Increases in international trade and global seafood consumption, along with fluctuations in the supply of different seafood species, have resulted in fraudulent product mislabeling. Grouper species, due to their high demand and varied commercial availability, are common targets for mislabeling by exploiting inefficient inspection practices. Compounding this problem is the fact that there are currently 64 species of fish from eleven different genera allowed to be labeled “grouper” per U.S. Food and Drug Administration guidelines. This wide diversity makes it difficult for regulators to discern legally salable groupers from restricted species. To obviate taxonomic misidentification when relying on external phenotypic characteristics, regulatory agencies are now employing genetic authentication methods which typically offer species-level resolution. However, standard genetic methods such as DNA barcoding require technical expertise and long turnover times, and the required instrumentation is not amenable for on-site analysis of seafood. To obviate some of these limitations, we have developed a handheld genetic sensor that employs a real-time nucleic acid sequence-based amplification assay (RT-NASBA) previously devised in our lab, for the analysis of fish tissue in the field. The base RT-NASBA assay was validated using a lab-based, benchtop RNA purification method as well as non-portable, commercial RT-NASBA analyzer. Described herein, is an uncomplicated method for purifying RNA from fish tissue in the field, which had similar efficiency to the benchtop method demonstrated through direct comparisons. We have also demonstrated that the field sensor is only slightly less sensitive than the benchtop instrument, and could discern 80.3% of groupers (no target sequence available for three species) on the 2014 FDA Seafood List from potential impostors. The complete field assay requires fewer than 80 min for completion and can be performed outside of the lab in its entirety.  相似文献

Human capital measurement and accounting has been under discussion for years without any satisfactory methodology emerging. The economic significance of today's knowledge-intensive organizations makes better HC measurement more pressing. We draw on insights from the knowledge-based theory of the firm and conclude we can only make sense of its human capital by looking in detail at its practices. Human capital is the value added at the level of the work practice—as traced by activity-base accounting. Overall the firm's human capital totals into its goodwill. The human capital can be estimated and then managed by allocating the goodwill to the activities taking place, a complex distribution process but one precisely complementary to that of activity-base accounting.  相似文献

Explaining how engineering devices work is important to students, engineers, and operators. In general, machine generated explanations have been produced from a particular perspective. This paper introduces a system called automatic generation of explanations (AGE) capable of generating causal, behavioral, and functional explanations of physical devices in natural language. AGE explanations can involve different user selected state variables at different abstraction levels. AGE uses a library of engineering components as building blocks. Each component is associated with a qualitative model, information about the meaning of state variables and their possible values, information about substances, and information about the different functions each component can perform. AGE uses: (i) a compositional modeling approach to construct large qualitative models, (ii) causal analysis to build a causal dependency graph, (iii) a novel qualitative simulation approach to efficiently obtain the system's behavior on large systems, and (iv) decomposition analysis to automatically divide large devices into smaller subsystems. AGE effectiveness is demonstrated with different devices that range from a simple water tank to an industrial chemical plant.  相似文献

Accurate identification of seafood in the marketplace is an issue of international concern, due to high rates of market substitution of cheaper or more widely available species for expensive or high-demand species. Salmon samples from stores and restaurants throughout western Washington, USA were tested using DNA sequencing of a short section of the mitochondrial cytochrome c oxidase I (COI) gene (DNA barcoding) to identify Atlantic salmon substituted for Pacific salmon. Of 99 salmon samples, 11 (11%) were Atlantic salmon sold as Pacific salmon. More than 38% of restaurant samples were mislabeled to species, while only 7% of store samples were mislabeled. Market substitution rates were significantly greater in restaurants compared to stores, and consistently greater in winter compared to spring, although not significantly. The high market substitution rate in restaurants documents a pressing need for more monitoring and enforcement specifically in restaurants. DNA barcoding is a valuable tool for rapid and definitive authentication of salmon in the marketplace, and should be more widely adopted to discourage market substitution.  相似文献

There has been an explosion of interest in health sciences applications of case-based reasoning (CBR), not only in the traditional CBR in medicine domain, but also in bioinformatics, enabling home health-care technologies, CBR integration, and synergies between CBR and knowledge discovery. This special issue features the best papers from the third workshop on CBR in the health sciences, held at ICCBR-05 in Madrid. It is the third in a series of exciting workshops, the first two of which were held at ICCBR-03, in Trondheim, Norway, and at ECCBR-04, in Madrid, Spain. The nine high-quality papers introduced here represent the research and experience of twenty-two authors working in eight different countries on a wide range of problems and projects. These papers illustrate some of the major trends of current research in CBR in the health sciences, and represent overall an excellent sample of the most recent advances of CBR in the health sciences.  相似文献

Snapping turtle (Chelydra serpentina) eggs were exposed to two concentrations of chemically- or physically-dispersed water accommodated fractions of weathered Arabian light crude oil (Low = 0.5 and High = 10 g oil/L water). Solutions were passed through nest substrate to simulate alterations in composition during percolation to egg depth. Hatchlings were raised for 13 months during which numerous endpoints were measured. Prior to percolation, total PAH (“tPAH”; the sum of 52 PAHs measured) in physically-dispersed oil fractions were similar (High, 43; Low 67 mg/L). Following percolation, tPAH was also similar in physically-dispersed fractions (High, 14; Low 24 mg/L). Addition of dispersant increased tPAH prior to percolation in the High treatment (302 mg/L) relative to Low (13 mg/L), but percolation resulted in nearly equal concentrations in both treatments (High, 30; Low, 22 mg/L) due to physical trapping of dispersed oil by the nest substrate. In both chemically- and physically-dispersed fractions, percolation reduced low molecular weight (MW) compounds such that embryos were exposed to primarily mid- to high MW compounds. Total PAH in eggs differed 15-fold between the chemically-dispersed High and physically-dispersed High treatments (560 and 36 µg/kg respectively), the former characterized by higher MW compounds than the latter. While eggs accumulated up to 560 µg/kg tPAH, we observed no effects on hatching success or hatchling/juvenile traits (DNA integrity, survival, growth, metabolism, energy storage, or behavior), our results demonstrate that PAH profiles are altered during percolation, suggesting that experiments with subsurface organisms should be designed to account for compositional changes that occur as the solutions percolate through the substrate.  相似文献

The behaviour of masonry under seismic loads has recently become of great interest. New methods for design and analysis, as well as for resistance, have been developed. The main focus has been on experimental and analytical research for the development of new brick material and mortar. With the introduction of Eurocode 8, the behaviour of old masonry structures under seismic load became an interesting topic for engineers. In particular, the available resistance capacity of existing masonry structures is highly relevant to the restructuring of existing buildings. Laboratory tests on masonry walls were performed in order to determine the mechanical behavioural characteristics of old brick material.  相似文献

This paper explains the remarkable restructuring of the Indianautomobile industry. It argues that firms have had to deploynew governance modes (flexible industrial practices) for economiccoordination to overcome supply bottlenecks and meet expandingdemand. Firms that failed to adopt these practices performedpoorly, while firms that attained economies of scale were ableto graduate to exploiting economies of scope.
