Blooms of toxic cyanobacteria such as Microcystis aeruginosa periodically occur within wastewater treatment lagoons in the warmer months, and may consequently cause contamination of downstream water and outages of the supply of recycled wastewater. Lab-scale sonication (20 kHz) was conducted on suspensions of M. aeruginosa isolated from a wastewater treatment lagoon, and two other algal strains, Anabaena circinalis and Chlorella sp., to investigate cell reduction, growth inhibition, release of microcystin and sonication efficiency in controlling the growth of the M. aeruginosa. For M. aeruginosa, for all sonication intensities and exposure times trialled, sonication led to an immediate reduction in the population, the highest reduction rate occurring within the initial 5 min. Sonication for 5 min at 0.32 W/mL, or for a longer exposure time (>10 min) at a lower power intensity (0.043 W/mL), led to an immediate increase in microcystin level in the treated suspensions. However, prolonged exposure (>10 min) to sonication at higher power intensities reduced the microcystin concentration significantly. Under the same sonication conditions, the order of decreasing growth inhibition of the three algal species was: A. circinalis > M. aeruginosa > Chlorella sp., demonstrating sonication has the potential to selectively remove/deactivate harmful cyanobacteria from the algal communities in wastewater treatment lagoons.  相似文献

Abstract: The production of grape seed flour (GSF) from a waste product generated during winemaking, is of interest in product development applications due to its potential health benefits. However, before GSF can used in baking as a source of additional antioxidants, research on its heat stability is required. The overall objective of this study was to assess changes in phenolic content and antioxidant activity of GSF during heating. Merlot GSF was heated at 5 temperatures (120 to 240 °C) for 0 to 90 min. At each time/temperature combination, total phenolic content (TPC), total flavanoid content (TFC), Trolox equivalent antioxidant capacity (TEAC), 2,2 diphenyl‐1‐picrylhydrazyl (DPPH) radical scavenging, and ferric ion reducing antioxidant power (FRAP) of the extracts were determined. Specific polyphenolic compounds, including catechin, gallocatechin, epicatechin, and gallic acid were also measured. Results showed that when Merlot GSF was heated to ≥180 °C, significant decreases in the TPC and antioxidant activity, measured using FRAP, DPPH, and TEAC, were observed. Longer heating times also caused a reduction in antioxidant capability. Catechin and epicatechin content decreased with increasing heating temperature while gallocatechin and gallic acid content increased. Both catechin and epicatechin content had strong positive correlations (r > 0.91) with TPC and TFC, as well with FRAP and TEAC, suggesting that the GSF antioxidant activity is related to the presence of these particular compounds. Overall, while a decrease in antioxidant content was observed during heating, this occurred at higher baking temperatures. Thus GSF may be suitable for use as an ingredient in baked goods to bolster antioxidant content. Practical Application: In order for Merlot grape seed flour (GSF) to be used in baking as a source of additional antioxidant compounds, the impact of heating on the polyphenolic compounds in the GSF needed to be examined. Thermal treatment of Merlot GSF caused significant decreases in the TPC, antioxidant power, and specific polyphenolic compounds when heated ≥180 °C. Thus while antioxidant content decreased with higher heating temperatures, this occurred at higher baking temperatures. Thus, GSF may be suitable for use as an ingredient in baked goods to bolster antioxidant content.  相似文献

The mechanism by which soya lipoxygenase enzyme action improves the Theological properties of wheat flour doughs during mechanical development in air has been investigated further. Free-lipid extraction, reconstitution and replacement experiments have shown that the rheological effect of lipoxygenase action, which is consistent with an oxidative improvement of the dough proteins and may also result in extended mixing tolerance, only occurred in the presence of an oxidisable, polyunsaturated, free-lipid substrate. Addition of this substrate in an oxidised state (produced either by autoxidation or enzyme-oxidation) to doughs mixed from fat-extracted flour under nitrogen resulted only in a small rheological improvement, greater for the autoxidised than the enzyme-oxidised lipid, but in no way comparable with the large rheological effect of lipoxygenase action during dough mixing in air. Furthermore, the presence of an antioxidant, nordihydroguaiaretic acid (NDGA), during dough development, although greatly inhibiting peroxide formation, only marginally impaired the rheological improvement due to lipoxygenase action. Additional evidence is therefore provided for a coupled oxidation mechanism being responsible for the rheological effect, since lipoxygenase-catalysed oxidation actively occurring in the dough during mixing appears to be the fundamental requirement, irrespective of whether the primary oxidation products lead to lipid peroxides or oxidised NDGA.  相似文献

The first self-assembled InAs quantum dash lasers grown by molecular beam epitaxy on InP (001) substrates are reported. Pulsed room-temperature operation demonstrates wavelengths from 1.60 to 1.66 μm for one-, three-, and five-stack designs, a threshold current density as low as 410 A/cm2 for single-stack uncoated lasers, and a distinctly quantum-wire-like dependence of the threshold current on the laser cavity orientation. The maximal modal gains for lasing in the ground-state with the cavity perpendicular to the dash direction are determined to be 15 cm-1 for single-stack and 22 cm-1 for five-stack lasers  相似文献

1.75 W CW power in AlGaInAs-InP strained QW lasers is demonstrated. Room temperature threshold current densities are 410 A/cm/sup 2/, and the characteristic temperature is 69 K. Natural Organic Matter (NOM) from Myponga Reservoir, South Australia, was separated into four organic fractions based on their hydrophobic and hydrophilic properties using a sequence of non-ionic and ionic resins. NOM fractions were isolated for the purpose of determining the impact of alum coagulation on removal of these fractions in conventional water treatment, and their potential as precursors in the formation of disinfection by-products (DBP) and in supporting microbial growth. The NOM comprised VHA (very hydrophobic acids), SHA (slightly hydrophobic acids), CHA (charged hydrophilics) and NEU (neutral hydrophilics) fractions. These fractions were then jar tested with alum using low (50 mg/L), operational (100 mg/L) and very high (200 mg/L) doses to assess the removal capacities for these fractions in a conventional treatment plant. High-performance size exclusion chromatography-UV-DOC (HPSEC-UV-DOC) revealed that alum removed more of the hydrophobic and higher molecular weight components of NOM, but less of the NEU fraction and lower molecular weight components of NOM. Determination of biodegradable dissolved organic carbon (BDOC) indicated that the NEU fraction had the highest biodegradability, followed by the CHA, SHA and VHA fractions. The VHA fraction had the highest total-trihalomethane formation potential (t-THMFP), followed by NEU, SHA and CHA. The NOM not removed by alum coagulation had the potential to support microbial growth (NEU fraction), and disinfection by-product (DBP) formation (VHA and NEU fractions). To obtain treated water with lower overall residual NOM, other treatment methods would need to be applied in addition to alum coagulation in order to reduce the concentration of the neutral fraction.  相似文献

Treatment of cultured carrot cells with dibutyryl cAMP or forskolin resulted in the appreciable decrease in extracellular K+ concentration. This decrease was found to be transient and the concentration of the ion in the culture medium restored to the original level within few minutes. The cAMP-induced decrease in K+ level in the medium was almost completely inhibited when carrot cells were incubated in the presence of K+ channel blockers, CsCl and tetraethylammonium chloride. Appreciable amounts of 45Ca2+ were discharged from 45Ca2+-loaded inside-out vesicles of carrot plasma membrane by the stimulation with cAMP, however, the release of the ion was significantly inhibited in the presence of the K+ channel blockers. The release of 45Ca2+ from the vesicles was also observed when K+ current was evoked with an ionophore, valinomycin, even in the absence of cAMP. These results suggest that the gating of some of the inward K+ channels located at plasma membrane of cultured carrot cells is controlled by cytoplasmic concentration of cAMP and the inward K+ current across the plasma membrane induced by the nucleotide elicits Ca2+ influx into the cells possibly by the activation of voltage-dependent Ca2+ channels.  相似文献

In four experiments we examined the relative contribution of different impulse parameters to response outcome variability in a unidimensional timing task. The findings indicate that (a) there is not a linear and proportional relation between force and force variability; (b) low rates of force production are proportionally more variable than high rates of force production; (c) adding a preload to the limb induces effects on movement error different from those induced by the addition of mass to the manipulandum; and (d) the space-time error function for movement control is strongly linked to the average rate of force production. Collectively, these findings indicate limitations to the extant impulse-variability models and suggest that rate of force production is an important parameter determining response outcome variability. Resistance of tumour cells to methylating and monochloroethylating agents in vitro and in vivo has been linked to levels of the DNA repair protein O6-methylguanine-DNA methyltransferase (MGMT). In a clinical trial of temozolomide in advanced malignant melanoma, the relationship between pretreatment MGMT levels in biopsies of cutaneous tumours and involved lymph nodes and clinical response to the drug has been studied. Among 50 evaluable patients, there were three complete responses (CR), four partial responses (PR), six with stable disease (SD) and 37 with progressive disease (PD), with an overall response rate of 14%. In 33 patients in whom MGMT level and clinical response could be evaluated, the tumour MGMT levels (fmol mg(-1) protein) were: CR, 158 +/- 119; PR, 607 +/- 481; NC, 171 +/- 101; PD, 185 +/- 42.3. Thus, measurements of pretreatment levels of MGMT in melanoma did not predict for response to temozolomide.  相似文献
