Self-assembling peptide nanofibrils (PNF) have gained increasing attention as versatile molecules in material science and biomedicine. One important application of PNF is to enhance retroviral gene transfer, a technology that has been central for gene therapy approaches. The best-investigated and commercially available PNF is derived from a 12-mer peptide termed EF-C. The mechanism of transduction enhancement depends on the polycationic surface of EF-C PNF, which bind to the negatively charged membranes of viruses and cells, thereby overcoming electrostatic repulsions and increasing virion attachment and fusion. To better understand how EF-C PNF interact with the cell surface, scanning electron and time-lapse confocal microscopy were performed. The fibrils are found to be actively engaged by cellular protrusions such as filopodia. Consequently, chemical suppression of protrusion formation abrogates fibril binding and virion delivery to the cell surface of immortalized and primary T cells. Vice versa, induction of plasma membrane blebs result in increased fibril binding. Thus, the mechanism of PNF-mediated viral transduction enhancement involves an active engagement of virus-loaded fibrils by cellular protrusions, which may explain its superior performance over soluble transduction enhancers. 相似文献
This paper presents methods used to perform discrete adjoint gradient evaluations for linear stress and vibration analysis. The methods are implemented within the framework of a discrete adjoint structural solver being developed for multidisciplinary adjoint optimizations of turbomachinery components. The code is differentiated using the algorithmic differentiation tool CoDiPack in tandem with manual treatment of the iterative solvers. Stress analysis leads to a linear system of equations that is typically solved by an iterative solver (e.g. GMRES). To ensure accuracy, the adjoint problem is formulated as a new linear system of equations to be solved. Vibration analysis results in a generalized eigenvalue problem that is also typically solved by an interative solver. The adjoint problem takes out the generalized eigenvalue solve and replaces it by one outer product per eigenfrequency, leading to significantly cheap eigenfrequency gradients for vibration analysis.

The process energy demand and the environmental indicators of two carbon fiber reinforced plastic process chains have been investigated. More precisely, the impact of different production set-ups for a standard textile preforming process using bindered non-crimp fabric (NCF) and a material efficient 2D dry-fiber-placement (DFP) process are analyzed. Both 2D preforms are activated by an infrared heating system and formed in a press. The resin-transfer-molding (RTM) technology is selected for subsequent processing. Within a defined process window, the main parameters influencing the process energy demand are identified. Varying all parameters, a reduction of 77% or an increase of 700% of the electric energy consumption compared to a reference production set-up is possible, mainly depending on part size, thickness, and curing time. For a reference production set-up, carbon fiber production dominates the environmental indicators in the product manufacturing phase with a share of around 72–80% of the total global warming potential (GWP). Thus, the reduction of production waste, energy efficient carbon fiber production, and the use of renewable energy resources are the key environmental improvement levers. For the production of small and thin parts in combination with long curing cycles, the influence of the processing technologies is more pronounced. Whereas for a reference production set-up, only 10% (NCF–RTM) and 15% (DFP–RTM) of the total GWP are caused by the processing technologies, a production set-up leading to a high process energy demand results in a share of 40% (NCF–RTM) and 49% (DFP–RTM), respectively. 相似文献
The fate of the green plant pigment chlorophyll (Chl) in de‐greening leaves has long been a fascinating biological puzzle. In the course of the last three decades, various bilin‐type products of Chl breakdown have been identified, named phyllobilins (PBs). Considered ‘mere’ leftovers of a controlled biological Chl detoxification originally, the quest for finding relevant bioactivities of the PBs has become a new paradigm. Indeed, the PBs are abundant in senescent leaves, in ripe fruit and in some vegetables, and they display an exciting array of diverse heterocyclic structures. This review outlines briefly which types of Chl breakdown products occur in higher plants, describes basics of their bio‐relevant structural and chemical properties and gives suggestions as to ‘why’ the plants produce vast amounts of uniquely ‘decorated’ heterocyclic compounds. Clearly, it is worthwhile to consider crucial metabolic roles of PBs in plants, which may have practical consequences in agriculture and horticulture. However, PBs are also part of our plant‐based nutrition and their physiological and pharmacological effects in humans are of interest, as well. 相似文献
Despite recent progress, the application of CRISPR/Cas9 in perennial plants still has many obstacles to overcome. Our previous results with CRISPR/Cas9 in apple and pear indicated the frequent production of phenotypic and genotypic chimeras, after editing of the phytoene desaturase (PDS) gene conferring albino phenotype. Therefore, our first objective was to determine if adding an adventitious regeneration step from leaves of the primary transgenic plants (T0) would allow a reduction in chimerism. Among hundreds of adventitious buds regenerated from a variegated T0 line, 89% were homogeneous albino. Furthermore, the analysis of the target zone sequences of twelve of these regenerated lines (RT0 for “regenerated T0” lines) indicated that 99% of the RT0 alleles were predicted to produce a truncated target protein and that 67% of RT0 plants had less heterogeneous editing profiles than the T0. Base editors are CRISPR/Cas9-derived new genome-editing tools that allow precise nucleotide substitutions without double-stranded breaks. Hence, our second goal was to demonstrate the feasibility of CRISPR/Cas9 base editing in apple and pear using two easily scorable genes: acetolactate synthase—ALS (conferring resistance to chlorsulfuron) and PDS. The two guide RNAs under MdU3 and MdU6 promoters were coupled into a cytidine base editor harboring a cytidine deaminase fused to a nickase Cas9. Using this vector; we induced C-to-T DNA substitutions in the target genes; leading to discrete variation in the amino-acid sequence and generating new alleles. By co-editing ALS and PDS genes; we successfully obtained chlorsulfuron resistant and albino lines in pear. Overall; our work indicates that a regeneration step can efficiently reduce the initial chimerism and could be coupled with the application of base editing to create accurate genome edits in perennial plants. 相似文献
