A new, fast, variation method (FVM) for determining an elastic strip response to stresses arbitrarily distributed on the flat side of the strip is proposed. The remaining surface of the strip may have an arbitrary form, and it is free of stresses. The FVM, as well as the wellknown finite element method, starts with the variational principle. However, it does not use the meshing of the strip. A comparison of FVM results with the exact analytical solution in the special case of shear stresses and a rectangular strip demonstrates an excellent agreement  相似文献

Biosensors are analytical devices that use a biological or biologically derived material immobilized at a physicochemical transducer to measure one or more analytes. Although there are a large number of reviews on biosensors in general, there has been little systematic information presented on the application of natural receptors in sensor technology. This perspective discusses broadly the fundamental properties of natural receptors, which make them an attractive option for use as biorecognition elements in sensor technology. It analyses the current situation by reference to typical examples, such as the application of nicotinic acetylcholine receptor and G protein-linked receptors in affinity sensors and analyses the problems that need to be resolved prior to any commercialization of such devices.  相似文献

We introduce instrumentation and methodology for two-channel chemical cytometry of sister cells-two cells born from division of the same mother cell. The method is based on capillary electrophoresis with laser-induced fluorescence detection and allows simultaneously probing multiple intracellular components in sister cells. To test the new technology, we compared the expression patterns of green fluorescent protein (GFP) between the sisters in cultured cancer cells stably transfected with a GFP-expressing construct. We found that all sister cells had detectable asymmetry in the GFP expression patterns with a confidence level of higher than 95%. To our best knowledge, this is the first reported observation of asymmetric patterns of protein expression in sister cells in a cancer cell line. The proposed technology can reliably detect minor differences in chemical contents between sister cells, which makes it a potentially indispensable tool in studying the molecular mechanisms of developmental processes. It will be especially valuable in quantitative studies of cells with complex proliferation kinetics (e.g., stem cells).  相似文献

Beam intensity scintillations, characterized by a refractive-index structure parameter and caused by variations of macrometeorological features of the coastal atmosphere such as air temperature, wind speed and direction, and relative humidity, are examined theoretically and experimentally. In our theoretical analysis we present two well-known models considered separately for over-water and over-land atmospheric optical communication or imaging channels. By means of comparison with our experiments carried out in midland coastal environments in southern and northern Israel, we show the limitations of the models to predict the refractive-index structure Cn2 parameter for both daytime and nighttime turbulent atmospheres in different coastal zone meteorological conditions. We also present an extension of an existing model with two different practical applications that, as is shown experimentally, can be a good predictor of Cn2 for optical atmospheric paths over midland coastal zones.  相似文献

Study of cell cycle-dependent protein expression is important in oncology, stem cell research, and developmental biology. In this paper, we report the first protein fingerprint from a single cell with known phase in the cell cycle. To determine that phase, we treated HT-29 colon cancer cells with Hoescht 33342, a vital nuclear stain. A microscope was used to measure the fluorescence intensity from one treated cell; in this form of image cytometry, the fluorescence intensity is proportional to the cell's DNA content, which varies in a predictable fashion during the cell cycle. To generate the protein fingerprint, the cell was aspirated into the separation capillary and lysed. Proteins were fluorescently labeled with 3-(2-furoylquinoline-2-carboxaldehyde, separated by capillary sieving electrophoresis, and detected by laser-induced fluorescence. This form of electrophoresis is the capillary version of SDS-PAGE. The single-cell electropherogram partially resolved approximately 25 components in a 30-min separation, and the dynamic range of the detector exceeded 5000. There was a large cell-to-cell variation in protein expression, averaging 40% relative standard deviation across the electropherogram. The dominant source of variation was the phase of the cell in the cell cycle; on average, approximately 60% of the cell-to-cell variance in protein expression was associated with the cell cycle. Cells in the G1 and G2/M phases of the cell cycle had 27 and 21% relative standard deviations in protein expression, respectively. Cells in the G2/M phase generated signals that were twice the amplitude of the signals generated by G1 phase cells, as expected for cells that are soon to divide into two daughter cells. When electropherograms were normalized to total protein content, the expression of only one component was dependent on cell cycle at the 99% confidence limit. That protein is tentatively identified as cytokeratin 18 in a companion paper.  相似文献

We present a new wide-aperture, line-focused ultrasonic material characterization system. The foci of the transmitting and receiving transducers are located in the specimen-immersion liquid interface; and the output voltage V(x, t) of the system is recorded as a function of the lateral position of the receiving transducer. The two-dimensional spectrum of V(x, t) can be expressed as a product of the transfer function of the system and the reflectance function of the interface. In comparison with a system based on scanning in the z direction, the angular resolution of the proposed technique increases with decreasing angle of incidence. There are no geometrical restrictions on the length of the recorded spatial data and the angle of incidence in the case of lateral scanning. The temperature coefficient of the measurement error is low because of the constancy of the propagation distance of ultrasound in the immersion fluid during data acquisition.  相似文献

Reliable, repeatable earthquake forecast is a subject surrounded by controversy and scepticism. What is clear is that reliable forecast would be a critical tool for effective earthquake disaster management. It is proposed that satellites and ground-based facilities may detect earthquake precursors in the ionosphere a few hours or days before the main shock. A low-cost 100 kg class satellite carrying a topside sounder is proposed, to make systematic measurements over seismically active zones. The mission aims to confirm or refute the hypothesis of ionospheric earthquake precursors, define the reliability and reproducibility, and enable further scientific understanding of their mechanisms.  相似文献

A theoretical study was conducted using a multiple-voltage probe sensor for detecting nonconducting inclusions in conducting media. Results show that the multiple-voltage probe sensor is capable of providing precise quantitative measurements of submerged nonconducting objects if the surface voltage response has a standard two-peak form. The standard response is observed for well-localized non-slender single inclusions below the sensor surface. In this case, the peak separation distance is associated with the inclusion depth whereas the peak magnitude is associated with the inclusion volume. Linear dependencies of the inclusion depth and the inclusion volume are observed for a wide variety of inclusion shapes. The predefined form of the surface voltage response makes it feasible to identify useful signal responses at very high noise levels. This is accomplished by using a 2D neural network classifier, based on the probabilistic neural network. A reasonable recognition error of less than 20 % is obtained if the signal-to-noise ratio is larger than or equal to 1/10. A metal casting example shows that the multiple-voltage probe sensor can measure inclusion concentrations in hot conducting melts (gas bubbles and sludge) with inclusion radii in the range from 100 to 1000 m. In contrast to existing particle counter technology, this sensor construction is simple to construct and does not require special aperture and vacuum treatment.  相似文献
