition. This paradox between high rice productivity and low nutritional status of children triggered an interest in analyzing the correlations between agricultural production of households in the Demak Regency and the prevalence of stunting of children under-five. In December 2014 to February 2015, a cross-sectional survey was carried out in three sub-districts in Demak (Karanganyar, Dempet and Gajah) having the highest rice production and at the same time poor nutritional status of under-five children. The oldest under-five children from farmer family backgrounds were selected. Data on general characteristics, anthropometry of the children, as well as household socio-economics, including agricultural characteristics were obtained. Of the 335 children, 31.9% of them were stunted. A higher proportion of households that grew rice solely or mainly for own consumption (62.4%) and had significantly smaller sized rice-fields (p = 0.041) had stunted rather than non-stunted children. In total, rice production, farm income and agriculture diversity of families with non-stunted children were not significantly different from those of stunted children. However, yearly expenditure for improving family health and environmental conditions were significantly higher among non-stunted than stunted children (p = 0.044). Family decisions about what to spend money on also seemed to influence the nutritional status of the children, rather than nutritional diversity or revenue from agriculture itself. Therefore, improvement of agricultural production and diversity should be accompanied by education on household expenditure.  相似文献
