Like other chromatographic techniques, retention factor, k, in micellar electrokinetic chromatography (MEKC) is directly related to solute partition coefficient and the chromatographic phase ratio as k = Kphi. Unlike conventional chromatography, however, the phase ratio and partition coefficient can be accurately determined in MEKC for a given micellar pseudostationary phase. This means that retention factor in MEKC can be predicted for solutes with known micelle-water partition coefficients without any prior experimentation. In this paper, the use of this simple relationship for prediction of retention behavior in MEKC is examined. The principle of additivity of functional group contribution to partitioning is used to calculate the micelle-water partition coefficient, Kmw, for SDS micellar pseudophase. The micellar substituent constants for 20 functional groups (training set) were determined. Using these substituent constants, the Kmw and retention factors for a group of 80 neutral solutes (test set) were predicted. The linear plot of predicted versus observed log k had an R2 = 0.97 and a slope equal to 1.01. It is shown that the retention times (thus chromatograms) in MEKC can be predicted from the calculated retention factors after only one initial experiment to measure teo and t(mc) under the experimental conditions.  相似文献

Newly developed software additions to the three-dimensional dynamic image analysis system, 3D-DIAS, are described for simultaneously reconstructing and motion analyzing in three dimensions the outer surface, nucleus and pseudopods of living, crawling cells. This new system is then used to describe for the first time a nuclear behavior cycle in translocating Dictyostelium discoideum amoebae and to investigate the role of pseudopod extension in this process. The nuclear behavior cycle is tuned to the two phases of the general cell behavior cycle [Wessels et al., 1994], and includes nuclear migration both in the z- and in the x,y-axes from the proximal border of the prior anterior pseudopod to the proximal border of a newly expanding anterior pseudopod. Nuclear migration is cued by pseudopod-substratum contact, achieves velocities in excess of 50 microm/min, and is accompanied by characteristic changes in nuclear shape. The rules and characteristics of nuclear behavior are demonstrated to be intact in two mutants affecting pseudopod formation, a myosin IB null mutant (myoB-) and a myosin II heavy chain phosphorylation mutant (3XALA). The rules and characteristics of nuclear migration, however, are disrupted upon dissolution of microtubules by colcemid. Together the above results demonstrate that the newly developed 3D-DIAS system can be used to gain new insights into the dynamic changes in the intracellular 3D architecture associated with cellular translocation.  相似文献

Anger may have greater effects on chronic pain severity than other negative emotions and may do so by increasing muscle tension near the site of injury (symptom-specific reactivity). For patients with chronic low back pain (CLBP), relevant muscles are lower paraspinals (LP). Ninety-four CLBP patients and 79 controls underwent anger and sadness recall interviews. EMG and cardiovascular activity were recorded. Patients exhibited greater LP tension increases during anger and slower recovery than controls. Only patients showed greater LP reactivity during anger than sadness. For both groups, trapezius reactivity during anger and sadness did not differ. LP reactivity to anger correlated with everyday pain severity for patients. Anger-induced symptom-specific LP reactivity may be linked to chronic pain aggravation among CLBP patients. Ras associated with diabetes (Rad), a new ras-related GTPase, was recently identified by subtractive cloning as an mRNA in skeletal muscle that is overexpressed in NIDDM. To better understand its metabolic significance, we measured skeletal muscle Rad expression in well-characterized insulin sensitive (IS) and insulin resistant (IR) subjects with normal glucose tolerance and in untreated NIDDM patients. We found no differences in expression of Rad mRNA levels among IS, IR, and NIDDM groups using a ribonuclease protection assay (0.22 +/- 0.06, 0.13 +/- 0.01, and 0.16 +/- 0.02 relative units, respectively; NS) and no differences in Rad protein expression using a specific anti-peptide Rad antibody (1.05 +/- 0.18, 1.14 +/- 0.08, and 1.08 +/- 0.21 units/mg protein, respectively; NS). However, Rad protein levels were positively correlated with BMI (r = 0.43, P = 0.03) and percentage body fat (r = 0.55, P < 0.005), two independent measures of obesity, and negatively correlated with resting metabolic rate (r = 0.49, P = 0.01). In multiple regression analyses, percentage body fat and resting metabolic rate independently accounted for 30 and 10% of individual variability in muscle Rad protein expression. In conclusion, Rad expression in skeletal muscle is not altered as a function of insulin resistance or NIDDM in humans. However, these data, for the first time, implicate a role for Rad in regulating body composition and energy expenditure and provide a framework for studies designed to elucidate Rad's cellular functions.  相似文献

Field-programmable gate arrays (FPGAs) are an important implementation medium for digital logic. Unfortunately, they currently suffer from poor silicon area utilization due to routing constraints. In this paper we present Triptych, an FPGA architecture designed to achieve improved logic density with competitive performance. This is done by allowing a per-mapping tradeoff between logic and routing resources, and with a routing scheme designed to match the structure of typical circuits. We show that, using manual placement, this architecture yields a logic density improvement of up to a factor of 3.5 over commercial FPGAs, with comparable performance. Field-programmable gate arrays (FPGAs) are becoming an increasingly important implementation medium for digital logic. One of the most important keys to using FPGAs effectively is a complete, automated software system for mapping onto the FPGA architecture. Unfortunately, many of the tools necessary require different techniques than traditional circuit implementation options, and these techniques are often developed specifically for only a single FPGA architecture. In this paper we describe automatic mapping tools for Triptych, an FPGA architecture with improved logic density and performance over commercial FPGAs. These tools include a simulated-annealing placement algorithm that handles the routability issues of fine-grained FPGAs, and an architecture-adaptive routing algorithm that can easily be retargeted to other FPGAs. We also describe extensions to these algorithms for mapping asynchronous circuits to Montage, the first FPGA architecture to completely support asynchronous and synchronous interface applications  相似文献

A telescope on the Moon is needed for astronomy and can be constructed in this decade or early in the next century. Design for this telescope will be fundamentally different from the design of free‐flying telescopes. Its design will be more like the new Keck telescope being completed on a mountaintop in Hawaii than the Hubble Space Telescope, in low Earth orbit. Success of the lunar‐based telescope will depend on an appropriately engineered structure, a suitable interface (foundation) in the lunar soil, and a carefully thought out construction process. Participation of engineers in identifying and resolving issues for this extraterrestrial engineering and construction project is a natural extension of the traditional engineering role, and will prepare the engineering and construction communities for the subsequent greater challenges associated with basing on the Moon. These communities need to document now the types of data and information that NASA should obtain in the next early lunar missions so that construction on the Moon will be facilitated.  相似文献

This paper describes early planning stages of a nurse-managed clinic to offer primary health care services in a nontraditional university setting. A community needs assessment was conducted with quantitative and qualitative findings reported. This unique setting triggered a number of considerations in planning for services.
