Catechol-O-methyltransferase-deficient mice exhibit sexually dimorphic changes in catecholamine levels and behavior

Catechol-O-methyltransferase (COMT) is one of the major mammalian enzymes involved in the metabolic degradation of catecholamines and is considered a candidate for several psychiatric disorders and symptoms, including the psychopathology associated with the 22q11 microdeletion syndrome. By means of homologous recombination in embryonic stem cells, a strain of mice in which the gene encoding the COMT enzyme has been disrupted was produced. The basal concentrations of brain catecholamines were measured in the striatum, frontal cortex, and hypothalamus of adult male and female mutants. Locomotor activity, anxiety-like behaviors, sensorimotor gating, and aggressive behavior also were analyzed. Mutant mice demonstrated sexually dimorphic and region-specific changes of dopamine levels, notably in the frontal cortex. In addition, homozygous COMT-deficient female (but not male) mice displayed impairment in emotional reactivity in the dark/light exploratory model of anxiety. Furthermore, heterozygous COMT-deficient male mice exhibited increased aggressive behavior. Our results provide conclusive evidence for an important sex- and region-specific contribution of COMT in the maintenance of steady-state levels of catecholamines in the brain and suggest a role for COMT in some aspects of emotional and social behavior in mice.     

Assessing Consequences of Substance Use: Psychometric Properties of the Inventory of Drug Use Consequences.

Negative consequences are an important component of the substance use change process, yet no standardized measure exists to assess consequences of use. This study evaluated the psychometric properties of the Inventory of Drug Use Consequences, a self-report measure assessing drinking and drug use consequences. Participants (N=252) were assessed at entry into outpatient substance abuse treatment. Items loaded strongly on 1 factor, and internal consistency of the total scale was high. Fifteen items with the highest correlations with the total scale were chosen to represent the construct. The shortened measure exhibited good internal consistency, convergent and discriminant validity, and detected change over time and may be the measure of substance use consequences that is lacking in the field. Electro-optical characteristics of indium tin oxide (ITO) films: effect of thermal annealing

The effect of thermal annealing on the electrical and optical characteristics of ITO films prepared by reactive sputtering and thermal evaporation have been studied. The effect of the thermal annealing is to improve the conductivity and the optical transmission in the shorter wavelength region. The conductivity of the films increases with annealing temperature, this behaviour is associated with grain growth in the film.     

Eigenmode analysis of dielectric loaded top-hat monopole antennas

Eigenmode expansions are used for computing the currents on a dielectric loaded top-hat monopole radiating above an infinite conducting ground plane. The approach is facilitated by adding a parallel ground plane above the antenna, thus allowing use of cylindrical harmonic field expansions in each of three regions. Expansion coefficients are found by enforcing boundary and continuity conditions at conducting surfaces and regional interfaces. Convergence and accuracy are assessed using comparisons with three other methodologies, namely an integral equation solution, a quasi-static analysis, and multi-octave experimental measurements     

200°C, 96-nm wavelength range, continuous-wave lasing fromunbonded GaAs MOVPE-grown vertical cavity surface-emitting lasers

We report record temperature and wavelength range attained using MOVPE-grown AlGaAs vertical cavity surface-emitting lasers (VCSEL's). Unbonded continuous-wave lasing is achieved at temperatures up to 200°C from these top-emitting VCSEL's and operation over a 96-nm wavelength regime near 850 nm is also achieved from the same nominal design. Temperature and wavelength insensitive operation is also demonstrated; threshold current is controlled to within a factor of 2 (2.5-5 mA) for a wavelength range exceeding 50 nm and to within ±30% (5-10 mA) for a temperature range of 190°C at 870 nm     

Meat tenderness and the calpain proteolytic system in longissimus muscle of young bulls and steers

The objectives of this study were to examine the effects of castration on the calpain proteinase system (mu-calpain, m-calpain, and calpastatin) activities and meat tenderness. Six each, MARC III bulls and steers were slaughtered at approximately 12 mo of age. Longissimus muscle samples were obtained for determining myofibril fragmentation index, Warner-Bratzler shear force, and sensory panel evaluation at 1, 7, and 14 d postmortem, and mu- and m-calpain and calpastatin activities at 24 h postmortem. Bulls produced leaner carcasses with lower (P < .05) quality grades than did steers. Meat from bulls had higher (P < .05) shear force values than meat from steers; however, sensory panelists were unable (P > .05) to detect differences in tenderness or other sensory traits between bulls and steers. Activities of mu- and m-calpain were not affected (P > .05) by castration; however, calpastatin was higher (P < .05) in muscles from the bull carcasses. Lower (P < .05) myofibril fragmentation index values indicate that less proteolysis occurred in muscle from bulls than in muscle from steers during the first 7 d postmortem. Greater calpastatin 24-h activity may be associated with the increased shear force of meat from bulls.     

Validation of an inhibition ELISA using a monoclonal antibody for foot-and-mouth disease (FMD) primary diagnosis

An inhibition ELISA (IH-ELISA) test for foot-and-mouth disease virus (FMDV) was validated using 106 epithelial samples from suspected cases of FMD in Argentina submitted to the Argentine National Diagnostics Laboratory (GELAB) over a period of 12 months and examined in parallel with the complement fixation test (CFT). IH-ELISA was found to be more sensitive, detecting 25% (26 samples) more FMDV positives than the CFT in original suspensions of field samples. The effect of storage conditions on 12S stability was examined. Plates stored at 4 degrees C blocked with 1% ovalbumin and plates stored at -20 degrees C with or without blocking buffer could be used for at least 90 days. When various brands of polystyrene plates were compared for 12ps adsorption it was found that those microplates of higher binding capacity were more efficient.     

Factors influencing learning through play in ICT settings

Using a mixed method approach of questionnaires, observations and field notes, the authors have studied a number of settings during the past two years which have focussed on the development of ICT capability through play. Some of these have involved children identified as disaffected or disadvantaged, whilst others have involved initial teacher education students. In this paper, we consider some of the results obtained from the most recent of these settings, relate these to those obtained from the other settings, and explore further a number of inter-related issues which have arisen from the analysis. We particularly focus on the effect of learners’ self-esteem and self-efficacy; the influence of gender-related characteristics; the role of interaction between learners during activity; and the value of reflection.
