Food Safety Objectives (FSOs) have been proposed as a practical tool to translate public health targets for food safety into tolerable levels of pathogens in a food product. The FSO concept is subject to intensive debate, and has not been developed in detail. We evaluate the proposed definition of FSOs and their implementation from the perspective of Quantitative Microbiological Risk Assessment (QMRA). The relationship between QMRA and FSOs is illustrated by a model for the public health risk of Shiga-producing Escherichia coli in steak tartare. We conclude that the proposed definition of FSOs needs to be modified to properly account for variability in and uncertainty about the contamination of food with pathogenic microorganisms and emphasize that both prevalence and concentration of pathogens must be considered. For this purpose, we propose the P-D equivalence curve, a simple graphical tool to separate "tolerable" from "non-tolerable" combinations of prevalence and concentration (dose).  相似文献

Quantitative structure-activity relationships (QSARs) have proved increasingly useful for predicting the biological activities of molecules (e.g., their binding affinities to different receptors) and can be used in environmental chemistry as a preliminary tool for screening the activities of untested molecules, producing valuable information on which compounds should be tested more thoroughly with experimental affinity assays or in animals. The predictive ability of the consensus kNN QSAR method is corroborated here using a diverse set of 245 compounds, which have been assayed for their relative binding affinities to the estrogen receptor of four species: human (ER alpha and ER beta), calf, mouse, and rat. Leave-one-out cross-validation (LOO-CV) and gamma-randomization tests were applied to the QSAR models for internal validation, and separate training and test sets were used for external validation. The internal predictive abilities of the consensus models for all five data sets were convincing, with cross-validated correlation coefficients (LOO-CV q2 values) varying from 0.69 (human ER beta data) to 0.79 (human ER alpha data). The external predictive abilities were also encouraging, as the predictive r2 scores (pr-r2 values) varied from 0.62 (human ER beta data) to 0.77 (calf and mouse data). The results indicate that consensus kNN QSAR is a feasible method for rapid screening of the estrogenic activity of organic compounds.  相似文献

The biosolids accumulation and biodegradation of domestic wastewater treatment plant (DWTP) sludge by filamentous fungi have been investigated in a batch fermenter. The filamentous fungi Aspergillus niger and Penicillium corylophilum isolated from wastewater and DWTP sludge was used to evaluate the treatment performance. The optimized mixed inoculum (A. niger and P. corylophilum) and developed process conditions (co-substrate and its concentration, temperature, initial pH, inoculum size, and aeration and agitation rate) were incorporated to accelerate the DWTP sludge treatment process. The results showed that microbial treatment of higher strength of DWTP sludge (4% w/w of TSS) was highly influenced by the liquid state bioconversion (LSB) process. In developed bioconversion processes, 93.8 g/kg of biosolids was enriched with fungal biomass protein of 30 g/kg. Enrichment of nutrients such as nitrogen (N), phosphorous (P), potassium (K) in biosolids was recorded in 6.2% (w/w), 3.1% (w/w) and 0.15% (w/w) from its initial values of 4.8% (w/w), 2.0% (w/w) and 0.08% (w/w) respectively after 10 days of fungal treatment. The biodegradation results revealed that 98.8% of TSS, 98.2% of TDS, 97.3% of turbidity, 80.2% of soluble protein, 98.8% of reducing sugar and 92.7% of COD in treated DWTP sludge supernatant were removed after 8 days of microbial treatment. The specific resistance to filtration (SRF) in treated sludge (1.4x10(12) m/kg) was decreased tremendously by the microbial treatment of DWTP sludge after 6 days of fermentation compared to untreated sample (85x10(12) m/kg).  相似文献

Potassium (K) content in 408 male workers of a rare earths processing plant was estimated using a shadow shield whole body counter. The average K content in the non-vegetarian and vegetarian group was found to be 1.5 g kg(-1) and 2.1 g kg(-1) respectively. The absorbed dose due to 40K is determined as 0.18 +/- 0.02 mGy. While the K content was found to be proportional to body build index, it is inversely proportional to slenderness. Body K was maximum in the middle aged group (35-45 y) and a minimum in younger and older persons. The correlation of potassium content with some disease patterns was also studied. Diabetes mellitus patients were observed to have low K content of 0.9 +/- 0.2 g kg(-1) and those who suffer from cardiovascular disorders were found to have high K content 2.7 +/- 0.3 g kg(-1). The studies showed that the depletion of body K content takes place a few years prior to the clinical detection of diabetes and it builds up a few years before the clinical detection of cardiovascular disorders. It is stressed that the medical information is preliminary as the number of subjects studied was not very large. Wider and in depth study by various groups can hopefully strengthen the observation.  相似文献

Design and functional aspects of PANSPEC, a panoramic-imaging chemical vapor sensor (PANSPEC is an abbreviation for infrared panoramic-viewing spectroradiometer), were advanced and its optical system reoptimized accordingly. The PANSPEC model unites camera and fused solid-state interferometer and photopolarimeter subsystems. The camera is an eye of the open atmosphere that collects, collimates, and images ambient infrared radiance from a panoramic field of view (FOV). The passive interferometer rapidly measures an infrared-absorbing (or infrared-emitting) chemical cloud traversing the FOV by means of molecular vibrational spectroscopy. The active photopolarimeter system provides a laser beam beacon. This beam carries identification (feature spectra measured by the interferometer) and heading (detector pixels disclosing these feature spectra) information on the hazardous cloud through a binary encryption of Mueller matrix elements. Interferometer and photopolarimeter share a common configuration of photoelastic modulation optics. PANSPEC was optimized for minimum aberrations and maximum resolution of image. The optimized design was evaluated for tolerances in the shaping and mounting of the optical system, stray light, and ghost images at the focal plane given a modulation transfer function metric.  相似文献

Heterosis and breed differences were estimated for milk yield traits, somatic cell score (SCS), and productive life (PL), a measure of longevity. Yield trait data were from 10,442 crossbreds and 140,421 purebreds born since 1990 in 572 herds. Productive life data were from 41,131 crossbred cows and 726,344 purebreds born from 1960 through 1991. The model for test-day yields and SCS included effects of herd-year-season, age, lactation stage, regression on sire's predicted transmitting ability, additive breed effects, heterosis, and recombination. The model for PL included herd-year-season, breed effects, and general heterosis. All effects were assumed to be additive, but estimates of heterosis were converted to a percentage of the parent breed average for reporting. Estimates of general heterosis were 3.4% for milk yield, 4.4% for fat yield, and 4.1% for protein yield. A coefficient of general recombination was derived for multiple-breed crosses, but recombination effects were not well estimated and small gains, not losses, were observed for yield traits in later generations. Heterosis for SCS was not significant. Estimated heterosis for PL was 1.2% of mean productive life and remained constant across the range of birth years. Protein yield of Brown Swiss x Holstein crossbreds (0.94 kg/d) equaled protein yield of purebred Holsteins. Fat yields of Jersey x Holstein and Brown Swiss x Holstein crossbreds (1.14 and 1.13 kg/d, respectively) slightly exceeded that of Holsteins (1.12 kg/d). With cheese yield pricing and with all traits considered, profit from these crosses exceeded that of Holsteins for matings at breed bases. For elite matings, Holsteins were favored because the range of evaluations is smaller and genetic progress is slower in breeds other than Holstein, in part because fewer bulls are sampled. A combined national evaluation of data for all breeds and crossbreds may be desirable but would require an extensive programming effort. Animals should receive credit for heterosis when considered as mates for another breed.  相似文献
