Pore size and connectivity of artificial bone scaffolds play key role in regulating cell ingrowth and vascularization during healing. The objective of this study was to develop a novel process for preparing 3D pore-interconnected open-cell bone substitutes with varying pore sizes. This was achieved by thermal-induced expansion, drying, then sintering the mixture of biphasic calcium phosphate (BCP) and a thermal responsive porogen comprising chitosan (CS) and hydroxypropyl methyl cellulose (HPMC). The interpolymer complexes (IPCs) of CS/HPMC were prepared and investigated by FT-IR. The mixtures of IPCs/BCP were heated up to 100 °C for analyzing their thermal expansion properties. This resulted in ~13% and ~42% volume increment for IPC-1/BCP and IPC-2/BCP, respectively, while ~230% volume increased in the case of IPC-3/BCP (therefore chosen for sintering bone blocks). Heating rate-dependent (0.20–0.25 °C/min range) sintering profiles for IPC-3/BCP were utilized to produce BCP bone blocks. Gasification of IPC during sintering resulted in the formation of interconnected porous structures, and the morphology was investigated by SEM, revealing varying sizes ranging from 106 ± 13 μm to 1123 ± 75 μm. The pore size range of bone blocks from 235 ± 46 μm to 459 ± 76 μm portrayed significantly high MC3T3-E1 cell viability with prominent filopodial extensions, and elongated cells, depicting efficient biocompatibility. Therefore, the process for preparing porous interconnected 3D bone blocks were feasible, thereby serving as an alternative for potential bone tissue engineering applications.  相似文献

There is growing interest in the use of structure-based virtual screening to identify small molecules that inhibit challenging protein–protein interactions (PPIs). In this study, we investigated how effectively chemical library members docked at the PPI interface mimic the position of critical side-chain residues known as “hot spots”. Three compound collections were considered, a commercially available screening collection (ChemDiv), a collection of diversity-oriented synthesis (DOS) compounds that contains natural-product-like small molecules, and a library constructed using established reactions (the “screenable chemical universe based on intuitive data organization”, SCUBIDOO). Three different tight PPIs for which hot-spot residues have been identified were selected for analysis: uPAR⋅uPA, TEAD4⋅Yap1, and CaVα⋅CaVβ. Analysis of library physicochemical properties was followed by docking to the PPI receptors. A pharmacophore method was used to measure overlap between small-molecule substituents and hot-spot side chains. Fragment-like conformationally restricted small molecules showed better hot-spot overlap for interfaces with well-defined pockets such as uPAR⋅uPA, whereas better overlap was observed for more complex DOS compounds in interfaces lacking a well-defined binding site such as TEAD4⋅Yap1. Virtual screening of conformationally restricted compounds targeting uPAR⋅uPA and TEAD4⋅Yap1 followed by experimental validation reinforce these findings, as the best hits were fragment-like and had few rotatable bonds for the former, while no hits were identified for the latter. Overall, such studies provide a framework for understanding PPIs in the context of additional chemical matter and new PPI definitions.  相似文献

We have cloned the Saccharomyces cerevisiae C-4 sterol methyl oxidase ERG25 gene. The sterol methyl oxidase performs the first of three enzymic steps required to remove the two C-4 methyl groups leading to cholesterol (animal), ergosterol (fungal), and stigmasterol (plant) biosynthesis. An ergosterol auxotroph, erg25, which fails to demethylate and concomitantly accumulates 4,4-dimethylzy-mosterol, was isolated after mutagenesis. A complementing clone consisting of a 1.35-kb Dra I fragment encoded a 309-amino acid polypeptide (calculated molecular mass, 36.48 kDa). The amino acid sequence shows a C-terminal endoplasmic reticulum retrieval signal KKXX and three histidine-rich clusters found in eukaryotic membrane desaturases and in a bacterial alkane hydroxylase and xylene monooxygenase. The sterol profile of an ERG25 disruptant was consistent with the erg25 allele obtained by mutagenesis.  相似文献

This paper details a compression system for stereoscopic (3-D) images that takes advantage of the disparity compensation effect by heavily compressing one image and a lightly compressing the other. Two methods were tried for the creation of the heavily compressed image: Lossy Pyramid coding and Pruned Discrete Cosine Transform (PDCT) with Variable Length Coding VLC. Both versions of the program used a lightly quantized PDCT image for the clear image.A detailed explanation of the algorithms, the effect of compression level, and the effect of compression method on stereoscopic perception is presented. In this work, Yb3+ was selected to replace the Y3+ in yttrium aluminum garnet (YAG) in order to reduce its thermal conductivity under high temperature. A series of (Y1-xYbx)3Al5O12 (x=0, 0.1, 0.2, 0.3, 0.4) ceramics were prepared by solid-state reaction at 1600 °C for 10 h. The microstructure, thermophysical properties and phase stability under high temperature were investigated. The results showed that all the Yb doped (Y1-xYbx)3Al5O12 ceramics were comprised of a single garnet-type Y3Al5O12 phase. The thermal conductivities of (Y1-xYbx)3Al5O12 ceramics firstly decreased and subsequently increased with Yb ions concentration rising from room temperature to 1200 °C. (Y0.7Yb0.3)3Al5O12 had the lowest thermal conductivity among investigated specimens, which was about 1.62 W m−1 K−1 at 1000 °C, around 30% lower than that of pure YAG (2.3 W m−1 K−1, 1000 °C). Yb had almost no effect on the coefficients of thermal expansion (CTEs) of (Y1-xYbx)3Al5O12 ceramics and the CTE was approximate 10.7×10−6 K−1 at 1200 °C. In addition, (Y0.7Yb0.3)3Al5O12 ceramic remained good phase stability when heating from room temperature to 1450 °C.  相似文献

Up-conversion phosphor is a potential candidate as non-contact temperature sensor because of its unjammable and unique detection abilities. In this work, we investigate the influence of Yb3+ concentration on the emission color, thermal sensing and optical heater behavior of Er3+ doped Y6O5F8 phosphor. Our results show that the emission color of Er3+ and Yb3+ co-doped Y6O5F8 powder changes from green to yellow with the Yb3+ concentration increasing. Importantly, the temperature sensing sensitivities of Er3+ and Yb3+ co-doped Y6O5F8 powder reach 0.008, 0.009, 0.010 and 0.011 K−1 as the sample doped with 2%, 5%, 8% and 11% Yb3+ at 476 K, respectively. Moreover, the temperature of high Yb3+ concentration sample shows preferable optical heating behavior, whose temperature is ascended by a large value of 94 K when the excitation pump power density changes from 1.0 to 13.1 W cm−2. These results suggest Er3+ and Yb3+ co-doped Y6O5F8 powder has great potential in colorful display, temperature sensing and optical heating.  相似文献

Fruits and vegetables have protective effects against many human cancers, including pancreatic cancer. Isoprenoids are one class of phytochemicals which have antitumor activity, but little is known about their effects on cancer of the pancreas. We tested the hypothesis that isoprenoids would inhibit the growth of pancreatic tumor cells. Significant (60–90%) inhibition of the anchorage-independent growth of human MIA PaCa2 pancreatic tumor cells was attained with 25 μM farnesol, 25 μM geranylgeraniol, 100 μM perillyl amine, 100 μM geraniol, or 300 μM perillyl alcohol. We then tested the relative in vivo antitumor activities of dietary farnesol, geraniol, and perilyl alcohol against transplanted PC-1 hamster pancreatic adenocarcinomas. Syrian Golden hamsters fed geraniol or farnesol at 20 g/kg diet exhibited complete inhibition of PC-1 pancreatic tumor growth. Both farnesol and geraniol were more potent than perillyl alcohol, which inhibited tumor growth by 50% at 40 g/kg diet. Neither body weights nor plasma cholesterol levels of animals consuming isoprenoid diets were significantly different from those of pair-fed controls. Thus, farnesol, geraniol, and perillyl alcohol suppress pancreatic tumor growth without significantly affecting blood cholesterol levels. These dietary isoprenoids warrant further investigation for pancreatic cancer prevention and treatment.  相似文献

Recent nursing graduates often find it challenging to prioritize competing tasks in the increasingly complex hospital environment. In order to develop a normative framework for task prioritization, Registered Nurses from a variety of settings were interviewed about how they prioritized their activities in the workplace. From 30 interviews, 422 prioritization decisions were coded when study participants were confronted with two tasks that could not be done simultaneously. The findings suggest a seven-level prioritization hierarchy of nursing activities: 1) addressing imminent clinical concerns, 2) high uncertainty activities, 3) significant, core clinical caregiving and managing pain, 4) relationship management, 5) documenting, helping others, and patient support, 6) system improvement and cleaning/preparing supplies, and 7) personal breaks and social interactions. Explicitly providing a normative framework for task prioritization during nursing education may help to accelerate learning this skill. For more experienced nurses, we believe that flexibility to deviate from this framework will be important for providing high-quality, personalized care that best matches the particular context.

Relevancy to industry

Nursing personnel comprises a significant portion of the human capital and operating budget for hospitals. Knowledge of what tasks are likely to be shed under severe workload conditions aids with optimizing hiring and allocation of nursing personnel.  相似文献
