DNA biosensor for the detection of hydrazines   总被引:1,自引:0,他引:1  
A double-stranded (ds) DNA-coated carbon paste electrode is employed as a remarkably sensitive biosensor for the detection of hydrazine compounds. The sensor relies on monitoring changes in the intrinsic anodic response of the surface-confined DNA resulting from its interaction with hydrazine compounds and requires no label or indicator. Short reaction times (1-10 min) are sufficient for monitoring part-per-billion levels of different hydrazines. Applicability to untreated natural water samples is illustrated. The response mechanism is discussed, along with prospects of using DNA biosensors for quantitaing other important molecules and elucidating DNA interactions and damage.  相似文献

Light and transmission electron microscopy techniques have been applied in observations of hypervelocity impact craters in two different copper targets: a 38 m grain size mill-processed target, and a 763 m grain size annealed target, the smaller grained target being impacted with a 1100 aluminium sphere and the larger grained target being impacted with a soda-lime glass sphere, at velocities near 6 km s–1. Both target craters exhibited dynamic recrystallization near the crater wall. The jetting associated with these two craters was very different. Considerably more plastic flow and a larger rim characterized the larger grained target. No significant melt-related phenomena were observed either near the crater wall or in the jetted rim for either crater. Consequently, the principal features of crater formation involve extreme plastic flow in the solid state. Microbands were observed to occur profusely in a zone below the smaller grained mill-processed target crater while more profuse and extremely long, unidirectional bundles of microbands (which were coincident with traces of {1 1 1} planes) occurred below the annealed larger grained target crater. These observations attest to the dominant and unique role played by deformation microbands in cratering in copper, because essentially no deformation twins were observed in either target.  相似文献

Extra-contractual referrals (ECRs) can be a cause of considerable anxiety to purchasing authorities, mainly because of their potential to generate unexpected expenditure. But ECRs can also be used as a tool for monitoring the demand for, and quality of, clinical services. ECRs were studied in the Darlington Health Authority district using a variety of methods including inter-disciplinary meetings, a series of interviews with local GPs, and a questionnaire to general practitioners on 230 consecutive ECRs. The methods and results of the questionnaire study are presented. The commonest reasons for making ECRs included the mistaken belief that a contract existed with the ECR provider, patient dissatisfaction with the local provider, and referral to benefit from shorter waiting lists. ECRs for bone-mass densitometry, orthopaedics, and ear nose and throat services were over-represented. Questionnaire results were validated by comparison with an interview study of all GPs in the district. We conclude that trends in ECRs can be monitored as a convenient "early warning system' to alert purchasing authorities to changes in demand or perceived problems with local provider units. ECR data must be interpreted in the context of further local background information from sources such as GPs and public health physicians. In the case of Darlington, scrutiny of ECRs has led to changes in services and contracts.  相似文献

LytA amidase is the best known bacterial autolysin. It breaks down the N-acetylmuramoyl-L-alanine bonds in the peptidoglycan backbone of Streptococcus pneumoniae and requires the presence of choline residues in the cell-wall teichoic acids for activity. Genetic experiments have supported the hypothesis that its 36-kDa chain has evolved by the fusion of two independent modules: the NH2-terminal module, responsible for the catalytic activity, and the COOH-terminal module, involved in the attachment to the cell wall. The structural organization of LytA amidase and of its isolated COOH-terminal module (C-LytA) and the variations induced by choline binding have been examined by differential scanning calorimetry and analytical ultracentrifugation. Deconvolution of calorimetric curves have revealed a folding of the polypeptide chain in several independent or quasi-independent cooperative domains. Elementary transitions in C-LytA are close but not identical to those assigned to the COOH-terminal module in the complete amidase, particularly in the absence of choline. These results indicate that the NH2-terminal region of the protein is important for attaining the native tertiary fold of the COOH terminus. Analytical ultracentrifugation studies have shown that LytA exhibits a monomer <--> dimer association equilibrium, through the COOH-terminal part of the molecule. Dimerization is regulated by choline interaction and involves the preferential binding of two molecules of choline per dimer. Sedimentation velocity experiments give frictional ratios of 1.1 for C-LytA monomer and 1.4 for C-LytA and LytA dimers; values that deviated from that of globular rigid particles. When considered together, present results give evidence that LytA amidase might be described as an elongated molecule consisting of at least four domains per subunit (two per module) designated here in as N1, N2, C1, and C2. Intersubunit cooperative interactions through the C2 domain in LytA dimer occur under all experimental conditions, while C-LytA requires the saturation of low affinity choline binding sites. The relevance of the structural features deduced here for LytA amidase is examined in connection with its biological function.  相似文献

Changes in steady-state fluorescence anisotropy of 1 -(4-trimethylaminophenyl)-6-phenyl-1,3,5-hexatriene TMA-DPH) are applied to the detection of lamellar-hexagonal transitions in egg phosphatidylethanolamine. Even low (2 mole%) proportions of diacylglycerol decrease the hexagonal transition temperature considerably, as confirmed by differential scanning calorimetry. Diacylglycerol is also found to promote a lamellar to "isotropic" (Q(224) cubic) transition in mixtures of phosphatidylcholine: phosphatidylethanolamine:cholesterol. This nonreversible transition is also observed by (31)P nuclear magnetic resonance and detected as a large increase in TMA-DPH steady-state anisotropy. The same technique reveals as well that lysophosphatidylcholine counteracts the effect of diacylglycerol and stabilizes the lamellar phase in both transitions. Diacylglycerol and lysophosphatidylcholine are known to respectively promote and inhibit membrane fusion in a variety of systems. These data are interpreted in support of the hypothesis of a highly bent structural fusion intermediate ("stalk"). They also show the interest of lipid-phase studies in predicting and rationalizing membrane fusion mechanisms.  相似文献

PURPOSE: We investigated the changes in sexuality and quality of life that evolve after lower urinary tract reconstruction in neurologically impaired women previously treated with an indwelling urethral catheter. MATERIALS AND METHODS: A total of 18 neurologically impaired women treated with an indwelling urethral catheter underwent bladder reconstruction. Pubovaginal sling urethral compression was required to restore perineal dryness in 13 patients and was the only operation required in conjunction with intermittent catheterization in 3. Eight patients underwent ileocystostomy, that is creation of a "bladder chimney," and 4 underwent augmentation cystoplasty with creation of a continent catheterizable stoma. In 3 patients ileocystoplasty alone with intermittent urethral catheterization was performed. All patients were followed 6 to 40 months (mean 18) after reconstructive surgery using a 9-part questionnaire to score numerically the effect of surgical reconstruction on sexuality and quality of life issues. RESULTS: On a scale of 0 (worst) to 5 (best) mean score for self-esteem improved from 1 preoperatively to 4 postoperatively, self-image from 1 to 4, sexual desire from 2 to 4 and ability to cope with disability from 1 to 4, respectively. In 4 of the 15 women who were sexually active preoperatively the frequency of sexual intercourse doubled from a mean of 3 to 6 times per month, respectively, and all 4 women reported improved sexual satisfaction. All 13 patients with pelvic pain and 5 with symptoms of autonomic dysreflexia noticed significant improvement if not complete resolution of the symptoms. CONCLUSIONS: Neurogenic lower urinary tract dysfunction treated with an indwelling urethral catheter is detrimental to sexuality and quality of life in neurologically impaired women. Urinary tract reconstruction restores not only quality of life but also sexuality by improving self-image, self-esteem and the ability to cope. Indwelling catheterization as a method of long-term urinary treatment should be avoided in women.  相似文献

Net radiation is a key component in the surface radiation budget. Numerous studies have developed frameworks to estimate net radiation or its components (upwelling or downwelling longwave and/or shortwave radiation) from remote sensing data for clear sky conditions. Application of existing methodologies to estimate net radiation for cloudy sky conditions from remote sensing sensors remains a significant challenge. In this paper, we present a framework to estimate instantaneous and daily average net radiation under all sky conditions from using the data from the MODerate Resolution Imaging Spectroradiometer (MODIS), onboard from the Terra satellites. Bisht et al. (2005) methodology is used for the clear sky portion of the MODIS overpass; while for cloudy portion of the MODIS overpass an extension of Bisht et al. (2005) methodology is applied. The extension of Bisht et al. (2005) methodology utilizes the MODIS cloud data product (MOD06_L2) for cloud top temperature, cloud fraction, cloud emissivity, cloud optical thickness and land surface temperature for cloudy days. The methodology is applied over the Southern Great Plains (SGP) for a time period covering all seasons of 2006. During the MODIS-Terra overpasses in 2006 over the SGP, only 24% of day-overpasses and 9% of night-overpasses had 75% or more of the study region as cloud free. Thus, this proposed study is applicable to a large portion of the MODIS-Terra overpasses. The root mean square errors (RMSE) of instantaneous and daily average net radiation estimated under cloudy conditions using the MOD06_L2 product, comparing to ground-based measurements are 37 W m− 2 and 38 W m− 2, respectively. The strength of the proposed methodology is that it can rely exclusively on remote sensing data in the absence of ancillary ground observations, thus it has a potential to estimate surface energy budget globally.  相似文献

In the recent past we have seen a boom of distributed interactive multimedia environments which use multiple correlated media sensors, multi-view displays, and advanced haptic-audio-visual user interfaces for teleimmersive gaming, business meetings and other collaborative activities. However, when we investigate the emerging teleimmersive environments closer, we realize that their overall session management, including end-to-end session setup, establishment and run-time management are not appropriate for the new demands that these environments present. These environments are cyber-physical rooms that demand (a) large scale of multi-sensory devices across geographically-distributed locations and (b) interaction with each other in synchronous and real-time manner. To deal with the new teleimmersive demands, we present a new session management design with (a) session initiation protocol(s) that understand media correlations, (b) view-based multi-stream topology establishment among multiple parties, (c) efficient, light-weight and distributed session monitoring with querying and debugging capabilities, (d) flexible view-based session adaptation with efficient topology adjustments, and (e) light-weighted and consistent session tear-down protocols. The presented design of the next generation session management protocols, services, algorithms and data structures is based on our extensive experiences with building 3D teleimmersive interactive systems, experimenting with high impact teleimmersive applications and deploying such environments at various venues.  相似文献

The localization of the components of an object near to a device before obtaining the real interaction is usually determined by means of a proximity measurement to the device of the object’s features. In order to do this efficiently, hierarchical decompositions are used, so that the features of the objects are classified into several types of cells, usually rectangular.In this paper we propose a solution based on the classification of a set of points situated on the device in a little-known spatial decomposition named tetra-tree. Using this type of spatial decomposition gives us several quantitative and qualitative properties that allow us a more realistic and intuitive visual interaction, as well as the possibility of selecting inaccessible components. These features could be used in virtual sculpting or accessibility tasks.In order to show these properties we have compared an interaction system based on tetra-trees to one based on octrees.  相似文献
